C. elegans of stage L4 were collected by exposing the nematodes to 100mg/mL of the M9 buffer solution of Platycodon grandiflorum extract for 30 minutes, then exposing the nematodes to LPS (100. mu.g/mL) for 24 hours, comparing the samples without the extract, after 24 hours, washing the individuals with M9 buffer, repeating this procedure twice, then transferring to NGM agar plates, then allowing them to acclimate for 30 minutes, and then performing survival analysis. All data were replicated 3 times, and the data were analyzed and graphed using GraphPad Prism 7. The results are shown in FIG. 1.
2. Effect of lipopolysaccharide on Switling Rate of caenorhabditis elegans in the Presence of Platycodon grandiflorum extract
C. elegans of the L4 stage of the synchronization were collected by exposing the nematodes to 100mg/mL of the M9 buffer solution of Platycodon grandiflorum extract for 30 minutes, then exposing the nematodes to LPS (100. mu.g/mL) for 24 hours, comparing samples without the extract, after 24 hours, washing the individuals with M9 buffer, repeating this procedure twice, then transferring to NGM agar plates, then allowing them to acclimate for 30 minutes, and then performing the swallowing rate analysis under the microscope. All data were replicated 3 times, and the data were analyzed and graphed using GraphPad Prism 7. The results are shown in FIG. 2.
3. Effect of lipopolysaccharide on the content of C.elegans malondialdehyde in the presence of Platycodon grandiflorum extract
C. elegans of stage L4 of the synchronization was collected by exposing the nematodes to 100mg/mL of the M9 buffer solution of Platycodon grandiflorum extract for 30 minutes, then exposing the nematodes to LPS (100. mu.g/mL) for 24 hours, comparing the samples without the extract, after 24 hours, washing the individuals with M9 buffer, repeating this step twice, collecting the nematodes and sonicating in ice-cold lysis buffer, and centrifuging to obtain the supernatant. The malonaldehyde measurement was performed using a malonaldehyde kit, according to the kit method. All data were replicated 3 times, and the data were analyzed and graphed using GraphPad Prism 7. The results are shown in FIG. 3.
4. Effect of lipopolysaccharide on glutathione content of caenorhabditis elegans in the presence of Platycodon grandiflorum extract
C. elegans of stage L4 of the synchronization was collected by exposing the nematodes to 100mg/mL of the M9 buffer solution of Platycodon grandiflorum extract for 30 minutes, then exposing the nematodes to LPS (100. mu.g/mL) for 24 hours, comparing the samples without the extract, after 24 hours, washing the individuals with M9 buffer, repeating this step twice, collecting the nematodes and sonicating in ice-cold lysis buffer, and centrifuging to obtain the supernatant. The content of the glutathione is measured by adopting a glutathione kit according to a kit method. All data were replicated 3 times, and the data were analyzed and graphed using GraphPad Prism 7. The results are shown in FIG. 4.
5. Biochemical indicator analysis of nematode
As shown in fig. 1, data statistics show that the 24-hour survival rate of nematodes in the LPS induction + platycodon grandiflorum extract treatment group is improved by 24% compared with that in a single LPS induction group; as shown in fig. 2, data statistics found that the 24-hour swallowing rate of the LPS-treated + platycodon extract-treated group was increased by 35% relative to that of the LPS-treated group alone; as shown in fig. 3, data statistics show that compared with a single LPS-induced group, the MDA content of nematodes in the lipopolysaccharide-induced + platycodon grandiflorum extract-treated group was reduced by 37%; as shown in fig. 4, data statistics show that the GSH content of the nematodes in the LPS-induced + platycodon grandiflorum extract-treated group was increased by 34.8% compared to the LPS-induced group alone. Statistical data show that the survival rate, the swallowing rate and the glutathione content of the nematodes treated by the platycodon grandiflorum extract are remarkably increased, the malondialdehyde content is remarkably reduced, the platycodon grandiflorum extract remarkably slows down the inflammation induction effect of lipopolysaccharide on the nematodes, and the platycodon grandiflorum extract has the effects of relieving and resisting inflammation.
