Abstract
本发明公开了一种具有抗衰老和抗蓝光功效的护肤组合物及其制备方法,属于化妆品技术领域,所述护肤组合物,至少包含:5~15%重量的来源于米糠的组分;2~4%重量的儿茶提取物;和1~3%重量的番红花提取物。本发明采用双频超声处理米糠获得了一种富含谷维素的米糠的组分,将该提取物与儿茶提取物、番红花提取物复合后呈现了惊人的抗衰老和抗蓝光效用。
Description
技术领域
本发明属于化妆品技术领域,尤其涉及一种具有抗衰老和抗蓝光功效的护肤组合物及其制备方法,该护肤组合物对蓝光具有较强的过滤作用,帮助肌肤抵御蓝光的侵害,同时对自由基具有较强的清除效果。
背景技术
蓝光(简称HEV light)为阳光的重要组成,通常指在可见光光谱上,处在400nm-480nm之间的具有较高能量的光线,也常见于各式的LED灯和电子屏幕等。蓝光对皮肤的损伤通常体现在氧化应激及持续的色素过度沉着,有研究指出,皮肤中的黑素细胞可以通过OPN3感知可见光中的蓝光(415nm),并生成酪氨酸酶和多巴色素互变酶的蛋白复合物,持续激活酪氨酸酶,让肌肤产生持久的色沉,而长期的色素沉着会使皮肤产生暗沉,失去光泽,产生色斑;蓝光可以穿透皮肤表层达到真皮层,在长时间的照射下,细胞中的线粒体会产生活性氧自由基,并与线粒体、DNA和脂肪类的细胞发生氧化反应,使线粒体和DNA受到损伤,对细胞膜功能起到抑制作用,破坏细胞结构,导致细胞受到损伤失去活力[2]。可见,蓝光可以诱导细胞产生自由基,让皮肤真皮层的纤维细胞受到伤害,导致细胞外的基质大量减少,进而加速皮肤的老化速度。
目前在化妆品领域,主要从以下几个方面来实现抗蓝光的功效:①蓝光吸收剂,主要为色粉或有机高分子材料,这些材料能够吸收蓝光,降低蓝光的穿透率,从而起到抗蓝光的作用;②抑制蓝光引起的色沉:通过采用一些多功能肽来降低黑色素含量,从而起到抑制黑色素沉着的作用。
米糠是稻谷加工成大米过程中的副产品之一,其含有大量的营养物质,如蛋白质、脂肪、膳食纤维、大量的矿物质、抗氧化剂(维生素E、生育三烯酚、谷维素、卵磷脂等),这些物质对预防人体心、脑血管疾病,抗癌、增强免疫力、降低血脂及预防便秘等具有显著的功效,并且米糠来源简单,易于获得,成本低。但是目前未见将其应用于抗蓝光研究的报道。
儿茶(Acacia catechu)为豆科合欢树的植物,儿茶的主要有效成分儿茶酚类化合物,如儿茶酚、儿茶酸、焦儿茶酚、根皮酚、原儿茶酸、槲皮素等;研究显示儿茶水提取物具有抑菌作用,对大肠杆菌和枯草杆菌具有良好的抑制作用;此外,30%丁二醇提取物对老鼠毛发具有促进的作用,但是目前未见研究儿茶提取物在抗蓝光方面的效用。
番红花(Crocus sativus)又名藏红花,是鸢尾科番红花属球根类多年生草本植物,其干燥的柱头部分含有类胡萝卜素及其苷类,其具有抗氧化作用,对超氧自由基和单线氧自由基的捕获能力强,可用作抗衰老剂、生发剂和保湿剂,但是未见研究番红花提取物在抗蓝光方面的效用。

发明内容
本发明以米糠为原料制备获得了一种富含谷维素的活性成分,惊人发现,该活性成分表现出卓越的皮肤防护效果,对蓝光及蓝光引起的色沉具有优异的抵御作用。
本发明是通过以下技术方案实现上述发明目的的:一种具有抗衰老和抗蓝光功效的护肤组合物,其特征在于,至少包含:
5~15%重量的来源于米糠的组分;
2~4%重量的儿茶提取物;和
