技术领域

背景技术
干细胞通过其自我更新能力和分化成各种体细胞类型的能力来定义。关于增殖、效力和细胞命运决定的细胞程序可由调节转录因子表达和/或激活的信号转导事件介导。
干细胞对于理解和治疗一系列疾病、损伤和其它健康相关疾患具有巨大的前景。也许人干细胞最重要的潜在应用是产生可用于基于细胞的疗法的细胞和组织。如今,捐赠的器官和组织经常用于替代生病或毁坏的组织,但是对可移植的组织和器官的需求远超过可用的供应。定向分化为特定细胞类型的干细胞提供了替代细胞和组织的可再生来源以治疗疾病的可能性,所述疾病包括但不限于黄斑变性、脊髓损伤、中风、烧伤、心脏病、糖尿病、骨关节炎和类风湿性关节炎。
为了实现新的基于细胞的疗法的前景,必须对干细胞进行操纵使得它们具有成功分化、移植和植入所必需的特征。例如,为了用于移植目的,必须可再现地使干细胞广泛增殖并产生足够量的细胞用于制备组织、分化成所需细胞类型、在移植后在受体中存活、在移植后整合到周围组织中、在接受者的一生中适当地发挥功能、以及避免免疫排斥。
发明内容
成功使用干细胞疗法的主要障碍之一是这些细胞一旦植入就可能变成癌性的。此外,本领域已知的干细胞疗法需要在输注或植入之前进行离体修饰以制备成功的治疗产品。虽然这通常在实验室中对分离的细胞进行,但本公开提供了用于原位(例如直接在器官中)产生诱导多能干细胞(iPSC)的方法。

图2A显示用治疗性基因体外转染后视网膜细胞(ARPE-19,ATCCTM)培养物中GFP+细胞计数的比较。所有GFP阳性细胞数基于103个细胞估计。

具体实施方式

干细胞疗法的临床试验已经显示出视网膜色素上皮(RPE)细胞移植的有希望的前景。例如,2011年,Advanced Cell Technology(Santa Monica,CA,USA)进行I/II期临床试验以阐明hESC衍生的RPE细胞移植在干性AMD和斯塔加特病患者中的功效。随后,Schwartz等人(2016)发表了18名干性AMD或斯塔加特病患者的两项I/II期试验的结果。四年后,半数以上的治疗患者经历了视力的持续改善,并证实了可能的细胞移植证据。然而,包括植入技术、免疫排斥和无异源组分的关键问题仍然需要进一步研究和改进。此外,需要考虑使用可携带诸如唾液酸或Neu5Gc的因子的人或动物来源的干细胞组分,所述因子可引起细胞的不希望的免疫原性或甚至使患者暴露于某些病原体。因此,通过iPSC技术和视网膜微环境调节基因表达的干细胞疗法代表了用于干性AMD治疗的潜在新方法。
用于产生iPSC的技术提供了将宿主(或患者)分化组织直接诱导为干细胞样细胞而无需外源施用干细胞诱导蛋白如Oct4、Sox2、cMyc或Klf4的新机会。虽然已知这些转录因子在诱导iPSC中起作用,但它们也可触发肿瘤发生。因此,本文公开的组合物和方法包括用抗癌基因如PCBP1转导细胞以减少或防止癌发生。

将治疗基因有效递送到靶组织或细胞是成功基因疗法的最重要的障碍。由于裸DNA在体内被吞噬免疫细胞或胞外核酸酶快速清除或降解,因此可能需要保护转基因的方法。此外,由于自发DNA摄取的效率低,因此还需要用于实现组织或细胞进入的媒介物。因此,DNA通常与一些类型的基因递送媒介物(通常称为载体)组合以保护和介导目的基因的有效组织或细胞进入。

基于质粒DNA的载体通常用于基因治疗,并且可以容纳大的DNA片段,并允许操纵影响基因转移和表达的各种调节元件。在最基本的情况下,表达质粒含有表达盒和骨架。表达盒是含有一个或多个目的基因和在靶细胞中表达所需的任何调节序列的转录单元。骨架可以含有选择标记物(例如抗生素抗性基因或营养缺陷型选择基因)和/或在细菌中产生质粒所需的复制起点。在一些实施方案中,质粒不含有选择标记物或传统复制组分(例如小环、自我复制小环、小串、线性DNA等)。

