Abstract
本发明属于生物医学技术领域,公开了一种近红外荧光小分子探针及其合成方法与应用,所述近红外荧光小分子探针是IR‑780‑F。本发明的近红外荧光小分子探针是在IR‑780分子基础上衍生得来,保留IR‑780分子的基础结构,改变与N原子相连的烷基碳链末端,在末端添加‑CF3基团。IR‑780‑F在光稳定、靶向癌细胞和荧光成像等方面表现出优异的性能,毒性小,可用于靶向肿瘤组织、体内近红外荧光成像、进行光热治疗等,实现诊疗一体化。

技术领域
本发明属于生物医学技术领域,尤其涉及一种近红外荧光小分子探针及其 合成方法与应用。
背景技术
目前,手术切除、放疗、化疗是临床治疗癌症的重要手段,但这些方法都 有一定的局限性,疗效仍有待提高,急需发展治疗癌症的新方法、新药物。
近年来新兴的癌症治疗策略包括免疫治疗、基因治疗、光动力治疗、光热 治疗等。其中,光热治疗(photothermal therapy,PTT)由于其非侵入性、时空控 制精确、特异性强、肿瘤破坏效率高等优点,已成为肿瘤治疗领域的研究热点。 近红外光(Near infrared,NIR)具有良好的组织穿透能力,在时间和空间上具有 高分辨率的可调性,在光热治疗过程中,肿瘤部位的温度变化可由光热药物将 NIR光能转化为热能引起,42℃以上的温度即可有效杀死肿瘤细胞,并且肿瘤 细胞的耐热性比正常细胞差,升高温度可以杀死肿瘤细胞,从而避免对正常细 胞产生明显的副作用。
当基态的光热剂受到光源照射并跃迁至S1态,一部分会以辐射的形式跃迁 回到基态,另一部分则以热的形式将能量传递给周围环境回到基态。光热治疗 中的所有激光传输模式旨在均匀地提高肿瘤组织中温度,同时防止对周围健康 组织的损害。然而,由于肿瘤的有效消融需要破坏每个肿瘤细胞,光热治疗通 常需要肿瘤中心达到更高的温度(≥50℃),并且形成温度梯度以使肿瘤边缘 达到治疗温度。这就要求光热剂具备较高的光热转换效率和较好的肿瘤靶向性。
目前,研究者们已经开发了很多具有高光热转换效率的光热剂用于肿瘤光 热治疗,主要包括贵金属纳米材料、碳基纳米材料、金属和非金属化合物纳米 材料以及有机小分子染料等。纳米颗粒虽然具有良好的光热转化效率,但其在 体内不易降解,制备过程复杂、潜在的长期毒性、尺寸不均等问题大大限制了 纳米光热材料的进一步临床转化,而有机小分子染料类光热剂在以上方面则具 有天然的优势。
当前文献报道的有机小分子光热剂,大多需要借助“纳米化”的策略来辅 助发挥其光热治疗性能,然而这却又使得药物体系面临着“纳米材料”体系的 共同局限性,难以进一步开展临床转化。IR-780具有天然的肿瘤靶向性功能, 然而其光热性能尚有待提高,在荷瘤小鼠模型中的给药方式为瘤内注射给药。
本发明拟解决的科学问题:通过CF3-修饰显著提高IR-780分子光热转化效 果,明确其光热增强机制,采用向荷瘤小鼠尾静脉注射的方式给药实现靶向光 热治疗其肿瘤。
发明内容
针对现有技术存在的问题,本发明提供了一种近红外荧光小分子探针及其 合成方法与应用,尤其涉及一种用于靶向乳腺癌治疗的近红外荧光小分子探针 及其合成方法与应用。
本发明是这样实现的,一种近红外荧光小分子探针的合成方法,所述近红 外荧光小分子探针的合成方法包括以下步骤:
步骤一,在100mL三口瓶中加入四溴化碳CBr4和三苯基磷PPh3,置于冰 水混合物中,待溶液温度降至0℃,缓慢滴加3,3,3-三氟-1-丙醇试剂;
