1. Tyrosinase activity inhibition assay: firstly, preparing a reagent, specifically as follows:
1) Sodium phosphate buffer (1/15 mol/L, pH = 6.8)
Accurately weighing 1.000g of sodium dihydrogen phosphate and 1.186g of disodium hydrogen phosphate, adding a small amount of deionized water to dissolve, fixing the volume to 500mL, and storing in a refrigerator at 4 ℃ for later use;
2) L-tyrosine solution (7.5 mmol/L)
Accurately weighing 0.272g of L-tyrosine, firstly adding 3-4 drops of concentrated hydrochloric acid, adding about 50mL of deionized water, after the L-tyrosine is completely dissolved by slight heating, adjusting the pH value to about 7 by using a sodium hydroxide solution, and adding deionized water to a constant volume of 200mL;
3) Test solution
Each 0.1g of the compositions of examples 1 to 16 and comparative examples 1 to 5 was accurately weighed, dissolved in 20mL of deionized water to give 5mg/mL of test solutions, and then diluted in duplicate to 2.5mg/mL, 1.25mg/mL, 0.625mg/mL, and 0.3125mg/mL. Table 3 in examples 1 to 13 and comparative examples 1 to 5, each test solution had a concentration of 0.5mg/mL;
4) Positive control (+ CK)
Accurately weighing 0.1g of arbutin powder, dissolving in 20mL of deionized water to obtain 5mg/mL of positive control mother liquor, and diluting to 2.5mg/mL, 1.25mg/mL, 0.625mg/mL and 0.3125mg/mL;
5) The preparation of the tyrosinase liquid uses fresh and intact potatoes to prepare the tyrosinase liquid, and the specific operations are as follows: cleaning potato, pre-cooling at 4 deg.C for about 4 hr, peeling, and cutting into 1.0cm pieces 3 D-form, and freeze at-20 ℃ overnight. Weighing, adding a 4 ℃ precooled sodium phosphate buffer solution according to the mass and volume ratio of 1The obtained tyrosinase crude enzyme solution is stored at 4 ℃ and is used up after 2 hours;
the test is carried out, the total reaction system of each group of tests is 5mL, and the specific design is shown in Table 1:
TABLE 1 5mL test System design
In this system, the final concentration (mg/mL) gradient of the test solution, the positive control mother solution, and arbutin was 0.03125, 0.0625, 0.125, 0.25, and 0.5.
During the experiment, phosphate buffer solution, test solutions of each group, positive control mother solution and enzyme solution are sequentially added into a test tube, and water bath is carried out at 30 ℃ for 10min. The substrate L-tyrosine was then added and the timer was started immediately. The absorbance of each test solution was measured at a wavelength of 475nm at 20min of reaction. During the determination, the inhibition rate of each group of test solution (including negative control) on tyrosinase is calculated by using the following formula with the corresponding negative control as a reference.
Inhibition = [ (A-B)/A ]. Times.100%
Wherein "A" is the absorbance of the standard control and "B" is the absorbance of the test solution (or positive control). Each experiment was done in 3 replicates. The high inhibition rate indicates that the inhibition intensity of the compound on tyrosinase activity is high. Note: when the absorbance is measured by a spectrophotometer, the test solution, the standard control and the positive control are respectively zeroed by the negative control 1, the negative control 2 and the negative control 3.
2. DPPH-free radical scavenging test: the compositions prepared in examples 1 to 16 and comparative examples 1 to 5 were diluted with deionized water to give a test solution having a mass fraction of 1%. Taking 4mL of DPPH-absolute ethanol solution with the concentration of 0.1mmol/L into a container, adding 2mL of test sample, uniformly mixing, keeping out of the sun for 30min at room temperature, measuring the absorbance at the wavelength of 517nm, and recording the absorbance as C1; taking 4mL of absolute ethyl alcohol solution and 2mL of test sample solution, and recording the absorbance value as C2 under the same condition; the absorbance values of 4mL of the anhydrous ethanol solution and 2mL of the anhydrous ethanol solution measured under the same conditions were recorded as C3. DPPH radical clearance was calculated as follows: DPPH radical clearance (%) = [1- (C1-C2)/C3 ] × 100%, averaged over three measurements. DPPH free radical is a stable nitrogen center organic free radical, strong absorption is achieved at 517nm, an ethanol solution of DPPH is purple, when the DPPH free radical scavenger is encountered, single electrons of DPPH are paired to enable the color of the solution to become light, the fading degree of the DPPH free radical is in quantitative relation with the quantity of the received electrons, therefore, a spectrophotometer can be used for carrying out quantitative analysis to detect the condition of scavenging the free radicals, and the higher the scavenging rate is, the stronger the oxidation resistance and the anti-aging capacity of the DPPH free radical is.