任何合适的启动子可用于驱动表达盒中基因的表达。调节和/或诱导型启动子是重要的,因为它们允许根据临床应用中可能需要的不同条件原位诱导干细胞。这些启动子能够阻止干细胞无限繁殖/诱导和不停地生长。

实施例
实施例1-PCPB1和转录因子在诱导视网膜干细胞中的作用
视网膜变性是由视网膜细胞的进行性和最终死亡引起的视网膜退化。这些视网膜病影响全世界约2000个个体中的一个。
目前,干细胞疗法和基因疗法由于其治愈疾病(包括影响视力的疾病)的潜力而成为头条,然而,这些干细胞疗法需要在输注或植入之前进行离体修饰以制备成功的治疗产品。这是由于许多原因,其中主要是需要递送重编程细胞基因表达模式和诱导多能干细胞(iPSC)的分子。虽然这通常在实验室中对分离的细胞进行,但是本文公开了用于原位(例如直接在眼中)产生iPSC的方法。
用于治疗视网膜变性疾病的基因治疗载体系统的构建:将参与iPSC产生的所有基因插入到一个载体中并转导到视网膜细胞中。图1显示了含有强人延伸因子-1α(EF)启动子以驱动PCBP1、GFP基因(图1A)以及转录因子SOX2、Oct4和KLF4基因(图1B)共表达的载体系统。最终产物是在单个递送载体系统中含有这些转录因子的所有盒的载体(图1C)。每个治疗性基因的开启由处于控制或诱导方式下的启动子驱动。
结果:将分化的视网膜细胞诱导为干细胞样细胞的体外治疗功效:从ATCCTM获得分化的视网膜细胞培养系(ARPE-19),并在不含FBS的培养基中生长。随后分离细胞培养物用于基因转染和感染。
PCBP1基因和转录因子基因在视网膜细胞中的表达:将视网膜细胞分离并且分成三组:第一组用含有PCBP1-GFP(绿色荧光蛋白)的载体转染以共表达PCBP1和GFP。24-28小时后,观察到GFP的表达,表明载体系统成功地将所有基因(转录因子)递送到视网膜细胞中(图2A和2B);第二组用于通过提取总RNA进行实时RT-PCR反应来证实视网膜细胞中PCBP1的表达水平(图3);第三组用于从分化的视网膜细胞诱导iPSC,如下所示。在图2A中,数字显示超过90%的GFP阳性细胞。该图显示了在荧光显微镜下与图2B相同的选定区域(左上(TL)、左下(BL)、右上(TR)、右下(BR)和中心(C))的比较研究。在每个孔中,由荧光显微镜的相机选择并捕获五个单独的区域(左上(TL)、左下(BL)、右上(TR)、右下(BR)和中心(C))。然后使用荧光显微镜观察/捕获5个区域中的每一个的照片。在每个区域中计数GFP阳性细胞,并在5个区域中计算平均值。这5个区域的平均值乘以103(因为显微镜捕获的面积约为整个细胞面积的0.1%)。因此,该方法的公式为:
用基因载体转染视网膜细胞后28-72小时,收获细胞,通过Qiagen提取试剂盒进行RNA提取。将分离的总RNA储存在-80C°以备将来使用,并使用标准实验室技术产生cDNA文库用于RT-PCR。
实时PCR证实PCBP1在视网膜细胞中的体外表达水平。如图3所示,PCBP1通过编码PCBP1的载体成功地在靶细胞中高度表达。
干细胞培养:在细胞培养板上培养视网膜细胞。将PCBP1(连同GFP)和其它转录因子(SOX2/OCT4/KLF4)通过lipofectamine转染到细胞中。将转染的细胞培养2-3天,接种在Matrigel上,并用干细胞生长培养基(StemRD目录号SPGro-free)培养。
转录因子诱导干细胞(PCBP1/OCT4/KLF4/SOX2):使用三种商业化的逆转录病毒转录因子(OCT4/KLF4/SOX2)。在病毒转导前2天,在37℃,5%CO2培养箱中将20μg C/EBPα蛋白加入到视网膜培养细胞上。在病毒转导当天,将适量(终浓度为10μM)的β-雌二醇加到细胞上,以激活C/EBPα-雌激素受体,并用转录因子(OCT4/KLF4/SOX2)通过逆转录病毒感染细胞。将细胞孵育2-3天,然后收集上清液并用500μl PBS洗涤培养物两次。然后将上清液/PBS以300g离心5分钟,将沉淀重悬于新鲜的重编程培养基(人iPS细胞生长培养基)中,并铺板于新的24孔板(用matrigel充分制备)上。将细胞在37℃培养箱中孵育4天,每天更换干细胞培养基。在第4天开始每天观察细胞。在第10-12天,菌落应生长至合适的大小用于分析、分离和再接种。(图4A)。
