Abstract
本发明公开了一种互叶白千层复合精油微乳液及其制备方法,包括复合互叶白千层精油、复合表面活性剂、助表面活性剂和水;复合互叶白千层精油为互叶白千层精油和维生素E的混合物,互叶白千层精油和维生素E的质量份数比为1∶0.0003;复合表面活性剂为食品级吐温20和司盘80复配混合物,吐温20和司盘80的质量份数比为8.95∶1.05;复合互叶白千层油和复合表面活性剂的质量份数比为1∶3.25;助表面活性剂为聚乙二醇400;复合表面活性剂和助表面活性剂的质量份数比为7∶3。有益效果是得到平均粒径在纳米乳液范围内、外观呈透明或半透明状的复合精油微乳液。
Description
技术领域
本发明属于精油微乳液制备技术领域,尤其涉及一种互叶白千层复合精油微乳液及其制备方法。
背景技术
互叶白千层的新鲜枝叶可提取精油,是互叶白千层的主要产物,具有良好的广谱抑菌、止痒、保健作用,且对皮肤灼伤的治疗有效,有宜人的肉豆蔻香气,是公认优良的天然芳香剂、抗菌剂、防腐剂,应用在日化、制药、食品等行业。然而,茶树油成分多且复杂,难溶于水、易挥发、易氧化、高浓度下具有强烈刺激性气味,暴露于高湿、高温、光、空气环境或长时间储存条件下生物活性极不稳定,易形成一系列氧化产物,大大限制其实际应用。现有技术中,采用精油与维生素E混合的方法,最大限度的延长茶树油储存时间,保证其良好的抗菌、消炎作用。中国专利申请,申请号CN201410442511.9,申请日2014.09.02,申请公开号CN104146157A,申请公开日2014.09.02,公开了一种互叶白千层树油饲料添加剂制备方法及制备产品使用方法,其步骤包括有∶将萜品醇-4含量是30-40%的精油经过三次升温、保温后,使精油降温至常温;加入自来水和维生素E混合,加热搅拌后,装入容器,用高速剪切机搅拌后,灌装入瓶制成添加剂。但在实际应用时,由于精油的高挥发性,上述技术方案提供的互叶白千层树油饲料添加剂稳定性较差,一些外界因素如水分、蛋白质、pH和酶类会对精油的抗菌活性造成影响。并且食物中较高的水分会降低植物精油的溶解性能和稳定性,难溶于水的精油与病原体的接触有限,使其抗菌效果减弱。当植物精油添加到食品体系中时,可能由于食品体系中富含的脂肪或蛋白质对被损坏的菌体具有一定的修复作用,从而降低了植物精油的抑菌效果。实际使用时的抗菌效果与实验室检测抗菌效果有较大出入。若想要达到实验室检测同样的抗菌效果,现有技术通常采用的方案是所需植物精油的浓度比体外抑菌测试的高出1%~3%,缺点是这种处理方式使得植物精油浓度往往超出感官可接受范围。因此,实现精油稳定性、抗氧化性的提高及缓释性能,对茶树油产品的开发利用及产业的发展具有重要意义。
发明内容
1.要解决的技术问题
针对现有技术中互叶白千层油为原料制备的精油稳定性和抗氧化性能差、无缓释性能,实际使用时的抗菌效果与实验室检测抗菌效果有较大出入等问题,本发明的目的在于提供一种互叶白千层复合精油微乳液及其制备方法,通过对表面活性剂、助表面活性剂类型及用量的选择,尽可能在减少表面活性剂及助表面活性剂用量的前提下,增溶更多的复合精油,得到平均粒径在纳米乳液范围内、外观呈透明或半透明状的复合精油微乳液,改善了精油与水溶性物质的相容性,提高了精油的稳定性、抗氧化性和缓释性能。
2.技术方案
为实现上述目的,达到上述技术效果,本发明采用如下技术方案:
一种互叶白千层复合精油微乳液,其特点是包括复合互叶白千层精油、复合表面活性剂、助表面活性剂和水;所述复合互叶白千层精油为互叶白千层精油和维生素E的混合物,所述互叶白千层精油和维生素E的质量份数比为1∶0.0003;所述复合表面活性剂为食品级吐温20和司盘80复配混合物,所述吐温20和司盘80的质量份数比为8.95∶1.05;所述复合互叶白千层油和复合表面活性剂的质量份数比为1∶3.25;所述助表面活性剂为聚乙二醇400;所述复合表面活性剂和助表面活性剂的质量份数比为7∶3;所述水的用量标准为∶水与助表面活性剂的体积和稍大于复合互叶白千层精油与复合表面活性剂的体积和;通过水的用量调节使最终乳液中互叶白千层精油的浓度至适应不同用途。
