The invention discloses an environment-friendly and efficient tilapia mossambica white feces prevention and treatment composition, a preparation method and application thereof, wherein the composition comprises bighead atractylodes rhizome, poria cocos, astragalus membranaceus, Chinese yam and liquorice, the preparation method is characterized in that a complex enzyme enzymolysis method and a water extraction and alcohol precipitation method are combined, starch polysaccharide and glycoprotein in cell membranes and cytoplasm are subjected to enzymolysis through complex enzyme, medicinal polysaccharide is released better, the medicinal polysaccharide is extracted, concentrated and precipitated and separated by the water extraction and alcohol precipitation method, and finally the extract which is high in titer, convenient to use and convenient to store is obtained. Background
Tilapia mossambica has the advantages of fast growth, poor feeding quality, low feed ratio, more meat yield and high yield, and is an important economic cultured fish in China. However, with the development of high-density intensive culture models, various diseases are frequently occurring. Tilapia mossambica often has the phenomenon of 'feces dragging' by anus of a fish body due to large food intake in spring and summer alternation and high-temperature seasons, and a large amount of white flocculent feces float around a fish pond feeding machine and at a downwind port, so that the problems of digestive tract occurrence, intestinal inflammation and intestinal mucosa falling off can be judged. The intestinal tract is not only an important digestion and absorption organ of the fish body, but also the largest immune and metabolic organ of the fish body, and is also the largest contact surface area organ with the external environment in the organism and is the initial organ of the organism injury, so the protection of the intestinal tract health plays an important role in protecting the health of the fish body. However, for common freshwater fish at present, terminal farmers do not pay attention to daily care of intestinal health of fish bodies, and often look for emergency specific drugs such as antibiotics after dead fish and outbreak fish are found, which causes environmental problems such as increase of drug-resistant strains, water pollution and drug residues and food safety problems. Research shows that the intestinal flora plays important physiological roles in nutrition absorption, immune response and growth and development of animals, so that intestinal injury can be repaired by increasing the number of beneficial microorganisms in the intestinal tract, maintaining the balance of the intestinal microflora. Related technology discloses a feed additive for preventing tilapia enteritis, which improves intestinal mucosa thickness, inhibits propagation of intestinal harmful microorganisms and promotes discharge of harmful substances by adding citric acid, bile acid, zeolite powder, streptococcus lactis, lactobacillus and bacillus licheniformis. The technology plays a role by mixing the additive into commercial feed, mainly because the activity of the live bacteria has the technical difficulty of activity retention, and the actual production application is limited to a certain extent.
Aiming at the problem of white feces of tilapia, the tilapia can be prevented and treated by adjusting intestinal flora. But probiotics require low-temperature storage conditions, are not suitable for being directly added into feed production, increase the labor force of farmers due to material mixing, and reduce the activity retention rate reaching intestinal tracts after being directly fed.
Disclosure of Invention
The present invention is directed to solving at least one of the problems of the prior art described above. The invention provides a composition for preventing and treating tilapia white feces with environmental protection and high efficiency, a preparation method and an application thereof, mainly by extracting plant polysaccharide from a traditional Chinese medicine composition as prebiotics, the composition is easy to store at normal temperature, has good stability, can be used for feed processing, directly reaches intestinal tracts, and is used as a carbon source of intestinal flora to promote the proliferation of intestinal beneficial bacteria and inhibit harmful bacteria, thereby improving intestinal microecological flora, maintaining balanced flora structure and being beneficial to preventing and treating tilapia white feces and enteritis.
According to one aspect of the invention, the invention provides an environment-friendly and efficient composition for preventing and treating white feces of tilapia, which comprises the following components: atractylodis rhizoma, Poria, radix astragali, rhizoma Dioscoreae, and Glycyrrhrizae radix; the composition is prepared by the following method:
s1: pulverizing Atractylodis rhizoma, Poria, radix astragali, rhizoma Dioscoreae and Glycyrrhrizae radix, sieving, soaking in water for a certain time to obtain composition slurry; adding warm water into the complex enzyme for activation to obtain a complex enzyme solution;
s2: heating the composition slurry, adding a citric acid-sodium citrate buffer system to adjust the pH value to 4.5-5.0, and then adding the complex enzyme solution for enzymolysis;
s3: after the enzymolysis is finished, heating to inactivate the enzyme, cooling, supplementing water, and performing water bath extraction to obtain an extracted solution;
s4: carrying out solid-liquid separation on the extracted liquid, concentrating the obtained liquid to obtain a concentrated solution, adding absolute ethyl alcohol into the concentrated solution, standing to obtain a precipitate, and drying to obtain the composition;
wherein the complex enzyme consists of cellulase, pectinase, intermediate temperature alpha-amylase and neutral protease.
