Abstract
本发明公开了一种磷钾矿区土壤微生物修复菌剂,微生物修复菌剂包括活化地衣芽孢杆菌、活化胶冻样芽孢杆菌、活化枯草芽孢杆菌、哈茨木霉菌、活化溶磷菌、硅酸盐细菌、天然柠檬酸提取物、电气石颗粒、陶粒、负离子材料颗粒和淀粉明胶薄膜。本发明的活化地衣芽孢杆菌、活化胶冻样芽孢杆菌、活化枯草芽孢杆菌、哈茨木霉菌撒入土壤之后可以迅速激活繁殖,进而形成种群优势压制有害菌的繁殖扩散,多种微生物菌在生长过程中产生多粘菌素、制霉菌素、短杆菌肽等活性物质,活性物质对致病菌或内源性感染的条件致病菌有明显的抑制作用,同时还能释放土壤中的养分,活化溶磷菌和硅酸盐细菌能够大幅度的降低土壤地区中的固态磷钾元素,促进磷钾元素的消化。

技术领域
本发明涉及一种微生物修复菌剂,特别涉及一种磷钾矿区土壤微生物修复菌剂、制备装置以及方法,属于微生物修复菌剂技术领域。
背景技术
矿产是经济发展的基础性资源,为我国的高速经济发展做出巨大贡献。然不合理的矿山开采行为给矿山周边带来严重的生态环境问题,诸如,土壤退化、自然景观破坏、地质灾害、水资源污染、生物多样性降低等。这些环境问题加剧矿山生态系统功能的丧失,最终导致矿山失去原有的环境价值。
磷、钾是农作物不可缺少的重要元素,它们对农作物的作用互不相同,彼此不可替代,在我国,肥料元素比例严重失调,多氮少磷缺钾现象十分严重,急需大力发展磷钾肥,我国的磷资源分布不均,90%以上的磷矿为中低品位矿,我国可溶性钾资源短缺,仅占世界已探明储量钾盐的1.74%,目前我国的钾肥依然主要依赖于进口,另一方面,我国以钾长石为主的难溶性钾资源却十分丰富且分布广,利用难溶性钾资源制取钾肥前景广阔。
磷钾矿区周围的土壤通常受到磷钾元素的影响,使得磷钾元素形成固态的难以降解的块状物,降低了土壤的肥力效果以及耕种性,国内外普遍采用的物化处理方法大都存在处理费用高昂、二次污染严重、普适性差等系列问题,微生物修复具有成本低,修复效果好,对环境及土壤结构破坏小,有利于生态恢复等优点,被广泛地应用到矿山的土壤修复,是一种比较有效的改良土壤的方式,但多数的微生物修复方法效果不显著、修复后容易出现复发效果,限制了该技术的推广应用,因此,开发研究一种新型的磷钾矿区土壤微生物修复菌剂迫在眉睫。
发明内容
本发明的目的在于提供一种磷钾矿区土壤微生物修复菌剂、制备装置以及方法,以解决上述背景技术中提出的问题。
为实现上述目的,本发明提供如下技术方案:一种磷钾矿区土壤微生物修复菌剂,所述微生物修复菌剂包括活化地衣芽孢杆菌、活化胶冻样芽孢杆菌、活化枯草芽孢杆菌、哈茨木霉菌、活化溶磷菌、硅酸盐细菌、天然柠檬酸提取物、电气石颗粒、陶粒、负离子材料颗粒、淀粉明胶薄膜。
作为本发明的一种优选技术方案,所述微生物修复菌剂包括如下重量百分比:活化地衣芽孢杆菌10-15份、活化胶冻样芽孢杆菌5-10份、活性枯草芽孢杆菌10-20份、哈茨木霉菌8-10份、活化溶磷菌5-8份、硅酸盐细菌5-8份、天然柠檬酸提取物2-5份、电气石颗粒6-10份、陶粒20-30份、负离子材料颗粒7-10份和淀粉明胶薄膜1-2份。
作为本发明的一种优选技术方案,所述微生物修复菌剂为多孔隙球体结构,且多孔隙球体结构表面包覆有淀粉明胶薄膜。
作为本发明的一种优选技术方案,一种磷钾矿区土壤微生物修复菌剂的制备装置,所述制备装置包括菌株培养组件、混合调配组件、喷雾干燥组件、高温煅烧组件和胚体锻压组件,所述菌株培养组件通过计量泵与混合调配组件连接,所述混合调配组件与喷雾干燥组件的进料管连通,所述胚体锻压组件与高温煅烧组件连接,其中:

作为本发明的一种优选技术方案,一种磷钾矿区土壤微生物修复菌剂的制备方法,包括以下方法:
S1:配置微生物培养液,微生物培养液每升溶液中包含:蔗糖25g/L,葡萄糖酸钠20g/L,蛋白胨15g/L,酵母粉8g/L,硫酸铵0.5g/L,硫酸锌0.5g/L,磷酸二氢钾1.0g/L,氯化钙0.5g/L,生长促进剂0.1g/L;
S2:选取如下重量百分比份数的活化地衣芽孢杆菌10份、活化胶冻样芽孢杆菌10份、活性枯草芽孢杆菌20份、哈茨木霉菌10份、活化溶磷菌5份、硅酸盐细菌5份,分别置于步骤S1制备得到的微生物培养液中培养,培养过程中过程中pH控制为6.8-7.2,培养温度控制在26-32℃,搅拌转速控制在200-300r/min,溶氧浓度控制在22%,培养时间8-12小时;
S3:取步骤S2培养后的活化地衣芽孢杆菌培养液、活化胶冻样芽孢杆菌培养液、活性枯草芽孢杆菌培养液、哈茨木霉菌培养液、活化溶磷菌培养液、硅酸盐细菌培养液,按照份量配比混合成微生物修复液;
S4:选取电气石颗粒10份、陶粒28份和负离子材料颗粒8份,精细研磨成粉状经过1100-1200°煅烧成颗粒,采用天然粘土材料将颗粒锻压成直径为10毫米的球形载体,再经过1300-1400°烧制定型;
S5:将2份的天然柠檬酸提取物添加到步骤S3得到微生物修复液,搅拌均匀,将均匀后的微生物修复液均匀喷洒在的球形载体上,通过控制喷洒量、烘干温度和时长,使得球形载体浸润有微生物修复液,再将淀粉明胶流体加入到喷雾干燥设备中,调节喷雾干燥温度,均匀喷洒到带有微生物修复液的球形载体上,使得球形载体表面形成淀粉明胶薄膜,得到制成符合标准要求的微生物修复剂。
作为本发明的一种优选技术方案,所述步骤S5中,控制喷洒量、烘干温度和时长的的参数值为(0.85ml/cm2)、烘干温度(T<40℃)和时长(t≤1.5h);喷雾干燥设备为离心式喷雾干燥设备,进风温度400℃,出风温度200℃。
与现有技术相比,本发明的有益效果是:
1.本发明一种磷钾矿区土壤微生物修复菌剂、制备装置以及方法,通过将活化地衣芽孢杆菌、活化胶冻样芽孢杆菌、活化枯草芽孢杆菌、哈茨木霉菌、活化溶磷菌和硅酸盐细菌的混合液混合成微生物修复液,再通过锻压成的球形载体浸润微生物修复液,淀粉明胶薄膜对球形载体进行包覆,起到保护效果,同时分解的淀粉明胶薄膜能够给活化地衣芽孢杆菌、活化胶冻样芽孢杆菌、活化枯草芽孢杆菌、哈茨木霉菌、活化溶磷菌和硅酸盐细菌提供能量,活化地衣芽孢杆菌、活化胶冻样芽孢杆菌、活化枯草芽孢杆菌、哈茨木霉菌撒入土壤之后可以迅速激活繁殖,进而形成种群优势压制有害菌的繁殖扩散,多种微生物菌在生长过程中产生多粘菌素、制霉菌素、短杆菌肽等活性物质,活性物质对致病菌或内源性感染的条件致病菌有明显的抑制作用,同时还能释放土壤中的养分,促进作物的生长,活化溶磷菌和硅酸盐细菌能够大幅度的降低土壤地区中的固态磷钾元素,促进磷钾元素的消化,防止过渡营养化,天然柠檬酸提取物、电气石颗粒、陶粒、负离子材料颗粒等物质的不仅仅能够提高微生物菌的活性,且能够增加土壤中的微量矿物质,缓慢均匀地释放在土壤中,借助土壤中水分带来的微弱的电流作用,矿物质更容易被植物吸收,实用性强。

S1:配置微生物培养液,微生物培养液每升溶液中包含:蔗糖25g/L,葡萄糖酸钠20g/L,蛋白胨15g/L,酵母粉8g/L,硫酸铵0.5g/L,硫酸锌0.5g/L,磷酸二氢钾1.0g/L,氯化钙0.5g/L,生长促进剂0.1g/L;