1~3%重量的番红花提取物。
进一步地,所述来源于米糠的组分的制备包括:
S1、往米糠中加入去离子水和乙酸乙酯,分散搅拌均匀,得料液;
S2、将料液置于双频超声波下进行处理,过滤,收集滤液,冻干,即得。
进一步地,所述步骤S1中,米糠、水和乙酸乙酯的重量比为1:1:1~3。更优选地,重量比为1:1:1,三者的重量比对产物中谷维素的含量影响不大,但是当料液中水分较低时,表现较为粘稠,影响后续超声波处理效果。
米糠中含有丰富的蛋白质、脂肪以及一些生物活性化合物,如维生素E和谷维素,其中谷维素的抗氧化活性是维生素E的4倍,具有降低胆固醇、抗炎、抗衰老、神经保护等生理功能。发明人惊人地发现,与其他加工方式如高温蒸煮等,经双频超声波处理后,产物中谷维素的含量不降低,反而成倍地增加,且试验显示,与单频超声相比,双频超声产生的杂乱的超声波更利于米糠中谷维素的释放,与单频相比,产物中谷维素的含量提高了1.5倍左右。
进一步地,双频超声处理中,超声波的频率大小显得尤为重要,出于谷维素含量增加的目的,经多试验筛选发现,将米糠同时置于超声波频率为28kHz和68kHz的超声下处理5~10min,能够在不破坏谷维素的情况下增加更多的谷维素。具体地,上述参数中双频可采用不同超声波频率的超声波振板组合实现,在处理时,两个超声波振板间隔为20~50cm之间,可分别对称放置于超声波槽内的上下侧或左右侧。
所述双频超声波处理具体到本申请中,是指同时采用超声波频率为28kHz和68kHz同时超声5~10min,而单频超声波是指只采用一个频率的超声波处理。
所述超声功率是指上下超声波振板的功率为500~600W。
进一步地,所述超声时间为8min,超声功率为600W。
进一步地,所述儿茶提取物是使用醇从儿茶中提取获得的产物。需要说明的是,所述醇可以是乙醇、甲醇等,优选为乙醇,更优选为70~85%体积分数的乙醇,更优选是采用70~85%体积分数的乙醇通过加热回流提取制备;更优选地是,加热回流提取的提取温度为80~95℃,提取次数为1~3次。相比,水溶性儿茶提取物,采用醇溶性儿茶提取物与配方中其余两种功效成分复合后抗衰老及抗蓝光功效更优异。
进一步地,所述番红花提取物是使用醇从番红花中提取获得的产物。需要说明的是,所述醇可以是乙醇、甲醇等,优选为甲醇,更优选为60~75%体积分数的甲醇,更优选是采用60~75%体积分数的60~75%甲醇通过加热回流提取制备;更优选地是,加热回流提取的提取温度为80~92℃,提取次数为1~3次。相比,水溶性儿茶提取物,采用醇溶性儿茶提取物与配方中其余两种功效成分复合后抗衰老及抗蓝光功效更优异。
本发明另一目的在于提供所述护肤组合物在制备具有抗衰老、抗蓝光功效的护肤品中的用途。
具体地,本发明所述护肤组合物可用于增加皮肤弹性,减少肌肤皱纹,提升肌肤亮泽度,抵抗蓝光伤害,清除蓝光自由基,祛除蓝光造成的黑色素沉着。
另外,本发明还提供一种具有抗衰老、抗蓝光功效的护肤品,其包含所述护肤组合物。具体地,在所述护肤品中,所述护肤组合物可作为主要的功效成分存在,同时,在所述护肤品中,所述护肤组合物以0.1~99.99%的重量存在,所述护肤组合物在所述护肤品中的重量可以是0.1~99.99%之间的任意值。除此之外,为了制备成稳定的护肤品制剂,所述护肤品还可以包括化妆品常用的添加剂,包括溶剂、增稠剂、乳化剂、防腐剂、保湿剂、香精等。该护肤组合物对蓝光形成的各种自由基,如羟自由基,超氧阴离子自由基等具有优异的清除效果,且对蓝光造成的黑色素沉着具有显著的抑制作用。