抗体染色和重组:生物标记物CRX-1可用作视网膜干细胞重组/分化的标记物。图4B显示本实验中诱导的iPSC细胞表达CRX-1。将人抗CRX抗体(AF7085,R&D系统)施加到培养细胞上,并将缀合的二抗(NL010,R&D系统)作为染色信号添加到人抗CRX抗体上。抗体的成功结合产生红色荧光信号。
载玻片上的细胞用PBS(0.145M NaCl,0.0027M KCl,0.0081M Na2HPO4,0.0015MKH2PO4,pH7.4)洗涤两次,然后用固定缓冲液(4%多聚甲醛的PBS溶液)固定。固定后,将含有固定细胞的载玻片在400μL洗涤缓冲液(0.1%BSA(牛血清白蛋白)的PBS溶液)中洗涤两次,然后在室温下在透化缓冲液(0.5%Triton X-100的PBS溶液)中孵育5分钟,然后通过加入400μL封闭缓冲液(5%BSA的PBS溶液)封闭非特异性染色。将载玻片在室温下孵育45分钟,除去封闭缓冲液。根据制造商的说明书,在稀释缓冲液(1%BSA,0.3%Triton X-100和0.01%叠氮化钠的PBS溶液)中稀释未缀合的一抗(或荧光缀合的一抗)。类似地稀释并加入二抗以说明与靶细胞结合的一抗。使用适用于所用标记的荧光显微镜和滤光片组对载玻片进行观察。将载玻片在<-20℃储存在载玻片盒中用于随后的检查。如图4B所示,用含有PCBP1和SOX2、OCT4、KLF4转录因子的载体转导后,一些细胞转变成视网膜干细胞样细胞。图5中所示的用一种视网膜干细胞标记物染色的抗体表明,在用含有PCBP1和SOX2、OCT4、KLF4转录因子的载体转染后,一些细胞已经转变成视网膜干细胞样细胞(对照组:不用PCBP、SOX2、OCT4、KLF4进行转染)。
此外,载体上PCBP1的存在阻止了癌症生物标记物的表达。图6显示基因载体产物能够体外转导皮肤癌细胞。图7显示PCBP1和PRL-3在胰腺癌细胞中的表达。用PCBP1转导胰腺癌细胞抑制转移标记物PRL-3表达。当与对照组比较时,PRL-3通过PCBP1 mRNA的过表达而下调。这也通过蛋白质印迹在蛋白质水平上得到证实(图6B)。
实施例2-将治疗载体注射到动物模型的眼中和体内治疗功效的研究:
移植的RPE细胞在人眼中显示有限的粘附和存活,并且老化的布鲁赫膜不可能支持移植的RPE细胞的粘附、存活、分化和功能。因此,使用基因工程在RPE细胞中过表达整合素或整合素激活剂或使用在支架上生长的RPE细胞可能显示出有希望的前景。此外,尽管视网膜下空间曾被认为具有免疫特权,但研究还表明移植的干细胞在宿主眼中的长期存活仍需要免疫抑制。在未来的临床应用中正在考虑免疫抑制的过程和用于免疫抑制的药物。
因此,原位使用iPSC技术促进视网膜细胞更新和再生以修复损伤将是治疗患有黄斑变性疾病的患者的实用且改进的策略,其具有最小或没有上述副作用。本文公开的系统用于治疗视网膜疾病的功效将在动物模型中测试。基因陷阱载体将用于体内研究干细胞。例如,AAV和慢病毒都可用于递送本公开的质粒。载体可以用本公开的质粒转染,然后使用标准实验室技术纯化。
动物模型:本研究将使用C57Bl/6和DBA/2J小鼠。对于C57Bl/6和DBA/2J小鼠,将动物置于三个主要剂量组中,包括对照(仅GFP),仅含PCBP1的载体和与三种转录因子SOX2、OCT4和KLF4组合的PCBP1载体(参见表3)。
所有动物将根据NIH指南在12小时光照/黑暗循环下饲养在动物设施中。
