Technical Field
The invention belongs to the technical field of biological experimental equipment, and particularly relates to a cell climbing sheet culture device and an application method thereof.
Background
Cell culture is an essential experimental process in biomedical research, and cell slide is needed when further observing the characteristics of cell morphology, protein expression and the like. In the conventional cell slide, a slide is placed in a culture dish/culture plate, conventional cells are spread before the slide, and after the cells grow on the slide substantially full, the slide is taken out, and after rinsing, residual culture medium and serum are sufficiently removed, and then cell fixation and subsequent experiments are performed. The traditional cell slide technology and the traditional cell slide device have the following defects: since cells generally grow on only one side of the cell slide, all experimental operations are performed on the side containing cells. In the process of immunofluorescence staining operation, researchers are easy to confuse one surface containing cells and one surface not containing cells in the glass sheet due to operation negligence, and the recognition error can cause wrong operation, so that the whole experiment is directly failed; secondly, the slide is easy to slide and difficult to fix; due to surface tension, the bottom surface of the climbing sheet can be tightly attached to the bottom surface of the culture pore plate, the climbing sheet is easily crushed in the process of taking out the climbing sheet, and even if the climbing sheet is not crushed, cells originally full on the climbing sheet can be damaged by tweezers, so that an experimental result is influenced; cells can gather around the slide due to the edge effect, and the central area is sparsely distributed, so that the experimental result is influenced; fifthly, in order to prevent the adjacent holes from being polluted by cells, only one cell culture bottle or device can be used for processing, but most of experiment designs need to process the same cells at different time, or the same stimulation needs to be carried out but the stimulation time is inconsistent, so that only a plurality of culture plates can be used for carrying out experiments, great resource waste is caused, the condition that the cell growth environment is inconsistent can also occur, and certain influence can be caused on the experiment results.
Therefore, it is necessary to provide a new cell-slide culture apparatus to solve the above problems.

The invention aims to provide a cell slide culture device which has multiple functions of cell culture, high-flux cell slide preparation and the like, and overcomes the defects in the traditional cell slide design.

According to the cell climbing sheet culture device provided by the invention, cells directly grow on a glass plate of the device, and are uniformly distributed; links such as plate laying, slide taking and the like are eliminated, the experimental time is saved, and cell slides with good growth state and uniform distribution can be obtained in 4 hours after the cell suspension is added, and the condition of cell slide failure is avoided; a plurality of glass plates can be placed in the device at the same time, a plurality of cell culture devices are arranged on each glass plate, and one device can obtain a plurality of cell crawl sheets, so that the preparation of the cell crawl sheets in large batch is facilitated; the method can be suitable for preparing complex experimental designs such as 'same 1 time point, multiple cells, multiple intervention treatments', 'multiple time points, multiple cells, multiple intervention treatments' and the like; the consistency of intervention conditions can be ensured, and the accuracy of research results is improved; the glass tubes are mutually independent, and integrated slide glass of various cells can be simultaneously prepared and subjected to various intervention treatments on a single glass slide; each cell culture device on the glass plate can be designed to have a diameter of 8mm and a height of 10mm, and the cell suspension required in each device is less than 500 microliters, so that the cell culture reagent, the cell number and the drug intervention cost are greatly saved.

The invention is simple and reliable in preparing cell climbing tablets, and the growth state and distribution condition of cells are superior to those of the traditional method.
