Abstract
The invention belongs to the technical field of soybean food processing, and discloses a preparation method of soybean protein isolate with high solubility, which comprises the steps of preparing defatted soybean meal, and preparing a soybean protein isolate solution from the defatted soybean meal by an alkali extraction and acid precipitation method; preheating the soybean protein isolate solution, carrying out ultrasonic treatment, and carrying out spray drying, separation and sieving to obtain a soybean protein isolate raw material; adding deionized water and corresponding enzyme into the soybean protein isolate raw material, and inactivating the enzyme by using a boiling water bath to obtain an enzymatic hydrolysate; and (3) performing microwave radiation on the enzymolysis liquid, freeze-drying, and adding phospholipid and sucrose to obtain the soybean protein isolate with high solubility. The preparation method of the soybean protein isolate with high solubility has the advantages of simple operation, easy control, good safety, wide source of used raw materials, low cost and suitability for large-scale industrial production and application. The soybean protein isolate prepared by the method has the advantages of high dissolving speed, high dissolving tendency, stable property, good effect and the like.
Description
Technical Field
The invention belongs to the technical field of soybean food processing, and particularly relates to a preparation method of a soybean protein isolate with high solubility.
Background
At present, the soybean is rich in protein, high in essential amino acid content and balanced in species, and can provide eight kinds of essential amino acids, multiple vitamins, minerals and the like which are required by a human body. The soybean protein isolate is a main protein component in soybeans, mainly consists of 7S and 11S globulin, is high-quality plant protein, is one of few nutritional products capable of replacing animal protein, and has multiple functions of good dispersibility, emulsifying property, foaming property, water retention property, oil retention property, viscoelasticity and the like. Therefore, soy protein isolate is widely used in food production. Practical studies show that the solubility, which is one of the main functional properties of the isolated soy protein, is a prerequisite and basis for the physiological functional properties and the applicability, the application range, the extraction and processing process conditions and the like of the isolated soy protein. However, the traditional method for preparing the isolated soy protein has poor effect, complex reaction, difficult control, poor safety and the like. Therefore, it is highly desirable to design a new method for preparing soybean protein isolate having high solubility.
Through the above analysis, the problems and defects of the prior art are as follows: the traditional method for preparing the soybean protein isolate has poor effect, and has the problems of complex reaction, difficult control, poor safety and the like.
Disclosure of Invention
Aiming at the problems in the prior art, the invention provides a preparation method of soybean protein isolate with high solubility.
The present invention is achieved by a method for preparing a soybean protein isolate having high solubility, comprising the steps of:
step one, preparing a soy protein isolate solution: after the defatted soybean flour is prepared, the defatted soybean flour is subjected to an alkali extraction and acid precipitation method to prepare a soybean protein isolate solution;
step two, pretreatment of the soy protein isolate solution is carried out: preheating the soybean protein isolate solution, carrying out ultrasonic treatment, and carrying out spray drying, separation and sieving to obtain a soybean protein isolate raw material;
step three, carrying out enzyme method modification treatment on the soybean protein isolate solution: adding deionized water and corresponding enzyme into the soybean protein isolate raw material, and inactivating the enzyme by using a boiling water bath to obtain an enzymatic hydrolysate;
step four, preparing the modified isolated soy protein: and (3) performing microwave radiation modification on the enzymolysis solution, freeze-drying, adding phospholipid and sucrose, and uniformly mixing to obtain the soybean protein isolate with high solubility.
Further, the preparation of the soy protein isolate solution in the first step comprises:
(1) crushing the defatted soybean meal, sieving the crushed defatted soybean meal with a sieve of 80-100 meshes, adding 8-10 times of deionized water by weight, performing ultrasonic treatment, adding chitosan to perform nucleic acid adsorption, and drying to obtain defatted soybean meal;
(2) adding the defatted soybean powder into deionized water, stirring and dispersing, adding 3mol/L sodium hydroxide solution to adjust the pH value to 8.2, stirring and leaching at room temperature for 2-4 h, centrifuging, and taking supernatant;
(3) adding 2mol/L hydrochloric acid solution into the supernatant, centrifuging, taking the lower layer of soybean protein curd, adding deionized water, and adjusting the pH value to 7.5 by using 3mol/L sodium hydroxide solution to obtain the soybean protein isolate solution.
Further, the power of ultrasonic treatment is 500-700W, the temperature of ultrasonic treatment is 40-45 ℃, and the time of ultrasonic treatment is 30-40 min.
Further, the pretreatment of the soy protein isolate solution in the second step comprises:
(1) preheating the soy protein isolate solution for 15-25 min at 55-65 ℃;
(2) putting the soy protein isolate solution into an ultrasonic cleaner for ultrasonic treatment for 60-80 min;
(3) and (3) carrying out spray drying, separation and sieving on the soybean protein isolate solution subjected to ultrasonic treatment to obtain the soybean protein isolate raw material.