表3:动物模型实验设计
体内药物递送到动物眼睛中:在大鼠和小鼠模型中,通过视网膜下注射将载体构建体施用到眼睛的视网膜层中(图9)。
空间视力:为了测试通过递送基因载体修复受损的视网膜细胞是否可以从黄斑生成恢复视力,将使用验光测试设备(Cerebral Mechanics,Lethbridge,Canada)测试动物的视力。该测试包括布置成正方形的四个计算机监视器,其投影衬有垂直正弦波光栅的旋转圆柱的虚拟三维空间。将不受限制的动物放置在正方形中心的平台上,在那里它们以反射性头部运动跟踪光栅。通过使动物头部上的“圆柱”重新定中心,光栅的空间频率被固定在观察位置。通过使用心理物理学阶梯级数增加光栅的空间频率直到失去随后的响应来量化敏锐度阈值,从而定义敏锐度。以每月间隔测试大鼠。还在注射后3、5、7和11周在该设备中测试了Elovl4小鼠(参见图10)。版本结果可以按周期/度进行调整/分析。更高的周期/度表示更好的版本恢复(即,正常大鼠版本为0.6周期/度)。
治疗动物中癌症发展的评估:在NIH III小鼠模型中测试畸胎瘤形成的长期风险,选择其免疫缺陷状态。裸鼠具有三种突变,使其缺乏T细胞、NK细胞和成熟的T非依赖性B淋巴细胞。然而,NIH III小鼠保留了眼色素沉着,这为视网膜下注射提供了更好的可视化。手术技术与RCS研究中的相同。该研究在三个时间点将治疗组与对照组比较:1个月、3个月和9个月(动物的大致寿命;每队列n=6。通过以下研究,在安全性研究中,在任何注射病毒基因载体的动物中均未发现畸胎瘤或肿瘤形成。畸胎瘤或肿瘤形成可通过如下所述的组织化学或药物分布研究来评估。
组织化学(癌症或肿瘤、组织结构):在功能研究结束时,用过量戊巴比妥钠杀死所有动物并用磷酸盐缓冲盐水灌注。将眼睛取出,浸入2%多聚甲醛中1小时,用蔗糖浸润,在光学切割温度下包埋,并在低温恒温器上切成15μm水平切片。每个载玻片收集4个切片(间隔50μm),收集每四个切片的五个系列。用甲酚紫染色以评估注射部位和视网膜层的完整性。剩余的载玻片用于抗体染色、癌症/肿瘤检测和组织结构分析。
药物分布和药物作用途径在视网膜组织中的分析:使用人和小鼠或大鼠Affymetrix HG-U133 Plus 2.0微阵列平台(Santa Clara,CA)对来自参与药物递送的不同器官/组织的PCBP1表达进行全局基因表达分析。另外,当与治疗比较时,分析ARPE-19(美国典型培养物保藏中心TM,Manassas,VA)和视网膜母细胞瘤(美国典型培养物保藏中心TM)细胞系和正常视网膜细胞作为对照。统计学软件将支持数据分析。
使用Bio-Rad Mini-Protean和Mini-Transblot Cell(Hercules,CA),使用标准SDS-PAGE方法进行PCBP1和其它转录因子的免疫印迹分析。使用Western LightningChemiluminescence Reagent(PerkinElmer Life and Analytical Sciences,Boston)和Kodak 4000MM数字成像站(Rochester,NY)使蛋白质条带可视化。
实施例3-PCBP1经由PRL3/STAT3途径与Sox2、Oct4、KLF4和其它相互作用:
为了研究PCBP1与其它干细胞转录因子的相互作用并确定它们相互作用的途径,可以如下进行视网膜细胞中的研究。用本公开的各种载体处理四种细胞培养物。如图11A所示,将癌细胞培养物用作对照,不用任何载体转染。该培养物的qRT-PCR将显示与癌症和转移相关的生物标记物(PRL-3/STAT3)的表达。如图11A所示,转染含有视网膜细胞的三种培养物。在用含有PCBP1和SOX2/KLF4/OCT4干细胞转录因子的载体转染的细胞培养物中,与癌细胞或未转染的视网膜细胞中的表达水平相比,预期PRL-3/STAT3的表达被抑制。在用含有SOX2/KLF4/OCT4干细胞转录因子的载体转染的细胞培养物中,与未转染的视网膜细胞和另外用PCBP1转染的那些相比,预期PRL-3/STAT3的表达增加。图11B显示了本实验中使用的代表性平板。
