Abstract

技术领域
本发明涉及用作皮肤抗炎剂的β-L-天冬氨酰-L-精氨酸。进一步地,本发明还提供了一种β-L-天冬氨酰-L-精氨酸用于熟龄肌肤的美容用途。特别地,所述β-L-天冬氨酰-L-精氨酸发挥抗氧化作用和/或改善衰老的皮肤细胞特别是衰老的成纤维细胞中的线粒体功能,和/或恢复皮肤的紧致度。进一步地,本发明还提供了一种用于处理熟龄肌肤的美容方法。本发明还涉及用于治疗受损熟龄肌肤的治疗方法中的β-L-天冬氨酰-L-精氨酸。特别地,将所述β-L-天冬氨酰-L-精氨酸用于一种促进伤口愈合的治疗方法。
背景技术
熟龄肌肤的特征是真皮层(真皮)中衰老细胞的数量不断增加,细胞停止复制和分裂。位于真皮层内的成纤维细胞是其基本成分。成纤维细胞的主要功能是通过细胞外基质成分,包括胶原蛋白和弹性蛋白的产生和分泌来提供拉伸强度和弹性。随着衰老,损伤和氧化应激导致成纤维细胞进入生长停滞和功能变异的状态,即细胞衰老。衰老的成纤维细胞分泌生长因子、细胞因子和降解酶,导致皮肤弹性丧失、伤口愈合延迟和表面形态改变(Krtolica et al.,PNAS October 9,2001,98(21),12072-12077;Sorell and Caplan,JCell Sci,2004 Feb 15;117:667-675;Ezure et al.,IUBMB Journal,BioFactors 2019;45:556-562;Sibilla et al.,The Open Nutraceuticals Journal,2015,8:29-42)。
线粒体被认为是细胞的“发电站”。线粒体的主要功能是从细胞中吸收营养(葡萄糖),将其分解并转化为能量(ATP)。然后细胞利用该能量来发挥各种生物功能。线粒体功能失调与许多年龄相关的疾病和光老化有关。功能失调的不健康线粒体会减少能量产生并增强氧化应激(Krutmann and Schroeder,J Investig Dermatol Symp Proc,2009;14(1):44-9)。
在功能性线粒体中,氧的摄取、ATP的产生和活性氧(ROS)的产生受到严格控制,以维持氧化还原平衡。线粒体稳态受损,诸如线粒体生物发生增加、线粒体自噬减少、ATP生成减少和ROS生成增加,会导致衰老细胞周期停滞。此外,ROS水平的增加会导致DNA损伤,从而导致永久性细胞周期停滞。衰老细胞产生并分泌生长因子、细胞外基质降解蛋白和促炎细胞因子(SASP),这些产物与ROS一起不仅能稳定衰老,还能诱导旁分泌衰老,进而可能加剧衰老过程中的有害影响(Korolchuk et al.,EBioMedicine,2017,21:7-13;Correira-Meloand Passos,Biochimica et Biophysica Acta,2015,Volume 1847,Issue 11,1373-1379)。此外,衰老细胞分泌的促炎细胞因子和生长因子(SASP为衰老相关分泌表型)导致慢性炎症,最终可能导致组织损伤。
在自然衰老过程中,皮肤不断积累衰老细胞,导致其外观和纹理恶化,进而引起诸如皮肤松弛和皱纹的美容问题。此外,伤口和抓伤等损伤在熟龄肌肤中的愈合速度明显慢于年轻肌肤。为了保持年轻肌肤的外观和纹理,需要干预甚至更为理想地逆转皮肤中细胞衰老的影响。
