2 method
2.1 Single factor experiment
The optimal extraction scheme is comprehensively evaluated by taking the cream yield and the total flavone content as indexes.
Three factors of water addition, extraction time and extraction times are investigated, and the following is a specific method step of a single-factor investigation experiment.
Investigation of the amount of water added: weighing 30g of lotus seed meat, 15g of dried medlar, 15g of cornus officinalis, 15g of rhizoma polygonati preparata and 15g of gordon euryale seed, respectively adding water with volume of 8 times, 10 times and 12 times of the lotus seed meat, soaking for 2 hours, extracting for 10min and extracting for 2 times.
Examination of extraction time: weighing 30g of lotus seed meat, 15g of dried medlar, 15g of cornus officinalis, 15g of rhizoma polygonati preparata and 15g of gordon euryale seed, adding 10 times of water by volume, soaking for 2 hours, and extracting for 10min, 20min and 30min respectively for 2 times.
Examination of extraction frequency: weighing 30g of lotus pulp, 15g of dried medlar, 15g of cornus officinalis, 15g of rhizoma polygonati preparata and 15g of gordon euryale seed, adding 10 times of water by volume, soaking for 2h, extracting for 20min, and extracting for 1, 2 and 3 times respectively.
2.2 Box-Behnken response surface method design experiment
According to a single-factor experiment, 3 factors of water addition amount (A), extraction time (B) and extraction times (C) are selected as objects to be examined, and each factor has three levels, as shown in table 1:

2.3 determination of cream yield
The method for measuring the cream yield comprises the following steps: precisely absorbing 20ml of sample extract, adding into an evaporating dish dried to constant weight, evaporating in water bath, drying in a 105 ℃ oven for 3h, transferring into a drier, cooling for 1h to room temperature, rapidly and precisely weighing the mass, measuring the mass of the dried product by adopting a weight reduction method, and calculating the cream yield according to the following formula (W is the cream mass, M is the medicinal material mass, and V is the volume of the sample solution)
The cream yield (%) = (W × V)/(20 × M) × 100%
2.4 calculation of composite score
Taking the cream yield and the total flavone content as indexes, adopting a weighted comprehensive scoring method, wherein the weight ratio of the cream yield to the total flavone content is 50 and 50 respectively, and calculating the comprehensive scoring according to the following formula:
composite score = (total flavone content/maximum total flavone content + cream yield/maximum cream yield) × 0.5
2.5 determination of Total Flavonoids content
2.5.1 preparation and content determination of Standard substance solution
Precisely weighing 1.7mg of rutin standard substance in a 10ml volumetric flask, and fixing the volume (the concentration is 0.17 mg/ml) by using methanol; respectively taking 0.1ml, 0.2ml, 0.4ml, 0.8ml, 1.6ml and 3.2ml of the solution in a 10ml volumetric flask, and fixing the volume by using methanol; measuring absorbance at 360nm, and drawing a standard curve;
2.5.2 preparation and content determination of test solution
Precisely weighing 10mg of a sample volatilized in a water bath, adding 10ml of methanol, carrying out ultrasonic treatment for 10 minutes, centrifuging for 1000r, carrying out 5min, taking 1ml of supernate, putting the supernate into a 10ml volumetric flask, carrying out constant volume treatment by using the methanol, and measuring the absorbance at 360 nm.
3 results of
3.1 drawing of rutin Standard Curve
Drawing a standard curve according to the absorbance values of rutin with different concentrations measured at 360nm to obtain an equation of the absorbance and the concentration, namely y =1.4346x-0.0217, R2=0.9987, the linear relation is good, and the concentration of the sample can be calculated according to the formula and the measured absorbance of the sample;
3.2 Box-Behnken response surface method design experiment scale
Calculating to obtain total flavone content according to the extract yield formula and rutin standard curve, and obtaining comprehensive score according to the formula, wherein the result is shown in Table 2:

The composite scores were subjected to multiple linear regression using software Design Expert 11, with results as given in table 3:

The P values of the interaction items AB and AC are both greater than 0.05, which indicates that the interaction between A and B is weak, the P value between B and C is less than 0.05, and indicates that the interaction between B and C is certain; A. b, C, wherein P values are all less than 0.05, the influence of A, B, C on comprehensive scores is obvious, wherein P (B) > P (A) > P (C), C (extraction times) has the largest influence on comprehensive scores of the water extraction process of the book-losing and old-fashioned square, A (water addition amount) is used, B (extraction time) has the smallest influence on comprehensive scores, and finally, the influence results of 3 factor levels on the extraction process of the book-losing and old-fashioned square are sequentially C (extraction times) > A (water addition amount) > B (extraction time), a regression model P < 0.0001, the model is obvious, r =0.995, the model fitting degree is good, the mismatching item is greater than 0.05, the model mismatching is not significant, the experimental error is small, and the optimal extraction process of the book-losing and old-fashioned square can be well predicted.
In order to more intuitively see the interaction influence of various factors on the comprehensive scoring of the water extraction process of the old-fashioned prescription, a response surface graph and a contour graph are drawn through software Design Expert 11, and are shown as fig. 1, fig. 2 and fig. 3.
As can be seen from the response surface graph and the contour graph, when one factor is fixed, the influence of the other two factors on the comprehensive score is fixed. As shown in FIG. 1, when C (number of extractions) is fixed, A (amount of water addition) and B (extraction time) are nearly in the same plane, and contour lines are nearly parallel, which indicates that the interaction between A and B is weak; as shown in fig. 2, when B (extraction time) is fixed, when both a (water addition amount) and C (extraction times) are close to the maximum value, the composite score is high; from fig. 3, it can be seen that when a (water addition amount) is fixed, B (extraction time) is small, and C (extraction frequency) is large, the composite score is high. In conclusion, while the influence of the comprehensive factors of the three is considered, the optimal water extraction scheme obtained by software is that the water addition amount is 12 times, the extraction time is 17.638 minutes, the extraction times are 3 times, the obtained paste yield is 37.53%, the total flavone content is 1.367mg/ml, and the comprehensive score is 96.16 highest. In combination, the optimal extraction process was determined to be 12 times the amount of water added, soaking for 2 hours followed by extraction for 18 minutes for a total of 3 times.
Meanwhile, the inventor finds that the device used in the improved water extraction process of the Shangshu Yilao formula in the example 4 adopts the specific device for preparing the paste formula to obtain the paste yield of 38.37 percent and the total flavone content of 1.396mg/ml, and the quality of the paste is better than that of the paste prepared in the examples 1-3.
Comparative example 1
The same as example 1, except that it did not contain polygonatum kingianum, and the number of extraction was 1. Through detection, the cream yield is 28.85%, and the content of the total flavonoids is 1.116 mg/ml.
Comparative example 2
Similar to example 2, except that 40g of lotus seed, 8g of dried medlar, 8g of cornus officinalis, 8g of alcohol solomonseal rhizome and 8g of gordon euryale seed are added, and then water with volume of 8 times is added for soaking for 0.5h and extraction is carried out for 10 min. Detection shows that the cream yield is 31.16% and the content of total flavonoids is 1.203 mg/ml.
Test example 1 acute toxicity test
Test materials:
animals: 70 mice are taken, each mouse weighs 18 g-22 g, and the male and female halves are respectively.
Medicine preparation: the invention relates to a pulverized traditional Chinese medicine composition powder.
Mice were randomly divided into 7 groups of 10 mice each, each half male and female. The cream formulation was adjusted to be pasty (maximum concentration is standard), each mouse was perfused with a maximum amount of the drug administered twice a day for 14 consecutive days and the animals were observed. In the observation period of 14 days, the mice were normal in appetite, activity, hair color, mental status, etc., developed normally, and had no death. The medicine of the present invention has high safety, no obvious side effect and very low acute toxicity.
Test example 2 drug efficacy test
The influence of the traditional Chinese medicine composition on the mouse cough model is as follows:
the mice were randomly divided into a model control group, and a blank control group, an example group and a comparative example group were set. The administration is carried out by intragastric administration for 5 days every day, and after the last administration for 2h, the mice are placed in a bell jar, and concentrated ammonia water is uniformly sprayed for 5s to stimulate the mice to cause cough, and the cough frequency of the mice within 10min is observed.

The experimental result shows that compared with the original old formula control group, the components of the formula of the ointment prepared from the raw materials have an inhibiting effect on the cough caused by strong ammonia water, but the formula of the traditional Chinese medicine composition in the examples 1 to 4 has a more obvious inhibiting effect on the cough caused by strong ammonia water, the effects of the examples 1 to 4 are better than those of the comparative examples 1 to 2, wherein the effect of the example 3 is better than those of the examples 1 and 2, and the effect of the example 4 is better than that of the example 3.
Test example 3 clinical test
General materials: 140 adult patients with spleen-lung qi deficiency diagnosed by traditional Chinese medicine diagnosis in recent two years are selected as study objects, 80 young people of 18-40 years old are selected, 40 male people and 40 female people are selected, 60 middle-aged people of 41-60 years old are selected, and 30 male people and 30 female people are selected.

As can be seen from the above table, the cream formula components formulated from the raw materials of the present invention have better effects on the cure rate and the significant therapeutic effect compared to the original old formula control group and comparative examples 1, 2, wherein the effects of examples 1-4 are better than those of comparative examples 1-2, wherein the effect of example 3 is better than those of examples 1 and 2, and the effect of example 4 is better than that of example 3.