图12显示了干细胞转录因子和PRL-3/STAT3途径相互作用的推定机制。在iPSC产生期间,PCBP1与干细胞转录因子(例如SOX2、OCT4、KLF4)一起作用以维持干细胞发育。然而,PCBP1还作用于与STAT3信号的下调相关的抑制PRL-3的途径以促进祖细胞分化为靶细胞或组织,而不增加肿瘤发生。
实施例4-体外心脏成纤维细胞干细胞诱导
美国每年有超过150万例心肌梗死病例,继发于供氧不足的心肌细胞(CMC)的不可逆死亡导致严重损伤。心脏中的CMC具有有限的再生能力,因此,约五百万存活于急性心肌梗死的患者患有缺血性心肌病,导致心力衰竭。因此,需要通过补充受损的CMC来修复瘢痕组织的急性心肌梗死的有效疗法。
直接成纤维细胞重编程首先通过心肌生成因子如GATA4、Mef2C和Tbx5(GMT)的病毒过表达来尝试(Chen JX等人(2012))。最近,Ma等人(2017)通过改变蛋白质化学计量进一步优化该策略以在成熟CMC的产生中实现更高的效率。治疗策略的后续修改通过小分子抑制促纤维化信号传导将重编程效率提高了高达60%。然而,GATA4基因表达的改变与几种癌症类型有关,因此增加了肿瘤发生可能性。
原位诱导以特异性靶向受损细胞的干细胞促进组织修复。原位诱导可避免与移植前非原位干细胞诱导相关的常见问题,如靶区不准确和病原体污染。本文显示心脏成纤维细胞(CF)直接转化为CMC,这显著降低副作用(包括肿瘤发生)的风险。
将心脏成纤维细胞体外诱导为干细胞样细胞
构建表达共表达靶基因下游的PCBP1和GFP(PCBP1-IRES-GFP)并分别表达Oct4、Klf4、Sox2和c-Myc的单独的慢病毒载体。人胎儿心脏成纤维细胞(FCF)在不含FBS的培养基中生长,然后分离用于慢病毒转导。随后,将它们分成三组。组1用含有或不含有Sox2/Oct4/Klf4的LVV PCBP1-IRES-GFP转导,用于GFP可视化,或用仅GFP载体作为阳性对照(图13A)。组2用于通过总RNA提取和qRT-PCR分析证实PCBP1 mRNA在心脏成纤维细胞中的表达(图13B)。图13B显示通过qRT-PCR比较PCBP1-IRES-GFP载体转导的CF和对照(未转染)的PCBP1mRNA水平。转染的细胞显示出高于99倍的PCBP1表达(ddCt=99.9;p<0.05)。
其它转录因子的mRNA也通过qRT-PCR证实(数据未显示)。组3用于将FCF iPSC诱导为心脏干细胞样细胞。
使来自组3的FCF连续生长,然后用慢病毒载体转导72小时,然后转移到含有matrigel作为底物的新平板上,并在存在c/EBP-α和β雌二醇下在无FBS的干细胞培养基中生长16天。
已显示c-Kit受体酪氨酸激酶能使培养的心脏细胞分化成CM、平滑肌和内皮细胞,并用作心脏干细胞标记物。使用c-Kit特异性抗体检测新出现的心脏干细胞生物标记物并鉴定心脏干细胞样细胞,如图14所示。转导后16天,FCF失去其典型的纺锤体结构并显示为表型心脏干细胞球状结构。这些FCF也被c-Kit抗体检测呈阳性(图14)。有趣的是,用单独表达PCBP1的LVV转导CF有效地诱导心脏成纤维细胞将表型从经典纺锤体样结构改变为类似于心脏球体(心脏干细胞来源)所述的结构,并被证明具有再生能力(图14C和14D)。
