本发明公开了一种近红外二区荧光探针分子,其特征在于,以吡嗪衍生物为受体,连接供体单元,具有供体‑受体‑供体的分子结构,所述的近红外二区荧光探针分子的结构式如式(I)所示,具有近红外二区荧光发射,荧光发射波长为958nm;且具有高达120倍的单线态活性氧的产生能力与具有高达70℃的光热温度,集近红外二区荧光发射、优异的活性氧产生能力与光热效果于一体。进一步可制备得到用于多模式诊疗一体化的纳米粒子,该纳米粒子对肿瘤细胞的杀灭能力强、组织穿透力强、成像质量优异、生物相容性好,能够在保证生物体健康的同时抑制其体内肿瘤的生长。
Description
技术领域
本发明属于生物医用材料领域,具体涉及一种近红外二区荧光探针分子及用于多模式诊疗一体化的纳米粒子。
背景技术
光学成像在当代医学中起着越来越重要的作用,荧光成像具有高灵敏度、高稳定性、高时间和空间分辨率以及实时收集的优点。由于生物组织的高吸收,散射和自发荧光作用,荧光成像在生物体内应用的主要瓶颈是组织穿透深度浅、信噪比低(SBR)、自发荧光、淬灭、光致漂白等问题。为了解决上述问题,近红外一区荧光材料被开发,但大多数仅仅具有单一的诊断成像或者治疗作用,且穿透深度与发光效率仍然有待提高,难以进一步推进荧光材料在临床中的应用。因此有必要继续开发发射波长更长,发光更亮的荧光材料,能更加有效地在深部肿瘤中成像,并且能够实现诊疗一体化。
第二个近红外窗口(NIR-II,1000-1700nm)比可见光(400-700nm)和第一个近红外窗口(NIR-I,700-900nm)具有更好的成像质量(高分辨率)和更深的组织穿透力而受到广泛关注。
当前研究中,主要的光疗手段包括两种:一种是活性氧治疗,利用的是单线态活性氧具有较高的氧化活性,能有效对肿瘤细胞进行杀伤消融;另一种是光热治疗,高温度的热量能有效对肿瘤细胞进行消融。近红外成像与治疗一体化的材料在临床无创诊疗方面具有很大潜力。
公开号为CN111689950A的中国专利文献公开了一种基于二苯胺基的有机双杂环近红外荧光探针,该近红外荧光探针的发射波长位于近红外光区,为690nm;公开号为CN111961040A的中国专利文献公开了一种基于咔唑基的有机双杂环近红外荧光探针,该近红外荧光探针的发射波长同样位于近红外光区,为716nm;上述两种近红外荧光探针都可作为荧光成像试剂使用,但成像质量以及组织穿透力有待进一步提高。
公开号为CN108864106A的中国专利文献公开了一种近红外二区有机小分子荧光探针,其主要成分为吡咯并吡咯二酮(DPP)衍生物,进一步通过纳米共沉淀方法制备得到具有优良的近红外二区荧光成像及光声成像能力、光动力及光热转换性能的纳米颗粒,但该近红外二区有机小分子荧光探针的结构较为复杂,合成困难。
发明内容
本发明提供了一种近红外二区荧光探针分子,以吡嗪衍生物为受体,连接相同或不同的供体单元,形成供体-受体-供体的分子结构,具有近红外二区荧光发射以及良好的活性氧产生能力与光热效果,能够制备得到用于多模式诊疗一体化的纳米粒子,该纳米粒子对肿瘤细胞的杀灭能力强、成像质量优异,能够在保证生物体健康的同时抑制其体内肿瘤的生长。
具体采用的技术方案如下:
一种近红外二区荧光探针分子,以吡嗪衍生物为受体,连接供体单元D1和D2,具有供体-受体-供体的分子结构,结构式如式(I)所示:
式(I)中,n为1~10的整数;X为元素S、Se或Te中的任意一种;Y为元素O、S、N或Se中的任意一种;
D1和D2分别选自式(II-1)-(II-8)中的任意一种:
式(II-1)-(II-8)中,x、y和z分别为1~20中任意整数,为取代的位置。
