With the development of social economy and the enhancement of health care consciousness of people, people strive to tonify food, so that health food is more and more favored. Therefore, the research of the traditional Chinese medicine health food by taking the traditional Chinese medicine theory as guidance accords with the requirements of modern people, and has good development prospect. How to improve sleep has become a popular problem which needs to be solved urgently, and research on safe, reliable and less-dependent sleep improvement products has become a hot spot of research and development.
The spina date seed is a dry mature seed of a Rhamnaceae plant Ziziphi Spinosae (Ziziphi Spinosae Semen) and is mainly produced in the vast northern area of China. The wild jujube recorded in Shen nong Ben Cao Jing can 'An Wuzang, lighten body and prolong life'. The spina date seed is sweet and sour in taste and neutral in nature, and has the functions of nourishing the liver, calming the heart, soothing the nerves, arresting sweating and the like. The spina date seed serving as a traditional Chinese medicine in China has the characteristics of complex chemical components and various pharmacological actions, so that the spina date seed has a potential development prospect in the aspects of medicines and foods.
Lycium barbarum L is a special Ningxia herb, which is recorded in Shennong Ben Cao Jing (Shennong's herbal), and has the effects of nourishing kidney and heart, nourishing heart and tranquilizing mind.
Arillus longan (Dimocarpus longan Long.) is a evergreen fruit tree and arbor of the genus Dimocarpus of the family Sapindaceae (Sapindaceae), is a plant used as both medicine and food, and is regarded as a precious tonic since ancient times. The arillus longan is nutritious, and has effects of nourishing heart, invigorating spleen, nourishing blood, tranquilizing mind, and strengthening body constitution.
Semen Platycladi (Arboruitae seed), also known as semen Platycladi, and semen Platycladi, is the seed of Platycladus orientalis of Cupressaceae. At present, the traditional Chinese medicine world generally considers that the platycladi seed has the effects of nourishing liver, calming heart, soothing nerves and the like.
At present, no report is found at home and abroad on the food for improving sleep prepared by taking spina date seeds, medlar, platycladi seeds and longan aril as raw materials, so that a health-care food for improving sleep and a preparation method thereof are urgently needed, and an important development direction is provided.

The purpose of the invention is realized by the following technical scheme: a composition with sleep improving effect is mainly prepared from semen Ziziphi Spinosae, fructus Lycii, semen Platycladi, and arillus longan extract; when the extract is prepared and extracted, the raw materials of the wild jujube seed, the medlar, the platycladi seed and the longan aril are calculated according to the parts by weight: 18-36 parts of spina date seed, 15-30 parts of medlar, 15-30 parts of platycladi seed and 15-30 parts of arillus longan.

The common auxiliary materials are selected from a filling agent, an acidity agent, a sweetening agent and an essence.
The composition with the sleep improving effect comprises, by weight, 19.32-37.64 parts of an extract, 57.96-92.92 parts of a filling agent, 0-3 parts of an acidulant, 0-0.4 part of a sweetener and 0-1 part of an essence.

The more preferable technical scheme is as follows: 9.32-27.64 parts of extract, 27.96-82.92 parts of filler, 0-3 parts of sour agent, 0-0.4 part of sweetener and 0-1 part of essence; the extract is prepared from the following raw materials in proportion: 18-36 parts of spina date seed, 15-30 parts of medlar, 15-30 parts of platycladi seed and 15-30 parts of arillus longan.
The filler is selected from one or more of mannitol, lactose, microcrystalline cellulose, isomalt and maltodextrin;
the sour agent is one or more of citric acid, ascorbic acid, lactic acid and DL-malic acid;
the sweetener is one or more selected from aspartame, acesulfame potassium and stevioside;
the essence is selected from one or more of juicy peach essence, strawberry essence, litchi essence, sweet orange essence, lavender essence and rose essence.
The preparation method of the composition with the effect of improving sleep comprises the following steps:
(1) extracting spina date seed, medlar, platycladi seed and longan pulp with water according to the solvent amount of 6-12 times, extracting for 1-3 times, 0.5-2h each time, concentrating and drying the extracting solution.
The preparation method of the composition with the effect of improving sleep further comprises the following steps:
(2) adding adjuvants into the obtained extract at a certain proportion, and mixing to obtain composition with sleep improving effect;
or:
(2) adding appropriate amount of filler, sour agent, and sweetener;
(3) preparing the mixture obtained in the step (2) into a soft material by using pure water, sieving and drying to prepare the oral preparation.