In some embodiments of the invention, the components are in parts by weight as follows: 22.5-25 parts of bighead atractylodes rhizome, 22.5-25 parts of poria cocos, 18-22 parts of astragalus membranaceus, 15-20 parts of Chinese yam and 10-15 parts of liquorice.

In some embodiments of the invention, in step S1, the slurry-to-liquid ratio of the composition slurry is 1: (10-15); the soaking time is 45-60 min.
In some embodiments of the present invention, in step S1, the screening mesh number is 100-150 meshes.
In some embodiments of the invention, in step S1, the amount of the complex enzyme is 0.3-0.5% of the total mass of the atractylodes macrocephala koidz, the poria cocos wolf, the astragalus membranaceus, the Chinese yam and the licorice.
In some embodiments of the invention, in step S1, the amount of the warm water is 5-10 times of the mass of the complex enzyme; the temperature of the warm water is 40-45 ℃, and the activation time is 10-15 min.

The invention also provides tilapia feed, which consists of basic feed and the composition, wherein the addition amount of the composition is 0.3-0.7% of the mass of the basic feed. Further, the basic feed comprises soybean meal, rapeseed meal, wheat, sunflower kernel meal, corn DDGS, flour, rice bran meal, barley, fish meal, soybean oil, monocalcium phosphate, tilapia premixed feed, salt and choline chloride.
According to a preferred embodiment of the present invention, at least the following advantages are provided:
(1) the components of the composition of the invention, including the white atractylodes rhizome, the tuckahoe, the astragalus root, the yam and the liquorice, are added and reduced on the basis of the traditional Chinese medicine 'four monarch decoction' (from Song Dynasty 'Taiping Huimin He Ji Ju Fang'), wherein the white atractylodes rhizome is sweet and bitter and warm in property and has the effects of tonifying qi, invigorating spleen and promoting diuresis; poria cocos is sweet and mild, and has the effects of excreting dampness, invigorating spleen and calming heart; the two herbs have the common efficacy of strengthening spleen and eliminating dampness, because spleen prefers dryness and dislikes dampness, white atractylodes rhizome can eliminate dampness, and poria cocos can promote diuresis with bland flavor, and when the two herbs are used together, one dryness can permeate into the other to eliminate water dampness and spleen qi is healthy. Radix astragali is sweet and warm in nature, has effects of invigorating middle warmer and qi, and tonifying spleen yang, and can be used for treating diarrhea due to spleen deficiency; the yam is slightly astringent, can tonify spleen and stomach, nourish yin and strengthen kidney, and can be used for treating diarrhea and loose stool; the yam and the astragalus root are combined to play the roles of inducing astringency and raising the body resistance, and the two medicines are combined to nourish yin and raise yang, so that the yam and the astragalus root have the mutual enhancing effect on treating the diarrhea. The liquorice is sweet in taste and neutral in nature, has the functions of tonifying spleen and qi, eliminating phlegm and stopping cough, relieving spasm and pain, clearing heat and detoxicating, and harmonizing the medicines, and enters heart, lung, spleen and stomach channels. The spleen is one of the main immune organs of an animal body, has an important function on resisting the pathogenic bacteria inflammation, and the bighead atractylodes rhizome and the poria cocos have the effects of strengthening the spleen, tonifying qi and excreting dampness and are monarch drugs; the astragalus root has the effects of tonifying spleen and middle energizer, and the Chinese yam has the effects of excreting dampness and strengthening spleen and inhibiting diarrhea, and the two medicines are used as ministerial medicines. Licorice root, radix Glycyrrhizae is used as a guiding drug to regulate the four drugs. The monarch drug and the ministerial drug are applied in proportion, so that the conductant drug makes the monarch drug and the ministerial drug combine to eliminate pathogenic factors, the prescription of the composition has good regulating effect on the aspects of regulating gastrointestinal tract movement, improving digestion and absorption of nutrient substances and the like, and can improve spleen-stomach qi deficiency and gastrointestinal dysfunction to different degrees.