本发明通过以吡嗪衍生物为受体,连接相同或不同的供体单元,构筑供体-受体-供体型(D-A-D)化合物。由于D-A-D结构,能有效促使分子内的电荷转移,使分子的LUMO与HOMO能级有效分离,促进CT态有效使分子的发射波长更长,并且通过理论计算结果可以得知,该结构的三重态能量为1.33ev,单重激发态的能量为1.65ev,可以知道该结构具有较小的△Est,更利于电子的跃迁,并且根据报道三重态能量高于0.98ev能有效地产生活性氧,可知该结构能有效地产生活性氧的能力,并且实验结果也证明了这一点。
本发明还提供了所述的近红外二区荧光探针分子的制备方法,包括以下步骤:
(1)将式(Ⅲ)所示的二硝基化合物还原,反应得到式(Ⅳ)所示的二胺化合物;
(2)将步骤(1)得到的二胺化合物与苯偶酰在酸性条件下发生环化反应,得到式(Ⅴ)所示的中间体;
(3)将步骤(2)得到的中间体与N-溴代丁二酰亚胺发生Wohl-Ziegler反应,得到式(Ⅵ)所示的二溴代化合物;
(4)将步骤(3)得到的二溴代化合物与供体共混,在钯催化剂、碱性条件下进行Suzuki偶联反应得到所述的近红外二区荧光探针分子;
其中,式(Ⅲ)-(Ⅵ)中,X、Y与n的定义同上;
优选的,步骤(2)中,酸性条件由冰醋酸提供,反应时间为8-12h。
优选的,步骤(3)中,采用氯仿和乙酸的混合溶剂,氯仿与乙酸的比例为1:1,反应原料在保护性氛围中,冰浴的下加入溶剂,随后在室温下继续反应,反应时间为8-12h。
优选的,步骤(4)中,碳酸钾提供碱性条件,采用四三苯基膦钯催化剂,在保护性氛围下反应,反应温度为80~95℃,反应时间为12-24h。
本发明还提供了一种用于多模式诊疗一体化的纳米粒子,成分包括所述的近红外二区荧光探针分子。
所述的近红外二区荧光探针分子具有近红外二区荧光发射以及良好的活性氧产生能力与光热效果,较高的氧化活性以及高温度的热量可以有效地对肿瘤细胞进行杀伤消融;制备得到的纳米粒子对肿瘤细胞的杀灭能力强、成像质量优异,能够在保证生物体健康的同时抑制其体内肿瘤的生长,可用于肿瘤的多模式诊疗一体化。
本发明还提供了所述纳米粒子的制备方法,包括以下步骤:将所述的近红外二区荧光探针分子与表面活性剂混合,共沉淀制备得到所述纳米粒子。
优选的,所述的表面活性剂为DSPE-PEG,所述的近红外二区荧光探针分子与表面活性剂的质量比为1:1~3。上述表面活性剂和原料比的选择使得制得的纳米粒子的作用效果更好。
本发明还提供了所述纳米粒子在肿瘤诊断和/或肿瘤治疗中的应用。
与现有技术相比,本发明的有益效果在于:
(1)本发明提供的近红外二区荧光探针分子具有近红外二区荧光发射,相对荧光量子产率达到2.92%;而现有技术中绝大多数近红外二区荧光材料的相对荧光量子产率低于2%,该近红外二区荧光探针分子在小鼠体内具有较亮的荧光信号。
(2)本发明提供的近红外二区荧光探针分子具有高达120倍的单线态活性氧的产生能力,能有效对肿瘤细胞进行杀伤作用,起到一定的治疗效果。
(3)本发明提供的近红外二区荧光探针分子具有高达70℃的光热温度,在小鼠体内也能有高达65.2℃的温度,能有效进行肿瘤细胞的消融。
(4)本发明提供的近红外二区荧光探针分子集荧光、光动力以及光热的效果于一体,进一步制备得到的纳米粒子可以实现细胞层面的荧光成像、细胞相容性好;且该纳米粒子在生物体内成像效果优异,且能够在保证生物体健康的同时杀灭或消融肿瘤细胞,抑制体内肿瘤的生长,可用于多模式诊断与治疗一体化,在临床肿瘤的无创诊断和治疗方面具有广泛的发展前景。

具体实施方式
下面结合附图与实施例,进一步阐明本发明。应理解,这些实施例仅用于说明本发明,而不用于限制本发明的范围。
实施例1
本实施例制得的近红外二区荧光探针分子的结构式如下式所示:
