1 materials and methods
1.1 animals
The SPF grade SD healthy rats are male, 50 in total, have the body mass of 180-220 g, and are provided by the animal experiment center of Shanghai medical university. Adaptive feeding for 1 week at room temperature of about 23 deg.C and relative humidity of 40-70%.
1.2 test drugs
The invention relates to a qi-tonifying and vitality-restoring ointment formula which comprises the following components in parts by weight: prepared as described in example 1. The preparation is administered by mixing with purified water.
The original qi-tonifying and vitality-restoring ointment formula comprises the following components: 150 g of white ginseng (decocted separately), 150 g of honey-fried astragalus root, 150 g of raw astragalus root, 150 g of codonopsis pilosula, 150 g of fried bighead atractylodes rhizome, 90 g of pericarpium citri reticulatae, 200 g of raw coix seed, 200 g of cooked coix seed, 150 g of whole angelica, 150 g of common yam rhizome, 200 g of white poria, 100 g of lotus seed pulp, 90 g of wide curcuma aromatica, 90 g of medlar, 90 g of vinegar-processed bupleurum, 150 g of Chinese taxillus twig, 90 g of raw liquorice, 100 g of longan pulp, 100 g of black sesame, 100 g of walnut pulp and 100 g of red date. The above medicines are decocted for concentration and dregs are removed, and the concentrated juice is added with 300 g of donkey-hide gelatin, 100 g of antler gelatin, 350 g of white honey and 350 g of yellow wine to be melted and collected into paste. 1.3 grouping and Molding
The 50 SD rats are randomly divided into 5 groups, including a normal group, a model group, a qi-tonifying and recovery paste formula group, an original qi-tonifying and recovery paste formula group and a ginsenoside group, wherein each group comprises 10 rats. The other 4 groups except the normal control group were molded by the complex stimulation method.
Specifically, a chronic binding method, a cold water swimming method and a random day and night inversion method are adopted for compound stimulation molding. Single day chronic restraint method: the restraint cylinder is manufactured by self, the size of the restraint cylinder is suitable for being filled into a rat but the head of the rat cannot rotate freely, the head of the rat is drilled into the restraint cylinder towards the ventilation opening, the rat does not have strong resistance, and the rat is then erected on a wood board and is restrained for 30min each time. A double-day cold water swimming method: a swimming pool with size of 200cm × 80cm × 80cm is prepared, water depth is set to 50cm, water temperature is 10 + -1 deg.C, and rats are placed into swimming for 8min every time, and the swimming pool is carried out every other day. Random day and night inversion method: the continuous fluorescent lamp irradiation is given at 18:00 nights to 6:00 next morning, and the sun is placed in a dark closed environment for 3 times. 2.2 rat tail suspension test results
