Abstract
本发明涉及一种水凝胶面膜制备方法及其产品和应用,采用静电纺丝的方式,制备抗菌纳米颗粒的丝素纤维膜,随后丝素纤维与甘蔗水解液共发酵,使细菌纤维与丝素纤维复合交联,获得复合水凝胶面膜。本发明采用静电纺丝的方式,将纳米抗菌颗粒直接纺织成纤维,使面膜基材本身具备抗菌能力,因此添加美颜成分时,无需另行添加任何抗菌成分,延长面膜的保质期。通过丝素纤维膜后续与甘蔗发酵液的共培养发酵,使细菌纤维素与丝素纤维复合交联,从而制备水凝胶面膜基材。该水凝胶面膜基材吸附富含甘蔗提取物的美颜活性成分后,可获得水凝胶面膜,该面膜具有美白、保湿、消炎、促进成纤维细胞生长的作用。

技术领域
本发明涉及一种水凝胶面膜制备方法及其产品和应用。具体涉及一种利用静电纺丝和发酵工艺制备的细菌纤维素/丝素纤维复合水凝胶面膜的制备方法。本发明属于化妆品技术领域。
背景技术
生活水平的提高促使更多的人关注美容领域,其中面膜是面部皮肤保养美颜的产品,由于其使用方便效果显著,深受到爱美人士的关注,在市场上具有广泛的认可度。传统的面膜通常为贴式型、泥膏型等,此类面膜中通常含有美颜液体,利用其与面部的直接接触,促使面膜中的营养物质进入皮肤表面的角质层,达到保湿、美白、祛痘的目的。然而,通常贴型面膜主要采用无纺布或相关纤维织物作为基质材料,存在对精华液吸附量小、保水力不足、皮肤贴合性差等缺点,造成美容活性成分的浪费。
水凝胶面膜以三维网络空间吸附精华液,美容成分不易流式同时具有保水性好的优点。然而部分水凝胶成分可能存在化学交联剂,如戊二醛等成分,可能造成皮肤的潜在危害,因此采用生物的成分制备水凝胶面膜极具应用前景。
甘蔗中含有乙醇酸、丁二醇,前者易渗入皮肤,促进废旧角质层脱落,加速沉积在角质层的黑色排出体外,防止毛孔堵塞。后者可提高皮肤含水量,使皮肤水润透亮,同时提取物还具有抗菌活性,是一种天然的防腐剂。甘蔗提取物可以抑制血小板活化因子(PAF)表达,降低皮肤炎症,存进皮肤血液循环和活化细胞,具有消炎、长效保湿、和美白功效。甘蔗是南方的主要能源作物,甘蔗榨汁后的甘蔗渣富含纤维素和半纤维素等多聚糖,可用于制备细菌纤维素。因此甘蔗是极佳的制备面膜基材的原料。
发明内容
本发明目的在于提供一种水凝胶面膜的制备方法。
本发明的再一目的在于:提供一种上述方法制备的水凝胶面膜产品。
本发明的又一目的在于:提供一种上述产品的应用。
本发明目的通过下述方案实现:一种水凝胶面膜的制备方法,其特征在于采用静电纺丝的方式,制备抗菌纳米颗粒的丝素纤维膜,随后丝素纤维与甘蔗水解液共发酵,使细菌纤维与丝素纤维复合交联,获得复合水凝胶面膜,包括以下步骤:
(1)0.1~1g纳米氧化锌(ZnO NPs)溶解于20%~50%浓度的丝素蛋白(SF)溶液,于60℃混合均匀,得到浓缩的纺丝液;将混合均匀的纺丝液注入10 mL注射器,设定电纺压为15~20 kv,以流速0.1~0.5 mL/h,在12~20 cm接受距离内进行静电纺丝,得到丝素纤维膜;上述蚕丝纤维膜121℃、0.07~0.1 MPa高温高压灭菌20min,烘干备用;
