Description
Technical Field
The invention belongs to the technical field of traditional Chinese medicine compositions, and particularly relates to a sleeping-aiding formula and sleeping essential oil.
Background
The data show that sleep dysfunction is present in about one third of all people worldwide, and sleep problems can have a large negative impact on the physiology and psychology of people. And as socio-economic development and competition become more intense, the number of patients with sleep disorders tends to increase every year. In order to solve the above problems, various medicines, health products, and daily products having functions of improving sleep quality and treating sleep disorders have been developed and applied in succession.
At present, methods for treating insomnia include drug therapy, muscle relaxation, hypnosis, suggestive therapy, behavioral therapy, and the like, and at present, drug therapy, i.e., administration of hypnotics or similar sedatives, is mainly used in medicine. Although hypnotics or similar sedatives have a sedative effect on the central nervous system of the brain, the drugs are short in duration, need to be taken for a long time, and cause side effects, and also cause toxic effects on the central nervous system after long-term administration, thereby causing morbid phenomena such as listlessness, difficult excitation, memory decline, inappetence and the like. In addition, the hypnotic enters the human body through oral administration, enters blood through gastric digestion, and then enters the brain through blood circulation to exert the drug effect, so that the whole process needs a long time and has great hysteresis. The traditional Chinese medicine mainly comprises plant extracts, can achieve the effect of improving sleep quality through traditional Chinese medicine therapy, does not cause medicine dependence, and does not cause iatrogenic diseases. The Chinese patent with publication number CN106729068A provides an external energy oil which can help sleep, the effect of improving sleep can be achieved only after the external energy oil is continuously used for a long time, and the improvement effect is not obvious. Therefore, the development of a sleep-aiding product which has small side effect and can quickly and effectively improve the sleep quality is urgently needed.

Example 3 efficacy testing of hypnotic essential oil
The positive effect of the present invention in improving sleep is further illustrated by animal experimental data.
1. Experimental animal ICR grade male BALB/C mouse, the weight average of body is between 20-22 g, provided by Guangdong province medical experimental animal center. Mice were housed in independent ventilated positive and negative pressure animal cages, maintained at controlled temperature (20 + -2 deg.C) and humidity (60 + -15%), and maintained on a 12 hour light/dark cycle. Food and water were freely available during the feeding period and acclimatized 7 days prior to the official experiment.
2. Drug management: 1-11 parts of sleep-improving essential oil prepared according to the formula shown in Table 2.
3. Animal administration: mice were randomly divided into 12 groups by weight, each: control group and experimental drug group. The experimental drug groups were separately gavage with the corresponding hypnotic essential oil, and the control group was gavage with distilled water, and the gavage volume was 0.2ml/10g body weight, once a day for 21 consecutive days.
4. The experimental method comprises the following steps: the influence of the hypnotic essential oil on the sleep incidence, sleep time and sleep latency of the pentobarbital sodium-induced mice is analyzed by using a pentobarbital sodium-induced mouse experiment (refer to handbook for health food inspection and evaluation technical specifications (2003 edition)).
a. Experiment on influence of sleep incidence on mice induced by subthreshold dose of sodium pentobarbital
1h after the last administration of each group, 0.1mg/10g of the body weight of each mouse is injected into the abdominal cavity of each mouse, 1min after the disappearance of the righting reflex is taken as an index of falling asleep, the mice are placed on a warm blanket to relieve the tension degree, and the sleep incidence rate of the mice within 30min is recorded.
And (4) analyzing results: compared with the control group, the sleep incidence rate of the mice in the essential oil 1-4 groups is very different (P <0.01), and the incidence rate of the mice in the essential oil 1 group reaches the highest value (see table 3).
TABLE 3 Effect of hypnotic essential oil in combination with sodium pentobarbital on the incidence of sleep in mice

Note: p <0.05 and P <0.01 (tables 3, 4, 5, supra) compared to the control group.
b. Experiment for prolonging influence of sodium pentobarbital on sleep time of mice induced by sodium pentobarbital
Each mouse was injected intraperitoneally with pentobarbital sodium solution (50mg/kg) at 0.1mg/10g body weight 1h after the last dose. Taking 1min after the righting reflex disappears as an index of falling asleep, placing the mice on a warm blanket to relieve the tension degree, and recording the sleep time of each group of mice, wherein the sleep time of the mice is calculated by 120min when the sleep time of the mice exceeds 120 min.
And (4) analyzing results: compared with the control group, the mice of the essential oil 1 group, the essential oil 2 group and the essential oil 3 group have very significant difference in sleeping time (P <0.01), and show significant sedation and hypnosis synergy (see Table 4).

c. Experiment on influence of barbiturate sodium on sleep latency of mice
1h after the last administration, each mouse was injected with 0.1mg/10g of body weight of barbital sodium solution (250mg/kg) intraperitoneally. Taking disappearance of righting reflex as an index, placing the mice on a warm blanket to relieve the tension degree of the mice, recording the time from administration to disappearance of righting reflex of each group of mice, and calculating the sleeping time of the mice for more than 60min by 60 min.
And (4) analyzing results: the combined effect of essential oil 1, essential oil 2 and essential oil 4 groups with barbiturate sodium had a very significant effect on the sleep latency of mice compared to the control group (P < 0.01). Mice had a reduction in sleep latency of approximately 10min in groups of essential oil 1, essential oil 2 and essential oil 4 compared to the control group (see table 5). The sleep latency time of the mice in the essential oil groups 1, 2 and 4 is shorter than that of the mice in other essential oil groups, and the results show that the hypnotic essential oil groups 1, 2 and 4 are more effective in shortening the sleeping time of the mice and have better hypnotic effect on the mice.