According to the analysis of the inventor, the composition contains the chamomile extract, the chamomile extract contains flavonoid compounds, amino acids and other components, can act together with the aloe extract, has a good moisturizing effect, regulates the water-oil balance of the skin to a certain extent, effectively replenishes moisture for the skin, and enables the skin to be in a relatively stable state, so that the growth difference of the composition in three time periods of 10min, 30min and 60min is obviously shortened; in addition, the chamomile extract has an anti-inflammatory effect, can reduce the generation of acnes, can resist oxidation and slow down skin aging.
Example 15, in which disodium EDTA was used as an adjuvant, the compositions prepared in example 15 exhibited slightly improved tyrosinase activity inhibition and DPPH-free radical scavenging as well as skin stratum corneum moisture levels as compared to the compositions prepared in example 2, indicating that the overall performance of the compositions prepared in example 15 was improved. The inventor analyzes that the disodium EDTA can play a role of a chelating agent in a composition system, so that the combination and compatibility of plant extracts, solvents and other components in the composition are improved, the components in the composition are uniformly matched, and the effect of the composition is improved better.
Example 16 the compositions prepared in example 16 have better improved properties than the compositions prepared in example 3 by combining and formulating the components and processes with beneficial effects in the above examples, and the compositions prepared in example 16 have better whitening and moisturizing effects through the combination of various substances in the composition system and the selection of more appropriate additives.
Comparative example 5 the bearberry leaf, the ruscus aculeatus and the glycyrrhiza glabra were all replaced with the same amount of water, the composition prepared in comparative example 5 did not have the bearberry leaf extract, the ruscus aculeatus extract and the glycyrrhiza glabra extract, and the composition prepared in comparative example 5 was inferior in each performance. In the comparative example 1, the Ruscus aculeatus and the Glycyrrhiza glabra are replaced by the same amount of water, the bearberry leaf extract is added in the composition of the comparative example 1 compared with the composition of the comparative example 5, the tyrosinase activity inhibition rate and the free radical clearance rate of the composition prepared in the comparative example 1 are improved compared with those of the composition of the comparative example 5, and the bearberry leaf extract is helpful for improving the whitening and anti-aging effects; the Ruscus aculeatus extract is more in the comparative example 2 than in the comparative example 5, and the tyrosinase activity inhibition rate and the free radical clearance rate of the composition prepared in the comparative example 2 are improved compared with those of the composition prepared in the comparative example 5, so that the Ruscus aculeatus extract is also beneficial to improving the whitening and anti-aging effects; comparative example 3 has more glycyrrhiza glabra extract than the composition of comparative example 5, and the composition prepared in comparative example 3 has improved tyrosinase activity inhibition rate and free radical scavenging rate compared with the composition of comparative example 5, which indicates that glycyrrhiza glabra extract is helpful for improving whitening anti-aging effect, but the improvement effect is not as good as that of bearberry leaf extract and Ruscus aculeatus extract.
The composition prepared in the comparative example 4 has a significantly weaker effect of improving the tyrosinase activity inhibition rate than the composition prepared in the comparative example 5 than the composition prepared in the comparative example 1 and the composition prepared in the comparative example 2, which shows that the combined use of the bearberry extract and the butcher's broom extract can generate a certain inhibition effect on the whitening effect of the composition; example 3 in comparison with comparative example 4, the glycyrrhiza glabra extract is added, and the tyrosinase activity inhibition rate and the increase amount of the skin moisture content of the composition prepared in example 3 are obviously improved in comparison with those of the composition prepared in comparative example 5, which shows that the addition of the glycyrrhiza glabra extract helps to relieve the inhibition effect between the bearberry leaf extract and the ruscus aculeatus extract, and further improves the moisturizing effect of the composition. The inventor analyzes that certain synergistic effect can be generated among the bearberry leaf extract, the Ruscus aculeatus extract and the Glycyrrhiza glabra extract, so that effective components playing a role in whitening in the bearberry leaf extract, the Ruscus aculeatus extract and the Glycyrrhiza glabra extract are converted, the inhibition problem is solved, and the moisturizing effect is further improved.
The above are preferred embodiments of the present application, and the scope of protection of the present application is not limited thereto, so: equivalent changes in structure, shape and principle of the present application shall be covered by the protection scope of the present application.