二肽已被记录用于皮肤美容。WO2017/162879 A1涉及用于皮肤护理和美容用途的β-天冬氨酰二肽。在皮肤模型中,显示二肽增加基底角质形成细胞的繁殖,并且发现大量的β-天冬氨酰二肽可改善成年志愿者的皮肤弹性。然而,在体内试验中没有显示志愿者的年龄,并且使用的β-天冬氨酰二肽的量非常大(5%水溶液),因此经济上不可行。
美国专利US 5478560 A披露L-精氨酸-L天冬氨酸能够保持皮肤水分并加速细胞增殖。
发明内容
本发明的一个目的是为熟龄肌肤提供美容治疗,干预甚至逆转引起衰老过程中皮肤中衰老细胞增加的效应以及上述相关的美容问题。
特别地,本发明的一个目的是提供一种用于熟龄肌肤的美容治疗,其减轻皮肤细胞中氧化应激的影响,特别是线粒体功能的丧失。
本发明的另一个目的是提供一种处理熟龄肌肤的美容方法,减少皮肤中衰老细胞的数量。
本发明的另一个目的是提供一种皮肤的抗炎治疗,特别是减少或防止衰老的皮肤细胞分泌促炎因子的治疗。
本发明的另一个目的是提供一种针对受损熟龄肌肤的治疗,特别是一种促进伤口愈合的治疗。
本发明涉及用作一种皮肤抗炎剂的β-L-天冬氨酰-L-精氨酸,特别用于防止或减少皮肤细胞、优选地为衰老的皮肤细胞分泌一种或多种促炎细胞因子和/或生长因子。
在本发明中令人惊讶地发现,当二肽局部施用于皮肤时,β-L-天冬氨酰-L-精氨酸能够显著减少皮肤细胞,特别是衰老的皮肤细胞分泌促炎细胞因子和/或生长因子。
优选地,所述促炎细胞因子和/或生长因子选自由白细胞介素6(IL-6)、白细胞介素8(IL-8)、血管内皮生长因子(VEGF)和转化生长因子(TGF)组成的组。
在一个实施例中,β-L-天冬氨酰-L-精氨酸用于一种治疗和/或预防皮肤慢性炎症的方法,优选为由衰老的皮肤细胞分泌促炎细胞因子和/或生长因子引起的慢性炎症。
本发明还涉及β-L-天冬氨酰-L-精氨酸对熟龄肌肤的美容用途,用于发挥抗氧化作用和/或改善衰老的皮肤细胞特别是衰老的成纤维细胞中的线粒体功能,和/或恢复皮肤的紧致度。
根据本发明中的上下文发现,β-L-天冬氨酰-L-精氨酸发挥减少熟龄肌肤中衰老细胞的数量的作用。因此二肽对熟龄肌肤具有显著的美容治疗效果。还观察到很强的抗氧化作用,其保护皮肤细胞免受氧化损伤,恢复氧化还原平衡以改善线粒体功能。亦可预见,所观察到的效果可适用于头发护理,特别通过对毛囊的再生作用防止脱发和头发变白。
β-L-天冬氨酰-L-精氨酸可以通过化学合成或酶降解从蓝藻中获得的藻青素来获得。蓝藻在丰富的环境条件下合成藻青素颗粒肽(CGP),用于暂时储存营养和能量。当环境条件变得恶劣,营养物质匮乏时,藻类利用储存在CGP生物聚合物中的能量和营养物质,将它们分解成二肽和游离氨基酸,确保自身的生存。绿色化学和生物技术(微生物发酵)可用于提供海洋衍生二肽。有利地,该产品不含转基因生物(GMO),而且可以通过白色生物技术生产高纯度的该产品。作为一种白色无味的粉末,该产品具有水溶性,且在高达50℃的酸碱度为5-9条件下稳定,符合化妆品的安全和纯度要求。WO2009/150252 A1详细描述了使用肽酶(CGPase)水解CGP来制备二肽的方法。
在本发明的上下文中,术语“熟龄肌肤”是指包含大量衰老细胞的皮肤。特别地,熟龄肌肤是至少40岁、至少45岁、至少50岁、至少55岁、至少60岁、至少65岁或至少70岁的个体的皮肤。
