溶瘤病毒(OV)是优先感染和杀死癌细胞的病毒。病毒生长并导致癌细胞裂解(溶瘤),或触发其他机制以干扰癌症免疫抑制微环境,并触发机体的免疫应答以清除癌细胞。最近成功的临床数据和药物批准增加了公众对溶瘤病毒疗法的关注。溶瘤病毒疗法可以与其他药物、免疫检查点抑制剂和T细胞疗法结合使用以改善癌症患者的预后。
OV的递送途径包括瘤内(i.t.)注射、静脉内(i.v.)递送和腹膜内递送,其中主要应用瘤内注射。

具体实施方式
目前,大多数溶瘤病毒疗法仅限于局部区域注射(如瘤内注射,i.t.),且单次注射是否足以达到治疗效果仍在研究中。众所周知,一些OV已经被证明具有急性、短暂的毒性特征,因此注射病毒可诱导针对病毒而不是针对肿瘤细胞的竞争性免疫应答。此外,静脉递送OV也存在问题,其中OV可能从血流中迅速清除,因此需要频繁或高剂量施用,导致治疗成本增加和潜在的安全问题。因此,需要提供包含OV的组合物,其可被施用并在循环中保持延长的时间段以将治疗剂最终递送到靶细胞。
本领域已知,为了在纳米颗粒上实现隐形部分,可采用聚乙二醇(PEG)。然而,抗PEG免疫应答可能被触发,因此这种方法是有问题的。已经提出了替代方法,例如利用两性离子材料(例如聚(羧基甜菜碱)和聚(磺基甜菜碱))。