该近红外二区荧光探针分子(TP-TPA)的合成路线包括以下4个步骤:
步骤(1):还原反应
在保护性气体氛围下,把化合物1-1与锌粉、氯化铵混合,加入10:1的甲醇:水作为溶剂,常温下反应4小时,还原为化合物1-2,化合物1-2可以不提纯直接投入下一步的反应,其中,化合物1-1的按照文献(Synthesis and photovoltaic performance of a seriesof small band gap polymers,Journal of Materials Chemistry,19(30),5336-5342;2009)中公开的合成方法合成。
步骤(2):环化反应
将步骤(1)得到的化合物1-2与苯偶酰(化合物1-3)混合,以1:1的乙醇:乙酸为溶剂,60℃反应12小时后,得到化合物1-4,化合物1-4为紫色固体。
步骤(3):Wohl-Ziegler反应
将步骤(2)制得的化合物1-4溶解在氯仿和乙酸的混合溶剂中,氯仿与乙酸的比例为1:1(冰浴条件下加入溶剂),随后在0℃下加入2倍当量的N-溴代丁二酰亚胺,将全部的反应原料常温搅拌12小时后,得到化合物1-5。
步骤(4):Suzuki偶联反应
将步骤(4)制得的化合物1-5与碳酸钾、化合物1-6混合,加入适量的四三苯基膦钯催化剂,使用甲苯:乙醇:水=8:1:1作为溶剂,在保护性氛围下,95℃反应24小时后,经过提纯可以得到所述的近红外二区荧光探针分子,该近红外二区荧光探针分子为墨绿色固体,核磁共振氢谱图和核磁共振碳谱图分别如图1和图2所示,证明其成功合成。
实施例2
将实施例1的近红外二区荧光探针分子溶液与DSPE-PEG溶液混合(近红外二区荧光探针分子与DSPE-PEG的质量比为1:2),超声的情况下加入到水中,利用共沉淀的方法进行制备纳米粒子(TP-TPANPs)。
样品分析
实施例1制得的近红外二区荧光探针分子的核磁共振氢谱图和核磁共振碳谱图分别如图1和图2所示,证明了该近红外二区荧光探针分子的成功合成。
实施例1制得的近红外二区荧光探针分子的紫外吸收光谱和荧光发射光谱如图3所示,荧光发射波长为958nm;属于近红外二区的发射。
实施例2制得的纳米粒子的直径分布图如图4中的A所示,平均直径约为98nm,TEM图如图4中的B所示。
实施例2制得的用于多模式诊疗一体化的纳米粒子的活性氧产生能力测试结果如图5所示,DCFH为活性氧检测剂,ICG为光敏剂吲哚菁绿,活性氧检测在660nm波长的激光器下进行,该近红外二区荧光探针分子制得的纳米粒子能产生120倍的活性氧。
将实施例1制得的近红外二区荧光探针分子利用表面活性剂包裹成实施例2的水溶性纳米粒子之后,在置于660nm,0.5W/cm2的激光器下,分别对200μg/mL,150μg/mL,100μg/mL,50μg/mL以及纯水的光热效果进行检测,如图6所示,该纳米粒子的光热效果优异,最高能产生70℃的温度,光热转换效率达到36.8%,且光热稳定性好(图7)。
分别利用0μg/mL,10μg/mL,20μg/mL,30μg/mL,40μg/mL,50μg/mL,60μg/mL,80μg/mL,100μg/mL浓度的纳米粒子与4T1癌细胞共培养,在相应的条件下测试该纳米粒子的暗毒性(TP-TPANPs-Dark)和光毒性(TP-TPANPs-Laser),结果如图8所示,该纳米粒子在660nm,0.5W/cm2的激光器下,60-100μg/mL的浓度范围内,对4T1癌细胞的杀伤力极强。
