Abstract
一种新型的抗衰保健食品双歧杆菌制剂及其液-固结合发酵工艺是指:以一定配比的动植物蛋白粉(膏),碳水化合物,食用氨基酸,医用B族维生素,生物活性物质,蔗糖和食盐等原料,运用液-固结合生产工艺,对特选的益生菌双歧杆菌进行培养发酵及其产品。本发明除具有国内外液体工艺生产的益生菌食品的抗衰保健,提高机体免疫力特点外,其优点在于:设备投资少,操作简便,成本较低;产品所含益生菌总数在100亿/克以上,是国内外同类品的100倍以上,成分丰富,保健功能齐全,抗病,免疫和延缓衰老的效果明显。
Description
本发明属于新型的抗衰保健食品一双歧杆菌制剂及其液一同结合发酵工艺,属于应用微生物和保健食品科技领域。
近几年以来,在国内外医药界,衰老生物学界和微生物技术领域中,人体益生菌的研制工艺有较迅速发展。由于益生菌作为一种微生态制剂(Micro-ecological Preparation)是指提自健康婴儿肠道菌群的益生菌制品,食用后可调整人体肠道菌群的微生态平衡,并产生可抑制有害微生物繁殖的有机酸,从而减缓“大肠中毒”;它可保护体内蛋白质和各种酶系的生物活性,并刺激免疫活性产生;它可清除人体代谢过程中产生的自由基,可重现中老年人机体年轻时的表观微生物环境,起到延缓衰老,防治疾病的作用;此外,由于它含有多种氨基酸,维生素和人体必需的微量元素,因此可补充人体营养,增进健康。正因如此,国内外近年来研制生产的含不同益生菌的抗老保健食品颇受人们特别是中老年人的青睐。
但目前市场上常见的国内外生产的益生菌制品中存在以下不足:①营养成分局限性很大;②现有益生菌制剂的生产工艺均为液体发酵(深层发酵);生产工艺复杂设备昂贵投资大,产品价格高,不易普及推广;③现有同类产品均为液体制剂,益生菌易衰老死亡,因此产品中活菌比例较低,产品不易保存。
本发明的目的在于革新发酵工艺,降低生产成本,提高双歧杆菌细胞总数,延长双歧杆菌的存活时间,从而充分发挥其抗衰老保健的作用。
本发明核心内容是以持选的益生菌一双歧杆菌(Bifidoac-terium sp.)为菌种,运用液一同结合的发酵工艺进行培养发酵,并在发酵产物中适量加入赖氨酸,蛋氨酸,谷氨酸,生物素和B族维生素配制而成。
本发明具体实施步骤为:
1,菌种培养:以特选的双歧杆菌(Bifidoacterium sp.)为菌种,在液态为主的环境下,进行三级培养。
①斜面培养
斜面采用牛肉膏,蛋白胨,蔗糖,NaCl和水组成常规培养基,PH7.0~7.2。经灭菌接种后,在30℃下培养24小时。
②三角瓶培养
三角瓶培养采用麸皮加水煮30分钟过滤,滤液加入适量的蛋白胨,牛肉膏,蔗糖,食盐为培养基,PH7~7.2,分装入三角瓶,经灭菌,接种后在室温下(25~30℃)培养36小时左右,定时摇动(每小时一次),当细胞数达到130×10个/克时终止培养。
③扩大培养
扩大培养仍采用三角瓶培养所用培养液,经灭菌后放入已消毒的单缸洗衣机内,接入三角瓶培养的菌种液,室温下培养(室内用紫外灯和甲醛加高锰酸钾做灭菌处理),一小时开启洗衣机45分钟,停机15分钟,待细胞数达135×10个/克时,终止培养,备用。
2,同体发酵:
①发酵原料配制:发酵原料分液态培养料和同体培养料两种。
A,液态培养料:取打浆土豆和麸皮约占总量3~5%,加水煮25分钟,再加入蛋白胨(占总量0.5~0.8%),生肉膏(0.2~0.4%),蔗糖(3~7%)和食盐(0.02~0.04%)溶解而成,调PH7~7.2。
