Abstract

提供一种人源化的抗VEGFR2抗体药物。还提供编码所述抗体的核酸序列(包括重/轻链可变区)、含有所述核酸序列的载体、药物组合物和试剂盒。所述抗体能特异性结合VEGFR2的第六结构域，独立于配体VEGF‑A而发挥作用，可直接抑制VEGFR2形成二聚体，从而阻断VEGFR2胞内区二聚化后引起的酪氨酸残基磷酸化及下游信号通路。同时，该抗体保留部分阻断VEGF‑C和VEGF‑D的作用，因而具有更佳的抑制血管生成作用。所述抗体还能激活NK细胞发生抗体依赖性细胞毒性(ADCC)反应，抑制肿瘤的生长和转移，可用于临床治疗黑色素瘤。