进行了制备带有溶瘤病毒核的RBC包覆的纳米颗粒的各种尝试,但除了经由超声处理使用特定方法外,其他已知方法均未成功。虽然不希望受到任何理论的约束,但由于某些物理和化学条件,病毒容易灭活的事实限制了本领域技术人员处理病毒的方式。例如,已知某些病毒可以通过压力、热量和/或化学物质(如高浓度醇溶液)而灭活。
挤压是一种用于创建具有固定横截面轮廓的对象的方法。将材料推过具有期望横截面的模具。与其他制造方法相比,该方法的两个主要优点是其能够制造非常复杂的横截面以及能够加工脆性材料的能力,因为材料只会遇到压缩应力和剪切应力。该方法还可以形成具有良好表面光洁度的零件。工业级挤压应用涉及压力、温度、加热和冷却控制。
就像处理活病毒的困难一样,利用细胞膜(如红细胞膜)的方法也不容易。提供了经由超声处理和挤压来制备RBC膜囊泡的方法。参见例如,Luk等人,Nanoscale,2014,6,2730-2737。在上述文献中,PLGA聚合物核通过溶剂置换法包覆有RBC膜,然后使用Avanti微型挤压机通过100nm聚碳酸酯多孔膜挤压。据报道,挤压的囊泡仅在一定的阈值挤压压力以上产生,并且其大小取决于挤压压力。最小压力似乎与脂质双层的裂解张力有关,而不是与系统的任何弯曲模量有关。在针对水的粘度进行校正后,等浓度脂质溶液通过孔隙的流速与脂质性质无关。参见Hunter等人,Biophysical Journal,74,1998,2996-3002。
然而,尽管使用挤压来制备RBC膜,但还没有发现用RBC膜制备包含病毒核的纳米颗粒的挤压方法。由于方法中涉及的压力、热量和/或其他因素,该方法很可能不适合通过RBC膜封装病毒。
事实上,本领域已知,基于压力的病毒灭活方法是一种有前景的替代方法和工业上成熟的技术。参见例如,Dumard等人,PLOS ONE,2013年11月,第8卷,第11期,e80785。例如,已知腺病毒对紫外线处理有抵抗力,但低压紫外线使腺病毒灭活。因此,本领域普通技术人员不会考虑在病毒上使用挤压工艺,因为该方法可能使病毒灭活。
根据本发明的实践,令人惊讶地发现,涉及挤压的条件可用于制备包含本文所述病毒核的本发明纳米颗粒而不会使病毒灭活,从而使此类方法成为制备所述纳米颗粒的可行方法。
在一些实施方案中,本发明方法涉及在挤压步骤之前,在适当溶剂(例如,水基溶剂)中制备包含病毒颗粒和细胞衍生的膜的预包覆溶液。病毒颗粒的示例为腺相关病毒、腺病毒、单纯疱疹病毒和牛痘病毒,但不限于此。在一些实施方案中,以基因组拷贝数测量的病毒浓度范围为每毫升104个基因组拷贝到每毫升1015个基因组拷贝。血液衍生的膜的浓度以总蛋白质浓度测量,在一些实施方案中,其范围为0.001mg/mL至10mg/mL。
根据制备方法中使用的病毒大小,可使用具有大约等于或大于病毒大小的孔的纳米孔膜。例如,已知大小为约100nm的病毒可与孔径为100nm或更大的纳米孔膜相匹配。
本领域技术人员容易识别用于挤压方法/工艺的任何合适装置。例如,根据本发明的实践,使用一种装置,该装置含有通向将纳米孔保持固定在适当位置而不允许泄漏的腔室的注入口,以及穿过纳米孔的挤压口。
然后,通过注入口将上述预包覆溶液加载到含有纳米孔膜的腔室中,令人惊讶的是,在施加一定压力(例如,约5-10psi)的情况下,溶液冲过纳米孔膜,在另一侧出现,并在挤压口上收集。在该方法中施加的物理力导致细胞衍生的膜的破坏。然后,被破坏的膜包裹并重新形成在病毒颗粒周围,从而包覆或封装一些病毒颗粒。有几次尝试利用了所讨论的本领域已知的挤压工艺。然而,令人惊讶地发现,仅根据本发明实践的低压挤压工艺提供了令人满意的病毒封装纳米颗粒。这种挤压工艺可以重复,通常重复15-30次,以确保溶液中几乎所有病毒颗粒的彻底封装。在一些实施方案中,挤压工艺重复10-30次、15-30次、15-25次或15-20次。在病毒上添加包覆层的物理行为会增加产生的颗粒的大小。因此,封装的验证由颗粒大小直径的增加证明。
本文提及的术语“溶瘤病毒”包括以下非限制性示例:疱疹病毒、牛痘病毒、呼肠孤病毒、腺病毒、麻疹病毒、细小病毒或其组合。

本发明提供了细胞膜衍生的纳米颗粒,用于递送治疗剂,例如溶瘤病毒。该药剂基本上通过用细胞膜脂质包覆治疗剂如溶瘤病毒来伪装。
在一些实施方案中,诸如溶瘤病毒的治疗剂包覆有与受试者(例如,患者)血流内的循环相容的脂质。这允许经由EPR(增强的渗透性和保留)效应将药剂递送到肿瘤血管系统,其中某些分子倾向于在肿瘤组织中比在正常组织中积聚更多。EPR效应通常用于描述对癌组织的纳米颗粒和脂质体递送。其中一个示例是关于金纳米颗粒的热消融的研究。因此,在一些实施方案中,本文公开的纳米颗粒将积聚在肿瘤的血管系统附近,并且治疗剂(例如,OV)将与癌细胞接触。OV随后感染细胞,导致细胞裂解、死亡或移除。