B,同态培养料:用精玉米粉(45~50%),精大米粉(12~15%),脱脂大豆粉(15~20%),全脂乳粉(25~30%)以及食用酵母膏(0.2~0.3%)混合而成。
②发酵:
同体培养料和液态培养料按1∶0.4的比例和料,高压灭菌一小时,冷却至40℃以下,备用。
将经三级培养的双歧杆菌,接种到经灭菌、冷却的发酵原料中,进行同体发酵。将接种后的发酵原料摊放在无菌洁净的带盖搪瓷盘中,平均每盘350克左右,在室温下发酵24~36小时,然后干燥粉碎,备用。
3,产品配制及成型:
将双歧杆菌发酵产物加入适量赖氨酸(0.02~0.2%),谷氨酸(0.04~0.2%),蛋氨酸(0.05~0.15%),生物素(0.0001%)和医用B族维生素(VB1,VB2,VB3,VB5,VB6,VB7等占0.002%)调质而成产品混合料。把产品混合料经粉碎后压制成糖衣片剂或装胶囊,再经包装成产品入库。
本产品的工艺流程为:
在液态为主环境下进行三级菌种培养,然后进行配制液态和同态培养料,再进行和料,经过接三级菌种后,进行同体发酵,发酵后真空干燥,再进行产品调质,经质检,粉碎,成型后包装入库。
本发明主要技术指标:去掉糖衣后,内为黄棕色粉末,或团粒,具有益生菌同态制剂的特殊气味,无异味,略甜,适口性好,无异物。
理化及微生物指标为:蛋白质≥19%;水分≤7%,灰分≤4%,砷≤0.2Mg/Kg,铅≤0.1Mg/Kg,益生菌总数(出厂时)≥100×10个/克;大肠菌群菌≤25个/克,霉菌总数≤90个/克,黄曲霉素符合规定,致病菌不得检出。
本发明优点:由于采用液一同体相结合的发酵工艺,而具有投资少,工艺简便,产品和成本价格低廉,便于推广应用等优点。产品有丰富的氨基酸,微生素和人体必需的微量元素,并有较强的生物活性。与液体发酵工艺生产的类似产品相比,其营养成分齐全,保质期长,抗病,免疫和延缓衰老的效果明显。
Claims (5)
1,一种双歧杆菌制剂是指:以一定配比的动植物蛋白粉(膏),碳水化合物,食用氨基酸,医用B族维生素,生物活性物质,蔗糖和食盐等为原料,运用液同结合生产工艺,对特选的益生菌双歧杆菌进行培养发酵并经调质而成产品。
2,按照权利要求1所述的双歧杆菌制剂其专门生产的液一固体相结合的发酵工艺,其特征是:对特选的双歧杆菌在液态(包括乳状)为主的环境下进行一,二,三级菌种培养,然后转入液态培养料与固态培养料混合而成的发酵原料中进行同体发酵。
3,按照权利要求2所述的双歧杆菌制剂其专门生产的液一固体相结合的发酵工艺,其特征是所说的对发酵产物进行调质是指将发酵产物加入占总量0.02~0.2%的赖氨酸,0.04~0.2%的谷氨酸,0.05~0.15%的蛋氨酸,0.002%的医用B族维生素和0.0001%的生物素。
4,按照权利要求2所述的双歧杆菌制剂其专门生产的液一固体相结合的发酵工艺,其特征是:所说的用于同体发酵的液态培养料是指:取打浆土豆和麸皮约占总量3~6%,加水煮25分钟,再加入蛋白胨(占总量0.5~0.8%),生肉膏(占总量0.2~0.4%),蔗糖(占3~7%)和食盐(占0.02~0.04%)溶解而成,调PH值7~7.2。
5,按照权利要求2所述的双歧杆菌制剂其专门生产的液一固体相结合的发酵工艺,其特征是:所说的用于固体发酵的固体培养料是指:用精玉米粉(占总量45~50%),精大米粉(占12~15%),脱脂大豆粉(占15~20%),全脂乳粉(占25~30%)以及食用酵母膏(占0.2~0.3%)混合而成,同体培养料与液态培养料的比例为1∶0.4。