在上述用途的优选实施例中,相应地,所述熟龄肌肤是至少40岁、至少45岁、至少50岁、至少55岁、至少60岁、至少65岁或至少70岁的个体的皮肤。
本发明上下文中的“抗氧化作用”是一种抑制氧化反应,诸如细胞环境中蛋白质羰基化的作用。氧化反应可以由例如紫外线辐射诱发,并且可以导致自由基的形成。自由基会产生连锁反应,对细胞,特别是对DNA造成严重损害,引发修复机制和氧化应激反应。最终导致细胞中活性氧(ROS)的数量增加。
如上所述,线粒体从细胞中吸收营养,将其分解并转化为能量(ATP)。在功能性线粒体中,氧的摄取、ATP的产生和活性氧(ROS)的产生受到严格控制,以维持氧化还原平衡。因此,“线粒体功能改善”是指改善或恢复细胞中的能量产生和氧化还原平衡的效果。
皮肤具有的塑性和弹性成分,其共同赋予皮肤粘弹性特征。当力作用在皮肤上时,皮肤不会立即恢复到其原始状态,而是最初保持轻微变形(滞后)的状态。在熟龄肌肤中,塑性势头在变形中起着较大的作用,不同皮肤区域表现出不同的塑性和弹性。测量方法依赖于吸力。皮肤弹性测试仪的测量头会产生真空,将皮肤吸入测量设备。由光源和光检测器组成的光学装置测量光强度,光强度取决于吸入的皮肤量。测量得到弹性和皮肤紧致度参数。
本发明上下文中的“皮肤紧致度”代表皮肤对真空吸力产生的阻力。另一方面,弹性是皮肤恢复原状所需的时间。
在本发明的上下文中,“衰老细胞”是由于损伤和氧化应激而进入生长停滞和功能变异状态的细胞。特别地,衰老细胞不再分裂,但仍有代谢活性。在衰老过程中,组织中衰老细胞的数量显著增加。上文已列出了衰老细胞的更多特征,下文中将进一步说明。
衰老细胞中巨大的功能失调线粒体是由于线粒体ATP缺乏补偿策略,线粒体尺寸可以作为评估细胞衰老变化的定量方法。衰老细胞中染色质的紊乱导致细胞核增大。因此,细胞核与细胞的尺寸比是衰老的另一个有效标志(et al.,Redox Biology,2017,13;550-567;Chandra et al.,Cell Reports,2015;10(4):471-483)。
线粒体含有仅从母体遗传而来的线粒体DNA(mtDNA)。线粒体DNA是一种在线粒体内发现的小型环状染色体。线粒体DNA的主要功能是指导参与氧化磷酸化的酶和膜结构的产生。在衰老细胞中,由于补偿机制,mtDNA数量增加,可以使用荧光标记可视化,并以细胞体中荧光点的密度(mtDNA含量)或mtDNA占据的细胞体积百分比(线粒体质量)进行测量。
纤连蛋白是将细胞与细胞外基质中的胶原纤维连接起来的糖蛋白,允许细胞在基质中移动。纤连蛋白对伤口愈合有很大影响,包括在肉芽组织的发育和组织过程中为细胞的迁移和生长形成合适的基质,以及结缔组织基质的重塑和合成。因此,纤连蛋白也对肌理产生积极影响,特别是皮肤的紧致度。
细胞以未折叠、非活性的形式分泌纤连蛋白。纤连蛋白与整合素结合会触发纤连蛋白分子的构象变化,使其形成二聚物。所述二聚物结合胶原蛋白和细胞表面整合素,引起细胞骨架的重组以促进细胞运动(Freeman and Hamilton;Pearson Prentice Hall,2005)。