与其他平台(例如,PEG包覆的纳米颗粒)相比,RBC膜衍生的纳米颗粒具有许多表面标志物,例如CD47、CD59(MAC抑制蛋白,避免补体细胞裂解)、CD55(DAF)、CD35(CR1)和免疫球蛋白超家族中的其他成员,以保护自身免受机体清除。在一些实施方案中,本文中用于包覆的细胞膜可进一步掺入非脂质组分,例如细胞表面标志物、MHC分子和糖蛋白。在其他实施方案中,细胞膜可进一步掺入亲水性组分,例如PEG。
如本文所用,术语“细胞膜”是指在细胞或突现病毒颗粒内或周围充当选择性屏障的生物膜封闭或分离结构。细胞膜对离子和有机分子具有选择性渗透性,并控制物质进出细胞的运动。细胞膜包括磷脂单层或双层,以及任选的相关蛋白质和碳水化合物。如本文所用,细胞膜是指从细胞或细胞器的天然存在的生物膜获得的膜,或从其衍生的膜。

如本文所用,术语“从其衍生的”是指天然膜的任何后续修饰,例如分离细胞膜、产生膜的部分或片段、从取自细胞或细胞器的膜移除和/或向其中添加某些组分,例如脂质、蛋白质或碳水化合物。膜可以通过任何合适的方法从天然存在的膜衍生而来。例如,可从细胞制备或分离膜,且制备的或分离的膜可与其他物质或材料结合以形成衍生的膜。在另一个示例中,细胞可被重组工程化以产生在体内掺入其膜中的“非天然”或“天然”物质,并且细胞膜或病毒膜可从该细胞制备或分离以形成衍生的膜。
可通过已知方法制备细胞膜。例如,可以使用微射流机(MF)或低渗溶液(如水)破碎细胞,然后用盐水或PBS进行超滤或透滤。在一些情况下,具有较高离子强度的溶液可用于去除猪肝中的血液,因此PBS或NaCl溶液可帮助通过过滤方法去除细胞内物质。例如在透滤过程中,也可以使用抗凝剂如EDTA,以去除杂质。在一些实施方案中,切向流过滤(TFF)装置或离心可用于纯化膜。纯化的膜组分可诸如通过BCA蛋白质测试来测试细胞蛋白质的存在。优选地,在包覆之前去除大多数非膜组分。
尽管做出了大量努力,但发现按照已知方法,如US2013/033 7066中的程序,病毒诸如溶瘤病毒或CRISPR(来自病毒的DNA序列)不能被细胞膜(例如,RBC膜)包覆或封装。令人惊讶地发现,仅在某些条件下,细胞膜(例如,RBC血影)包覆病毒或类似物。制备本文公开的纳米颗粒的方法需要挤压在溶液中的RBC血影和OV的混合物。在某些实施方案中,根据使用的多孔膜的孔径,本发明挤压工艺中施加的压力为约1psi至300psi。在某些实施方案中,工艺中施加的压力为约5psi至300psi、5psi至200psi、5psi至100psi、5psi至50psi、5psi至40psi、5psi至30psi、5psi至20psi或5至10psi。在某些实施方案中,工艺中施加的压力为约5psi至10Psi。
根据本发明的实践,适合于包覆的病毒包括溶瘤病毒以及可感染癌细胞的其他病毒。病毒可以具有有包膜或未包膜的形式。在一些实施方案中,病毒载体是优选的,因为它们可以感染癌细胞并复制(复制能力)。

实施例
实施例1.细胞膜包覆的纳米颗粒经由挤压的示例性制备
细胞膜的制备(例如,RBC血影制备)为本领域所知。在此,遵循制备细胞膜、RBC细胞膜的非限制性示例,以提供用于本发明纳米颗粒制备的示例性细胞膜。
材料:
·病毒(具有已知粒径,例如腺病毒为10^12至10^13PFU,疱疹病毒为10^8至10^9PFU,牛痘病毒为10^8至10^9PFU,腺相关病毒为10^11至10^13个病毒颗粒/mL)。