(2) Most of the traditional Chinese medicines are orally taken by water decoction, in recent years, some terminal farmers also take certain curative effects by decocting the traditional Chinese medicines by the method for health care and disease treatment of fishes, and actually the soluble polysaccharide is an important component of the traditional Chinese medicine water decoction and an important material basis for the traditional Chinese medicines to exert the curative effects. The preparation method combines a complex enzyme enzymolysis method and a water extraction and alcohol precipitation method, firstly breaks cell walls by cellulase and pectinase, and then carries out enzymolysis on cell membranes and amylopolysaccharide and glycoprotein in cytoplasm by medium temperature alpha-amylase and neutral protease so as to better release medicinal polysaccharide, and then carries out extraction, concentration and precipitation separation on the medicinal polysaccharide by the water extraction and alcohol precipitation method, so that the extract with high titer, convenient use and convenient storage is finally obtained.
(3) After the components of the invention are extracted by the preparation method, the main components are plant polysaccharide substances, animals lack polysaccharide hydrolase, the polysaccharide except starch can not be directly digested and absorbed, the intestinal flora plays an important bridge role, and the polysaccharide is used as a prebiotic of the intestinal flora, promotes the proliferation of intestinal beneficial bacteria and inhibits the proliferation of harmful bacteria. Example 1
The embodiment prepares an environment-friendly and efficient composition for preventing and treating white feces of tilapia, and each 100 parts of the composition comprises the following components in parts by weight: 22.5 parts of bighead atractylodes rhizome, 22.5 parts of poria cocos, 20 parts of astragalus membranaceus, 20 parts of Chinese yam and 15 parts of liquorice, and the preparation process comprises the following steps:
respectively crushing the raw materials, sieving the crushed raw materials with a 100-mesh sieve, weighing the composition according to the component proportion, uniformly stirring, and mixing the materials according to a material-liquid ratio of 1: 12 adding 12 times of distilled water, and soaking for 30min to obtain composition slurry; meanwhile, weighing the complex enzyme according to 0.4% of the dry weight of the composition, and adding 5 times of warm water at 40 ℃ for activation for 10-15min to obtain a complex enzyme solution; adjusting the temperature of the slurry of the composition to 45-50 ℃ and the pH value to 4.5-5.0 by using a water bath mode and a citric acid-sodium citrate buffer system, keeping the reaction condition, slowly adding a complex enzyme solution, slightly stirring, and performing enzymolysis at constant temperature for about 120min (according to the weight of the composition); and after enzymolysis is finished, heating to 85 ℃, inactivating enzyme in water bath for 15min, cooling to room temperature, and supplementing distilled water to ensure that the solid-to-liquid ratio of the slurry is 1: 40, stirring uniformly, extracting in water bath at 80 ℃ for 180min, filtering to obtain filtrate, vacuum concentrating to about 1/10, adding 4 times of absolute ethyl alcohol into the concentrated solution, standing for 24h, collecting precipitate, washing twice with absolute ethyl alcohol, acetone and diethyl ether in sequence, and vacuum drying at 60 ℃ until the water content is below 5% for storage to obtain the composition.
The complex enzyme is prepared from cellulase, pectinase, intermediate temperature alpha-amylase and neutral protease according to the proportion of 2: 1: 0.5: 0.5 proportion.
Test example 1
(1) Experimental fish and breeding management
The experimental tilapia was from Guangdong Shuncye aquatic products Co., Ltd, and was trained for 2 weeks in a temporary rearing pond using a basal diet before formal start of the experiment. Selecting strong and uniform tilapia (average weight 170.53 +/-2.31 g), randomly distributing to 20 circular breeding barrels (diameter 180cm, height 100cm, water containing volume 1800L), and repeating for 4 fodder groups with 30 tails in each barrel. During the experiment, 8: 30 and 16: 30 feeding once after being saturated, wherein the feeding amount of each time is based on that the experimental fish is eaten within 20 min. Adopts an indoor circulating water culture system, the water temperature range is 28.3-32.6 ℃, and the dissolved oxygen is more than 6.8mg.L-1Ammonia nitrogen is less than 0.5mg.L-1The illumination period is natural illumination. The culture period is 8 weeks.