III型胶原蛋白是皮肤的重要结构成分,因此对保持肌理,特别是皮肤的紧致度很重要。此外,III型胶原蛋白还参与血液凝结。在伤口愈合的后期,细胞外基质的重建是一个重要的过程,III型胶原蛋白在其中发挥作用。
在上述美容用途的优选实施例中,β-L-天冬氨酰-L-精氨酸实现一种或多种选自由下述效果组成的组的效果:
a)避免衰老的皮肤细胞遭受线粒体活性氧(ROS)诱导的损伤;
(b)避免真皮中受UVA诱导的活性氧(ROS);
(c)防止衰老的皮肤细胞中紫外线诱导的氧化,特别是蛋白质羰基化;
(d)减少衰老的皮肤细胞中的线粒体活性氧(ROS);
(e)减少衰老的皮肤细胞中的线粒体DNA(mtDNA);
(f)减小衰老的皮肤细胞的细胞尺寸;
(g)减少衰老的皮肤细胞数量;
(h)降低衰老的皮肤细胞中细胞核与细胞的尺寸比;
(i)刺激衰老的皮肤细胞中产生III型胶原蛋白;以及
(j)刺激衰老的皮肤细胞中产生纤连蛋白。
优选地,在衰老的成纤维细胞中获得上述(a)至(j)的效果。在上述美容用途的优选实施例中,所述衰老的皮肤细胞是衰老的成纤维细胞。
在下面的实施例中已经证明了(a)至(j)的效果在衰老细胞中显著,因此借助二肽可对熟龄肌肤进行有效的特别美容治疗。
在上述美容用途的一个实施例中,将β-L-天冬氨酰-L-精氨酸局部施用于皮肤。
已经证明,通过在皮肤上局部施用β-L-天冬氨酰-L-精氨酸可以获得上述效果。所述二肽可以应用于例如化妆品制剂,如面霜、喷雾、乳液、软膏或凝胶,其包含一种或多种化妆品可用的载体和赋形剂。技术人员知晓如何制备这种化妆品制剂。
在上述美容用途的另一个实施例中,β-L-天冬氨酰-L-精氨酸用于一种化妆品组合物中,在每种情况下相对于该组合物的总重量,所述化妆品组合物包含0.001wt%-2wt%,优选地为0.05wt%-0.5wt%,特别优选地为0.1wt%-0.3wt%的β-L-天冬氨酰-L-精氨酸。
上述规定的剂量通常用于化妆品制剂中,并提供所需的效果。有利地,所述二肽的使用量不必很高,因此避免生产成本的增加。
本发明还涉及一种用于处理熟龄肌肤的美容方法,包括以下步骤:

如上所述,并经以下实施例的证明,二肽在熟龄肌肤上的应用产生了许多有益效果。
在上述美容方法的一个实施例中,β-L-天冬氨酰-L-精氨酸发挥抗氧化作用和/或改善衰老的皮肤细胞特别是衰老的成纤维细胞中的线粒体功能,和/或恢复皮肤的紧致度,特别地,β-L-天冬氨酰-L-精氨酸实现一种或多种选自由下述效果组成的组的效果:
a)避免衰老的皮肤细胞遭受线粒体活性氧(ROS)诱导的损伤;
(b)避免真皮中受UVA诱导的活性氧(ROS);
(c)防止衰老的皮肤细胞中紫外线诱导的氧化,特别是蛋白质羰基化;
(d)减少衰老的皮肤细胞中的线粒体活性氧(ROS);
(e)减少衰老的皮肤细胞中的线粒体DNA(mtDNA);
(f)减小衰老的皮肤细胞的细胞尺寸;

优选地,在衰老的成纤维细胞中获得上述(a)至(j)的效果。在上述美容方法的优选实施例中,所述衰老的皮肤细胞是衰老的成纤维细胞。
由于已经证明了上述效果在含有大量衰老细胞的熟龄肌肤中显著,在上述美容方法中,所述熟龄肌肤是至少40岁、至少45岁、至少50岁、至少55岁、至少60岁、至少65岁或至少70岁的个体的皮肤。

如下例所示,将β-L-天冬氨酰-L-精氨酸应用于皮肤,可刺激纤连蛋白和III型胶原蛋白的产生,从而加速伤口愈合。在一个优选的实施例中,β-L-天冬氨酰-L-精氨酸可促进伤口愈合。