步骤二,滴加完后,反应液变为黄色粘稠液体;将反应装置移至室温后, 60℃加热回流1h;降温搭蒸馏装置,升温将产物蒸出,升温到100℃有液体蒸 出,继续升温,直至无液体蒸出,得到无色液体化合物1;
步骤三,将化合物1,2,3,3-三甲基-3H吲哚和碘化钾KI溶到乙腈中,在闷 罐中加热到150℃,搅拌过夜;
步骤四,使用薄板层析法判断反应程度,显示剩余原料余量较少,反应液 由浅黄色变成橙红色;将反应液过滤,用乙腈洗涤滤饼;滤液浓缩,经硅胶柱 层析得化合物2;
步骤五,将化合物2和2-氯-1-甲酰基-3-羟甲亚基环己烯溶到正丁醇/甲苯溶 液中,110℃搅拌过夜;
步骤六,使用薄板层析法判断反应程度,显示原料未剩余,并且有新点生 成;停止反应,将反应液浓缩干,经硅胶柱层析纯化得目标产物IR-780-F。
进一步,步骤一中,所述四溴化碳CBr4的添加量为13.95g,42.08mmol; 所述三苯基磷PPh3的添加量为11.04g,42.08mmol;所述3,3,3-三氟-1-丙醇的添 加量为4.0g,35.07mmol。
进一步,步骤二中,所述无色液体化合物1的质量为0.75g,12.1%。
进一步,步骤三中,所述化合物1的添加量为0.65g,4.24mmol;所述2,3,3- 三甲基-3H吲哚的添加量为0.45g,2.83mmol;所述碘化钾KI的添加量为0.47g, 2.83mmol;所述乙腈的体积为20mL。
进一步,步骤四中,所述薄板层析法的展开剂为石油醚/乙酸乙酯=10/1;所 述硅胶柱层析的条件为:石油醚:乙酸乙酯=v:v=100:1-5:1;所述化合物2 的质量为60mg,8.48%。
进一步,步骤五中,所述化合物2的添加量为0.02g,0.08mmol;所述2- 氯-1-甲酰基-3-羟甲亚基环己烯的添加量为0.01g,0.04mmol;所述正丁醇/甲苯 为7/3,2mL。

本发明的另一目的在于提供一种在靶向肿瘤组织、体内近红外荧光成像、 肿瘤光热以及靶向乳腺癌测试用试剂盒,所述试剂盒包含所述近红外荧光小分 子探针。

本发明提供的近红外荧光小分子探针,是在IR-780分子的基础上进行衍生 得来,保留IR-780分子的基础结构,改变与N原子相连的烷基碳链末端,在末 端添加-CF3基团,可以用于靶向肿瘤组织、体内近红外荧光成像、肿瘤光热治 疗,降低了毒性,增强了光热转换效率。
本发明创新性的用三氟甲基对IR-780分子进行修饰,发现修饰后的分子 IR-780-F具有比IR-780更加优异的光热剂性能,尤其是可通过向荷瘤小鼠进行 瘤内注射的给药方式,展现了较好的肿瘤光热治疗效果。其高肿瘤靶向性及高 光热性能,是本项目的创新之处,相比无肿瘤靶向性的光热药物分子,这种突 出的性质尤其有利于开展散在性、未检出的肿瘤的光热治疗研究。
本发明通过CF3-修饰,获得稳定性好、靶向性强、光热转换效率显著增高 的IR-780衍生物,为其在肿瘤的光热治疗应用进行前期基础研究,从而推动有 机小分子光热剂的进一步临床研究。

针对现有技术存在的问题,本发明提供了一种近红外荧光小分子探针及其 合成方法与应用,下面结合附图对本发明作详细的描述。
本发明实施例提供的近红外荧光小分子探针是IR-780-F。
如图1所示,本发明实施例提供的近红外荧光小分子探针的合成方法包括 以下步骤:
S101,在100mL三口瓶中加入四溴化碳CBr4和三苯基磷PPh3,置于冰水 混合物中,待溶液温度降至0℃,缓慢滴加3,3,3-三氟-1-丙醇试剂;