Further, the temperature of ultrasonic treatment is 45-55 ℃, and the power of ultrasonic treatment is 100-120W.
Further, the enzymatic modification treatment of the soy protein isolate solution in the third step comprises:
(1) adding deionized water into the soybean protein isolate raw material to prepare 5-35% of protein isolate liquid;
(2) adding flavor protease, cellulase and papain into the soybean protein isolate liquid, and carrying out enzymatic hydrolysis treatment for 2-4 h to obtain enzymatic hydrolysate of the soybean protein isolate liquid;
(3) and (3) inactivating enzyme of the enzyme hydrolysis liquid of the soybean protein isolate liquid after enzymolysis for 20-30 min by using a boiling water bath, and quickly cooling to room temperature to obtain the enzyme hydrolysis liquid.
Further, before enzymolysis of the soybean protein isolate liquid, heating the soybean protein isolate liquid to 35-37 ℃.
Further, the preparation of the modified isolated soy protein in the fourth step comprises:
(1) placing the soybean separation proteolytic liquid under 550-560W power for microwave radiation for 8-15 min to obtain a modified soybean separation protein solution;
(2) freeze-drying the obtained modified isolated soy protein solution to obtain a modified isolated soy protein primary product;
(3) and crushing the primary soybean protein isolate product, and uniformly mixing the crushed primary soybean protein isolate product with phospholipid and cane sugar to obtain the soybean protein isolate with high solubility.
It is another object of the present invention to provide a soybean protein isolate having high solubility prepared by the method for preparing a soybean protein isolate having high solubility as described above.
Another object of the present invention is to provide a use of the soy protein isolate in the preparation of a soy protein isolate product.
By combining all the technical schemes, the invention has the advantages and positive effects that: the preparation method of the soybean protein isolate with high solubility provided by the invention has the advantages of simple operation, easy control, good safety, wide source of used raw materials, low cost and suitability for large-scale industrial production and application. The soybean protein isolate prepared by the preparation method of the soybean protein isolate with high solubility has the advantages of high dissolution speed, easy dissolution, stable property, good effect and the like, and can meet the processing requirements of the food industry.

In order to make the objects, technical solutions and advantages of the present invention more apparent, the present invention is further described in detail with reference to the following embodiments. It should be understood that the specific embodiments described herein are merely illustrative of the invention and are not intended to limit the invention.
In view of the problems of the prior art, the present invention provides a method for preparing soybean protein isolate having high solubility, which is described in detail below with reference to the accompanying drawings.
As shown in fig. 1, the preparation method of soybean protein isolate with high solubility provided by the embodiment of the present invention comprises the following steps:
s101, preparing a soy protein isolate solution: after the defatted soybean flour is prepared, the defatted soybean flour is subjected to an alkali extraction and acid precipitation method to prepare a soybean protein isolate solution;
s102, pretreatment of the isolated soy protein solution: preheating the soybean protein isolate solution, carrying out ultrasonic treatment, and carrying out spray drying, separation and sieving to obtain a soybean protein isolate raw material;
s103, carrying out enzymatic modification treatment on the soybean protein isolate solution: adding deionized water and corresponding enzyme into the soybean protein isolate raw material, and inactivating the enzyme by using a boiling water bath to obtain an enzymatic hydrolysate;
s104, preparing the modified soybean protein isolate: and (3) performing microwave radiation modification on the enzymolysis solution, freeze-drying, adding phospholipid and sucrose, and uniformly mixing to obtain the soybean protein isolate with high solubility.
As shown in fig. 2, the preparation of the soy protein isolate solution in step S101 according to the embodiment of the present invention includes:
s201, after the defatted soybean meal is crushed and sieved by a sieve with 80-100 meshes, adding 8-10 times of deionized water by weight, carrying out ultrasonic treatment, adding chitosan for adsorbing nucleic acid, and drying to obtain defatted soybean meal;
s202, adding the defatted soybean powder into deionized water, stirring and dispersing, adding 3mol/L sodium hydroxide solution to adjust the pH value to 8.2, stirring and leaching at room temperature for 2-4 hours, centrifuging, and taking supernatant;
s203, adding 2mol/L hydrochloric acid solution into the supernatant, centrifuging, taking the lower layer of soybean protein curd, adding deionized water, and adjusting the pH value to 7.5 by using 3mol/L sodium hydroxide solution to obtain the soybean protein isolate solution.
Further, the power of ultrasonic treatment is 500-700W, the temperature of ultrasonic treatment is 40-45 ℃, and the time of ultrasonic treatment is 30-40 min.