具体实施方式
实施例1:衰老的成纤维细胞中线粒体ROS的减少(体外研究)
用荧光染料量化β-L-天冬氨酰-L-精氨酸对线粒体活性氧(ROS)的抗氧化能力。用β-L-天冬氨酰-L-精氨酸孵育衰老细胞24小时。随后,用荧光染料将处理过的和未经处理的衰老细胞以及处理过的和未经处理的非衰老细胞在37℃下孵育30分钟。洗涤后,用氧化参考物质H2O2(100μM)处理部分非衰老细胞。在60分钟内,每5分钟立刻测量线粒体ROS的产生量。微量滴定板读数器(Variskan-Thermo)用作测量装置。在动力学测量过程中,在37℃下进一步孵育细胞。使用荧光法,平行测量抗氧化活性与细胞活力。实验包括空白、阴性和阳性对照。阳性对照由非衰老细胞组成,这些细胞仅用媒介物(DMSO)处理。将这些细胞与未经处理的衰老细胞(阴性对照)进行比较。以用白藜芦醇处理的衰老细胞为参照,比较β-L-天冬氨酰-L-精氨酸在降低衰老细胞线粒体ROS内源性生成中的活性。β-L-天冬氨酰-L-精氨酸以四重测定法进行测定。同时将β-L-天冬氨酰-L-精氨酸的效果与阴性和阳性对照进行比较。
从图1可以推断,参考未经处理的衰老细胞,用10μMβ-L-天冬氨酰-L-精氨酸处理的细胞显示线粒体(mtROS)的显著降低,与用白藜芦醇达到的效果相当。众所周知,白藜芦醇能够减少ROS,并被用作阳性对照。图中示出相对于代表100%的未经处理复制细胞的数值。
在图2中,示出了mtROS减少效应的剂量依赖性。增加β-L-天冬氨酰-L-精氨酸的用量显示出mtROS大幅度的减少。在该图中,示出了相对于代表100%的作为阴性对照的未经处理衰老细胞的数值。
实施例2:衰老的成纤维细胞中细胞尺寸的减小(体外研究)

实施例4:衰老的成纤维细胞中线粒体DNA(mtDNA)的减少(体外研究)
通过检测细胞核片段、线粒体DNA含量、线粒体质量和平均细胞密度,分析、研究了细胞核和线粒体DNA的破坏,后者代表活性。
用β-L-天冬氨酰-L-精氨酸处理细胞5天。孵育后拍摄细胞图像。为了标记细胞核和线粒体DNA,该细胞用荧光染料孵育30分钟。去除染料后,用荧光显微镜拍摄图像。图像分析以三重测定法进行。每个实验条件或每次重复分别取样10张图片。
数据收集:图像用蔡司axioplan显微镜拍摄。用国际衍射数据中心(ICDD)开发的软件进行图像分析。
数据解读:收集下述数据:平均细胞尺寸、mtDNA含量、线粒体密度、线粒体生物发生、线粒体质量和细胞核尺寸。每个区域的平均细胞数用作每个实验条件下细胞活性的对照。与未经处理的细胞相比,活性物质的作用也被认为是保护作用。纳入阳性对照的效果用作比较。图5示出了衰老细胞中的mtDNA被β-L-天冬氨酰-L-精氨酸降低到与年轻复制性细胞相当的量。
图5示出了衰老细胞中的mtDNA被β-L-天冬氨酰-L-精氨酸25降低到与年轻复制性细胞相当的量。参考化合物叠氮胸苷(AZT)是一种γ聚合酶抑制剂,减少mtDNA量。
实施例5:人体皮肤外植体经UVA照射后ROS水平的降低(离体研究)
皮肤模型的培养:将皮肤切成约8mm×3mm的大块(模型的直径×层厚),并培养至计划的实验结束。每次处理使用六个模型。该皮肤模型作为气-液界面培养物在穿孔不锈钢环中培养,与培养基(改良的Williams`E培养基)接触至期望的终点。