当CF变成干细胞样时,其逐渐丧失成纤维细胞特异性生物标记物并在细胞表面表达心脏干细胞生物标记物。例如,COL1a1是心脏成纤维细胞特异性标记基因。编码心肌肌钙蛋白T的TNNT2是CMC的生物标记物,并且在心脏新月期祖细胞中富集。如图15所示,用含有PCBP1、Sox2、Oct4和Klf4的LVV转导导致COL1a1 mRNA水平降低5倍以上。此外,用LVV转导诱导c-Kit表达增加30倍以上,TNN2表达增加近8倍。
这些结果表明,CF可由含有PCBP1、Sox2、Oct4和Klf4的LVV体外诱导以产生心脏干细胞样细胞。
实施例5-心脏成纤维细胞干细胞原位诱导
分化的成纤维细胞可以重新编程以诱导多能性并成为干细胞。目前的干细胞疗法在输注或植入之前需要离体修饰,主要是由于需要递送重新编程基因表达模式并诱导多能干细胞(iPSC)的分子。虽然这通常在实验室中对分离的细胞进行,但在离体操作期间难以维持干细胞状态。这里,不是离体诱导iPSC,诱导将原位发生,例如使用PCPB1和一种或多种转录因子的共表达直接在心脏中进行。PCBP1和iPSC转录因子的组合表达将直接诱导心脏成纤维细胞转化为干细胞样细胞,并最终转化为CMC用于治疗心肌梗塞。
使用分子克隆技术,将iPSC中涉及的四种转录因子工程化以由单一载体系统表达,促进转录因子的递送并实现可再现的结果。通过qRT-PCR检测每个单独的基因的mRNA表达,通过蛋白印迹进行蛋白分析。
手术处理小鼠心脏组织以产生心肌梗塞。然后将本公开的载体系统原位转导到分化的成纤维细胞中以触发CF转化为干细胞,这将通过干细胞特异性标记物来检测。将如下进行几种体外功能测定:
免疫细胞化学:将在培养的CF中诱导iPSC,通过明场可视化并通过检测心脏干细胞生物标记物确认。FCF将被LVV载体系统体外转导,该系统表达混合的个体病毒和多顺反子PCBP1、Sox2、Oct4和Klf4病毒。另外,将测试仅含有PCBP1(即没有其它转录因子)的LVV载体,因为从实施例4的研究中观察到心脏球结构。转导后4天,将细胞转移到基质胶包被的培养皿上并培养15天以诱导心脏干细胞。在心脏干细胞簇可见后(与对照组相比),用c-Kit特异性抗体进行免疫组织化学以在荧光显微镜下检测c-Kit表达水平。
体外心脏重编程的评价-CMC搏动测定:将iPSC如上所述诱导。细胞内钙水平的荧光成像将通过自动信号传导分析软件进行分析。通过分析指示心脏生理学的参数,预期CMC显示自发收缩细胞的表型反应。
体外肿瘤发生潜力的评价-信号传导途径分析:再生肝磷酸酶3(PRL3)是一种致癌因子,其激活通常与肿瘤发生相关。同样,Stat3参与多种癌症的致癌作用。对这些癌蛋白的评价在绘制由当前LVV系统调节的信号传导途径中是重要的。提取总RNA和蛋白质,然后使用qRT-PCR和蛋白质印迹分析总蛋白质和磷酸化/激活形式的特异性生物标记物如c-Kit、COL1a1、TNNT2、PRL3和Stat3。PRL3总蛋白和激活蛋白水平的下调以及下游靶标STAT3的激活降低表明肿瘤发生潜力降低。
在心肌梗塞鼠模型中的功效:将进行动物试验以确认系统在体内的适用性(参见图16)。将慢病毒载体注射到MI动物模型的心脏组织中以诱导CMC再生。通过行为研究、生理和组织学分析监测成功的心功能恢复。还将在分子水平上评估信号传导途径成分。
体内模型:MI的左前降支(LAD)动脉结扎鼠模型用于再现人类心脏病发作中典型的局部缺血性心肌组织损伤。先前使用小鼠MI模型的研究通过逆转录病毒载体在单个mRNA中递送Gata4、Mcf2c和Tbx5。在此,使用允许递送较大基因有效负载的慢病毒载体。在Balb/C小鼠中使用LAD连接的动物研究将有四个队列:1)PCBP1;2)PCBP1/Oct4/Klf4/Sox2;3)c-Myc/Oct4/Klf4/S0x2;和4)表达GFP的对照载体作为阴性对照。每个队列包括5只动物,实验重复三次。注射病毒后20天,对于从心脏病发作中恢复,记录动物的行为。然后将动物麻醉并处死。通过免疫组织化学分析移植的心脏在缺血区域的恢复情况。将监测PCBP1和Sox2、Oct4、Klf4转录因子下游的信号传导途径成分的整体微阵列基因表达谱。