(2)甘蔗渣经水解、脱毒后,加入1%酵母浸膏和0.5%胰蛋白胨,121℃、0.07~0.1MPa高温高压灭菌20min,作为培养基;随后以8~12%接种量接种醋酸杆菌于培养基中,混合均匀后,得到发酵液;发酵液注入装有步骤1制备的纤维膜的培养器皿中,25~30℃静置培养6~10天;将发酵产物采用去离子水彻底清洗,60℃烘干,即可得到细菌纤维素/蚕丝纤维复合水凝胶面膜基材。
所述的丝素蛋白膜厚度0.1~0.2 mm,纤维间隙范围为20~100 µm。
所述的细菌纤维素/蚕丝纤维复合水凝胶面膜厚度为0.5~5mm。
本发明提供一种水凝胶面膜,根据权利要求1-3上述任一所述方法制备得到。
本发明提供一种根据权利要求4所述凝胶面膜在美颜中的应用,所制备的细菌纤维素/蚕丝纤维复合水凝胶面膜基材与美颜活性成分混合,制备具备美颜成分的水凝胶面膜;其中美颜活性成分为:0.05~1g甘蔗提取物、0.1~1.5g透明质酸钠、2~10g 甘油、1~5 mL积雪草提取物、0.1~1g丝素蛋白、去离子水95~99 mL。
本发明的优点在于:
(1)采用静电纺丝的方式,将纳米抗菌颗粒直接纺织成纤维,使面膜基材本身具备抗菌能力,因此添加美颜成分时,无需另行添加任何抗菌成分,延长面膜的保质期。
(2)通过丝素纤维膜后续与甘蔗发酵液的共培养发酵,使细菌纤维素与丝素纤维复合交联,从而制备水凝胶面膜基材。
(3)该水凝胶面膜基材吸附富含甘蔗提取物的美颜活性成分后,可获得水凝胶面膜,该面膜具有美白、保湿、消炎、促进成纤维细胞生长的作用。

实施例1

(1)0.5g纳米氧化锌ZnO NPs溶解于浓度35%的丝素蛋白SF溶液中,于60℃混合均匀,得到浓缩的纺丝液;将混合均匀的纺丝液注入10 mL注射器,设定静电纺丝的电压为18kv,以流速0.25 mL/h、在18 cm接受距离内进行静电纺丝,得到含纳米氧化锌丝素纤维膜;所得的丝素纤维膜121℃、0.07~0.1 MPa高温高压灭菌20min,烘干备用;
(2)甘蔗渣经水解、脱毒后,加入1%酵母浸膏和0.5%胰蛋白胨,121℃、0.07~0.1MPa高温高压灭菌20min,作为培养基;随后以10%接种量接种醋酸杆菌于培养基中,混合均匀后,得到发酵液;
(3)发酵液注入装有步骤(1)制备的含纳米氧化锌丝素纤维膜的培养器皿中,25~30℃静置培养8天;将发酵产物采用去离子水彻底清洗,60℃烘干,即可得到细菌纤维素/丝素纤维复合水凝胶面膜基材;
所得的细菌纤维素/丝素纤维膜厚度0.1~0.2 mm,纤维间隙范围为20~100 µm;
将0.5g甘蔗提取物、0.5g透明质酸钠、8g 甘油、3 mL积雪草提取物、0.5g丝素蛋白、去离子水97 mL混合均匀,γ-射线辐射灭菌后,得到美颜溶液;将剪裁后的细菌纤维素/丝素纤维复合水凝胶面膜基材浸入美颜溶液,充分浸泡后,即可获得细菌纤维素/丝素纤维复合水凝胶面膜,所得的细菌纤维素/丝素纤维复合水凝胶面膜厚度为0.5~5mm。