本发明的有益效果在于:
本发明采用双频超声处理米糠获得了一种富含谷维素的米糠超声组分,将该组分与儿茶提取物、番红花提取物复合后呈现了惊人的抗衰老和抗蓝光效用。

具体实施方式
为更好地说明本发明的目的、技术方案和优点,下面将结合具体实施例对本发明作进一步说明。
实施例一、具有抗衰老和抗蓝光功效的护肤组合物
由12%重量的来源于米糠的组分、3%重量的儿茶提取物和2%重量的番红花提取物组成;其中:
来源于米糠的组分的制备:
S1、按照1:1:1的重量比往米糠中加入去离子水和乙酸乙酯,分散搅拌均匀,得料液;
S2、将上述料液置于设置有上下超声波振板的超声波槽内,控制上下超声波振板的超声波频率分别为28kHz和68kHz,超声处理8min,上下超声波振板的功率为600W;过滤,收集滤液,旋转蒸发除去乙酸乙酯,冻干,即得。
儿茶提取物的制备:取儿茶的茎叶,洗净,干燥,粉碎,加入其重量2倍量的75%体积分数的乙醇溶液,在90℃下加热回流提取2次,过滤,合并滤液,浓缩,冻干,即得。
番红花提取物的制备:取番红花柱头蕊花,洗净,干燥,粉碎,加入其重量2倍量的65%体积分数的甲醇溶液,在85℃下加热回流提取2次,过滤,合并滤液,浓缩,冻干,即得。
按照配方比例将来源于米糠的组分、儿茶提取物及番红花提取物混合均匀,即得护肤组合物。
实施例二、具有抗衰老和抗蓝光功效的护肤组合物
由5%重量的来源于米糠的组分、4%重量的儿茶提取物和3%重量的番红花提取物组成;其中:
来源于米糠的组分的制备:
S1、按照1:1:1的重量比往米糠中加入去离子水和乙酸乙酯,分散搅拌均匀,得料液;
S2、将上述料液置于设置有上下超声波振板的超声波槽内,控制上下超声波振板的超声波频率分别为28kHz和68kHz,超声处理5min,上下超声波振板的功率为600W;过滤,收集滤液,旋转蒸发除去乙酸乙酯,冻干,即得。
儿茶提取物的制备:取儿茶的茎叶,洗净,干燥,粉碎,加入其重量2倍量的80%体积分数的乙醇溶液,在95℃下加热回流提取2次,过滤,合并滤液,浓缩,冻干,即得。
番红花提取物的制备:取番红花柱头蕊花,洗净,干燥,粉碎,加入其重量2倍量的60%体积分数的甲醇溶液,在80℃下加热回流提取2次,过滤,合并滤液,浓缩,冻干,即得。
按照配方比例将来源于米糠的组分、儿茶提取物及番红花提取物混合均匀,即得护肤组合物。
实施例三、具有抗衰老和抗蓝光功效的护肤组合物
由15%重量的来源于米糠的组分、2%重量的儿茶提取物和1%重量的番红花提取物组成;其中:
来源于米糠的组分的制备:
S1、按照1:1:1的重量比往米糠中加入去离子水和乙酸乙酯,分散搅拌均匀,得料液;
S2、将上述料液置于设置有上下超声波振板的超声波槽内,控制上下超声波振板的超声波频率分别为28kHz和68kHz,超声处理10min,上下超声波振板的功率为500W;过滤,收集滤液,旋转蒸发除去乙酸乙酯,冻干,即得。
儿茶提取物的制备:取儿茶的茎叶,洗净,干燥,粉碎,加入其重量2倍量的85%体积分数的乙醇溶液,在80℃下加热回流提取2次,过滤,合并滤液,浓缩,冻干,即得。