S102,滴加完后,反应液变为黄色粘稠液体;将反应装置移至室温后,60℃ 加热回流1h;降温搭蒸馏装置,升温将产物蒸出,升温到100℃有液体蒸出, 继续升温,直至无液体蒸出,得到无色液体化合物1;
S103,将化合物1,2,3,3-三甲基-3H吲哚和碘化钾KI溶到乙腈中,在闷罐 中加热到150℃,搅拌过夜;
S104,使用薄板层析法判断反应程度,显示剩余原料余量较少,反应液由 浅黄色变成橙红色;将反应液过滤,用乙腈洗涤滤饼;滤液浓缩,经硅胶柱层 析得化合物2;
S105,将化合物2和2-氯-1-甲酰基-3-羟甲亚基环己烯溶到正丁醇/甲苯溶液 中,110℃搅拌过夜;
S106,使用薄板层析法判断反应程度,显示原料未剩余,并且有新点生成; 停止反应,将反应液浓缩干,经硅胶柱层析纯化得目标产物IR-780-F。
下面结合具体实施例对本发明的技术方案作进一步描述。
实施例1
1、发明概述
本发明涉及一种用于靶向乳腺癌治疗的近红外荧光小分子探针及其双模态 成像和光热治疗。本发明提供的用于双模态成像及治疗的近红外小分子探针是 在IR-780分子的基础上进行衍生得来,保留IR-780分子的基础结构,改变与N 原子相连的烷基碳链末端,在末端添加-CF3基团,可以用于靶向肿瘤组织、体 内近红外荧光成像、肿瘤光热治疗,增强光热转换效率。
2、设计思路
经验表明,在医药方面,当氟原子或含氟基团(尤其是CF3基团)引入化合物 中,其电效应和模拟效应改变了分子内部电子密度的分布,影响了化合物内部 结构的酸碱性,进而改变了其活性,而且还能提高化合物的脂溶性。氟原子取 代了化合物中的氢原子,其类酯化合物在生物膜上的溶解性得到了增强,促进 其在生物体内吸收的传递速度,使生理作用发生变化。所以不少含氟化合物比 不含氟化合物在医药、农药等药物性能上具有用量少、毒性低、药效高、代谢 能力强的优点。
对IR-780分子进行结构修饰,容易使得分子靶向性降低,为了保留分子靶向 性,本发明仅用化学活性较为惰性的三氟甲基替代了分子中原有的甲基,且C-F 键的键长与C-H键的键长相比增加很小,分子的几何构型极为相像。在分子结构 中,保留了IR-780分子的基础结构,改变与N原子相连的烷基链末端,添加-CF3基团,获得IR-780衍生物IR-780-F,为其在肿瘤的光热治疗应用进行前期基础研 究,以期推动有机小分子光热剂的进一步临床研究。
2.1合成步骤
IR-780-F分子探针的合成步骤如图2所示,具体步骤如下:
在100mL三口瓶中加入四溴化碳(CBr4)(13.95g,42.08mmol)和三苯基 磷(PPh3)(11.04g,42.08mmol)。将其置于冰水混合物中,待溶液温度降至0℃, 然后缓慢滴加3,3,3-三氟-1-丙醇(4.0g,35.07mmol)试剂。滴加完之后,反应 液变为黄色粘稠液体。将反应装置移至室温,然后60℃加热回流1h。降温搭蒸 馏装置,升温将产物蒸出,升温到100℃左右有液体蒸出,继续升温,直至无液 体蒸出。最后得到无色液体化合物1(0.75g,12.1%)。
将化合物1(0.65g,4.24mmol),2,3,3-三甲基-3H吲哚(0.45g,2.83mmol) 和碘化钾(KI)(0.47g,2.83mmol)溶到乙腈(20mL)中,在闷罐中加热到150℃, 搅拌过夜。使用薄板层析法判断反应程度,展开剂为石油醚/乙酸乙酯=10/1,显 示剩余原料余量较少,反应液由浅黄色变成橙红色。将反应液过滤,用乙腈洗 涤滤饼。滤液浓缩,经硅胶柱层析(石油醚:乙酸乙酯=v:v=100:1-5:1)得 化合物2(60mg,8.48%)。