将水凝胶面膜从美颜溶液中取出,灭菌后,在DMEM培养基中充分浸泡48 hr。收集对数生长期HSF成纤维细胞,按照细胞密度为3x103cell/孔接种至96孔板;设置空白对照(DMEM)、阳性对照(DMEM+10%FBS)、样品组(水凝胶面膜的DMEM浸泡液),对细胞进行处理,培养箱(37℃,5%CO2)中培养24h,采用CCK-8测定细胞活 ,根据公式计算,细胞比活力(100%)=(样品组-调零孔)/(空白组-调零孔)x100%。实施例1所制备的水凝膜面膜的细胞活性图。由附图1可知,与对照组相比,本发明所制备的水凝胶面膜生物相容性良好,可促进皮肤成纤维细胞增殖。
实施例2
一种水凝胶面膜基材,与实施例1步骤近似,按以下步骤制备:
(1)1g纳米氧化锌ZnO NPs溶解于浓度40%的丝素蛋白SF溶液中,于60℃混合均匀,得到浓缩的纺丝液;将混合均匀的纺丝液注入10 mL注射器,设定静电纺丝的电压为18 kv,以流速0.25 mL/h、在18 cm接受距离内进行静电纺丝,得到含纳米氧化锌丝素纤维膜;所得的丝素纤维膜121℃、0.07~0.1 MPa高温高压灭菌20min,烘干备用;
(2)甘蔗渣经水解、脱毒后,加入1%酵母浸膏和0.5%胰蛋白胨,121℃、0.07~0.1MPa高温高压灭菌20min,作为培养基;随后以10%接种量接种醋酸杆菌于培养基中,混合均匀后,得到发酵液;
(3)发酵液注入装有步骤(1)制备的含纳米氧化锌丝素纤维膜的培养器皿中,25~30℃静置培养9天;将发酵产物采用去离子水彻底清洗,60℃烘干,即可得到细菌纤维素/丝素纤维复合水凝胶面膜基材。
将0.5g甘蔗提取物、0.5g透明质酸钠、8g 甘油、3 mL积雪草提取物、0.5g丝素蛋白、去离子水97 mL混合均匀,γ-射线辐射灭菌后,得到美颜溶液;将剪裁后的细菌纤维素/蚕丝纤维复合水凝胶面膜基材浸入美颜溶液,充分浸泡后,及可获得水凝胶面膜。
实施例3
一种水凝胶面膜基材,与实施例2步骤相同,按以下步骤制备:
(1)1g纳米氧化锌ZnO NPs溶解于浓度40%的丝素蛋白SF溶液中,于60℃混合均匀,得到浓缩的纺丝液;将混合均匀的纺丝液注入10 mL注射器,设定静电纺丝的电压为18 kv,以流速0.25 mL/h、在18 cm接受距离内进行静电纺丝,得到含纳米氧化锌丝素纤维膜;所得的丝素纤维膜121℃、0.07~0.1 MPa高温高压灭菌20min,烘干备用;
(2)甘蔗渣经水解、脱毒后,加入1%酵母浸膏和0.5%胰蛋白胨,121℃、0.07~0.1MPa高温高压灭菌20min,作为培养基;随后以10%接种量接种醋酸杆菌于培养基中,混合均匀后,得到发酵液;
(3)发酵液注入装有步骤(1)制备的含纳米氧化锌丝素纤维膜的培养器皿中,25~30℃静置培养9天;将发酵产物采用去离子水彻底清洗,60℃烘干,即可得到细菌纤维素/丝素纤维复合水凝胶面膜基材。
水凝胶面膜制备:
将1g甘蔗提取物、0.5g透明质酸钠、8g 甘油、3 mL积雪草提取物、0.5g丝素蛋白、去离子水97 mL混合均匀,γ-射线辐射灭菌后,得到美颜溶液;将剪裁后的细菌纤维素/蚕丝纤维复合水凝胶面膜基材浸入美颜溶液,充分浸泡后,及可获得水凝胶面膜。
实施例4
一种水凝胶面膜基材,与实施例1步骤相近,按以下步骤制备:
(1)1g纳米氧化锌ZnO NPs溶解于浓度40%的丝素蛋白SF溶液中,于60℃混合均匀,得到浓缩的纺丝液;将混合均匀的纺丝液注入10 mL注射器,设定静电纺丝的电压为18 kv,以流速0.25 mL/h、在15 cm接受距离内进行静电纺丝,得到含纳米氧化锌丝素纤维膜;所得的丝素纤维膜121℃、0.07~0.1 MPa高温高压灭菌20min,烘干备用;
(2)甘蔗渣经水解、脱毒后,加入1%酵母浸膏和0.5%胰蛋白胨,121℃、0.07~0.1MPa高温高压灭菌20min,作为培养基;随后以8%接种量接种醋酸杆菌于培养基中,混合均匀后,得到发酵液;
(3)发酵液注入装有步骤(1)制备的含纳米氧化锌丝素纤维膜的培养器皿中,25~30℃静置培养9天;将发酵产物采用去离子水彻底清洗,60℃烘干,即可得到细菌纤维素/丝素纤维复合水凝胶面膜基材。
水凝胶面膜制备:
将0.5g甘蔗提取物、1g透明质酸钠、8g 甘油、3 mL积雪草提取物、0.5g丝素蛋白、去离子水97 mL混合均匀,γ-射线辐射灭菌后,得到美颜溶液;将剪裁后的细菌纤维素/蚕丝纤维复合水凝胶面膜基材浸入美颜溶液,充分浸泡后,及可获得水凝胶面膜。
Claims (9)
1.一种水凝胶面膜基材的制备方法,其特征在于,采用静电纺丝的方式制备含抗菌纳米颗粒的丝素纤维膜,随后丝素纤维与甘蔗水解液共发酵,使细菌纤维与丝素纤维复合交联,获得复合水凝胶面膜,包括以下步骤:
(1)0.1~1g纳米氧化锌ZnO NPs溶解于20%~50%浓度的丝素蛋白SF溶液,于60℃混合均匀,得到浓缩的纺丝液;将混合均匀的纺丝液注入10 mL注射器,设定静电纺丝的电压为15~20 kv,以流速0.1~0.5 mL/h、在12~20 cm接受距离内进行静电纺丝,得到含纳米氧化锌丝素纤维膜;所得的丝素纤维膜121℃、0.07~0.1 MPa高温高压灭菌20min,烘干备用;
(2)甘蔗渣经水解、脱毒后,加入1%酵母浸膏和0.5%胰蛋白胨,121℃、0.07~0.1 MPa高温高压灭菌20min,作为培养基;随后以8~12%接种量接种醋酸杆菌于培养基中,混合均匀后,得到发酵液;
(3)发酵液注入装有步骤(1)制备的丝素纤维膜的培养器皿中,25~30℃静置培养6~10天;将发酵产物采用去离子水彻底清洗,60℃烘干,即可得到细菌纤维素/丝素纤维复合水凝胶面膜基材。
2.根据权利要求1所述水凝胶面膜基材的制备方法,其特征在于:所述的丝素纤维膜厚度0.1~0.2 mm,纤维间隙范围为20~100 µm。
3.根据权利要求1或2所述水凝胶面膜基材的制备方法,其特征在于:按以下步骤制备:
(1)0.5g纳米氧化锌ZnO NPs溶解于浓度35%的丝素蛋白SF溶液中,于60℃混合均匀,得到浓缩的纺丝液;将混合均匀的纺丝液注入10 mL注射器,设定静电纺丝的电压为18 kv,以流速0.25 mL/h、在18 cm接受距离内进行静电纺丝,得到含纳米氧化锌丝素纤维膜;所得的丝素纤维膜121℃、0.07~0.1 MPa高温高压灭菌20min,烘干备用;
(2)甘蔗渣经水解、脱毒后,加入1%酵母浸膏和0.5%胰蛋白胨,121℃、0.07~0.1 MPa高温高压灭菌20min,作为培养基;随后以10%接种量接种醋酸杆菌于培养基中,混合均匀后,得到发酵液;