S2:选取如下重量百分比份数的活化地衣芽孢杆菌15份、活化胶冻样芽孢杆菌5份、活性枯草芽孢杆菌20份、哈茨木霉菌10份、活化溶磷菌6份、硅酸盐细菌4份,分别置于步骤S1制备得到的微生物培养液中培养,培养过程中过程中pH控制为6.9-7.1,培养温度控制在26-30℃,搅拌转速控制在220r/min,溶氧浓度控制在22%,培养时间9小时;
S3:取步骤S2培养后的活化地衣芽孢杆菌培养液、活化胶冻样芽孢杆菌培养液、活性枯草芽孢杆菌培养液、哈茨木霉菌培养液、活化溶磷菌培养液、硅酸盐细菌培养液,通过,按照份量配比混合成微生物修复液;
S4:选取电气石颗粒10份、陶粒28份和负离子材料颗粒8份,精细研磨成粉状经过1100-1200°煅烧成颗粒,采用天然粘土材料将颗粒锻压成直径为10毫米的球形载体,再经过1300-1400°烧制定型;
S5:将2份的天然柠檬酸提取物添加到步骤S3得到微生物修复液,搅拌均匀,将均匀后的微生物修复液均匀喷洒在的球形载体上,通过控制喷洒量、烘干温度和时长,使得球形载体浸润有微生物修复液,再将淀粉明胶流体加入到喷雾干燥设备中,调节喷雾干燥温度,均匀喷洒到带有微生物修复液的球形载体上,使得球形载体表面形成淀粉明胶薄膜,得到制成符合标准要求的微生物修复剂。
步骤S5中,控制喷洒量、烘干温度和时长的的参数值为(0.85ml/cm2)、烘干温度(T<40℃)和时长(t≤1.5h);喷雾干燥设备为离心式喷雾干燥设备,进风温度400℃,出风温度200℃

S103:取步骤S102培养后的活化地衣芽孢杆菌培养液、活化胶冻样芽孢杆菌培养液、活性枯草芽孢杆菌培养液、哈茨木霉菌培养液、活化溶磷菌培养液、硅酸盐细菌培养液,通过抽取管道109以及计量泵按照份量配比混合成微生物修复液;
S104:选取电气石颗粒10份、陶粒28份和负离子材料颗粒8份,精细研磨成粉状经过1100°煅烧成颗粒,采用天然粘土材料将颗粒锻压成直径为10毫米的球形载体,再经过1300°烧制定型;
S105:将2份的天然柠檬酸提取物添加到步骤S103得到微生物修复液,搅拌均匀,将均匀后的微生物修复液均匀喷洒在的球形载体上,通过控制喷洒量(0.85ml/cm2)、烘干温度(T<40℃)和时长(t≤1.5h),使得球形载体浸润有微生物修复液,再将淀粉明胶流体加入到离心式喷雾干燥设备中,调节喷雾干燥温度,均匀喷洒到带有微生物修复液的球形载体上,使得球形载体表面形成淀粉明胶薄膜,得到制成符合标准要求的微生物修复剂。
实施例二
S201:配置微生物培养液,微生物培养液每升溶液中包含:蔗糖25g/L,葡萄糖酸钠20g/L,蛋白胨15g/L,酵母粉8g/L,硫酸铵0.5g/L,硫酸锌0.5g/L,磷酸二氢钾1.0g/L,氯化钙0.5g/L,生长促进剂0.1g/L;
S202:选取如下重量百分比份数的活化地衣芽孢杆菌15份、活化胶冻样芽孢杆菌10份、活性枯草芽孢杆菌15份、哈茨木霉菌10份、活化溶磷菌5份、硅酸盐细菌5份,分别置于步骤S201制备得到的微生物培养液中培养,培养过程中过程中pH控制为7.0-7.2,培养温度控制在30-32℃,搅拌转速控制在260-280r/min,溶氧浓度控制在22%,培养时间10小时;
S203:取步骤S202培养后的活化地衣芽孢杆菌培养液、活化胶冻样芽孢杆菌培养液、活性枯草芽孢杆菌培养液、哈茨木霉菌培养液、活化溶磷菌培养液、硅酸盐细菌培养液,按照份量配比混合成微生物修复液;
S204:选取电气石颗粒10份、陶粒28份和负离子材料颗粒8份,精细研磨成粉状经过1200°煅烧成颗粒,采用天然粘土材料将颗粒锻压成直径为10毫米的球形载体,再经过1400°烧制定型;
