本发明属于微生物领域,公开了一种可表达乳糖酶的弧菌属菌株,该菌株为弧菌属菌株(Vibrio sp),菌株编号BHNC007003,保藏日期为2021年10月11日,保藏单位为中国典型培养物保藏中心(CCTCC),地址为中国.武汉.武汉大学,保藏编号为CCTCC NO:M 20211248。本发明的弧菌属菌株(Vibrio sp)从海水中筛选获得。所筛选获得的弧菌属菌株(Vibrio sp)所产的乳糖酶对酶活性适应温度和pH范围较广,在工业生产中有较大的优势。
Description
技术领域
本发明涉及微生物领域,具体讲是一种可表达乳糖酶的弧菌属菌株。
背景技术
弧菌属菌株(Vibrio sp)是不产孢子的短杆菌,形状短小,约0.5×(1~5)微米,因弯曲如弧而得名。分散排列,偶尔互相连接成S状或螺旋状。革兰氏染色阴性,菌体一端有单鞭毛,运动活泼。无芽胞,无荚膜。是一种化能有机营养菌,既是呼吸型又是发酵型代谢。碳水化合物类的代谢发酵有混杂的产物而无CO2或H2。见于淡水和海水,以及人或动物的消化道中。
而其所产的乳糖酶的工业生产方法主要是微生物发酵法,目前只有来源于微生物的乳糖酶有工业应用价值,在工业上主要用于乳制品工业。利用其水解作用分解乳制品中的乳糖,生产低乳糖或无乳糖乳制品,供乳糖不耐受的广大人群食用。而不同微生物来源的乳糖酶的性质差别较大。这些不同来源的乳糖酶分解乳糖的活性受pH值和温度的影响。因此筛选出一种能表达出温度范围广、拥有高活性的乳糖酶的菌株对工业化生产具有重大意义。
发明内容
本发明所要解决的技术问题是,克服以上现有技术的缺点:提供一种能产出适应pH值及温度范围广,高活性的乳糖酶的弧菌属菌株。
本发明的技术解决方案如下:一种可表达乳糖酶的弧菌属菌株,该菌株为弧菌属菌株(Vibrio sp),菌株编号BHNC007003,保藏日期为2021年10月11日,保藏单位为中国典型培养物保藏中心(CCTCC),地址为中国.武汉.武汉大学,保藏编号为CCTCC NO:M20211248。
其表达乳糖酶酶活力适应温度范围为25-45℃。
其表达乳糖酶最适的酶反应温度为33℃。
其表达乳糖酶酶活力适应pH值范围为4.0-8.0
其表达乳糖酶在pH 6.0时的酶活力最高。
其核苷酸测序序列如SEQ ID NO:1所示。
本发明的有益效果是:本发明的弧菌属菌株(Vibrio sp)从海水中筛选获得。所筛选获得的弧菌属菌株(Vibrio sp)所产的乳糖酶对酶活性适应温度和pH范围较广,在工业生产中有较大的优势。
本发明的弧菌属菌株(Vibrio sp)保藏日期2021年10月11日、保藏单位中国典型培养物保藏中心(CCTCC)和保藏编号CCTCC NO:M 20211248。

具体实施方式
下面用具体实施例对本发明做进一步详细说明,但本发明不仅局限于以下具体实施例。
实施例1
一、菌株培养、鉴定及构建系统发育树
1、菌株培养
LB培养基:氯化钠10g/L,蛋白胨10g/L,酵母浸粉5g/L,琼脂20g/L,纯水配制,添加20mg/ml X-gal。
YPD培养基(选择性培养基):蛋白胨20g/L,酵母浸粉10g/L,葡萄糖20g/L,琼脂20g/L,纯水配制,添加20mg/ml X-gal。
LB深孔板:氯化钠10g/L,蛋白胨10g/L,酵母浸粉5g/L,纯水配制。
采样:从海南采集一些海水和沙土样本,并将采集到样本逐一进行培养,分别涂布于LB和YPD平板培养基中,对应的平板分别编号,贴上封口膜,将平板置于30℃恒温倒置培养48h,之后平板上挑出了显蓝色单菌落,将挑出的单菌落分别接到LB摇瓶中,置于震荡培养箱培养,培养条件为30℃,避光,转速为245r/min。培养24h后,取样,添加50%甘油保存于-80℃冰箱。分别取培养24h,48h,72h的菌液,测定酶活。
、菌株鉴定
对该菌株进行16S rDNA的PCR扩增鉴定,具体步骤如下:
1)PCR扩增:通用引物515F和806R进行PCR扩增,其中,
引物515F序列为:5′- GTGYCAGCMGCCGCGGTAA -3′;
引物806R序列为:5′- GGACTACNVGGGTWTCTAAT -3′。
PCR反应体系为:25×TAQ酶5μL,515F引物2μL,806R引物2μL,菌液模板5μL,无菌水16μL。PCR扩增程序为:94℃预变性5min;94变性30s,49℃退火60s,72℃延伸60s,30个循环;72℃延伸5min,4℃终止反应。
2)凝胶电泳及PCR产物柱回收:PCR产物经1.5%琼脂糖凝胶电泳鉴定。利用快捷型琼脂糖凝胶DNA回收试剂盒(北京全式金生物技术有限公司)对目的PCR产物进行回收纯化,进行16S rRNA测序。
对所测得的16S rDNA,运用NCBI中的Blast 程序与数据库中的细菌16S rDNA序列进行相似性比对。
比对结果:该菌株BHNC007003为弧菌属菌株(Vibrio sp)的菌株。
、构建菌株BHNC007003的系统发育树
获得菌株BHNC007003的16S rDNA后,用MEGA 4软件(Neighbor-Joining)构建该菌株的系统发育树(见图1),分析各菌株的进化关系。
分析结果:菌株BHNC007003为弧菌属菌株(Vibrio sp)的菌株。
二、菌株保藏
本发明筛选得到的弧菌属菌株,分类命名为弧菌属菌株(Vibrio sp),菌株编号BHNC007003,其保藏编号为CCTCC NO:M 20211248,保藏日期为2021年10月11日,保藏单位为中国典型培养物保藏中心(CCTCC),地址为中国.武汉.武汉大学。
三、关于乳糖酶酶活力的最适温度
酶活力单位的定义:一个酶活单位(NLU)定义为在反应条件下释放1.30 μmol/min的邻硝基苯酚所需的酶量。
酶活测定方法:邻硝基苯酚β-D半乳糖苷(ONPG)溶于P-E-M缓冲液中(pH值为6.5)配制成质量分数为0.25%的底物溶液。取5ml底物溶液加入1ml酶液中,30℃保温10min。加入2mL碳酸钠溶液显色,测定420nm光吸收值。计算水解产物邻硝基酚(ONPG)的含量和酶活性。
在pH值为5.5条件下,分别在不同温度(20℃、25℃、30℃、33℃、35℃、37℃、40℃、50℃、60℃)下,测定酶活力,测定的相对酶活力变化曲线如图2所示,此乳糖酶的适应温度范围较广,在25-45℃之间相对酶活都在75%以上,最适的酶反应温度为33℃。
可见,本发明筛选得到的菌株BHNC007003其所产的乳糖酶酶具有较广的适应温度范围,菌株BHNC007003工业中具有一定的优势。
四、关于乳糖酶酶活力的最适PH.
取酶液分别用pH为(4.0、4.5、5.0、5.5、6.0、6.5、6.8、 7.0、7.2、7.5、8.0)的缓冲液稀释,测定酶活力,相对酶活力变化曲线如图3所示,乳糖酶在pH 6.0时的酶活力最高,pH适用范围为4.0-8.0。
