Abstract

本发明公开了甘蔗叶和八角茴香叶止痒组合物及其制备方法。试验证明,由甘蔗叶和八角茴香叶制得的止痒药物对4‑AP致痒小鼠止痒效果明显,为深入开发甘蔗叶和八角茴香叶治疗皮肤瘙痒、妇女外阴瘙痒的复方药物提供了科学依据。此外,本发明充分利用农作物废弃物,变废为宝,提高甘蔗、八角茴香经济价值。

Description

甘蔗叶和八角茴香叶止痒组合物及其制备方法
技术领域
本发明属于中医药技术领域,尤其涉及甘蔗叶和八角茴香叶止痒组合物及其制备方法。
背景技术
甘蔗叶是禾本科植物甘蔗(Saccharum Officinarum)的干燥叶,八角茴香叶为木兰科的植物八角(Illicium verum Hook.f.)的新鲜叶子均为农业生产中的农作物废弃物。目前,国内外对甘蔗叶、八角茴香叶等农作物废弃物的处理方法多为焚烧和丢弃,造成严重的资源浪费和环境污染。文献显示,这两种农作物废弃物均具有较大的药用价值及发展潜力,却一直停留在基础研究阶段,巨大的经济价值尚未被转化。
申请人前期研制了一种复方甘桑抑菌洗液(专利申请号2018106330889公开日2018.10.23),主要由甘蔗叶、桑白皮、柚叶、八角茴香叶、救必应、毛两面针和薄荷油制成。实验证明,复方甘桑抑菌洗液对大肠埃希菌、金黄色葡萄球菌、铜绿假单胞杆菌、乙型溶血性链球菌、白色念珠菌等其明显抑制作用。
发明内容
本发明要解决的技术问题是提供一种效果良好的甘蔗叶和八角茴香叶止痒组合物及其制备方法,该法还实现了两种农作物废弃物的资源化利用,为深入发掘甘蔗和八角茴的医药应用提供了依据,提高其经济价值其。
为解决上述问题,本发明采用以下技术方案:
甘蔗叶和八角茴香叶在制备止痒药物方面的应用。
止痒组合物,由甘蔗叶和八角茴香叶制得。
甘蔗叶和八角茴香叶的重量份分别为500-1000份、200-800份。
甘蔗叶和八角茴香叶的重量份分别为750份、450份。
止痒药物的制备方法,将甘蔗叶和八角茴香叶分别除杂、洗净、烘干;取八角茴香叶加水提取挥发油,药渣备用,挥发油另存;将药渣加入甘蔗叶后加水回流提取,合并提取液,过滤,浓缩成稠膏;将稠膏离心除去不溶性及其他大分子的杂质,离心后的稠膏中加入前述挥发油并加水搅拌混合均匀,即得止痒药液。
甘蔗叶和八角茴香叶的重量分别为750g、450g。
烘干为烘房内于60℃下干燥至含水量为8~12%。
八角茴香叶加水提取中药材:水重量比1:6;药渣加入甘蔗叶后加水回流提取中原料:水重量比1:8,回流提取两次,每次1h。
止痒药液含生药量为0.24g/mL。
发明人在前期复方甘桑抑菌洗液(七味药材)的基础上,结合临床用药经验,进一步研究发现,与复方甘桑抑菌洗液的抑菌作用完全不同,甘蔗叶和八角茴香叶两味药材联合使用具有一定的止痒功效,据此获得了本发明。试验证明,由甘蔗叶和八角茴香叶制得的止痒药物对4-AP致痒小鼠止痒效果明显,为深入开发甘蔗叶和八角茴香叶治疗皮肤瘙痒、妇女外阴瘙痒的复方药物提供了科学依据。此外,本发明充分利用农作物废弃物,变废为宝,提高甘蔗、八角茴香经济价值。
具体实施方式
一、实验材料
甘蔗叶、八角茴香叶、洁尔阴洗液(四川恩威制药,批号:170417)
SPF级ICR种小鼠60只
二、实验方法
2.1止痒药液的制备
将甘蔗叶750g、八角茴香叶450g除去杂质,用纯净水冲冼干净,稍润,切段,收集在托盘中,置烘房内于60℃下干燥至含水量为8~12%之间。取出,装入洁净的容器中备用。
取八角茴香叶加入6倍药材量的水,提取挥发油(2T多功能提取罐,水蒸气蒸馏法,1次,1h/次),另存。药渣加入甘蔗叶后8倍药材量的水,回流提取两次,每次1h(煮沸开始计时),合并提取液,过滤,浓缩成稠膏状(相对密度不低于1.10)。取出置干燥洁净的容器内备用,中检。将浓缩后的稠膏置离心机中离心除去不溶性及其他大分子的杂质,取出置洁净容器内备用。将离心后的稠膏加入配制罐中,加入纯化水至5L,过滤,加挥发油搅拌混合均匀,灌装,辐射灭菌,制成含生药量为0.24g/mL组合物溶液。
2.2对4-氨基吡啶(4-Aminopyridine,4-AP)致痒小鼠的影响
取ICR雌性小鼠50只,适应性喂养后,用10%硫化钠(新鲜制备的纯水配制)对每只小鼠颈背部脱毛,脱毛区域面积约为2cm*2cm。24h后选取脱毛区域无创口的小鼠40只,随机分为4组,分别为高剂量组、低剂量组、洁尔阴阳性对照组及模型组。将小鼠称重后,按照小鼠面积0.2ml/cm2剂量用棉签涂抹给药,高剂量组为100%浓度(止痒药液原液)的组合物溶液,低剂量组为50%浓度(止痒药液原液稀释一倍)的组合物溶液,洁尔阴阳性药组为100%浓度的洁尔阴洗液,模型组涂纯水,间隔2h给药一次,一天5次,给药2d,末次给药后1h按0.01mg/10g(用生理盐水配制成0.1mg/ml)剂量给每只小鼠背部给药区域中心皮下注射4-AP,注射后观察10min内小鼠舔背部的次数,以连续舔体后出现短暂的间隔为一次,结果以各组舔体次数均值±标准差