将化合物2(0.02g,0.08mmol)和2-氯-1-甲酰基-3-羟甲亚基环己烯(0.01g,0.04mmol)溶到正丁醇(n-BuOH)/甲苯(toluene)(7/3,2mL)溶液中,110℃ 搅拌过夜。使用薄板层析法判断反应程度,展开剂为石油醚/乙酸乙酯=10/1,显 示原料未剩余,并且有新点生成。停止反应,将反应液浓缩干,经硅胶柱层析 纯化(石油醚:乙酸乙酯=v:v=1:1)得目标产物IR-780-F。
2.2分子光谱测试
称取一定质量的IR-780-F溶解在DMSO中,取定量溶解液加入到HEPES 缓冲液中配成浓度为10.0μM(含10%DMSO)的溶液并测试样品的吸收光谱。 采用Agilent的Uv-Vis分光光度计获得紫外吸收光谱,扫描波长从500~900nm, 扫速为1002nm/min。再取定量溶解液加入到甲醇、乙腈、PBS缓冲液、HEPES 缓冲液、牛血清、成人血清中,使其终浓度至10.0μM(含10%DMSO),测试 样品的发射光谱。采用型号F-4600,Hitachi的FT-IR光谱仪获得FT-IR发射光 谱,扫描波长从750~900nm,激发波长:λex=740nm,激发狭缝宽度5nm,发射 狭缝宽度5nm。
2.3 IR-780-F分子的体外光热效率
为了评估IR-780及IR-780-F溶液的体外产热能力,使用溶解在DMSO中 的10mMIR-780-F溶液,取一定量用PBS稀释至0.2mg/mL、0.4mg/mL、 0.8mg/mL、1.2mg/mL并对这些溶液及PBS进行评估,IR-780溶液的配制同上。
使用红外热像仪(FLIR-E6)记录IR-780-F和PBS的温度变化及IR-780和PBS 的温度变化。将PBS作为对照、0.2mg/mLIR-780-F溶液和IR-780溶液、 0.4mg/mLIR-780-F溶液和IR-780溶液、0.8mg/mLIR-780-F溶液和IR-780溶液、 1.2mg/mLIR-780-F溶液和IR-780溶液暴露于808nm激光照射,功率密度为 2.0W/cm2用于测量并记录温度变化。
3、细胞实验
3.1细胞毒性体外评估
用CCK-8实验测定浓度为10-60μM的IR-780-F及IR-780对4T1细胞的毒 性。收集已孵育好的4T1细胞进行离心,将其制成细胞悬浮液,并调整细胞浓 度至8×104个/mL。将4T1细胞转移到96孔板中,每孔加入100μL,使得待测 细胞的密度约为8000/孔,铺设两个96孔板用于实验。将接种好待测细胞的96 孔板放入37℃恒温培养箱中培养12h,一个板加入浓度梯度为10-60μM的 IR-780-F孵育15min 1h。一个板加入浓度梯度为10-60μM的IR-780孵育1h。 孵育时间结束,立即向每孔加入CCK-8试剂(10μL,5mg/mL,即0.5%CCK-8), 继续孵育细胞40min。最后使用酶标仪测定各孔的吸光度强度,以对照组细胞的 存活率为100%,试验组的细胞存活数值为对照组的百分比。
4、动物实验
4.1肿瘤模型
将孵育好的4T1细胞进行离心收集,PBS缓冲溶液清洗3次后制成细胞悬 浮液,细胞计数仪计数,用PBS缓冲溶液稀释成所需浓度。经过预处理的4T1 细胞(约1×106个/100μL)皮下注射到BALB/c Nude小鼠的右腿后部,大约一 周后可获得肿瘤模型小鼠。
