本发明公开了一种富含多酚的膳食组合物及其在改善肥胖引起的精液品质降低方面的应用,所述组合物按重量份计算,由以下原料制备而成:迷迭香酸提取物1‑5份,原儿茶酸提取物3‑7份,白藜芦醇提取物1‑3份,优选的,迷迭香酸提取物3份,原儿茶酸提取物5份,白藜芦醇提取物2份,形成3:2:5的配比。实验发现日粮添加0.1%的本膳食组合物对缓解高脂诱导的精液品质下降,改善精子活率和活力效果最强,即日粮添加迷迭香酸:原儿茶酸:白藜芦醇=3:5:2这一比例的复配物,且添加剂量在0.1%时改善精液品质的效果最佳。

本发明属于天然植物提取产品领域,一种富含多酚的膳食组合物及其在改善肥胖引起的精液品质降低方面的应用。
背景技术
肥胖已成为男性生育能力低下或不育的主要原因之一。根据世界卫生组织的数据,2014年超重和肥胖已达到人群的23%。预测肥胖人数将从2005年的3.96亿人增加到2030年的5.73亿人。我国的超重或肥胖的成年男性比例从2002年到2010年增加了25%以上。20岁及以上人群中超重和肥胖检出率为42.3%,中心性肥胖检出率为44%,且呈现出年轻化趋势。肥胖不仅会干扰身体健康,包括心血管疾病、2型糖尿病、恶性肿瘤、神经退化和加速衰老,同样也会对男性的生殖功能产生影响。肥胖肥胖会带来睾丸激素水平下降,引起系统系炎症和造成机体氧化应激,导致机体导致精子数量降低,活力降低,直接影响男性生殖能力。急需研发出一种天然的膳食组合物,改善由肥胖引起的精液品质下降问题。
发明内容
植物多酚是一类具有多种生物学活性的化合物,广泛存在于植物的根、茎、叶、花和果实中,在表皮含量尤为丰富。植物多酚具有抗氧化、抗炎、调节脂质代谢等生物学活性。迷迭香酸、原儿茶酸和白藜芦醇是植物界中分布广泛的天然酚类物质,具有极强的抗氧化和抗炎能力。我们前期研究表明,迷迭香酸、原儿茶酸和白藜芦醇在体外和体内都是氧化应激和炎症的有效抑制剂,且具有调节脂质代谢,降低脂肪沉积的作用。肥胖及应激是引起机体炎症和氧化损伤,导致精液品质下降的重要原因,因此,迷迭香酸、原儿茶酸和白藜芦醇作为强有力的缓解氧化应激和炎症的天然活性物质,具有改善由肥胖和应激引起的精子活力下降,精子死亡率增高,提高精液质量的潜在功效。本专利发明在前期研究基础上,探究了迷迭香酸、原儿茶酸、白藜芦醇及其组合物对精液质量的改善作用,并筛选出合适的添加比例。具体涉及迷迭香酸、原儿茶酸、白藜芦醇及其组合物的制备及改善肥胖引起的精液品质下降或用于提高精液品质的用途。

本发明通过不同配比不同添加量筛选出一种可用于降低肥胖或高能饮食引起的精子死亡率增高和存活率下降相关症状的膳食组合物,通过研究精子活率、精子死亡率、精子活力、直线速率、路径速率、曲线速率和侧摆幅值,发现日粮添加0.1%的膳食多酚组合物对缓解高脂诱导的精液品质下降,改善精子活率和活力效果最强,即日粮添加迷迭香酸:原儿茶酸:白藜芦醇=3:5:2这一比例的组合物,且添加剂量在0.1%时改善精液品质的效果最佳。

选取24只C57BL/6J试验小鼠(SPF级,雄性,4-6周龄)及辐照杨木垫料均从湖南斯莱克景达实验动物有限公司购买,低脂日粮、高脂日粮参照research diets配制;所用试剂或仪器未注明生产厂商者,均为可以通过市购获得的常规试剂产品。
将实施例2制备的复配物A按1g/kg(0.1%)和0.5g/kg(0.05%)的比例添加至粉末状高脂日粮中;采用逐级预混原则(至少三级),混合标准品及高脂饲料,待标准品完全溶于高脂饲料(用手摊平混合饲料看不到标准品细碎颗粒);将混合饲料揉成适宜大小的圆状混合饲料,并分袋标记,于-20℃冰箱冷冻保存。
小鼠预饲一周后用Experimental Animal Allotment Program(EAAP)随机分组软件,将小鼠随机分为4组,每组6个重复,开始正式试验。试验期间保证自由采食与饮水,保持室温恒温(23℃),通风良好单笼饲养,12小时白天(18:00-6:00系统自动关灯),12小时黑夜(6:00-18:00,系统自动开灯)。试验处理共分为4组,空白组饲喂低脂日粮,对照组饲喂高脂日粮,膳食多酚干预组分别饲喂添加0.05%、0.1%上述复配物A的高脂日粮。试验期间,注意观察小鼠的精神状态,每7天更换垫料一次、更换饮用水一次,每天记录小鼠的采食量,试验期为12周。
结果分析
实施例2制备的膳食多酚复配物A对小鼠精液品质的影响如表1所示,日粮中添加膳食多酚复配物A能提高精子存活率,降低死亡率,提高精子活力、路径速率、曲线速率、直线频率和侧摆幅值,可以缓解由于高脂饮食导致的精子质量降低。