(3)发酵液注入装有步骤(1)制备的含纳米氧化锌丝素纤维膜的培养器皿中,25~30℃静置培养8天;将发酵产物采用去离子水彻底清洗,60℃烘干,即可得到细菌纤维素/丝素纤维复合水凝胶面膜基材。
4.根据权利要求1或2所述水凝胶面膜基材的制备方法,其特征在于:按以下步骤制备:
(1)1g纳米氧化锌ZnO NPs溶解于浓度40%的丝素蛋白SF溶液中,于60℃混合均匀,得到浓缩的纺丝液;将混合均匀的纺丝液注入10 mL注射器,设定静电纺丝的电压为18 kv,以流速0.25 mL/h、在18 cm接受距离内进行静电纺丝,得到含纳米氧化锌丝素纤维膜;所得的丝素纤维膜121℃、0.07~0.1 MPa高温高压灭菌20min,烘干备用;
(2)甘蔗渣经水解、脱毒后,加入1%酵母浸膏和0.5%胰蛋白胨,121℃、0.07~0.1 MPa高温高压灭菌20min,作为培养基;随后以10%接种量接种醋酸杆菌于培养基中,混合均匀后,得到发酵液;
(3)发酵液注入装有步骤(1)制备的含纳米氧化锌丝素纤维膜的培养器皿中,25~30℃静置培养9天;将发酵产物采用去离子水彻底清洗,60℃烘干,即可得到细菌纤维素/丝素纤维复合水凝胶面膜基材。
5.根据权利要求1或2所述水凝胶面膜基材的制备方法,其特征在于:按以下步骤制备:
(1)1g纳米氧化锌ZnO NPs溶解于浓度40%的丝素蛋白SF溶液中,于60℃混合均匀,得到浓缩的纺丝液;将混合均匀的纺丝液注入10 mL注射器,设定静电纺丝的电压为18 kv,以流速0.25 mL/h、在15 cm接受距离内进行静电纺丝,得到含纳米氧化锌丝素纤维膜;所得的丝素纤维膜121℃、0.07~0.1 MPa高温高压灭菌20min,烘干备用;
(2)甘蔗渣经水解、脱毒后,加入1%酵母浸膏和0.5%胰蛋白胨,121℃、0.07~0.1 MPa高温高压灭菌20min,作为培养基;随后以8%接种量接种醋酸杆菌于培养基中,混合均匀后,得到发酵液;
(3)发酵液注入装有步骤(1)制备的含纳米氧化锌丝素纤维膜的培养器皿中,25~30℃静置培养9天;将发酵产物采用去离子水彻底清洗,60℃烘干,即可得到细菌纤维素/丝素纤维复合水凝胶面膜基材。
6.一种水凝胶面膜基材,根据权利要求1-3任一所述的方法制备得到的。
7.一种根据权利要求6所述水凝胶面膜基材在美颜中的应用,所制备的细菌纤维素/丝素纤维复合水凝胶面膜基材与美颜活性成分混合,制备具备美颜成分的水凝胶面膜;其中,美颜活性成分为:0.05~1g甘蔗提取物、0.1~1.5g透明质酸钠、2~10g 甘油、1~5 mL积雪草提取物、0.1~1g丝素蛋白、去离子水95~99 mL。
8.根据权利要求7所述水凝胶面膜基材在美颜中的应用,其特征在于:所述的细菌纤维素/丝素纤维复合水凝胶面膜厚度为0.5~5mm。
9.根据权利要求1所述水凝胶面膜基材在美颜中的应用,其特征在于:所述的美颜溶液,将0.5~1g 甘蔗提取物、0.5~1g 透明质酸钠、8g 甘油、3 mL积雪草提取物、0.5g丝素蛋白、去离子水97 mL混合均匀,γ-射线辐射灭菌后得到的。