体内直接重编程的评价-瘢痕尺寸的测定:MI后8周,用标准Masson-Trichrome染色法对心脏组织进行染色。梗塞区域显示为蓝色,而存活心肌为红色。使用Image J软件分析一系列横切面(第一层位于连接正下方)上的瘢痕区域。
体内直接重编程的评价-心脏功能:将分析试验动物心脏的超声心动图、血流动力学和MRI,以评价注射或未注射LVV的MI后4、8和12周时的心脏功能。
体内直接重编程的评价-基因调节:将从MI后的不同心脏区域和时间点分离CMC,并通过消化和FACS处理。收集总RNA和蛋白质并分析信号传导途径成分如Stat3和/或PRL3。与c-Myc、Sox2、Oct4和Klf4组相比,用含有PCBP1、Sox2、Oct4和Klf4的LVV注射的组将显示出相当的或甚至更好的心脏功能恢复。预计LVV转录因子组都比对照组恢复得更好,并且GFP和盐水对照组之间应该没有显著差异。
替代载体构建策略:由于四种转录因子蛋白的累积大尺寸,因此它们由于有效负载能力的限制可能需要单独递送。因此,可以使用以下替代的构建选项。(1)重设计构建体,通过产生“PCBP1-2A-Sox2”和“Oct4-2A-KLF4”两种融合蛋白,这两种融合蛋白的表达将由两个单独的EF启动子驱动,每种蛋白在递送后同样在细胞内释放;(2)分别构建4个表达各转录因子的单独的载体。
如果不能使用单个载体诱导体内模型中的干细胞样细胞,则可应用以下替代方案:(1)使用不同的构建体-将含有四个单独载体的病毒汇集在一起以递送到靶向组织中,因为已经证明该单独载体在先前制备的构建体中表达转录因子;(2)选择例如腺相关病毒载体(AAV)的另一种递送系统是另一种选择。可以构建四种AAV载体,每种编码一种转录因子。AAV载体的优点是它们可以递送高得多的病毒滴度和局部蛋白浓度,这可能导致对靶细胞的更高功效。此外,AAV载体已被FDA批准用于治疗晚期心力衰竭患者的临床试验;(4)添加促纤维化信号传导如TGFβ和ROCK信号传导的抑制剂以提高效率。
实施例6-MCF-7(CSC)乳腺癌干细胞(BCSC)与MCF-7癌细胞中CD44和CD24的FACS检测
TGF-β培养:MCF-7乳腺癌细胞干细胞和MCF-7癌细胞在孵育后使用补充有TGF-β(5ng/mL)的完全培养基处理7天,以通过选择CD44+和CD24-亚群增强癌症干细胞产生和纯化。培养所选择的细胞(例如用TGF-β+培养基)直到细胞数目足以用于实验(通常在分离后约4至5天)。对照细胞(即未施用TGF=β的细胞)用推荐的培养基正常处理。用Lipofectamine 3000进行所有转染。使用没有载体DNA的Lipofectamine处理进行模拟转染。
将收获的细胞用抗人CD44 PE-缀合的单克隆抗体(10μL/106细胞)和抗人CD24APC-缀合的单克隆抗体(10μL/106细胞)共染色。非特异性抗体用作同种型对照。
图17显示MCF-7(CSC)(BCSC)具有较高的CD44表达,但CD24表达降低,从而证实了使用该方法富集乳腺癌干细胞群(CD24-;CD44+)。

实施例7-PCBP1过表达在癌症干细胞中的作用
如本文所述,用PCBP1构建体、LV-GFP、wPCBP1_0、m_PCBP1_1、mPCBP1_2转染或模拟转染各种癌细胞系。通过qRT-PCR分析转染细胞中PCBP1转录物的量。图22。转染后48小时从细胞中提取总RNA,然后使用逆转录PCR转化为cDNA。通过qPCR用PCBP1特异性Taqman探针测量PCBP1和变体。GAPDH作为内部对照。在所有三种细胞系中,LV-wPCBP1-GFP(野生型)、LV-PCBP1_突变体_1-GFP和LV-PCBP1_突变体_2-GFP中PCBP1 mRNA表达没有显著差异。模拟转染用作计算的基线。