Application example 1
A skin care emulsion containing platycodon grandiflorum extract comprises the following raw materials in percentage by mass:

The preparation method of the skin care emulsion comprises the following steps:
the method comprises the following steps: respectively weighing the phase A and the phase B, heating to 80 ℃, and stirring for dissolving;
step two: slowly add phase a to phase B, homogenize in a homogenizer at 13000rpm for 10min to complete emulsification, then continue mechanical stirring.
Step four: adding phase C when the temperature is reduced to 50 deg.C, stirring for 10min, and cooling to room temperature to obtain skin care emulsion.
Application example 2
A skin care cream containing platycodon grandiflorum extract comprises the following raw materials in percentage by mass:

The preparation method of the skin lotion comprises the following steps:
the method comprises the following steps: respectively weighing the phase A and the phase B, heating to 65 ℃, and uniformly stirring;
step two: add phase A to phase B, homogenize for 10min in a homogenizer at 13000rpm, then continue mechanical stirring.
Step four: adding the C phase when the temperature is reduced to 50 ℃, continuously stirring for 10min, and cooling to room temperature to obtain the skin lotion.
While the invention has been described with respect to a preferred embodiment, it will be understood by those skilled in the art that various changes in form and details may be made therein without departing from the spirit and scope of the invention.
Claims (10)
1. A preparation method of a platycodon grandiflorum extract with a soothing or anti-inflammatory effect is characterized by comprising the following steps:
step 1: sequentially cleaning fresh radix Platycodi, sun drying, pulverizing, and sieving to obtain radix Platycodi powder;
step 2: adding the platycodon grandiflorum powder into a solvent absolute ethyl alcohol or an ethanol water solution, and fully mixing to obtain a mixed solution;
and step 3: filtering the mixed solution after ultrasonic treatment, and collecting filtrate;
and 4, step 4: carrying out vacuum rotary evaporation on the filtrate to obtain a concentrated solution;
and 5: drying the concentrated solution to obtain radix Platycodi extract powder.
2. The method for preparing a platycodon grandiflorum extract having a soothing or anti-inflammatory effect according to claim 1, wherein the mixing ratio of the platycodon grandiflorum powder to the solvent in the step 2 is 1: 1-10.
3. The method for preparing platycodon grandiflorum extract having a soothing or anti-inflammatory effect according to claim 1, wherein the ultrasonic conditions in the step 3 are as follows: the temperature is 35-55 ℃, the power is 100-200W, the frequency is 30-60kHz, and the time is 25-35 min; the filtration is as follows: filtering with 0.4-0.5 μm microporous filter paper membrane.
4. The method for preparing platycodon grandiflorum extract having a soothing or anti-inflammatory effect according to claim 1, wherein the drying process parameters in the step 5 are as follows: the temperature is 30-50 ℃ and the time is 12-24 h.
5. Use of the platycodon grandiflorum extract prepared by the method for preparing a platycodon grandiflorum extract having a soothing or anti-inflammatory effect according to any one of claims 1 to 4 in cosmetics.
6. Use according to claim 5, comprising use in a model for the assessment of soothing or anti-inflammatory efficacy of a cosmetic product.
7. The use of claim 6, wherein the model for evaluating soothing or anti-inflammatory efficacy of a cosmetic is caenorhabditis elegans as a biological carrier, wherein the effective evaluation concentration of the Platycodon grandiflorum extract is 80-100 mg/mL.
8. Use according to claim 5, comprising the use in the preparation of a cosmetic product with soothing or anti-inflammatory effects.
9. Use according to claim 8, wherein the soothing or anti-inflammatory cosmetic agents include skin lotions, creams and lotions.
10. The use according to claim 9, wherein the platycodon grandiflorum extract is added in a concentration of 0.5 to 2 wt% in the cosmetic with soothing or anti-inflammatory effects.