三、实验结果
如表1所示,背部涂抹药液后的小鼠舔体次数显著减少,给药浓度对小鼠舔体次数影响较小,高剂量组小鼠舔体次数略低于低剂量组,止痒药液高低剂量组显著优于阳性对照组(洁尔阴洗液)。

2.一种止痒组合物,其特征在于由甘蔗叶和八角茴香叶制得。
3.根据权利要求2所述的止痒组合物,其特征在于:所述甘蔗叶和八角茴香叶的重量份分别为500-1000份、200-800份。
4.根据权利要求3所述的止痒组合物,其特征在于:所述甘蔗叶和八角茴香叶的重量份分别为750份、450份。
5.一种止痒药物的制备方法,其特征在于:将甘蔗叶和八角茴香叶分别除杂、洗净、烘干;取八角茴香叶加水提取挥发油,药渣备用,挥发油另存;将药渣加入甘蔗叶后加水回流提取,合并提取液,过滤,浓缩成稠膏;将稠膏离心除去不溶性及其他大分子的杂质,离心后的稠膏中加入前述挥发油并加水搅拌混合均匀,即得止痒药液。
6.根据权利要求5所述的止痒药物的制备方法,其特征在于:所述甘蔗叶和八角茴香叶的重量分别为750g、450g。
7.根据权利要求5所述的止痒药物的制备方法,其特征在于:所述烘干为烘房内于60℃下干燥至含水量为8~12%。
8.根据权利要求5所述的止痒药物的制备方法,其特征在于:所述八角茴香叶加水提取中药材:水重量比1:6;所述药渣加入甘蔗叶后加水回流提取中原料:水重量比1:8,回流提取两次,每次1h。
9.根据权利要求5所述的止痒药物的制备方法,其特征在于:所述止痒药液含生药量为0.24g/mL。
