仔猪腹泻是集约化养猪生产条件下的一种典型的高发疾病,导致仔猪腹泻的原因有多种,如病毒性腹泻、细菌性腹泻、营养和饲养管理引起的腹泻、生理性腹泻、应激性腹泻和环境变化引起的腹泻,其中生理性腹泻只是因为仔猪生理系统发育还不够完善,消化系统和体温调节技能不健全,对外界的影响抵抗力差,仔猪免疫力下降,消化功能失调,引发肠道内致病性微生物繁殖而引起的腹泻。其中,病毒感染是引起病毒性腹泻的主要原因之一;研究表明,感染轮状病毒后,仔猪小肠绒毛发生了大量脱落和广泛坏死,最终引起仔猪的水样腹泻和脱水性死亡,轻则延长猪的出栏时间,重则导致疾病恶化、群体性感染和死亡,给养殖户们造成很大的经济损失。

本发明的目的是为了缓解仔猪腹泻的问题,提供预防猪腹泻的饲料添加剂及其制备方法,该预防猪腹泻的饲料添加剂能够提高仔猪抵御猪流行性腹泻病毒的能力,降低仔猪腹泻的概率。
为了实现上述目的,本发明第一方面提供一种预防猪腹泻的饲料添加剂,所述预防猪腹泻的饲料添加剂包含硫代烟酰胺腺嘌呤二核苷酸和单甘脂。
本发明的发明人发现,硫代烟酰胺腺嘌呤二核苷酸和单甘脂复合得到的预防猪腹泻的饲料添加剂,在和饲料调配后,能够有效缓解仔猪腹泻,尤其是猪流行性腹泻病毒感染导致的仔猪腹泻的情况。
根据本发明,为了优化所述预防猪腹泻的饲料添加剂对猪流行性腹泻病毒的抵抗能力,优选条件下,所述硫代烟酰胺腺嘌呤二核苷酸和所述单甘脂的重量比为1:10-100,例如可以是1:10、1:15、1:20、1:25、1:30、1:35、1:40、1:45、1:50、1:60、1:70、1:80、1:90、1:100或上述任意两个数值组成的范围中的任意值,更优选为1:25-45。
根据本发明,为了进一步优化所述预防猪腹泻的饲料添加剂对猪流行性腹泻病毒的抵抗能力,优选条件下,所述单甘脂选自十二酸单甘油酯、1-癸酸单甘油酯和单油酸甘油酯中的至少一种。
根据本发明,为了提高猪对所述饲料添加剂的吸收效果,优选条件下,所述预防猪腹泻的饲料添加剂还包含载体,所述载体选自蒙脱石。
本发明第二方面提供一种制备预防猪腹泻的饲料添加剂的方法,所述方法包括:
将蒙脱石分散于氯化钠溶液中,得到悬浮液;
将硫代烟酰胺腺嘌呤二核苷酸、甘油酯和乳化剂加入所述悬浮液中,然后进行超声分散,得到预防猪腹泻的饲料添加剂。
通过将饲料添加剂与蒙脱石进行复合,能够延长预饲料添加剂在猪肠胃中的停留时间,进而提高猪对所述饲料添加剂的吸收效果,并最终达到提高缓解猪腹泻的效果。
根据本发明,优选条件下,所述氯化钠溶液的重量浓度为3-15wt%。
根据本发明,优选条件下,所述硫代烟酰胺腺嘌呤二核苷酸与所述甘油酯的重量比为1:10-100,优选为1:25-45。
本发明中,为了使所述饲料添加剂能够充分的分散在蒙脱石的表面,优选条件下,所述乳化剂选自司盘80、吐温80、聚氧乙烯蓖麻油、聚氧乙烯十二羟硬脂酸酯、聚氧乙烯甘油三油酸脂、山梨酸单硬脂酸脂、硬脂酰乳酸钠-钙、硬脂酰乳酸钠、三聚甘油酯、丙二醇脂肪酸酯、蔗糖酯和大豆磷脂中的至少一种。
本发明第三方面提供一种预防猪腹泻的饲料,包含基础饲料和饲料添加剂,所述饲料添加剂包含前述第一方面所述的预防猪腹泻的饲料添加剂或根据前述第二方面所述的方法制备得到的预防猪腹泻的饲料添加剂。
根据本发明,优选条件下,所述预防猪腹泻的饲料添加剂的用量为所述基础饲料的0.1-3重量%。
本发明对所述基础饲料的种类没有特殊的要求,可以为本领域技术人员所致,所述基础饲料可以是市售的猪饲料,也可以是根据现有的方法制备得到。
通过上述技术方案,本发明通过将硫代烟酰胺腺嘌呤二核苷酸和单甘脂进行复合,能够有效缓解由轮状病毒引起的仔猪腹泻,且所述预防猪腹泻的饲料添加剂的制备方法简单。
具体实施方式