处理:局部施用试验物质和对照物质。为此,将该皮肤模型用棉垫仔细清洁。随后,将每种测试物质或对照物分别按4μL的量施用到模型上,并用膜滤器(直径为6毫米)覆盖。
该皮肤模型分别用测试物质和阳性对照物孵育18小时。随后,将检测试剂(例如DCFH-DA)添加到培养基中,类似于Marionnet等人(Plos One,9,2014)的描述。再过30分钟后,去除DCFH-DA试剂并进行氧化刺激(例如UVA)。二氯荧光素二乙酸酯与ROS反应形成荧光产物。
紫外线照射:使用的太阳模拟器是Vilber Lourmat公司的BIO-SUN系统。该系统基于可编程微处理器,其控制紫外线照射。紫外光发射始终处于监控下,当达到预设能量时,该系统自动停止运行。因此,照射周期总是可重复的,与紫外线灯的强度损失无关。为了确定紫外线照射剂量,使用了“生物有效剂量(BED)”,参考Del Bino等人(Pigment CellResearch,19,2006)的相关描述。
ROS的测定:实验结束时,采收所述皮肤模型,低温保存,并使用低温恒温器切割,用于随后的图像采集和分析。荧光分析在真皮区域进行。用于分析的区域选择从真皮的上部,沿着基底层延伸到真皮的下部,同时确保避免血管、汗腺或毛囊等不规则结构。
在每个皮肤模型中,两个样本用于荧光分析的图像采集。通过使用Image-J分析软件(美国国立卫生研究院)测定荧光来分析每张图片。测量值按照选定表面的尺寸标准化。
统计:对于所述皮肤模型的每次处理,确定定量数据的平均值。为了计算变异,即均值标准偏差和标准误差(SEM),使用了原始数据。经处理的皮肤模型之间的差异通过排列检验的单向方差分析以及随后的进行排列的Turkey检验和T检验来计算。
从图6可以推断,β-L-天冬氨酰-L-精氨酸是真皮中非常有效的紫外线诱导的ROS清除剂,与未用β-L-天冬氨酰-L-精氨酸处理的经紫外线照射皮肤相比,显著降低了经紫外线照射皮肤的ROS水平。因此,β-L-天冬氨酰-L-精氨酸强效保护避免真皮内UVA诱导的ROS损伤。
实施例6:真皮中刺激纤连蛋白(离体研究)
I型胶原蛋白、III型胶原蛋白、基质金属蛋白酶1、弹性蛋白、原纤维蛋白和纤连蛋白
在37℃、5%CO2和100%湿度是条件下,在6天内每天在皮肤上施用4μl的β-L-天冬氨酰-L-精氨酸。用选定的抗体对12个皮肤切口进行免疫组织化学染色。选择乳头状真皮进行分析,因为它是真皮中对之前处理的反应中变化最大的部分。
在确保避开诸如血管、汗腺或毛囊的不规则结构的同时,仔细选择要分析的区域。
为了评估结果,使用Image-J分析软件(美国国立卫生研究院)测量了颜色强度和颜色分布,因此可以进行半定量评估。
图7示出了与未经处理的皮肤相比,β-L-天冬氨酰-L-精氨酸能够刺激真皮中纤连蛋白的产生。因此,β-L-天冬氨酰-L-精氨酸能够加速皮肤更新和伤口愈合。
图12示出了与未经处理的皮肤相比,β-L-天冬氨酰-L-精氨酸能够刺激III型胶原蛋白的产生。因此,β-L-天冬氨酰-L-精氨酸能够加速皮肤更新和伤口愈合。
实施例7:减少皱纹深度(临床研究)
该研究对26名年龄在53+/-3岁的女性志愿者进行。在脸的一侧涂抹产品,另一侧涂抹安慰剂,每天两次,持续两个月。