Preferably, the step (1) is:
extracting semen Ziziphi Spinosae, fructus Lycii, semen Platycladi, and arillus longan with water at a ratio of 6-12 times of solvent amount for 0.5-2 hr for 1-3 times at 100 deg.C, and mixing filtrates; concentrating the water extract to relative density of 1.30-1.50, cooling, and sealing and refrigerating for 12-24 hr to obtain concentrated extract.
The preparation method of the composition with the effect of improving sleep, more preferably the method comprises the following steps: extracting semen Ziziphi Spinosae, fructus Lycii, semen Platycladi, and arillus longan with water at 6-12 times of solvent amount for 1-3 times (0.5-2 hr each time) at 100 deg.C, and mixing filtrates. Concentrating the water extractive solution to relative density of 1.30-1.50, cooling, sealing and refrigerating for 12-24 hr to obtain concentrated extractive solution extract, adding appropriate amount of filler or sour agent and sweetener, making soft mass with pure water, sieving, drying, and making into oral preparation.
The composition with the sleep improving effect can be used for common foods and health-care foods. Preferably used for preparing meal replacement biscuits, gel soft candies, solid drinks or oral preparations. The oral preparation is preferably powder, granules and tablets.
The composition provided by the invention is mostly medicinal and edible products, can play a role in improving sleep after scientific and reasonable compatibility, can provide energy, protein, various mineral substances, amino acids and other nutritional ingredients for human bodies, and is a natural good health-care product.
Detailed Description
The present invention will be described in detail with reference to specific examples, but the scope of the present invention is not limited to the following examples.
Example 1
Pulverizing semen Ziziphi Spinosae, fructus Lycii, semen Platycladi, and arillus longan into 20 mesh powder, oven drying in oven at 50 deg.C, and making into teabag.
Example 2
(1) Extraction: mixing the spina date seed, the medlar, the platycladi seed and the longan pulp in proportion, adding water in an amount which is 12 times of the weight of the mixture, adjusting the temperature to be 80-100 ℃, and extracting for 1.5 hours for 1 time. Concentrating and drying the extract to obtain the concentrated extract.
(2) Molding: taking the concentrated extract obtained in the step (1), freeze-drying into 80-mesh powder, drying in an oven at 50 ℃, and preparing into powder.
Example 3
(1) Extraction: mixing the spina date seed, the medlar, the platycladi seed and the longan pulp in proportion, adding water in an amount which is 12 times of the weight of the mixture, adjusting the temperature to be 80-100 ℃, and extracting for 1.5 hours for 1 time. Concentrating and drying the extract to obtain the concentrated extract.
(2) Molding: and (2) taking the concentrated extract obtained in the step (1), freeze-drying the extract into powder of 80 meshes, adding flour and eggs, uniformly mixing, pouring the mixture into a mould, and drying the mixture in an oven at 120 ℃ to prepare the meal replacement biscuit.
Example 4
(1) Extraction: mixing the spina date seed, the medlar, the platycladi seed and the longan pulp in proportion, adding water in an amount which is 12 times of the weight of the mixture, adjusting the temperature to be 80-100 ℃, and extracting for 1.5 hours for 1 time. Concentrating and drying the extract, and concentrating the water extract until the relative density is 1.50 to obtain the concentrated extract.
(2) Molding: and (2) taking the concentrated extract obtained in the step (1), adding 5 parts of agar powder, 5 parts of Arabic gum and 18 parts of white granulated sugar, decocting at 100 ℃, cooling, pouring into a mold for molding, and preparing the gel soft candy.
Example 5
Mixing the spina date seed, the medlar, the platycladi seed and the longan pulp in proportion, adding water in an amount which is 12 times of the weight of the mixture, adjusting the temperature to be 80-100 ℃, and extracting for 1.5 hours for 1 time. Concentrating the extractive solution, drying, concentrating the water extract to relative density of 1.30, and making into granule.
Example 6
(1) Mixing spina date seed, medlar, platycladi seed and longan pulp in proportion, adding water in an amount which is 12 times of the weight of the mixture, adjusting the temperature to be 80-100 ℃, extracting for 1 hour and extracting for 2 times;
(2) concentrating and drying the extract, and concentrating the water extract until the relative density is 1.35 to obtain the concentrated extract.
(3) According to the weight parts, 35 parts of concentrated extract, 58 parts of filler, 1 part of sour agent, 1 part of sweetener and 1 part of essence are taken and mixed evenly. Making soft mass with pure water, sieving with 14 mesh sieve, granulating, drying, and packaging to obtain solid beverage.
