Based on the diagnosis standard of kidney yang deficiency in Chinese diagnostics, after the modeling treatment, the rats have reduced food intake and water intake, reduced activity, slow response, reduced body temperature, cold contraction of limbs, reduced physical quality, and rare hair loss, etc., which prompts the success of modeling, and the rats are divided into four groups according to a random digital table method after the success of modeling: a model group, a traditional Chinese medicine control group, a yang-warming and qi-tonifying paste formula group 2 and a yang-warming and qi-tonifying paste formula group 1.
1.4 modes of administration and observations and statistics
After successful modeling, rats in each group were weighed and different interventions were performed on each group starting on day 2.
The normal control group was perfused with 2mL of physiological saline daily for 28 consecutive days, anesthetized in the morning on day 29, weighed and sacrificed.
Model groups, 2mL of saline was gavaged daily for 28 consecutive days, rats were anesthetized in the morning on day 29 and weighed for sacrifice.
A traditional Chinese medicine control group, which is to administer corresponding liquid medicine for intragastric administration according to the dose of 10g crude drugs/(kg. d) -1, wherein the crude drugs are raw materials except sugar and yellow wine; the gavage was continued for 28 days, and the rats were anesthetized in the morning on day 29 and weighed and sacrificed.
The invention relates to a yang-warming and qi-tonifying paste formula 2, which is prepared by taking 10g of crude drugs/(kg.d) -1 according to the dose of (example 2) and then administering the corresponding liquid medicine for intragastric administration, wherein the crude drugs are raw materials except sugar and yellow wine; the gavage was continued for 28 days, and the rats were anesthetized in the morning on day 29 and weighed and sacrificed.
The invention relates to a yang-warming and qi-tonifying paste formula set 1, which is characterized in that 10g of crude drugs/(kg.d) -1 are given to corresponding liquid medicines according to the dose of (example 1) and the crude drugs are filled into stomach, wherein the crude drugs are raw materials except sugar and yellow wine; the gavage was continued for 28 days, and the rats were anesthetized in the morning on day 29 and weighed and sacrificed.
Observation indexes are as follows:
1, measuring the contents of T, ACTH and CORT in serum by an ELISA method, adopting a heart blood taking method, taking blood, placing in a centrifuge tube, controlling the temperature to be 4 ℃, standing for 2h, centrifuging for 10min (rotating speed is 3000r/min), separating serum, and placing in a refrigerator at-20 ℃ for detection. Serum T, ACTH, CORT levels were measured by enzyme-linked immunosorbent assay (ELISA) and performed exactly according to the procedures of the ELISA kit instructions.
2. And measuring the body mass of the rat and the weights of the hypothalamus, the pituitary and the adrenal gland, and recording the body mass when each group of rats successfully model and the body mass after the 29 th gastrodia elata is drunk. After sacrifice, the rats were dissected and the adrenal, pituitary, hypothalamic tissues were peeled off and weighed.

1.5 results:
1. comparison of T, ACTH and CORT contents in serum of rats of each group
Compared with a normal control group, the contents of serum T, ACTH and CORT of the kidney-yang deficiency model rat after model building are obviously reduced (P is less than 0.05), and the success of model building is prompted. Compared with a model group, the traditional Chinese medicine control group, the yang-warming and qi-tonifying paste formula group 2 and the yang-warming and qi-tonifying paste formula group 1 have improved serum T content (P is less than 0.05), and the yang-warming and qi-tonifying paste formula group 1 has the best effect; compared with the model group, the traditional Chinese medicine control group has no obvious effect on improving the ACTH level, and the Yang warming and Qi benefiting paste formula group 2 and the Yang warming and Qi benefiting paste formula group 1 can obviously improve the ACTH level (P is less than 0.05); the yang-warming and qi-tonifying paste formula 1 can improve the CORT level (P is less than 0.05) to a certain extent. Compared with the normal control group, the differences of the serum T, ACTH and CORT in the yang-warming and qi-tonifying paste formula group 2 and the yang-warming and qi-tonifying paste formula group 1 have statistical significance (P is less than 0.05). The results are shown in Table 1.

2. Compared with a normal control group, the weight increase of rats in the weight ratio of the body weight of rats to the weight of hypothalamus, pituitary gland and adrenal gland of each group is obviously slowed down; compared with the model group, the body mass changes of rats in the traditional Chinese medicine control group, the yang-warming and qi-tonifying paste formula group 2 and the yang-warming and qi-tonifying paste formula group 1 are increased to different degrees (P is less than 0.05), and the yang-warming and qi-tonifying paste formula group 1 is most obvious. Compared with a normal control group, the weight of the adrenal gland, the pituitary gland and the hypothalamus of the rat in the model group is obviously reduced (P is less than 0.05); compared with a model group, the medicine dry prognosis shows that the yang-warming and qi-tonifying paste formula group 1 shows obvious difference (P is less than 0.05). The results are shown in Table 2.