程序:为了用血液衍生的膜封装病毒,将含有高浓度膜(例如,RBC膜)和病毒的溶液通过纳米孔膜反复(约20次)挤压。首先用较大孔径(1600nm或800nm)的纳米孔膜进行挤压,然后收集所得洗脱液。然后,使用较小孔径(例如,800->400->200)对洗脱液进行更多挤压,并使用比病毒大小略大的孔径完成挤压。例如,如果病毒直径为120-150nm,则使用孔径为200nm的纳米孔膜。
下面详细说明挤压的示例性方法。
挤压前准备
·组装中间夹有纳米孔膜过滤器的挤压装置。
·将100uL的无菌去离子水装入气密注射器。
·使用气密注射器使100uL无菌去离子水穿过膜,以将膜预湿。
通过纳米孔膜的挤压
1.向500uL-1000uL的膜(例如,RBC膜)添加足够的按体积计约1-2%(500uL中5-10ul)的病毒(例如,腺病毒)并均匀混合。
2.将病毒/膜溶液(“V/M溶液”)加载到气密注射器中。
3.将气密注射器插入挤压机构中。
4.在挤压机构中,在加载的注射器对面插入空的气密注射器。
5.通过约5psi至10psi的低压,推动V/M溶液从加载注射器穿过膜到达空注射器。
6.将V/M溶液推回到现在空的(先前加载的)注射器。
7.重复步骤5-6至少10次,以确保V/M溶液穿过纳米孔膜,在一些情况下,不少于20次。
通过透射电子显微术(TEM)对所得的包覆的纳米颗粒进行成像,并测量粒径。图1显示了裸腺病毒与通过本文所述的示例性挤压处理得到的包覆的腺病毒相比的平均粒径的图表。包覆的病毒的较大直径清楚地证明了经由本发明方法制备包覆病毒的过程是成功的。

实施例2.RBC膜包覆的病毒纳米颗粒的荧光显微术研究结果
荧光显微术用于证明病毒在挤压工艺后保持传染性的能力。将A529癌细胞铺板到24孔组织培养板上的孔中,并允许其生长。当癌细胞生长时,我们用EDM包覆的腺病毒将其处理10分钟,在去除含有病毒的培养基之前,用PBS冲洗细胞,然后替换为常规生长培养基。所使用的EDM包覆的腺病毒含有一种基因,当病毒感染细胞时,该基因会表达荧光蛋白。

Claims (14)
1.一种制备纳米颗粒的方法,所述方法包括在水溶液中将包含病毒的内核和包含衍生自细胞的细胞膜的外表面组合;将挤压工艺应用于所述混合物溶液以形成包含包覆有所述外表面的所述内核的纳米颗粒。
2.根据权利要求1所述的方法,其中在所述应用挤压工艺时施加5-300psi的压力。
3.根据权利要求2所述的方法,其中所述挤压步骤需要5-100psi的压力。
4.根据权利要求1所述的方法,其中所述挤压步骤重复至少10次。
5.根据权利要求1所述的方法,其中所述水溶液是盐水溶液。
6.根据权利要求1所述的方法,其中所述水溶液是非盐水溶液,所述非盐水溶液包含糖或在水溶液中具有类似于糖的性质的组分。
7.根据权利要求6所述的方法,其中所述非盐水溶液是糖溶液。
8.根据权利要求7所述的方法,其中所述糖溶液是蔗糖或含葡萄糖的溶液。
9.根据权利要求1所述的方法,其中所述病毒是溶瘤病毒。
10.根据权利要求9所述的方法,其中所述溶瘤病毒是疱疹病毒、牛痘病毒、呼肠孤病毒、腺病毒、麻疹病毒、细小病毒或其组合。
11.根据权利要求10所述的方法,其中所述病毒是腺病毒。
12.根据权利要求1所述的方法,其中所述细胞是血细胞、脂肪细胞、干细胞、内皮细胞、外泌体、分泌小泡或突触小泡。
13.根据权利要求12所述的方法,其中所述血细胞是红细胞、白细胞或血小板。
14.根据权利要求13所述的方法,其中所述血细胞是红细胞。