(2) Experiment feed
The experimental soybean meal, the seed added rapeseed meal, the wheat, the sunflower kernel meal, the corn DDGS, the flour, the rice bran meal, the barley, the fish meal, the soybean oil, the monocalcium phosphate, the tilapia premixed feed, the salt and the choline chloride are all from Wanghai feed industry Co., Ltd in the cis-Deland of Foshan city; atractylodis rhizoma, Poria, radix astragali, rhizoma Dioscoreae, and Glycyrrhrizae radix are obtained from Guangzhou city medicinal materials. Cellulase, pectinase, medium temperature alpha-amylase, neutral protease were purchased from enzyme preparations. The experimental feed formulation and chemical composition are shown in table 1.

Wherein the addition content of the composition is 0 (control group), 0.1%, 0.3%, 0.5%, 0.7%, and the composition is used as 1 control group and 4 experimental groups for culture experiment.
(3) Sample collection and processing
After the culture experiment is finished, randomly taking 5 fish tails from each barrel, narcotizing the fish tails by MS-222, respectively weighing the fish tails, taking blood from cardiac venous sinus by using a scalp needle, placing the blood in a 2ml sterile centrifuge tube, standing the blood at 4 ℃ for coagulation, centrifuging the blood for 10min at 3000r/min, taking supernatant, storing the supernatant at-20 ℃ and determining the activity of the serological enzyme (using Nanjing to build a kit). Blood-bled fish were taken, spleens were separated and weighed and recorded to calculate spleen index for each experimental group.
Taking 3 fish tails from each barrel, quickly dissecting and taking out fish intestines beside an alcohol lamp on a clean workbench, placing on a sterilized clean tray, quickly cutting open the middle intestines, scraping 0.5g of intestinal contents, placing in a sterilized 10mL centrifuge tube, adding 4.5mL of sterile water, shaking and mixing uniformly to obtain the fish sausage with the concentration of 10-1Sequentially diluting the sample with sterile water to 10 times-8And (3) sucking 100uL of a sample with a proper dilution by using a pipette gun, coating the sample on a selective culture medium of escherichia coli and bifidobacterium, performing parallel culture at 37 ℃ for 24h, and counting viable bacteria (the viable bacteria number Ig CFU/g per gram of fish intestine specimen) after the culture is finished.
(4) Data processing
The data obtained were subjected to independent sample T-test with SPSS 19.0 for windows and compared for differences in significance of the mean, with a significance level P < 0.05.

The results in tables 2-4 show that the immune organ (spleen index) and the serum nonspecific immune enzyme activities of tilapia cultured by the feed of the experimental group added with the composition are higher than those of a control group without the composition, and the highest activity is obvious when the addition amount is 0.3-0.7%. The intestinal microbial flora of tilapia is also influenced, the content of beneficial bacteria (bifidobacteria) in intestinal tracts and the content of harmful bacteria (escherichia coli) in experimental fishes added with the composition are increased, and the ratio of the beneficial bacteria to the harmful bacteria is improved. Therefore, after the composition is prepared, the immune function of tilapia can be enhanced, the environment of intestinal flora of fish bodies can be improved, and the intestinal health can be promoted.
Test example 2
After the preliminary evaluation of test example 1, the composition was added to the commercial feed formulation in an amount of 0.5%, and a large pond test material was produced, using a normal commercial feed without the composition added to the same formulation as a control group. Respectively inviting 10 farmers to simultaneously try out and compare in Heizhou and Caragana, namely respectively selecting 5 ponds to feed test materials and 5 ponds to feed normal commercial feeds (a control group), feeding the fishes from the middle and the last ten days of 6 months to 10 months, feeding the fishes from about 20 tails to 1 jin, recording the number of ponds in which the streptococcus occurs in the high-temperature period of 7 and 8 months as shown in Table 5, recording the severity degree of the streptococcus occurring in the whole breeding period as shown in Table 6, and finally outputting the fishes and producing the fish in bags as shown in Table 5.