进一步的,所述互叶白千层复合精油微乳液按质量份数包括如下成分:复合互叶白千层精油0.1-11,吐温20 15-30,司盘80 2-4,聚乙二醇400 13-25,水30-45。
进一步的,所述互叶白千层复合精油微乳液按质量份数包括如下成分:互叶白千层精油5,维生素E 0.0015,吐温20 17.65,司盘80 2.35,聚乙二醇400 20,水54.9985。
进一步的,所述互叶白千层复合精油微乳液按质量份数包括如下成分:互叶白千层精油7,维生素E 0.0021,吐温20 24.72,司盘80 3.28,聚乙二醇400 28,水36.9979。
进一步的,所述互叶白千层复合精油微乳液按质量份数包括如下成分:互叶白千层精油10,维生素E 0.003,吐温20 28.69,司盘80 3.81,聚乙二醇400 21.7,水35.797。
进一步的,所述互叶白千层复合精油微乳液按质量份数包括如下成分:互叶白千层精油10,维生素E 0.0003,吐温20 28.69,司盘80 3.81,聚乙二醇400 13.9,水43.5997。
本发明的另一个目的在于提供一种互叶白千层复合精油微乳液的制备方法,其特点是包括如下步骤:
步骤1:将萜品醇-4含量30-40%的互叶白千层成品精油与维生素E混合,互叶白千层精油和维生素E的质量份数比为1∶0.0003,将复合互叶白千层精油加热至30℃,搅拌均匀;
步骤2:将吐温20和司盘80按质量份数比搅拌混合均匀得到复合表面活性剂,在30℃下与复合互叶白千层精油按比例混合、搅拌,待用;
步骤3:按比例混合适量助表面活性剂和水,得到助表面活性剂水溶液,将步骤2中的待用混合物缓缓加入助表面活性剂水溶液,用磁力恒温搅拌机在30℃下搅拌均匀后,灌装入瓶。
3.有益效果
与现有技术相比,本发明的有益效果是:本发明通过对表面活性剂、助表面活性剂类型及用量的选择,表面活性剂选择稳定性高、相容性好、无毒性的食品乳化剂吐温和司盘;助表面活性剂选择稳定效果好的药物常用助表面活性剂聚乙二醇400,尽可能在减少表面活性剂及助表面活性剂用量的前提下,增溶更多的复合精油,得到平均粒径在纳米乳液范围内、外观呈透明或半透明状的复合精油微乳液,改善了精油与水溶性物质的相容性,提高了精油的稳定性、抗氧化性以及缓释性能。

具体实施方式
为加深对本发明的理解,下面结合附图并通过实施例及对比例对本发明作进一步详细说明。
实施例1互叶白千层复合精油微乳液按质量份数包括如下成分:复合互叶白千层精油(互叶白千层精油及维生素E混合物)0.1-11,吐温20 15-30,司盘80 2-4,聚乙二醇40013-25,水30-45;将上述成分按照如下方法进行制备互叶白千层复合精油微乳液:
步骤1:将萜品醇-4含量30-40%的互叶白千层成品精油与维生素E混合,互叶白千层精油和维生素E的质量份数比为1∶0.0003,将复合互叶白千层精油加热至30℃,搅拌均匀;
步骤2:将吐温20和司盘80按质量份数比搅拌混合均匀得到复合表面活性剂,在30℃下与复合互叶白千层精油按比例混合、搅拌,待用;
步骤3:按比例混合适量助表面活性剂和水,得到助表面活性剂水溶液,将步骤2中的待用混合物缓缓加入助表面活性剂水溶液,用磁力恒温搅拌机在30℃下搅拌均匀后,灌装入瓶。得到肉眼可见澄清透明,符合纳米微乳液特点的互叶白千层复合精油微乳液。
对上述方法得到的互叶白千层复合精油微乳液进行乳液类型测定:
测试一.高速离心法测定,取100g互叶白千层复合精油微乳液,在离心力场下转动5min,不出现相分离,表明所得乳液质量稳定,符合微乳液特点。
测试二.染色法测定,如图1所示,从左到右一次为对照加苏丹红三、互叶白千层复合精油微乳液加亚甲基蓝、水加亚甲基蓝加入染料,可见水溶性亚甲基蓝在互叶白千层复合精油微乳液中扩散速度远大于脂溶性苏丹三,证明乳液为水包油溶液。