总结:使用TGFβ处理产生并分离癌症干细胞,并分选CD24-和CD44+群体(通过FACS和蛋白印迹证实)。用野生型或突变形式的PCBP1转染这些分离的癌症干细胞后,在表达野生型或突变形式的PCBP1后,癌症干细胞中的CD44和CD133显著降低。CD44可用作癌症干细胞生物标记物,CD133可用作癌细胞生物标记物。然而,如此处所证明的,野生型或突变PCBP1的表达可以阻断CD44和CD133的表达,以防止癌症干细胞的肿瘤发生潜力和干细胞性。这表明PCBP1可能在iPCS的产生过程中作为调节剂起重要作用。
实施例8-氯硝柳胺或PCBP1表达下调BCL-2和c-Myc
已经发现氯硝柳胺对涉及各种途径的癌症干细胞具有抑制潜力。因此,我们研究了氯硝柳胺作为癌症干细胞抑制剂。许多相关途径在癌症干细胞和干细胞之间共享(例如,维持未分化状态,而不是诱导PSC的产生)。在此,研究了氯硝柳胺对NF-kB途径的作用。图25。

总结:BCL-2和c-Myc通过用PCBP1(和突变体)或氯硝柳胺处理显著下调。由于氯硝柳胺已被用作癌症干细胞的抑制剂,因此这些数据支持PCBP1基因治疗的可比效果。因此,这些数据表明PCBP1也用作癌症干细胞抑制剂,因为它可以阻断CD44和CD133的表达,可能通过下调BCL-2和c-Myc表达,类似于氯硝柳胺对NF-kB途径的作用。这些数据支持PCBP1和突变体在iPSC产生期间抑制和预防肿瘤发生中的用途。

Claims (24)
1.一种从分化细胞诱导多能干细胞的方法,所述方法包括引入包含PCBP1或其突变体或变体的核酸序列或PCBP1或其突变体或变体和一种或多种其它转录因子的载体,其中所述多能干细胞的诱导不伴有肿瘤发生。
2.如权利要求1所述的方法,其中所述方法在体外进行。

4.如权利要求1所述的方法,其中所述方法在体内进行。
5.如权利要求1所述的方法,其中所述分化细胞是哺乳动物的。
6.如权利要求5所述的方法,其中所述哺乳动物细胞是人的。
7.如权利要求5所述的方法,其中所述哺乳动物细胞是器官细胞、组织细胞或血细胞。
8.如权利要求7所述的方法,其中所述组织细胞是视网膜或心脏细胞。
9.如权利要求1所述的方法,其中所述载体包含PCBP1或其突变体或变体的核酸序列和至少一种选自由SOX2、OCT4和KTL4组成的组的转录因子。
10.如权利要求1所述的方法,其中所述载体包含PCBP1或其突变体或变体的核酸序列和至少两种选自由SOX2、OCT4和KTL4组成的组的转录因子。
11.如权利要求1所述的方法,其中所述载体包含PCBP1或其突变体或变体的核酸序列、SOX2、OCT4和KTL4。
12.一种从分化细胞诱导多能干细胞的方法,所述方法包括引入包含PCBP1或其突变体或变体的载体,其中所述多能干细胞的诱导不伴有肿瘤发生。
13.如权利要求12所述的方法,其中所述方法在体外进行。
14.如权利要求12所述的方法,其中所述方法离体进行。
15.如权利要求12所述的方法,其中所述方法在体内进行。
16.如权利要求12所述的方法,其中所述分化细胞是哺乳动物的。

20.如权利要求12所述的方法,其中所述载体包含PCBP1或其突变体或变体的核酸序列和一种或多种其它转录因子。
21.如权利要求20所述的方法,其中所述载体包含PCBP1或其突变体或变体的核酸序列和至少一种选自由SOX2、OCT4和KTL4组成的组的转录因子。
22.如权利要求20所述的方法,其中所述载体包含PCBP1的核酸序列和至少两种选自由SOX2、OCT4和KTL4组成的组的转录因子。
23.如权利要求20所述的方法,其中所述载体包含PCBP1的核酸序列、SOX2、OCT4和KTL4。
24.如权利要求20所述的方法,其中所述载体不包含另外的转录因子。