Note: the severity of white feces in the whole culture process is automatically evaluated by farmers, the severity is represented by the number of particles, the severity is the most 5 particles, and the normal or negligible condition is considered as no.
The results of the large pond trials in tables 5 and 6 show that the incidence of streptococcus in two areas in the high temperature period is lower than that of the control group, the incidence degree of white feces is lower than that of the control group, the tilapia mossambica white feces can be effectively prevented from occurring, and the feed effect expression is favorably influenced finally when the feed group added with 0.5% of the composition is added. The incidence of streptococcus suis and white feces is high in temperature in this year, and the farmer of the test material group shows that the fish-making eating rate is better than that of the surrounding farmer, the incidence rate is greatly reduced than that of the farmer in the past year, the health care amount of extra medicines is greatly reduced, and medicines for treating diseases are basically not needed.
The embodiments of the present invention have been described in detail, but the present invention is not limited to the embodiments, and various changes can be made without departing from the gist of the present invention within the knowledge of those skilled in the art. Claims (10)
1. The environment-friendly and efficient composition for preventing and treating white feces of tilapia is characterized by comprising the following components: atractylodis rhizoma, Poria, radix astragali, rhizoma Dioscoreae, and Glycyrrhrizae radix; the composition is prepared by the following method:
s1: pulverizing Atractylodis rhizoma, Poria, radix astragali, rhizoma Dioscoreae and Glycyrrhrizae radix, sieving, soaking in water for a certain time to obtain composition slurry; adding warm water into the complex enzyme for activation to obtain a complex enzyme solution;
s2: heating the composition slurry, adding a citric acid-sodium citrate buffer system to adjust the pH value to 4.5-5.0, and then adding the complex enzyme solution for enzymolysis;
s3: after the enzymolysis is finished, heating to inactivate the enzyme, cooling, supplementing water, and then carrying out water bath extraction to obtain an extracted liquid;
s4: carrying out solid-liquid separation on the extracted liquid, concentrating the obtained liquid to obtain a concentrated solution, adding absolute ethyl alcohol into the concentrated solution, standing to obtain a precipitate, and drying to obtain the composition;
wherein the complex enzyme consists of cellulase, pectinase, medium temperature alpha-amylase and neutral protease.
2. The composition of claim 1, wherein the components are in parts by weight as follows: 22.5-25 parts of bighead atractylodes rhizome, 22.5-25 parts of poria cocos, 18-22 parts of astragalus membranaceus, 15-20 parts of Chinese yam and 10-15 parts of liquorice.
3. The composition of claim 1, wherein the components are in parts by weight as follows: 22.5 parts of bighead atractylodes rhizome, 22.5 parts of poria cocos, 20 parts of astragalus membranaceus, 20 parts of Chinese yam and 15 parts of liquorice.
4. The composition according to claim 1, wherein the mass ratio of the cellulase, pectinase, meso-alpha-amylase and neutral protease is (2-4): (1-2): (0.5-1): (0.5-1).
5. The composition as claimed in claim 1, wherein in step S1, the slurry ratio of the composition slurry is 1: (10-15); the soaking time is 45-60 min.
6. The composition as claimed in claim 1, wherein in step S1, the amount of the complex enzyme is 0.3-0.5% of the total mass of the rhizoma atractylodis macrocephalae, the poria cocos, the astragalus membranaceus, the Chinese yam and the licorice.
7. The composition as claimed in claim 1, wherein the heating temperature is 45-50 ℃ in step S2; the enzymolysis time is 120-180 min.
8. The composition as claimed in claim 1, wherein in step S3, the temperature of the water bath extraction is 80-85 ℃, and the time of the water bath extraction is 180-210 min.
9. The composition as claimed in claim 1, wherein the amount of the absolute ethanol used in step S4 is 3-5 times of the concentrated solution.
10. A tilapia feed, characterized in that it consists of a basal feed and a composition according to any one of claims 1 to 9, said composition being added in an amount of 0.3 to 0.7% by mass of said basal feed.