S205:将2份的天然柠檬酸提取物添加到步骤S203得到微生物修复液,搅拌均匀,将均匀后的微生物修复液均匀喷洒在的球形载体上,通过控制喷洒量、烘干温度和时长,使得球形载体浸润有微生物修复液,再将淀粉明胶流体加入到喷雾干燥设备中,调节喷雾干燥温度,均匀喷洒到带有微生物修复液的球形载体上,使得球形载体表面形成淀粉明胶薄膜,得到制成符合标准要求的微生物修复剂。

本发明通过将活化地衣芽孢杆菌、活化胶冻样芽孢杆菌、活化枯草芽孢杆菌、哈茨木霉菌、活化溶磷菌和硅酸盐细菌的混合液混合成微生物修复液,再通过锻压成的球形载体浸润微生物修复液,淀粉明胶薄膜对球形载体进行包覆,起到保护效果,同时分解的淀粉明胶薄膜能够给活化地衣芽孢杆菌、活化胶冻样芽孢杆菌、活化枯草芽孢杆菌、哈茨木霉菌、活化溶磷菌和硅酸盐细菌提供能量,活化地衣芽孢杆菌、活化胶冻样芽孢杆菌、活化枯草芽孢杆菌、哈茨木霉菌撒入土壤之后可以迅速激活繁殖,进而形成种群优势压制有害菌的繁殖扩散,多种微生物菌在生长过程中产生多粘菌素、制霉菌素、短杆菌肽等活性物质,活性物质对致病菌或内源性感染的条件致病菌有明显的抑制作用,同时还能释放土壤中的养分,促进作物的生长,活化溶磷菌和硅酸盐细菌能够大幅度的降低土壤地区中的固态磷钾元素,促进磷钾元素的消化,防止过渡营养化,天然柠檬酸提取物、电气石颗粒、陶粒、负离子材料颗粒等物质的不仅仅能够提高微生物菌的活性,且能够增加土壤中的微量矿物质,缓慢均匀地释放在土壤中,借助土壤中水分带来的微弱的电流作用,矿物质更容易被植物吸收,实用性强。

Claims (10)
1.一种磷钾矿区土壤微生物修复菌剂,其特征在于,所述微生物修复菌剂包括活化地衣芽孢杆菌、活化胶冻样芽孢杆菌、活化枯草芽孢杆菌、哈茨木霉菌、活化溶磷菌、硅酸盐细菌、天然柠檬酸提取物、电气石颗粒、陶粒、负离子材料颗粒和淀粉明胶薄膜。
2.根据权利要求1所述的一种磷钾矿区土壤微生物修复菌剂,所述微生物修复菌剂包括如下重量百分比:活化地衣芽孢杆菌10-15份、活化胶冻样芽孢杆菌5-10份、活性枯草芽孢杆菌10-20份、哈茨木霉菌8-10份、活化溶磷菌5-8份、硅酸盐细菌5-8份、天然柠檬酸提取物2-5份、电气石颗粒6-10份、陶粒20-30份、负离子材料颗粒7-10份和淀粉明胶薄膜1-2份。
3.根据权利要求1所述的一种磷钾矿区土壤微生物修复菌剂,所述微生物修复菌剂为多孔隙球体结构,且多孔隙球体结构表面包覆有淀粉明胶薄膜。
4.一种磷钾矿区土壤微生物修复菌剂的制备装置,其特征在于,所述制备装置包括菌株培养组件(1)、混合调配组件(2)、喷雾干燥组件(3)、高温煅烧组件(4)和胚体锻压组件(5),所述菌株培养组件(1)通过计量泵与混合调配组件(2)连接,所述混合调配组件(2)与喷雾干燥组件(3)的进料管连通,所述胚体锻压组件(5)与高温煅烧组件(4)连接,其中:
所述喷雾干燥组件(3)为喷雾干燥设备为离心式喷雾干燥设备。
5.根据权利要求4所述的一种磷钾矿区土壤微生物修复菌剂的制备装置,其特征在于:所述菌株培养组件(1)包括六间隙培养箱体(101)和加热底座(108),所述加热底座(108)的顶端与六间隙培养箱体(101)固定连接,所述六间隙培养箱体(101)的顶端安装有防菌盖(102),所述防菌盖(102)的顶端固定安装有六个传动齿盘(104),六个所述传动齿盘(104)之间通过传动齿带传动连接,其中一个所述传动齿盘(104)的顶端安装有驱动马达(103),所述六间隙培养箱体(101)正面的底部和背面的底部均安装有三个抽取管道(109),所述六间隙培养箱体(101)正面的顶部和背面的顶部均安装有加注管道(107),所述防菌盖(102)一侧的中部安装有进气管道(105),所述进气管道(105)的一端固定安装有抽气泵(106)。