测试三.粒径检测,如图2所示,粒径及电位检测平均粒径为(35.75±3.16)nm,如图3所示,Zeta电位(-5.74±2.40)m V。
测试四.互叶白千层复合精油微乳液制备后在25℃下储存的第1、7、14、21和28天对液滴粒径进行测量,经比较,不同储存天数间结果差异不显著,放置两个月未出现分层,如图所示4,表明制成的微乳液储存稳定性较好。同样条件下存储的现有技术精油与维生素E复合精油为乳白色不透明状,且有少许浮沫,如图5所示。对照实验试用的现有技术精油与维生素E复合精油成分配比为互叶白千层精油10,维生素E 0.003,吐温20 28.69,司盘803.81,聚乙二醇400 21.7,水35.797;现有技术的茶树精油乳液为互叶白千层精油及表面活性剂等。
测试五.透射电镜观察,如图6所示,透射电镜下可见液滴分布较为均一。
测试六.体外抑菌实验检测抑菌作用,如图7所示,右侧孔加入为10%互叶白千层复合精油微乳液,左上侧空为空白乳液对照,左下侧孔为青链霉素药物对照(青霉素浓度100ug/mL、链霉素浓度100ug/mL)。可见互叶白千层微乳孔有明显抑菌圈,十字交叉法测量约为23.7±0.2mm。
测试七.互叶白千层复合精油微乳液临床试用效果
如图8所示,左上为只进行皮损操作,未接种细菌;右上为造模后用10%互叶白千层复合精油微乳液治疗;左下为造模后不进行治疗的对照组;右下为造模后用除不含互叶白千层外成分同上的空白对照组。可见用10%互叶白千层复合精油微乳液治疗的区域在3d后已出现明显好转,7d后已基本恢复,对比未治疗的对照组及用除不含互叶白千层外成分同上的空白对照组,治疗效果显著。
如图9所示,比格犬A在使用互叶白千层复合精油微乳液第0d、第3d、第7d后患病处的各自效果;可见治疗前皮肤患处大面积发红,有破损及少量渗出;治疗三天后患处泛红好转,并且已有结痂;七天后患处基本好转,皮肤表面轻微不平整。
如图10所示,比格犬B在使用互叶白千层复合精油微乳液第0d、第3d、第7d后患病处的各自效果;治疗前患处已有部分结痂及红疹;三天后患处只有部分小丘疹;七天后无热痛反应基本痊愈。
对比例1
助表面活性剂分别采用聚乙二醇400(PEG-400)、无水乙醇(E)、甘油(DG)、丙二醇(PG),得到的互叶白千层复合精油微乳液效果作为对比测试的拟三元相图,如图11-14所示,聚乙二醇400微乳区域最大,故选择聚乙二醇400为助表面活性剂。
实施例2
互叶白千层复合精油微乳液按质量份数包括如下成分:互叶白千层精油5,维生素E 0.0015,吐温20 17.65,司盘80 2.35,聚乙二醇400 20,水54.9985。将上述成分按照如下方法进行制备互叶白千层复合精油微乳液:
步骤1:将萜品醇-4含量30-40%的互叶白千层成品精油与维生素E混合,互叶白千层精油和维生素E的质量份数比为1∶0.0003,将复合互叶白千层精油加热至30℃,搅拌均匀;
步骤2:将吐温20和司盘80按质量份数比搅拌混合均匀得到复合表面活性剂,在30℃下与复合互叶白千层精油按比例混合、搅拌,待用;
步骤3:按比例混合适量助表面活性剂和水,得到助表面活性剂水溶液,将步骤2中的待用混合物缓缓加入助表面活性剂水溶液,用磁力恒温搅拌机在30℃下搅拌均匀后,灌装入瓶。得到肉眼可见澄清透明,符合纳米微乳液特点的互叶白千层复合精油微乳液。
实施例3
互叶白千层复合精油微乳液按质量份数包括如下成分:互叶白千层精油7,维生素E 0.0021,吐温20 24.72,司盘80 3.28,聚乙二醇400 28,水36.9979。将上述成分按照如下方法进行制备互叶白千层复合精油微乳液:
步骤1:将萜品醇-4含量30-40%的互叶白千层成品精油与维生素E混合,互叶白千层精油和维生素E的质量份数比为1∶0.0003,将复合互叶白千层精油加热至30℃,搅拌均匀;
步骤2:将吐温20和司盘80按质量份数比搅拌混合均匀得到复合表面活性剂,在30℃下与复合互叶白千层精油按比例混合、搅拌,待用;