以下实施例中,硫代烟酰胺腺嘌呤二核苷酸为青岛克斯特生物科技有限公司型号为硫代烟酰胺腺嘌呤二核苷酸(氧化型),4090-29-3的市售品。
实施例1
将100g蒙脱石分散于50mL 9wt%的氯化钠溶液中,得到悬浮液;
将0.5g硫代烟酰胺腺嘌呤二核苷酸、30g单油酸甘油酯和0.3g聚氧乙烯甘油三油酸脂加入所述悬浮液中,然后在真空条件下,在65℃下进行搅拌1h,然后造粒、真空干燥,得到预防猪腹泻的饲料添加剂A1。
实施例2
将100g蒙脱石分散于50mL 9wt%的氯化钠溶液中,得到悬浮液;
将0.5g硫代烟酰胺腺嘌呤二核苷酸、25g单油酸甘油酯和0.3g聚氧乙烯甘油三油酸脂加入所述悬浮液中,然后在真空条件下,在65℃下进行搅拌1h,然后造粒、真空干燥,得到预防猪腹泻的饲料添加剂A2。
实施例3
将100g蒙脱石分散于50mL 9wt%的氯化钠溶液中,得到悬浮液;
将0.5g硫代烟酰胺腺嘌呤二核苷酸、45g单油酸甘油酯和0.3g聚氧乙烯甘油三油酸脂加入所述悬浮液中,然后在真空条件下,在65℃下进行搅拌1h,然后造粒、真空干燥,得到预防猪腹泻的饲料添加剂A3。
实施例4
将100g蒙脱石分散于50mL 9wt%的氯化钠溶液中,得到悬浮液;
将0.5g硫代烟酰胺腺嘌呤二核苷酸、10g单油酸甘油酯和0.3g聚氧乙烯甘油三油酸脂加入所述悬浮液中,然后在真空条件下,在65℃下进行搅拌1h,然后造粒、真空干燥,得到预防猪腹泻的饲料添加剂A4。
实施例5
将100g蒙脱石分散于50mL 9wt%的氯化钠溶液中,得到悬浮液;
将0.5g硫代烟酰胺腺嘌呤二核苷酸、100g单油酸甘油酯和0.3g聚氧乙烯甘油三油酸脂加入所述悬浮液中,然后在真空条件下,在65℃下进行搅拌1h,然后造粒、真空干燥,得到预防猪腹泻的饲料添加剂A5。
实施例6
将100g蒙脱石分散于50mL 9wt%的氯化钠溶液中,得到悬浮液;
将0.5g硫代烟酰胺腺嘌呤二核苷酸、50g 1-癸酸单甘油酯和0.3g聚氧乙烯甘油三油酸脂加入所述悬浮液中,然后在真空条件下,在65℃下进行搅拌1h,然后造粒、真空干燥,得到预防猪腹泻的饲料添加剂A6。
对比例1
按照实施例1的方法,不同的是,采用β-烟酰胺腺嘌呤二核苷酸代替硫代烟酰胺腺嘌呤二核苷酸,得到预防猪腹泻的饲料添加剂B1。
对比例2
按照实施例1的方法,不同的是,采用三油酸甘油酯代替单甘脂,得到预防猪腹泻的饲料添加剂B2。
对比例3
按照实施例1的方法,不同的是,硫代烟酰胺腺嘌呤二核苷酸和所述单油酸甘油酯的重量比为1:1,得到预防猪腹泻的饲料添加剂B3。
对比例4
按照实施例1的方法,不同的是,硫代烟酰胺腺嘌呤二核苷酸和所述单油酸甘油酯的重量比为1:150,得到预防猪腹泻的饲料添加剂B4。
对比例5
采用硫代烟酰胺腺嘌呤二核苷酸作为预防猪腹泻的饲料添加剂B5。
对比例6
采用单油酸甘油酯作为预防猪腹泻的饲料添加剂B6。
实验例1
预防猪腹泻的饲料添加剂对大鼠腹泻模型的缓解作用
选取选用健康28日龄健康仔猪260只,灌胃PEDV病毒(活菌数为1×109CFU/mL),实验组分别喂养添加实施例1-6和对比例1-6的预防猪腹泻的饲料添加剂的饲料(预防猪腹泻的饲料添加剂的添加量为0.5重量%),对照组的饲料中不含有任何添加剂,所有仔猪自由采食和饮水,试验为期28天,每天记录仔猪采食量,观察每头仔猪粪便情况并进行评分以计算腹泻率,试验开始和结束时对仔猪进行称重,实验结果如表1所示。

从表1中可以看出,本发明实施例得到的预防猪腹泻的饲料添加剂,能够明显降低仔猪腹泻的概率。

Claims (5)
1.一种预防猪腹泻的饲料添加剂,其特征在于,所述预防猪腹泻的饲料添加剂包含硫代烟酰胺腺嘌呤二核苷酸和单甘酯;
所述硫代烟酰胺腺嘌呤二核苷酸和所述单甘酯的重量比为1:20或1:50或1:60或1:90;
所述单甘酯选自1-癸酸单甘油酯和单油酸甘油酯中的至少一种。
2.根据权利要求1所述的预防猪腹泻的饲料添加剂,其特征在于,还包含载体,所述载体选自蒙脱石。
3.一种制备预防猪腹泻的饲料添加剂的方法,其特征在于,所述方法包括:
将蒙脱石分散于氯化钠溶液中,得到悬浮液;
将硫代烟酰胺腺嘌呤二核苷酸、甘油酯和乳化剂加入所述悬浮液中,然后进行超声分散,得到预防猪腹泻的饲料添加剂;
所述硫代烟酰胺腺嘌呤二核苷酸和所述单甘酯的重量比为1:20或1:50或1:60或1:90;
所述单甘酯选自1-癸酸单甘油酯和单油酸甘油酯中的至少一种。
4.根据权利要求3所述的方法,其中,所述氯化钠溶液的重量浓度为3-15wt%。
5.根据权利要求3所述的方法,其中,所述乳化剂选自司盘80、吐温80、聚氧乙烯蓖麻油、聚氧乙烯十二羟硬脂酸酯、聚氧乙烯甘油三油酸脂、山梨酸单硬脂酸脂、硬脂酰乳酸钠-钙、硬脂酰乳酸钠、三聚甘油酯、丙二醇脂肪酸酯、蔗糖酯和大豆磷脂中的至少一种。
