Description
(Rogach, A., S. V. Kershaw, M. Burt, M. T.Harrison, A. Kornowski, A. Eychmüller and H. Weller (1999). "Colloidallyprepared HgTe nanocrystals with strong room-temperature infraredluminescence." (Harrison, M. T., S. V.Kershaw, A. L. Rogach, A. Kornowski, A. Eychmüller and H. Weller (2000). "WetChemical Synthesis of Highly Luminescent HgTe/CdS Core/Shell Nanocrystals."Advanced Materials 12(2): 123-125.)
1)将还原型谷胱甘肽超声溶于超纯水中,调节pH值为9.511.5,加入Hg(ClO4)2溶液并混合均匀,得到透明澄清的GSH-Hg前驱体溶液,通N2气流将GSH-Hg前驱体溶液除氧;
2)在乙醇和水的混合液中,加入Te粉和过量的硼氢化钠,在氮气保护下加热至50~ 70℃进行反应,待黑色Te粉消失后,注入过量的稀硫酸产生H2Te气体;用N2气流将生成的H2Te气体带入步骤1)制备好的GSH-Hg前驱体溶液,在70 ~ 85℃和搅拌条件下反应5 ~ 60min,获得黄色透明的GSH配位的HgTe量子点溶液;加入可溶性Zn盐和谷胱甘肽,调节pH值为9.5~11.5,备用;
1)本发明设计了一种仅使用玻璃仪器和氮气瓶,一锅合成具有稳定近红外荧光的HgTe/ZnSe纳米量子点的装置路线图,既往文献采用电化学合成或在真空手套箱中合成HgTe量子点,需要额外的抽真空装置和多个步骤完成,而本发明的制备方法简单易行,一锅合成,成本较低;
2)与既往文献采用有致癌毒性的巯基乙醇稳定剂相比,本发明所采用的是细胞组成中的谷胱甘肽为稳定剂。合成的HgTe/ZnSe量子点具有量子尺寸效应,控制加热时间可以获得不同尺寸(2~5 nm)的量子点,荧光稳定性高,在室温下可放置3个月,其量子产率接近50%,荧光寿命达到50 ns以上;
3)HgTe量子点外层包裹生物相容性高的ZnSe壳层,减小HgTe或HgTe/CdS量子点的毒性,细胞毒性和细胞溶血实验表明,合成的HgTe/ZnSe量子点环境友好性和生物相容性显著提高;与HgTe量子点相比,所合成的近红外荧光纳米量子点具有光谱波长可调范围宽,发光性能稳定,荧光量子产率高,荧光寿命长等优点;

1)制备GSH-Hg前驱体溶液:在图中I处的三颈烧瓶中加入的超纯水200 mL,加入1.8 g(5.86 mmol)的GSH固体,搅拌使其完全溶解,氢氧化钠(0.5 M)调节溶液的pH值达到11.2,磁子搅拌下逐滴加入2.0 mL浓度为1 mol·L-1的Hg(ClO4)2(2.0 mmol)水溶液并混合均匀,得到透明澄清的GSH-Hg前驱体溶液,通N2气流(20 min以上)将GSH-Hg前驱体溶液除氧;
2)制备H2Te气体:在图中II处的单颈烧瓶中加入乙醇和水,获得体积比2:1的乙醇和水的混合液,然后加入80 mg(0.64 mmol) Te粉和50 mg硼氢化钠,在氮气保护下磁子搅拌下加热至60℃反应约20 min,此时黑色Te粉消失,变为无色溶液,然后向体系中缓慢注入2.0 mL、0.5 M稀硫酸(20~30滴/60秒),产生H2Te气体,整个实验步骤需要氮气氛围保护;
3)制备HgTe量子点:用N2气流将步骤2)生成的H2Te气体带入步骤1)制备好的GSH-Hg前驱体溶液,在75℃和搅拌条件下反应5 ~ 60 min,获得粒径1.9 ~ 3.0 nm的黄色透明的GSH配位的HgTe量子点溶液;加入1 mL、0.6 M (0.6 mmol)ZnSO4和3 mL的0.4 M(1.2mmol)配体谷胱甘肽水溶液,调节pH值为11.2,备用;
4)制备H2Se气体:在图中III处的单颈烧瓶中加入乙醇和水,获得体积比2:1的乙醇和水的混合液,然后加入40 mg(0.5 mmol)的Se粉和40 mg硼氢化钠,在氮气氛磁子搅拌作用下加热至50℃反应约20 min,此时黑色Se粉消失,转变为无色溶液,然后向体系中缓慢注入2.0 mL、0.5 M稀硫酸(20~30滴/60秒),产生H2Se气体,整个实验步骤需要氮气氛围保护;