步骤3:按比例混合适量助表面活性剂和水,得到助表面活性剂水溶液,将步骤2中的待用混合物缓缓加入助表面活性剂水溶液,用磁力恒温搅拌机在30℃下搅拌均匀后,灌装入瓶。得到肉眼可见澄清透明,符合纳米微乳液特点的互叶白千层复合精油微乳液。
实施例4
互叶白千层复合精油微乳液按质量份数包括如下成分:互叶白千层精油10,维生素E 0.003,吐温20 28.69,司盘80 3.81,聚乙二醇400 21.7,水35.797。将上述成分按照如下方法进行制备互叶白千层复合精油微乳液:

步骤2:将吐温20和司盘80按质量份数比搅拌混合均匀得到复合表面活性剂,在30℃下与复合互叶白千层精油按比例混合、搅拌,待用;
步骤3:按比例混合适量助表面活性剂和水,得到助表面活性剂水溶液,将步骤2中的待用混合物缓缓加入助表面活性剂水溶液,用磁力恒温搅拌机在30℃下搅拌均匀后,灌装入瓶。得到肉眼可见澄清透明,符合纳米微乳液特点的互叶白千层复合精油微乳液。
实施例5

步骤2:将吐温20和司盘80按质量份数比搅拌混合均匀得到复合表面活性剂,在30℃下与复合互叶白千层精油按比例混合、搅拌,待用;
步骤3:按比例混合适量助表面活性剂和水,得到助表面活性剂水溶液,将步骤2中的待用混合物缓缓加入助表面活性剂水溶液,用磁力恒温搅拌机在30℃下搅拌均匀后,灌装入瓶。得到肉眼可见澄清透明,符合纳米微乳液特点的互叶白千层复合精油微乳液。
将实施例2至5做测试一至测试六,可以得到与实施例1相同的技术效果。

1.一种互叶白千层复合精油微乳液,其特征在于:包括复合互叶白千层精油、复合表面活性剂、助表面活性剂和水;所述复合互叶白千层精油为互叶白千层精油和维生素E的混合物,所述互叶白千层精油和维生素E的质量份数比为1∶0.0003;所述复合表面活性剂为食品级吐温20和司盘80复配混合物,所述吐温20和司盘80的质量份数比为8.95∶1.05;所述复合互叶白千层油和复合表面活性剂的质量份数比为1∶3.25;所述助表面活性剂为聚乙二醇400;所述复合表面活性剂和助表面活性剂的质量份数比为7∶3;所述水的用量标准为∶水与助表面活性剂的体积和稍大于复合互叶白千层精油与复合表面活性剂的体积和;通过水的用量调节使最终乳液中互叶白千层精油的浓度至适应不同用途。
2.根据权利要求1所述一种互叶白千层复合精油微乳液,其特征在于∶所述互叶白千层复合精油微乳液按质量份数包括如下成分:复合互叶白千层精油0.1-11,吐温20 15-30,司盘802-4,聚乙二醇40013-25,水30-45。
3.根据权利要求1所述一种互叶白千层复合精油微乳液,其特征在于∶所述互叶白千层复合精油微乳液按质量份数包括如下成分:互叶白千层精油5,维生素E 0.0015,吐温2017.65,司盘802.35,聚乙二醇40020,水54.9985。
4.根据权利要求1所述一种互叶白千层复合精油微乳液,其特征在于∶所述互叶白千层复合精油微乳液按质量份数包括如下成分:互叶白千层精油7,维生素E 0.0021,吐温2024.72,司盘803.28,聚乙二醇40028,水36.9979。
5.根据权利要求1所述一种互叶白千层复合精油微乳液,其特征在于∶所述互叶白千层复合精油微乳液按质量份数包括如下成分:互叶白千层精油10,维生素E 0.003,吐温2028.69,司盘803.81,聚乙二醇40021.7,水35.797。

7.一种互叶白千层复合精油微乳液的制备方法,其特点是包括如下步骤∶
步骤1:将萜品醇-4含量30-40%的互叶白千层成品精油与维生素E混合,互叶白千层精油和维生素E的质量份数比为1∶0.0003,将复合互叶白千层精油加热至30℃,搅拌均匀;
步骤2:将吐温20和司盘80按质量份数比搅拌混合均匀得到复合表面活性剂,在30℃下与复合互叶白千层精油按比例混合、搅拌,待用;
步骤3:按比例混合适量助表面活性剂和水,得到助表面活性剂水溶液,将步骤2中的待用混合物缓缓加入助表面活性剂水溶液,用磁力恒温搅拌机在30℃下搅拌均匀后,灌装入瓶。