6.根据权利要求4所述的一种磷钾矿区土壤微生物修复菌剂的制备装置,其特征在于:所述混合调配组件(2)包括静态混合器(202),所述静态混合器(202)的一端固定安装有六管道端口(201),所述静态混合器(202)的顶端固定安装有加注料斗(203),所述静态混合器(202)的另一端固定安装有出口法兰(204)。
7.根据权利要求4所述的一种磷钾矿区土壤微生物修复菌剂的制备装置,其特征在于:所述高温煅烧组件(4)包括煅烧箱体(401),所述煅烧箱体(401)内腔的顶部固定安装有连接框架(406),所述连接框架(406)的内部安装有精细研磨辊(405),所述精细研磨辊(405)的一端穿过煅烧箱体(401)并固定安装有驱动电机(407),所述连接框架(406)的底端固定安装有精细筛网(404),所述煅烧箱体(401)内腔的底部固定安装有煅烧台(402),所述高温煅烧台体(402)的顶端安装有煅烧盒(403)。
8.根据权利要求4所述的一种磷钾矿区土壤微生物修复菌剂的制备装置,其特征在于:所述胚体锻压组件(5)包括锻压箱(501),所述锻压箱(501)内腔的顶部固定安装有驱动油缸(502),所述驱动油缸(502)的输出端连接有上锻压模具(503),所述锻压箱(501)内腔的底部固定安装有下锻压模具(504),所述下锻压模具(504)与上锻压模具(503)对应连接。
9.一种磷钾矿区土壤微生物修复菌剂的制备方法,其特征在于,包括以下方法:
S1:配置微生物培养液,微生物培养液每升溶液中包含:蔗糖25g/L,葡萄糖酸钠20g/L,蛋白胨15g/L,酵母粉8g/L,硫酸铵0.5g/L,硫酸锌0.5g/L,磷酸二氢钾1.0g/L,氯化钙0.5g/L,生长促进剂0.1g/L;
S2:选取如下重量百分比份数的活化地衣芽孢杆菌10份、活化胶冻样芽孢杆菌10份、活性枯草芽孢杆菌20份、哈茨木霉菌10份、活化溶磷菌5份、硅酸盐细菌5份,分别置于步骤S1制备得到的微生物培养液中培养,培养过程中过程中pH控制为6.8-7.2,培养温度控制在26-32℃,搅拌转速控制在200-300r/min,溶氧浓度控制在22%,培养时间8-12小时;
S3:取步骤S2培养后的活化地衣芽孢杆菌培养液、活化胶冻样芽孢杆菌培养液、活性枯草芽孢杆菌培养液、哈茨木霉菌培养液、活化溶磷菌培养液、硅酸盐细菌培养液,按照份量配比混合成微生物修复液;
S4:选取电气石颗粒10份、陶粒28份和负离子材料颗粒8份,精细研磨成粉状经过1100-1200°煅烧成颗粒,采用天然粘土材料将颗粒锻压成直径为10毫米的球形载体,再经过1300-1400°烧制定型;
S5:将2份的天然柠檬酸提取物添加到步骤S3得到微生物修复液,搅拌均匀,将均匀后的微生物修复液均匀喷洒在的球形载体上,通过控制喷洒量、烘干温度和时长,使得球形载体浸润有微生物修复液,再将淀粉明胶流体加入到喷雾干燥设备中,调节喷雾干燥温度,均匀喷洒到带有微生物修复液的球形载体上,使得球形载体表面形成淀粉明胶薄膜,得到制成符合标准要求的微生物修复剂。
10.根据权利要求9所述的一种磷钾矿区土壤微生物修复菌剂的制备方法,其特征在于:所述步骤S5中,控制喷洒量、烘干温度和时长的的参数值为(0.85ml/cm2)、烘干温度(T<40℃)和时长(t≤1.5h);喷雾干燥设备为离心式喷雾干燥设备,进风温度400℃,出风温度200℃。