番红花提取物的制备:取番红花柱头蕊花,洗净,干燥,粉碎,加入其重量2倍量的75%体积分数的甲醇溶液,在92℃下加热回流提取2次,过滤,合并滤液,浓缩,冻干,即得。
按照配方比例将来源于米糠的组分、儿茶提取物及番红花提取物混合均匀,即得护肤组合物。
对比例一护肤组合物:与实施例一相比,区别在于,来源于米糠的组分制备过程中,在超声处理步骤,采用单频超声波处理替换双频超声波处理,具体操作为:按照1:1:1的重量比往米糠中加入去离子水和乙酸乙酯,分散搅拌均匀,得料液,将料液置于超声波槽内,控制超声波频率为68kHz,超声功率为600W,超声处理8min。
对比二护肤组合物:与实施例一相比,区别在于,采用高温蒸煮替代双频超声波处理,具体操作为:按照1:1的重量比往米糠中加入去离子水,蒸煮2h,低温烘干,即得。
采用紫光分光光度计法对实施例一~三以及对比例一~二护肤组合物中所述的来源于米糠的组分中谷维素的含量进行测定:
谷维素标准曲线制备:精密称取谷维素对照品0.02g,置于50ml容量瓶中,加入40ml三氯甲烷,60℃水浴使谷维素溶解,冷却至室温,加入三氯甲烷稀释至刻度,摇匀,得谷维素标准溶液;精确吸取谷维素标准溶液0.25、0.5、1.0、1.25、1.5ml,分别置于50ml容量瓶中,加入三氯甲烷稀释至刻度,摇匀,制成一系列浓度的对照品溶液,用紫外分光光度法,以三氯甲烷为空白,在327nm波长处测定吸收度A,绘制并记录标准曲线,以吸光度对质量浓度进行线性回归,得回归方程y=0.0465x+0.0072,R2=0.9995,标准曲线见图1。
样品液制备:分别取1g各组样品,置于50ml容量瓶中,加入40ml三氯甲烷,60℃水浴使谷维素溶解,冷却至室温,加入三氯甲烷稀释至刻度,摇匀,得储备液;取1ml上述储备液,置于50ml容量瓶中,加入三氯甲烷稀释至刻度,摇匀,得样品液;于327nm波长处测定其吸光度,根据吸光度对质量浓度的标准曲线回归方程计算谷维素的含量,结果如表1所示。
式中,ρ为根据标准曲线所得谷维素质量浓度,μg/mL;m为样品质量,g。

由表1分析可知,双频、单频超声波均可促进米糠中谷维素的溶出,因为超声产生的高空化作用能够使米糠中分子结构更为疏松,大量释放出谷维素,而高温蒸煮处理谷维素含量较其他两种方式低,其中,对谷维素含量影响最大的是双频超声波处理,获得的产物中谷维素的含量成倍地增加,与其余组存在极显著的差异(P<0.01)。
实施例一~三以及对比例一~二中所述来源于米糠的组分抗蓝光效果评价
取6只比色皿,在其透光一侧贴附大小为1cm×4cm的3M医用透气胶带,其中5只比色皿分别在胶带上均匀涂抹5mg实施例一~三以及对比例一~二护肤组合物中所述的来源于米糠的组分,另一只比色皿作为空白组,将其置于避光处,室温下放置30min,然后用空白组调零后,通过紫外分光光度计测定样品在400~500nm处的紫外吸光度值,每组做5个重复,结果如表2所示。

由表2可看出,蓝光吸收能力与来源于米糠的组分中谷维素含量大致呈现正相关关系,谷维素含量越高,其蓝光吸收能力越强,其中,实施例一~三在400~480nm时的吸光度值均大于1,表示其能够完全吸收此波段的蓝光。
抗蓝光和清除自由基功效评价
按照上文抗蓝光试验步骤对实施例一~三及对比例一~二护肤组合物的抗蓝光效果进行测试,结果如表3所示。

由表3可知,将经双频超声处理得到的米糠成分与儿茶提取物及番红花提取物复配抗蓝光效果有所增强。