4.2 IR-780-F在荷瘤小鼠中的体内成像
为了确定使用IR-780-F进行体内光热治疗的最佳时间,本发明通过整个动 物NIR成像系统检查了IR-780-F在荷瘤小鼠中的组织渗透性和分布。使用PBS 缓冲溶液配制IR-780-F的注射液(含12.4%DMSO),当小鼠肿瘤生长至一定体 积后,尾静脉注射4mg/kgIR-780-F到荷瘤小鼠体内。将麻醉后的裸鼠置于活体 成像仪器,分别在0h,2h,4h,8h,12h,24h,48h,72h,96h,120h对其进 行荧光成像。5天后将裸鼠解剖,取出小鼠包括肠,肺,脾,肾,心脏,胃和肿 瘤部分在内的主要器官,将各器官置于活体成像系统中观察其荧光分布情况。
4.3 IR-780-F在肿瘤部位的光热效率
使用PBS缓冲溶液配制IR-780-F的注射液(含12.4%DMSO),进行瘤内 注射。12个小时后,将小鼠进行麻醉,使用光热成像仪对其成像,记录温度。 然后用激光照射小鼠肿瘤部位4分钟,再对其肿瘤部位进行光热成像,记录肿 瘤部位的温度。
4.4光热治疗效果
小鼠荷瘤成功后,选取肿瘤平均直径约为10×10mm的小鼠,随机分为四 组(每组5只小鼠):(1)对照组:瘤内注射0.1mL生理盐水;(2)激光组:瘤内 注射0.1mL生理盐水后行激光照射)808nm,2.0W/cm2,4min);(3)IR-780-F 组:只注射0.1mL即80μgIR-780-F溶液于肿瘤内,不进行激光照射;(4)IR-780-F +激光组:瘤内注射80μgIR-780-F溶液后行激光照射(808nm,2.0W/cm2,4min)。 将小鼠给予药物后麻醉,将小鼠肿瘤区域放置于激光下照射。在激光照射小鼠 肿瘤时,同时用红外热像仪测量激光照射时肿瘤区域的温度并记录。
5、结果与讨论
5.1 IR-780-F在不同溶剂中的光谱性质
针对IR-780-F的光物理性质进行了实验。如图3所示,本发明可以从吸收 光谱看出IR-780-F在HEPES中的最大吸收波长为773nm,从发射光谱可以看 出IR-780-F在HEPES、MeOH、FBS、HAS和MeCN中的最大发射波长分别为 804nm、814nm、820nm、818nm、816nm,发射波长均位于近红外光谱区段。 经计算,斯托克位移值即最大发射波长与最大吸收波长的差值为38nm,更有利 于IR-780-F在生物体中产生的荧光与背景荧光、激发波长分离,避免显影中的 背景干扰信号,提高荧光成像的灵敏度。IR-780-F无论在牛血清或成人血清中, 它的发射波长较于在其他溶剂中有红移的现象,这可能是因为血清中的某些蛋 白与IR-780-F结合,使得分子的光谱特性有所改变。
5.3 IR-780-F的体外光热效率
为了研究IR-780-F作为光热剂的潜力,以IR-780的光热转换效果进行对照, 系统研究了IR-780-F的光热性能,。因此,记录了808nm激光照射下IR-780-F 溶液及IR-780溶液各自的温度变化。图4A为在808nm激光照射下IR-780的光 热曲线,IR-780溶液的温度以浓度依赖的方式迅速升高,最高温度为69.3℃。 如图4B所示,在808nm激光照射下,IR-780-F溶液的温度以浓度依赖的方式 迅速升高。在功率为2.0W/cm2的808nm激光照射下,不同浓度的IR-780-F溶 液(Control、0.2mg/mL、0.4mg/mL、0.8mg/mL、1.2mg/mL)的光热曲线。IR-780-F 的温度以浓度依赖的方式升高到最大值69.3℃,而PBS的温度没有表现出明显 的升高。因此,可以得出结论,本发明设计的IR-780-F分子是一种有效的PTT 光热转换剂,比IR-780有着更好的光热转换效率。
