Experimental study on influence of yin-nourishing and jade-moistening ointment formula on vascular calcification and blood pressure of yin-deficiency and yang-hyperactivity type hypertension rat
1.1 animals
Healthy and clean SD rats (20 weeks old), female, with a body mass of 200-220 g and a body mass of 180-220 g, provided by the animal experiment center of Shanghai medical university. Adaptive feeding for 1 week at room temperature of about 23 deg.C and relative humidity of 40-70%.
1.2 test drugs
The invention relates to a yin nourishing and jade moistening ointment formula: prepared as described in example 1. The preparation is administered by mixing with purified water.
The original yin-nourishing and jade-moistening ointment formula comprises: american ginseng 90 g, starwort root 60 g, vinegar processed bupleurum 90 g, fertile anemarrhena 60 g, Guanhuangbai bark 60 g, shrunken peach 90 g, dried rehmannia root, prepared rehmannia root 120 g each, cornus officinalis 100 g, curcuma aromatica 90 g, finger citron 90 g, green calyx prune 90 g, Chinese rose 60 g, angelica root 120 g, red peony root, white peony root 100 g each, fried ligusticum wallichii 150 g, prepared cyperus tuber 90 g, fried bitter orange 90 g, poria cocos 120 g, albizia bark 90 g, wild jujube seed 60 g, tuber fleeceflower 150 g, trichosanthes root 90 g, dendrobium 60 g, polygonatum 90 g, honey-fried licorice root 60 g, red jujube 100 g, walnut meat 100 g, lotus seed meat 100 g, wolfberry 100 g and black sesame 100 g. 1.3 grouping and Molding
40 SD rats were randomly divided into 4 groups, including model group, inventive yin-nourishing and jade-moistening plaster formula group, former yin-nourishing and jade-moistening plaster formula group and enalapril group, each group containing 10 rats. All 4 groups were given for molding.
A model building method for yin deficiency and yang excess type hypertension rats comprises the following steps: rats were fasted and kept without water for 12h, and placed in an artificial climate chamber (dry 40 ℃ and wet 32 ℃) for heat stress for 3 h. Preoperative abdominal cavity injection chloral hydrate (30mg/kg) is anesthetized, the abdomen is upwards fixed on an operation table, local skin is disinfected, a knife is placed at a position of about 1.5cm below the xiphoid process along the abdominal midline, an abdominal muscle layer is cut, the right finger is inserted into the abdomen to search for the kidney, the left hand is matched with the body to gently extrude the kidney, gauze soaked by prepared normal saline is wrapped and fixed by hands, the left renal artery at the position close to the renal hilum on the left side is separated, a line is taken to cross from the lower part, the thickness level of the artery is compared, a proper thin line is selected and placed according to the trend of blood vessels, a transverse line is knotted, the thin line is taken out after ligation, the wound is recovered, and the operation table is disinfected by iodine. After the rat is raised for 4 weeks in a conventional manner, the molding condition is evaluated, and the blood pressure measurement shows that the blood pressure of the rat is increased by 22.6mmHg (3kPa) and reaches a reference value of 135mmHg (18kPa) or more.
The activity, mood, hair, diet, body temperature, etc. of the rats were observed at regular time every day, and the irritability level of the rats was observed and classified at 1 week (before administration) and 5 weeks (4 weeks) after the molding. Grade I (1) screaming and twitching while catching neck; level II (2 points) of irritability and biting when catching the neck; grade III (3), the tail is lifted, i.e. screaming and frightening, even people want to bite the tail or rats in the same cage frequently fight and bite the iron cage frequently. The above case is not obvious and is 0 grade (0 point). And eliminating the mould after grading if the mould is not successfully made. According to the fact that the appearance hair color becomes hard, the glossiness is reduced, the temperament is easy to irritate, the food intake and the water intake are increased, the model of yin deficiency and yang excess is determined to be successful. I.e. indicating that the molding was successful.