拍照:评估基于受控条件下的高分辨率数字摄影。在研究开始时、28天后和产品应用56天后拍摄照片。
受试者被要求位于光学装置的前面,拍摄其面部肖像或特写。受试者坐于灯光受控的环境中进行拍摄。拍摄条件保持不变,以确保可比性。
减少皱纹和线条:在受试者眼睛周围的有限区域进行皱纹和线条的评估。皱纹和线条的深度被测量为参数R。R的值越大,皱纹和线条越明显。皱纹的相对减少量根据以下公式计算:
R%=100(Ri-Rf)/Ri(R=皱纹深度,i=初始;f=最终)
数据分析和解读:数据分析使用了以下软件包:
从图8可以推断,每天两次使用0.1%的β-L-天冬氨酰-L-精氨酸,仅4周就能明显抚平鱼尾纹和细纹。八周后,与安慰剂相比,观察到85%的志愿者皱纹深度显著降低了6.8%。
实施例8:防止蛋白质羰基化(离体研究)
羰基化检测:去除脂肪和结缔组织后,称重并切割冷冻所述皮肤样品。将测试样品的皮肤碎片在冷提取缓冲液中混合并均质化。通过离心收集细胞裂解物。
根据生产商的方案,使用“密理博公司的OxyBlot Protein_Oxidation试剂盒”(#S7150)进行羰基化蛋白质的测量。计算中只考虑了尺寸在50kDa到150kDa之间的蛋白条带。获得的羰基化蛋白质的量通过肌动蛋白带(恒定)归一化,并相对于未经处理的样品给出。
从图9可以推断,比较未经处理的经紫外线刺激的皮肤和用0.2%和0.5%β-L-天冬氨酰-L-精氨酸处理的经紫外线刺激的皮肤,β-L-天冬氨酰-L-精氨酸在蛋白质水平上是一种非常有效的抗氧化剂,并且能够在离体模型中防止羰基化蛋白质的形成。阳性对照用具有抗氧化作用的维生素C和维生素B处理。
实施例9:皮肤紧致度和弹性的改善(临床研究)
该研究对26名年龄在53+/-3岁的女性志愿者进行。在脸的一侧涂抹产品,另一侧涂抹安慰剂,每天两次,持续两个月。
测量方法依赖于吸力。皮肤弹性测定仪的测量头会产生真空,将皮肤吸入测量设备。由光源和光检测器组成的光学装置测量光强度,光强度取决于吸入的皮肤量。所得的参数是弹性和皮肤紧致度。
皮肤紧致度代表皮肤对真空吸力产生的阻力。弹性是皮肤恢复原状所需的时间。
图10示出了在每天两次使用0.1%的β-L-天冬氨酰-L-精氨酸四周后,皮肤紧致度明显的改善效果。八周后,与安慰剂相比,观察到92%的志愿者皮肤紧致度显著增加(+10.1%)。
图11示出了在每天两次使用0.1%的β-L-天冬氨酰-L-精氨酸四周后,皮肤弹性的明显改善效果。八周后,与安慰剂相比,观察到100%的志愿者皮肤弹性显著增加(+8.5%)。
Claims (15)
1.一种用作皮肤抗炎剂的β-L-天冬氨酰-L-精氨酸,特别用于防止或减少皮肤细胞、优选地为衰老的皮肤细胞分泌一种或多种促炎细胞因子和/或生长因子。
2.根据权利要求1所述的β-L-天冬氨酰-L-精氨酸,其中,所述促炎细胞因子和/或生长因子选自由白细胞介素6(IL-6)、白细胞介素8(IL-8)、血管内皮生长因子(VEGF)和转化生长因子(TGF)组成的组。
3.根据权利要求1或2所述的β-L-天冬氨酰-L-精氨酸,其中,所述β-L-天冬氨酰-L-精氨酸用于一种治疗和/或预防皮肤慢性炎症的方法,优选为由衰老的皮肤细胞分泌促炎细胞因子和/或生长因子引起的慢性炎症。