Example 7
(1) Mixing the spina date seed, the medlar, the platycladi seed and the longan pulp in proportion, adding water in an amount which is 12 times of the weight of the mixture, adjusting the temperature to be 80-100 ℃, and extracting for 1.5 hours for 1 time;
(2) concentrating and drying the extract, and concentrating the water extract to relative density of 1.30 to obtain concentrated paste.
Example 8
(1) Mixing the spina date seed, the medlar, the platycladi seed and the longan pulp in proportion, adding water in an amount which is 12 times of the weight of the mixture, adjusting the temperature to be 80-100 ℃, and extracting for 1.5 hours for 1 time;
(2) concentrating and drying the extract, and concentrating the water extract until the relative density is 1.40 to obtain the concentrated extract.
(3) Taking 40 parts of concentrated extract, 52 parts of mannitol, 1 part of DL-malic acid and 1 part of orange essence according to the weight parts, uniformly mixing, preparing a soft material, sieving, drying, granulating, adding 0.5 part of magnesium stearate, uniformly mixing, tabletting and preparing into tablets.
Example 9
(1) Mixing the spina date seed, the medlar, the platycladi seed and the longan pulp in proportion, adding water in an amount which is 12 times of the weight of the mixture, adjusting the temperature to be 80-100 ℃, and extracting for 1.5 hours for 1 time;
(2) concentrating and drying the extract, and concentrating the water extract until the relative density is 1.50 to obtain the concentrated extract.
(3) Taking 19 parts of concentrated extract, 28 parts of mannitol, 28 parts of lactose, 3 parts of DL-malic acid, 0.4 part of aspartame and 1 part of juicy peach essence according to the weight parts, uniformly mixing, preparing soft materials, sieving, drying, finishing granules and preparing into granules.
A health food for improving sleep prepared according to example 9 was subjected to the following tests:
first, maximum drug tolerance experiment
40 healthy ICR (18-22 g) mice were selected, half male and half female. After adaptive feeding for 3-5 days, fasting for 12 hours without water prohibition, dividing the mice into four groups of blank females, blank males, administration females and administration males, administering the maximum solubility of 1.13g/mL and the maximum gavage capacity of 0.4mL/10g · bw to the mice, gavage once, continuously observing for two weeks, and weighing and killing the mice. Second, health food animal function test for improving sleep
The evaluation test of the health care function of the product is carried out according to the requirements related to the technical Specification for health food inspection and evaluation (2003 edition), and the test results are as follows:
1. the test animals were: healthy male ICR mice (18-22 g), 192.
2. Grouping experimental animals: the experimental animals are divided into four experimental groups, namely a direct sleep experimental group, an experimental group for prolonging the sleep time of the sodium pentobarbital, an experimental group for hypnotizing the sodium pentobarbital at the subthreshold dose and an experimental group for the sleep latency period of the sodium pentobarbital, wherein the four experimental groups are divided into four subgroups according to the weight of the mice, and each group is 12.
3. Dose selection and sample preparation: the low, medium and high three dose groups are set according to 1/6, 2/6 and 3/6 of the maximum tolerance. The sample prepared in example 9 was taken and the mice were gavaged once daily for 30 consecutive days at a volume of 0.2mL/10g · bw. The control group was gavaged with an equal volume of distilled water.
4. Method steps and results
The test results of the health food with sleep improving effect are shown in table 1, table 2 and table 3.
Direct sleep experiment
The experimental steps are as follows: and observing whether the animals in the test group are administered with the test samples at three doses, and whether the blank control group is administered with the solvent at the same volume to have sleep phenomenon. The sleep uses the disappearance of the righting reflex as an index, and when the mouse is placed in the dorsal position, the mouse can righting the body immediately. If the patient can not be righted for more than 30-60 s, the righting reflex is considered to disappear, and the patient enters sleep. The recovery of the righting reflex is the awakening of the animal, the time from disappearance of the righting reflex to recovery is the sleeping time of the animal, and the number of sleeping animals and the sleeping time are recorded.
The experimental results are as follows: the disappearance of the positive turning reflex is taken as a standard, the sleeping quantity of the tested mice in the negative control group and each dosage group is 0, and the product is proved to have no direct sleeping effect.
Experiment for prolonging sleep time of sodium pentobarbital
The experimental steps are as follows: the preliminary experiment is carried out before the formal experiment, the dose of the sodium pentobarbital which enables the animals to fall asleep for 100 percent but does not lead the animals to sleep for too long is determined, and the formal experiment is carried out by using the dose.