实验例2
选取36只C57BL/6J试验小鼠(SPF级,雄性,4-6周龄)及辐照杨木垫料均从湖南斯莱克景达实验动物有限公司购买,低脂日粮、高脂日粮参照research diets配制;所用试剂或仪器未注明生产厂商者,均为可以通过市购获得的常规试剂产品。
将实施例4制备的复配物B按1g/kg(0.1%)和0.5g/kg(0.05%)的比例添加至粉末状高脂日粮中;采用逐级预混原则(至少三级),混合标准品及高脂饲料,待标准品完全溶于高脂饲料(用手摊平混合饲料看不到标准品细碎颗粒);将混合饲料揉成适宜大小的圆状混合饲料,并分袋标记,于-20℃冰箱冷冻保存。

结果分析
膳食多酚复配物B对小鼠精液品质的影响如表2所示,日粮中添加膳食多酚复配物A能提高精子存活率,降低死亡率,提高精子活力、路径速率、曲线速率、直线频率和侧摆幅值,可以缓解由于高脂饮食导致的精子质量降低。

综上所述,日粮添加0.1%的膳食多酚复配物A对缓解高脂诱导的精液品质下降,改善精子活率和活力效果最强,即日粮添加迷迭香酸:原儿茶酸:白藜芦醇=3:5:2这一比例的复配物,且添加剂量在0.1%时改善精液品质的效果最佳。
以上所述实施例仅表达了本发明的1种实施方式,其描述较为具体和详细,但并不能因此而理解为对本发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。
Claims (10)
1.一种富含多酚的膳食组合物,其特征在于,所述组合物按重量份计算,由以下原料制备而成:迷迭香酸提取物1-5份,原儿茶酸提取物3-7份,白藜芦醇提取物1-3份。
2.根据权利要求1所述的一种富含多酚的膳食组合物,其特征在于,所述迷迭香酸提取物通过逆流提取技术提取自迷迭香茎叶,其中迷迭香酸含量大于20%。
3.根据权利要求2所述的一种富含多酚的膳食组合物,其特征在于,所述原儿茶酸提取物提取自五味子,其中的原儿茶酸含量大于95%。
4.根据权利要求3所述的一种富含多酚的膳食组合物,其特征在于,所述白藜芦醇提取物提取自虎杖,其中的白藜芦醇含量大于95%。
5.根据权利要求4所述的一种富含多酚的膳食组合物,其特征在于,所述组合物按重量份计算,由以下原料制备而成:迷迭香酸提取物3份,原儿茶酸提取物5份,白藜芦醇提取物2份。
6.如权利要求5所述的一种富含多酚的膳食组合物的制备方法,其特征在于,包括以下步骤:
S1迷迭香酸提取物通过逆流提取技术提取自迷迭香茎叶,真空冷冻干燥后,制成冻干粉,其中迷迭香酸含量大于20%;
S2所述原儿茶酸提取物提取自五味子,真空冷冻干燥后,制成冻干粉,其中的原儿茶酸含量大于95%;
S3所述白藜芦醇提取物提取自虎杖,真空冷冻干燥后,制成冻干粉,其中的白藜芦醇含量大于95%;
S4将步骤S1-S3获得的冻干粉均匀混合后,真空包装。
7.如权利要求1-5任一项所述的富含多酚的膳食组合物在制备改善肥胖引起的精液品质降低方面的膳食/保健品/饲料中的应用。
8.如权利要求7所述的应用,其特征在于,将所述膳食组合物添加于膳食/保健品/饲料中,添加量为0.05%-0.1%。
9.如权利要求8所述的应用,其特征在于,所述添加量为0.1%。
10.如权利要求6所述的一种富含多酚的膳食组合物的制备方法在制备改善肥胖引起的精液品质降低方面的膳食/保健品/饲料中的应用。