5.4 IR-780-F的细胞毒性
对于靶向肿瘤细胞的有机小分子显影剂,它对癌细胞的毒性大小也是重要 的特性之一。用CCK-8法检测不同浓度IR-780-F对4T1细胞的毒性,以IR-780 作为对照。如图5所示,从整体来看,在10~60μM浓度范围内,IR-780或IR-780-F 孵育1小时候,4T1细胞的细胞活力都随着探针浓度的增加而减小。
5.5 IR-780-F在小鼠体内分布情况
为了确定IR-780-F在进行小鼠光热治疗的最佳时机,本发明通过近红外活 体成像成像技术检测IR-780-F在荷瘤小鼠体内的组织渗透性和药物分布情况。 IR-780-F注射液经尾静脉注射至4T1肿瘤细胞荷瘤裸鼠模型中,间隔不同时间 段对小鼠进行麻醉,置于小动物活体成像系统中观察体内近红外荧光分布情况。 如图6所示,随着时间延长,肿瘤区域的荧光信号逐渐增加,前8h注入的IR-780-F 主要集中小鼠的心、肺、胸腔部位。随着时间的增加,IR-780-F在体内多次循 环,逐渐在肿瘤部位累积,肿瘤部位荧光强度逐渐增加,明显高于其他器官组 织。5天后去除背景荧光,发现小鼠体内荧光主要集中在肿瘤部位,其它位置几 乎无荧光。实验结果说明IR-780-F具有显著的肿瘤靶向积聚特性。
根据实验需求及动物处理准则,安乐死小鼠后,将其解剖并分离其各主要 器官和肿瘤。荧光主要聚集在肿瘤部位,肺部有弱荧光,可能是由于少许IR-780-F 聚集在肺部,没有完全代谢。解剖后的实验结果表明与体内荧光成像结果相一 致,进一步说明IR-780-F具有靶向肿瘤部位的特性,且在肿瘤处积聚,不易代 谢出体外,适于进行肿瘤部位的近红外荧光成像。
5.6 IR-780-F的体内光热效率
如图7所示,用红外热成像相机记录肿瘤组织中的热成像图像和温度变化。 近红外激光照射后,IR-780-F+激光治疗组肿瘤表面温度显着升高至52.9±0.2℃, 但PBS+激光组肿瘤表面温度略微升高至35.9±2.7℃。IR-780-F在暴露于NIR激 光照射后在体内诱导高光热效率,因此表明它们在实现体内PTT方面具有很大 的前景。
5.7 IR-780-F的体内治疗效果
本发明检测IR-780-F作为光热剂的肿瘤治疗效果。图8A显示为四组荷瘤小 鼠在实验处理后0天、7天、12天的肿瘤拍照图。图8B为四组肿瘤生长曲线图。 从图中得知:小鼠在IR-780-F加上激光照射时,肿瘤生长收到明显的抑制 (P<0.01),在光热治疗的第12天肿瘤大小的平均值是93.614±10.753mm3。相比 之下,IR-780-F组和单独激光组和实验对照PBS组相比,并没有表现出明显的抑 制肿瘤生长的效果(P>0.05),三组在第12天肿瘤大小分别为559.494±49.427mm3、 563.702±106.947mm3、694.046±48.45mm3,在本发明的实验过程中并没有观察到 四组小鼠体重的明显改变(P>0.05)(见图8C)。图8D所有小鼠在12天时处死获 取肿瘤后,用电子天平进行称重,可以发现IR-780-F加激光照射组的肿瘤体积明显小于其他三组,此外图8E的肿瘤解剖直观图也说明了这一点。
实施例2