After the animals are subjected to last intragastric administration for 15min, pentobarbital sodium (55 mg/kg · bw according to the pre-experimental result) is injected into the abdominal cavity of each group of animals, the injection amount is 0.2mL/20g, the disappearance of the righting reflex of the mice is taken as a sleep index, and the sleep time is recorded.
Note: indicates that the difference was significant (p <0.05) and indicates that the difference was very significant (p <0.01) compared to the control group.
The experimental results are as follows: compared with a blank control group, the sleeping time of the medium and high dose groups is obviously increased, the difference is obvious, and the statistical significance is achieved (p is less than 0.01), so that the product can prolong the sleeping time of mice induced by pentobarbital sodium.
③ Pentabarbital sodium subthreshold dose hypnosis experiment
The experimental steps are as follows: and determining the subthreshold hypnotic dose of the sodium pentobarbital, namely the maximum subthreshold dose of the sodium pentobarbital, wherein 80-90% of mice have positive turning reflexes and do not disappear. The dosage was used for the official experiments.
After the last administration for 15min, pentobarbital sodium (32 mg/kg · bw according to the pre-experiment result) is injected into the abdominal cavity of each group of animals, the injection amount is 0.2mL/20g, the number of the animals falling asleep and the sleep incidence rate of each group within 30min are recorded by taking the disappearance of the mouse positive turning reflex for more than 1min as the sleep-falling judgment standard.

The experimental results are as follows: compared with a blank control group, the number of the animals in the low, medium and high dose groups is increased, and compared with the control group, the number of the animals in the medium and high dose groups is obviously increased, the sleep incidence rate is improved, the statistical significance is achieved (p is less than 0.05), and the product can increase the subliminal dose hypnosis number of the pentobarbital sodium.
Experiment of sleep latency period of barbital sodium
The experimental steps are as follows: the dose of barbiturate sodium that caused 100% of the animals to fall asleep but did not cause too long a sleep time was determined and used for the official test.
After the last administration for 15min, barbital sodium (286 mg/kg · bw according to the pre-experiment results) was injected into the abdominal cavity of each group of animals at an injection amount of 0.2mL/20g, and the sleep latency was observed with the disappearance of the mouse positive turning reflex as an index.
Note: indicates that the difference was significant (p <0.05) and indicates that the difference was very significant (p <0.01) compared to the control group.
The experimental results are as follows: compared with a blank control group, the sleep latency of animals in the low, medium and high dose groups is in a reduction trend, compared with the control group, the medium and high doses are in a significant reduction trend, the sleep latency of mice is shortened, the difference is significant, and the statistical significance (p <0.05) is achieved, so that the barbiturate sodium sleep latency can be reduced by the product.
In conclusion, the composition of the spina date seed, the medlar, the platycladi seed and the longan pulp has the function of improving the sleep, and animal effect experiments prove that the composition is safe and reliable, does not have the direct sleep function, can prolong the sleep time of the pentobarbital sodium induced mice, increase the number of the pentobarbital sodium subliminal dose hypnotic mice falling asleep, and reduce the sleep latency of the pentobarbital sodium, so that the health-care function of improving the sleep is achieved.

A composition with sleep improving effect is characterized in that the composition mainly comprises extracts of spina date seeds, medlar, platycladi seeds and longan aril; when the extract is prepared and extracted, the raw materials of the wild jujube seed, the medlar, the platycladi seed and the longan aril are calculated according to the parts by weight: 18-36 parts of spina date seed, 15-30 parts of medlar, 15-30 parts of platycladi seed and 15-30 parts of arillus longan.

9. The composition with sleep improvement effect according to any one of claims 1 to 7, wherein the dosage form of the composition comprises teabags; powder; granules; a tablet; meal replacement biscuits; gel soft candy; granules; solid beverages or oral formulations; and (4) ointment.
10. A method for preparing a composition for improving sleep as claimed in any one of claims 1 to 7, comprising the steps of:
(1) extracting spina date seed, medlar, platycladi seed and longan pulp with water according to the solvent amount of 6-12 times of that of the spina date seed, medlar, platycladi seed and longan pulp at 80-100 ℃ for 1-3 times, 0.5-2h each time, and concentrating and drying the extracting solution.
11. The method for preparing a composition with sleep improvement assisting effect according to claim 10, characterized by further comprising the steps of:
(2) adding adjuvants into the extract at a certain proportion, and mixing to obtain the composition with sleep improving effect.