1.4 modes of administration
The model group is that the stomach is irrigated with 0.9 percent sodium chloride solution with the volume equal to that of the traditional Chinese medicine;
the invention relates to a yin-nourishing and jade-moistening paste formula which is prepared by feeding corresponding liquid medicine into stomach according to the dose of 10g crude drugs/(kg.d) -1, wherein the crude drugs are raw materials except sugar and yellow wine;
the original Yin nourishing and Jade moistening ointment formula comprises the following steps of administering corresponding liquid medicine for intragastric administration according to the dose of 10g crude drugs/(kg. d) -1, wherein the crude drugs are raw materials except sugar and yellow wine;
enalapril group, wherein the Enalapril is Enalapril 10mg/kg/d, dissolved in 0.9% sodium chloride solution with volume equal to that of the traditional Chinese medicine, and then administered to stomach;
rat tail artery blood pressure was measured before and after 1 treatment course (4 weeks) using a DSWY-1 rat tail artery blood pressure measuring device and blood pressure differences between groups were observed. Measuring calcium content in aortic smooth muscle cells, washing rat aorta with Hanks solution to remove blood stain on the surface of tissue, removing connective tissue, cutting blood vessel, scraping vascular endothelial cells, cutting into 3 mm with size of 0.5-1 mm, washing Hanks solution until the solution is not turbid, sucking out Hanks solution, adding collagenase II solution with volume fraction of 0.2, digesting for 15min at 37 ℃, filtering to remove undigested tissue, centrifuging for 5min at 1000r/min, removing supernatant, washing with Hanks solution, adding DMEM culture solution containing fetal calf serum with volume fraction of 10 to prepare cell suspension, inoculating into a 24-hole culture plate for culture, taking out cover glass of adherent PASMC after cell adherence, adding appropriate Hanks solution, bathing for 10min at 37 ℃, adding Furu-2/AM with final concentration of 5mg/L, shaking for 45min at 37 ℃, washing with Hanks solution of bovine serum albumin with volume fraction of 0.2 for 2 times, and (3) rewarming for 10min at 37 ℃ before measurement, putting the PASMC loaded with Furu-2/AM into a measuring pool, adding D-Hanks liquid, selecting single cells, and performing real-time measurement on Ca by using a fluorescence microscopy technology.
The statistical method adopts SPSS10.0 medical statistical software package to perform statistical processing of data, and the measured data is expressed by (x +/-s); ANOAV analysis of variance is adopted, parallel homogeneity of variance tests are carried out, pairing t tests are adopted for the homogeneity of variance, and otherwise, t' tests are adopted; multiple comparisons were performed using analysis of variance. The difference is statistically significant when P is less than 0.05.
1.5 results:
compared differences among various groups of models before treatment of the change of the tail arterial blood pressure of rats, the yin-nourishing and jade-moistening plaster formula group, the original yin-nourishing and jade-moistening plaster formula group and the enalapril group have no statistical significance (P is more than 0.05);
after treatment, the blood pressure of the tail artery of rats in the yin-nourishing and jade cream formula group, the original yin-nourishing and jade cream formula group and the enalapril group of the invention are all obviously reduced (P is less than 0.05), wherein the blood pressure reduction range of the yin-nourishing and jade cream formula group is larger than that of the original yin-nourishing and jade cream formula group (P is less than 0.05) in the table 1)

Calcium content in aortic smooth muscle cells is measured in real time by a fluorescence microscopy, and calcium content in the rat aortic smooth muscle cells of the yin-nourishing and jade-moistening cream formula group, the original yin-nourishing and jade-moistening cream formula group and the enalapril group is reduced to different degrees (P is less than 0.05), wherein the calcium content of the yin-nourishing and jade-moistening cream formula group is reduced more than that of the original yin-nourishing and jade cream formula group (P is less than 0.05), and the calcium content is shown in table 2.