肿瘤模型建立成功的荷瘤裸鼠,在尾静脉注射0.1mLIR-780-F溶液96h后进 行18F-FDGmicro-PET以及CT扫描。扫描前裸鼠禁食12h,正常饮水。扫描当 天,从裸鼠尾静脉快速注射400uCi18F-FDG。代谢30分钟后,2%异氟烷诱导麻 醉裸鼠,并将裸鼠固定于小动物检查床,调整裸鼠位置确定位于扫描线圈正中 后扫描10分钟。扫描期间以1.5%异氟烷维持麻醉。实验结果如图9~15所示。

Claims (10)
1.一种近红外荧光小分子探针在制备靶向肿瘤组织、体内近红外荧光成像、肿瘤光热治疗药物上的用途。
3.一种在靶向肿瘤组织、体内近红外荧光成像、肿瘤光热以及靶向乳腺癌测试用试剂盒,其特征在于,所述试剂盒包含权利要求1~2任意一项所述近红外荧光小分子探针。
4.一种近红外荧光小分子探针的合成方法,其特征在于,所述近红外荧光小分子探针的合成方法包括以下步骤:
步骤一,在100mL三口瓶中加入四溴化碳CBr4和三苯基磷PPh3,置于冰水混合物中,待溶液温度降至0℃,缓慢滴加3,3,3-三氟-1-丙醇试剂;
步骤二,滴加完后,反应液变为黄色粘稠液体;将反应装置移至室温后,60℃加热回流1h;降温搭蒸馏装置,升温将产物蒸出,升温到100℃有液体蒸出,继续升温,直至无液体蒸出,得到无色液体化合物1;
步骤三,将化合物1,2,3,3-三甲基-3H吲哚和碘化钾KI溶到乙腈中,在闷罐中加热到150℃,搅拌过夜;
步骤四,使用薄板层析法判断反应程度,显示剩余原料余量较少,反应液由浅黄色变成橙红色;将反应液过滤,用乙腈洗涤滤饼;滤液浓缩,经硅胶柱层析得化合物2;
步骤五,将化合物2和2-氯-1-甲酰基-3-羟甲亚基环己烯溶到正丁醇/甲苯溶液中,110℃搅拌过夜;
步骤六,使用薄板层析法判断反应程度,显示原料未剩余,并且有新点生成;停止反应,将反应液浓缩干,经硅胶柱层析纯化得目标产物IR-780-F。
5.如权利要求4所述的近红外荧光小分子探针的合成方法,其特征在于,步骤一中,所述四溴化碳CBr4的添加量为13.95g,42.08mmol;所述三苯基磷PPh3的添加量为11.04g,42.08mmol;所述3,3,3-三氟-1-丙醇的添加量为4.0g,35.07mmol。
6.如权利要求4所述的近红外荧光小分子探针的合成方法,其特征在于,步骤二中,所述无色液体化合物1的质量为0.75g,12.1%。
7.如权利要求4所述的近红外荧光小分子探针的合成方法,其特征在于,步骤三中,所述化合物1的添加量为0.65g,4.24mmol;所述2,3,3-三甲基-3H吲哚的添加量为0.45g,2.83mmol;所述碘化钾KI的添加量为0.47g,2.83mmol;所述乙腈的体积为20mL。
8.如权利要求4所述的近红外荧光小分子探针的合成方法,其特征在于,步骤四中,所述薄板层析法的展开剂为石油醚/乙酸乙酯=10/1;所述硅胶柱层析的条件为:石油醚:乙酸乙酯=v:v=100:1-5:1;所述化合物2的质量为60mg,8.48%。
9.如权利要求4所述的近红外荧光小分子探针的合成方法,其特征在于,步骤五中,所述化合物2的添加量为0.02g,0.08mmol;所述2-氯-1-甲酰基-3-羟甲亚基环己烯的添加量为0.01g,0.04mmol;所述正丁醇/甲苯为7/3,2mL。
10.如权利要求4所述的近红外荧光小分子探针的合成方法,其特征在于,步骤六中,所述薄板层析法的展开剂为石油醚/乙酸乙酯=10/1,所述硅胶柱层析纯化的条件为石油醚:乙酸乙酯=v:v=1:1。