清除自由基试验
采用75%的乙醇溶液将实施例一~三护肤组合物及对比例一~二配制成1%浓度的样品液,对其清除羟自由基、超氧阴离子自由基的能力进行测试:
超氧阴离子自由基清除能力测试:取0.05mol/L pH=8.2的Tris-HCl缓冲液4.5ml,于25℃水浴锅中加热20min,加入1ml样品溶液和0.4ml 25mol/L的邻苯三酚溶液,混匀,25℃水浴锅中反应5min,加入8mol/L的HCl1.0ml终止反应。以Tris-HCl缓冲液作为参照,在299nm处测定其吸光度值,空白对照用1ml乙醇溶液来替代样品,结果如表4所示。
超氧阴离子自由基清除率(%)=[1-(A2/A1)]×100%
式中,A1为空白对照组的吸光度值;A2为样品的吸光度值。
羟自由基清除能力测试:在25ml比色管中加入2mmol/L FeSO4 3mL,1mmol/LH2O23mL,摇匀,然后加入6mmol/L水杨酸3mL,摇匀,37℃水浴加热15min,测定其吸光度;分别加入2mL样品液,摇匀水浴加热15min,取出测定其吸光度,结果如表4所示。
羟自由基清除率(%)=[A1-A2-(A1-A3)]/A1×100%
式中,A1为未加样品前反应体系的吸光度值;A2为样品清除羟自由基后体系的吸光度值;A3为空白对照组清除羟自由基后体系的吸光度值。

由表4可知,本发明实施例一~三护肤化合物具有良好的抗氧化能力,对超氧阴离子自由基和羟自由基具有较高的清除率。
最后所应当说明的是,以上实施例用以说明本发明的技术方案而非对本发明保护范围的限制,尽管参照较佳实施例对本发明作了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者同等替换,而不脱离本发明技术方案的实质和范围。
Claims (10)
1.一种具有抗衰老和抗蓝光功效的护肤组合物,其特征在于,至少包含:
5~15%重量的来源于米糠的组分;
2~4%重量的儿茶提取物;和
1~3%重量的番红花提取物。
2.根据权利要求1所述护肤组合物,其特征在于,所述来源于米糠的组分的制备包括:
S1、往米糠中加入去离子水和乙酸乙酯,分散搅拌均匀,得料液;
S2、将料液置于双频超声波下进行处理,过滤,收集滤液,冻干,即得。
3.根据权利要求2所述护肤组合物,其特征在于,所述步骤S1中,米糠、水和乙酸乙酯的重量比为1:1:1~3。
4.根据权利要求2或3所述护肤组合物,其特征在于,所述双频超声波处理的操作参数为:超声频率为28kHz和68kHz,超声时间为5~10min,超声功率为500~600W。
5.根据权利要求4所述护肤组合物,其特征在于,所述超声时间为8min,超声功率为600W。
6.根据权利要求1所述护肤组合物,其特征在于,所述儿茶提取物是使用醇从儿茶中提取获得的产物。
7.根据权利要求7所述护肤组合物,其特征在于,所述番红花提取物是使用醇从番红花中提取获得的产物。
8.根据权利要求7所述护肤组合物,其特征在于,所述儿茶提取物是使用乙醇从儿茶中提取获得的产物;所述番红花提取物是采用甲醇从番红花中提取获得的产物。
9.根据权利要求1~8任一所述护肤组合物在制备具有抗衰老、抗蓝光功效的护肤品中的用途。
10.一种具有抗衰老、抗蓝光功效的护肤品,其特征在于,包含根据权利要求1~8任一所述护肤组合物。