4.一种β-L-天冬氨酰-L-精氨酸对熟龄肌肤的美容用途,用于发挥抗氧化作用和/或改善衰老的皮肤细胞特别是衰老的成纤维细胞中的线粒体功能,和/或恢复皮肤的紧致度。
5.根据权利要求4所述的美容用途,其中,所述熟龄肌肤是至少40岁、至少45岁、至少50岁、至少55岁、至少60岁、至少65岁或至少70岁的个体的皮肤。
6.根据权利要求4或5所述的美容用途,其中,所述β-L-天冬氨酰-L-精氨酸实现一种或多种选自由下述效果组成的组的效果:
a)避免衰老的皮肤细胞遭受线粒体活性氧(ROS)诱导的损伤;
(b)避免真皮中受UVA诱导的活性氧(ROS);
(c)防止衰老的皮肤细胞中紫外线诱导的氧化,特别是蛋白质羰基化;
(d)减少衰老的皮肤细胞中的线粒体活性氧(ROS);
(e)减少衰老的皮肤细胞中的线粒体DNA(mtDNA);
(f)减小衰老的皮肤细胞的细胞尺寸;
(g)减少衰老的皮肤细胞数量;
(h)降低衰老的皮肤细胞中细胞核与细胞的尺寸比;
(i)刺激衰老的皮肤细胞中产生III型胶原蛋白;以及
(j)刺激衰老的皮肤细胞中产生纤连蛋白。
7.根据权利要求4-6中任一项所述的美容用途,其中,将所述β-L-天冬氨酰-L-精氨酸局部施用于所述皮肤。
8.根据权利要求4-7中任一项所述的美容用途,其中,所述β-L-天冬氨酰-L-精氨酸用于一种化妆品组合物中,在每种情况下相对于所述组合物的总重量,所述化妆品组合物包含0.001wt%-2wt%,优选地为0.05wt%-0.5wt%,特别优选地为0.1wt%-0.3wt%的β-L-天冬氨酰-L-精氨酸。
9.一种用于处理熟龄肌肤的美容方法,包括以下步骤:
(i)将β-L-天冬氨酰-L-精氨酸局部施用于熟龄肌肤上。
10.根据权利要求9所述的美容方法,其中,所述β-L-天冬氨酰-L-精氨酸发挥抗氧化作用和/或改善衰老的皮肤细胞特别是衰老的成纤维细胞中的线粒体功能,和/或恢复皮肤的紧致度,特别地,其中,所述β-L-天冬氨酰-L-精氨酸实现一种或多种选自由下述效果组成的组的效果:
a)避免衰老的皮肤细胞遭受线粒体活性氧(ROS)诱导的损伤;
(b)避免真皮中受UVA诱导的活性氧(ROS);
(c)防止衰老的皮肤细胞中紫外线诱导的氧化,特别是羰基化;
(d)减少衰老的皮肤细胞中的线粒体活性氧(ROS);
(e)减少衰老的皮肤细胞中的线粒体DNA(mtDNA);
(f)减小衰老的皮肤细胞的细胞尺寸;
(g)减少衰老的皮肤细胞数量;
(h)降低衰老的皮肤细胞中细胞核与细胞的尺寸比;
(i)刺激衰老的皮肤细胞中产生III型胶原蛋白;以及
(j)刺激衰老的皮肤细胞中产生纤连蛋白。
11.根据权利要求9或10所述的美容方法,其中,所述熟龄肌肤是至少40岁、至少45岁、至少50岁、至少55岁、至少60岁、至少65岁或至少70岁的个体的皮肤。
12.根据权利要求9-11中任一项所述的美容方法,其中,所述β-L-天冬氨酰-L-精氨酸用于一种化妆品组合物中,在每种情况下相对于所述组合物的总重量,所述化妆品组合物包含0.001wt%-2wt%,优选地为0.05wt%-0.5wt%,特别优选地为0.1wt%-0.3wt%的β-L-天冬氨酰-L-精氨酸。
13.一种用于治疗受损熟龄肌肤的治疗方法中的β-L-天冬氨酰-L-精氨酸。