实施例2制得的用于多模式诊疗一体化的纳米粒子辅以激光照射在实验小鼠体内产生的温度变化情况图如图9所示,由实施例1的近红外二区荧光探针分子制得的多模式诊疗一体化的纳米粒子的光热作用优异,辅以激光照射的条件,能够短时间迅速升温,能够消融肿瘤,5min时,照射肿瘤部分的最高温度(Max)为65.7,平均温度(Ave)为58.2。
将浓度为2mg/mL的纳米粒子尾静脉注射150μL至荷瘤实验小鼠内,分别利用活体成像仪对小鼠0h,2h,4h,8h,12h,24h活体成像拍照,观察小鼠肿瘤部位的富集情况,以发光强弱的大小去判定纳米粒子在肿瘤部位的富集情况;结果可知,该纳米粒子在8h时在小鼠肿瘤部位富集情况明显。
经过该纳米粒子对荷瘤实验小鼠20天的观察和治疗,结果得出:该多模式诊疗一体化的纳米粒子能够有效地对肿瘤进行消融,且其他对照组肿瘤正常生长;并且通过对小鼠解剖后,取血、主要器官(心、肝、脾、肺、肾、肠)和肿瘤进行化验,结果表明接受纳米粒子治疗的小鼠,在血生化、血常规、以及主要器官切片等各个指标都在正常范围值内,并且与空白组的结果相似,可见利用该分子进行治疗生物相容性良好,不会对小鼠产生不良的副作用。
实施例2制得的用于多模式诊疗一体化的纳米粒子辅以激光照射对实验小鼠体重与对实验小鼠肿瘤体积的影响图分别如图10和图11所示,较于单独的激光处理和空白对照组,激光+纳米粒子处理方式对肿瘤体积有明显的抑制作用,且三种处理方式下,小鼠的体重均有上涨的趋势,说明该近红外二区荧光探针分子制得的纳米粒子的生物相容性好,能够在保证小鼠健康的同时显著抑制其体内肿瘤的生长。

Claims (9)
1.一种近红外二区荧光探针分子,其特征在于,以吡嗪衍生物为受体,连接供体单元D1和D2,具有供体-受体-供体的分子结构,结构式如式(I)所示:
式(I)中,n为1;X为元素S;Y为元素O、S或N中的任意一种;D1和D2分别选自式(II-1)、(II-2)、(II-6)、(II-8)中的任意一种:
为取代的位置。
2.根据权利要求1所述的近红外二区荧光探针分子的制备方法,其特征在于,包括以下步骤:
(1)将式(Ⅲ)所示的二硝基化合物还原,反应得到式(Ⅳ)所示的二胺化合物;
(2)将步骤(1)得到的二胺化合物与苯偶酰在酸性条件下发生环化反应,得到式(Ⅴ)所示的中间体;
(3)将步骤(2)得到的中间体与N-溴代丁二酰亚胺发生Wohl-Ziegler反应,得到式(Ⅵ)所示的二溴代化合物;
(4)将步骤(3)得到的二溴代化合物与供体共混,在钯催化剂、碱性条件下进行Suzuki偶联反应得到所述的近红外二区荧光探针分子;
其中,式(Ⅲ)-(Ⅵ)中,X、Y的定义同上;
3.根据权利要求2所述的近红外二区荧光探针分子的制备方法,其特征在于,步骤(2)中,酸性条件由乙酸提供,反应时间为8-12h。
4.根据权利要求2所述的近红外二区荧光探针分子的制备方法,其特征在于,步骤(3)中,使用氯仿和乙酸作为溶剂,反应条件为室温,反应时间为8-12h。
5.根据权利要求2所述的近红外二区荧光探针分子的制备方法,其特征在于,步骤(4)中,碳酸钾提供碱性条件,采用四三苯基膦钯催化剂,在保护性氛围下反应,反应温度为80~95℃,反应时间为12-24h。
6.一种用于多模式诊疗一体化的纳米粒子,其特征在于,成分包括权利要求1所述的近红外二区荧光探针分子。
7.根据权利要求6所述的用于多模式诊疗一体化的纳米粒子的制备方法,其特征在于,包括以下步骤:将所述的近红外二区荧光探针分子与表面活性剂混合,共沉淀制备得到所述纳米粒子。
8.根据权利要求7所述的用于多模式诊疗一体化的纳米粒子的制备方法,其特征在于,所述的表面活性剂为DSPE-PEG,所述的近红外二区荧光探针分子与表面活性剂的质量比为1:1~3。
9.根据权利要求6所述的用于多模式诊疗一体化的纳米粒子在制备肿瘤诊断和/或肿瘤治疗产品中的应用。