As shown in fig. 3, the pretreatment of the soy protein isolate solution in step S102 according to the embodiment of the present invention includes:
s301, preheating the soy protein isolate solution for 15-25 min at the temperature of 55-65 ℃;
s302, placing the soy protein isolate solution in an ultrasonic cleaner for ultrasonic treatment for 60-80 min;
s303, carrying out spray drying, separation and sieving on the soybean protein isolate solution after ultrasonic treatment to obtain a soybean protein isolate raw material.
Further, the temperature of ultrasonic treatment is 45-55 ℃, and the power of ultrasonic treatment is 100-120W.
As shown in fig. 4, the enzymatic modification treatment of the soy protein isolate solution in step S103 according to the embodiment of the present invention includes:
s401, adding deionized water into a soybean protein isolate raw material to prepare 5-35% of protein isolate liquid;
s402, adding flavor protease, cellulase and papain into the soybean protein isolate liquid, and carrying out enzymatic hydrolysis treatment for 2-4 h to obtain enzymatic hydrolysate of the soybean protein isolate liquid;
and S403, inactivating enzyme of the enzyme hydrolysate of the soybean protein isolate liquid after enzymolysis for 20-30 min by using a boiling water bath, and rapidly cooling to room temperature to obtain the enzyme hydrolysate.
Further, before the enzymolysis of the soybean protein isolate liquid, the soybean protein isolate liquid is heated to 35-37 ℃.
As shown in fig. 5, the preparation of the modified soy protein isolate in step S104 according to the embodiment of the present invention includes:
s501, placing the soybean separation proteolytic liquid under 550-560W power for microwave radiation for 8-15 min to obtain a modified soybean separation protein solution;
s502, freeze-drying the obtained modified soybean protein isolate solution to obtain a modified soybean protein isolate primary product;
s503, crushing the primary soybean protein isolate product, and uniformly mixing the crushed primary soybean protein isolate product with phospholipid and sucrose to obtain the soybean protein isolate with high solubility.

1. A method for preparing soybean protein isolate with high solubility, which is characterized by comprising the following steps:
step one, preparing a soy protein isolate solution: after the defatted soybean flour is prepared, the defatted soybean flour is subjected to an alkali extraction and acid precipitation method to prepare a soybean protein isolate solution;
step two, pretreatment of the soy protein isolate solution is carried out: preheating the soybean protein isolate solution, carrying out ultrasonic treatment, and carrying out spray drying, separation and sieving to obtain a soybean protein isolate raw material;
step three, carrying out enzyme method modification treatment on the soybean protein isolate solution: adding deionized water and corresponding enzyme into the soybean protein isolate raw material, and inactivating the enzyme by using a boiling water bath to obtain an enzymatic hydrolysate;
step four, preparing the modified isolated soy protein: and (3) performing microwave radiation modification on the enzymolysis solution, freeze-drying, adding phospholipid and sucrose, and uniformly mixing to obtain the soybean protein isolate with high solubility.
2. The method for preparing a soy protein isolate having high solubility as set forth in claim 1, wherein the preparation of the soy protein isolate solution in the first step comprises:
(1) crushing the defatted soybean meal, sieving the crushed defatted soybean meal with a sieve of 80-100 meshes, adding 8-10 times of deionized water by weight, performing ultrasonic treatment, adding chitosan to perform nucleic acid adsorption, and drying to obtain defatted soybean meal;
(2) adding the defatted soybean powder into deionized water, stirring and dispersing, adding 3mol/L sodium hydroxide solution to adjust the pH value to 8.2, stirring and leaching at room temperature for 2-4 h, centrifuging, and taking supernatant;
(3) adding 2mol/L hydrochloric acid solution into the supernatant, centrifuging, taking the lower layer of soybean protein curd, adding deionized water, and adjusting the pH value to 7.5 by using 3mol/L sodium hydroxide solution to obtain the soybean protein isolate solution.
3. The method for preparing a soybean protein isolate with high solubility according to claim 2, wherein the power of the ultrasonic treatment is 500 to 700W, the temperature of the ultrasonic treatment is 40 to 45 ℃, and the time of the ultrasonic treatment is 30 to 40 min.
4. The method for preparing soybean protein isolate having high solubility according to claim 1, wherein the pretreatment of the soybean protein isolate solution in the second step comprises:
(1) preheating the soy protein isolate solution for 15-25 min at 55-65 ℃;
(2) putting the soy protein isolate solution into an ultrasonic cleaner for ultrasonic treatment for 60-80 min;
(3) and (3) carrying out spray drying, separation and sieving on the soybean protein isolate solution after ultrasonic treatment to obtain the soybean protein isolate raw material.
