CN1686337A discloses an antidepressant Chinese medicinal preparation, in particular to a Chinese medicinal preparation prepared from white peony root, hypericum perforatum, acanthopanax root, schisandra fruit, cortex albiziae, polygala root, morinda officinalis, caltrop and other Chinese medicaments. CN1381242A discloses a capsule for treating depression, which is a new traditional Chinese medicine preparation for treating depression and consists of two traditional Chinese medicines of hypericum perforatum and acanthopanax. The medicine is prepared through the processes of medicine screening, processing, extracting, mixing, subpackaging and warehousing. The medicine has advantages of sufficient raw materials, simple preparation method, and convenient administration, and can be used for treating depression, clearing heat and purging fire, resolving stagnation and tranquilization, invigorating kidney and invigorating spleen, and has advantages of reliable curative effect, low side effect, and few contraindications.
None of the above patent documents adopts fermentation technology, and the taste of the Chinese medicinal preparation is poor and the biotransformation and utilization of the active ingredients of the Chinese medicinal preparation are relatively low.
Disclosure of Invention
Aiming at the defects in the prior art, the invention provides the health food with the functions of resisting anxiety and depression, which has high safety, small dosage and definite curative effect, generates synergistic action among the components and can be used as daily health food for people with mild and moderate depression.
A health food with an anxiolytic and antidepressant function comprises the following components in parts by weight: 20-30 parts of first composite fermentation powder and 10-20 parts of second composite fermentation powder;
the first compound fermentation powder is prepared by mixing and solid-state fermenting radix astragali, cortex Magnoliae officinalis, Atractylodis rhizoma, Poria, radix Angelicae sinensis, radix aucklandiae, cortex et radix Polygalae, arillus longan, semen Ziziphi Spinosae, fructus Jujubae, fructus Citri Sarcodactylis, and fructus Citri;
the second compound fermentation powder is prepared by mixing and fermenting rosemary, sage, saffron, basil, valerian root, passion flower, chamomile and balm.
The above components are all dry products with moisture content not more than 10 wt%.
The invention has the beneficial effects that: animal experiments prove that the formulation of the invention can reduce the immobility time of tail suspension and forced swimming of mice through the synergistic effect of the components, can be applied to health food with the functions of resisting anxiety and depression, and can improve the health condition of organisms. In addition, the formula of the invention is high in safety and small in dosage, and can be used as a daily health food for people with mild and moderate depression.
Preferably, the first composite fermentation powder is 28 parts, and the second composite fermentation powder is 15 parts.

(c) sterilizing the fermentation product obtained in the step (b) at the temperature of 130-135 ℃ for 5-10s, filtering, taking the filtrate, and drying to obtain the second composite fermentation powder.
Still further, in the step (b), the lactobacillus acidophilus colony content in the fermentation system is 9 × 106CFU/mL, the colony content of Lactobacillus reuteri is 6.5X 106CFU/mL, the bacterial colony content of the lactobacillus plantarum is 8 multiplied by 106CFU/mL。
Still further, the water used in the preparation is purified water.
Furthermore, the daily dosage of the health food composition is 2.1-3.3 g.
Furthermore, the obtained health food composition can be combined with auxiliary materials to be processed into various clinically acceptable dosage forms, such as powder, tablets, granules or hard capsules.

(3) the second composite fermentation powder adopts a liquid composite fermentation process, so that the fermentation efficiency is improved, the operation is easy, and the quality of the product is effectively guaranteed. The active ingredients are enriched, the anxiolytic and antidepressant effects are enhanced, and meanwhile, the effects of antioxidation and the like are achieved, free radicals can be effectively eliminated, and the effects of beautifying, regulating and the like are achieved.
Detailed Description
The principles and features of this invention are described below in conjunction with examples, which are set forth only to illustrate the invention and are not intended to limit the scope of the invention, the embodiments being described in parts by weight.
Example 1
A health food with an anxiolytic and antidepressant function comprises the following components in parts by weight: 28 parts of first composite fermentation powder and 15 parts of second composite fermentation powder;
the preparation method comprises the following steps:
1. preparing the first compound fermentation powder:
(1) accurately weighing 3 parts of astragalus, magnolia officinalis, bighead atractylodes rhizome, poria cocos, angelica sinensis, elecampane, polygala tenuifolia, arillus longan, spina date seed, jujube, fingered citron and citron, 1.5 parts of the astragalus, magnolia officinalis, bighead atractylodes rhizome, poria cocos, angelica sinensis, elecampane, polygala tenuifolia, 1.5 parts of the arillus longan, 0.5 part of the jujube, 0.5 part of the fingered citron and 1.5 parts of the citron respectively; pulverizing accurately weighed radix astragali, cortex Magnolia officinalis, Atractylodis rhizoma, Poria, radix Angelicae sinensis, radix aucklandiae, cortex et radix Polygalae, arillus longan, semen Ziziphi Spinosae, fructus Jujubae, fructus Citri Sarcodactylis, and fructus Citri, and sieving with 40 mesh sieve; proportionally adding into a fermentation tank, adding a certain amount of water, uniformly mixing, and sterilizing with high-pressure steam;
(2) and (2) cooling the product obtained in the step (1), and then inoculating a zymocyte powder solution, wherein the ratio of the bacillus subtilis to the saccharomyces cerevisiae to the lactobacillus acidophilus is 1: 2: 2, the inoculation amount is 8 percent, and the mixture is uniformly mixed and fermented for 7 days at the temperature of 35 ℃ to obtain a fermentation product;
(3) and (3) drying and crushing the fermentation product obtained in the step (2) at 50 ℃, and sieving the fermentation product with a 40-mesh sieve to obtain the first compound fermentation powder.
2. Preparing a second compound fermentation powder:
(a) accurately weighing 1 part of rosemary, 1.5 parts of sage, 1 part of saffron crocus, 1 part of basil, 1 part of valerian root, 1.5 parts of passion flower, 1.5 parts of chamomile and 1 part of balm; pulverizing herba Rosmarini officinalis, herba Salvia officinalis, stigma croci Sativi, herba Ocimi, rhizoma et radix Valerianae, herba Passiflorae Caeruleae, flos Matricariae Chamomillae, and herba Melissae axillaris, sieving with 30 mesh sieve, adding into fermentation tank, adding water, mixing, sterilizing with high pressure steam, wherein the mass ratio of total dry matter raw materials to water is 1: 30, of a nitrogen-containing gas;
(b) adjusting the pH of the product obtained in step (a) to 6.5, adding 9X 10 colony6CFU/mL Lactobacillus acidophilus with colony content of 6.5 × 106CFU/mL Lactobacillus reuteri and colony content of 8X 106Carrying out compound fermentation on the CFU/mL lactobacillus plantarum at the temperature of 37 ℃ for 96 hours to obtain a fermentation product;
(c) and (c) sterilizing the fermentation product obtained in the step (b) for 10s at 130 ℃, filtering, taking the filtrate, and drying to obtain the second compound fermentation powder.
3. Mixing materials:
and taking 28kg of the first composite fermentation powder and 15kg of the second composite fermentation powder obtained by the preparation method, wherein the raw materials are undersize materials which are sieved by a 40-mesh sieve, and obtaining mixed powder.
4. Preparing granules:
and (3) adding 25kg of isomaltose hypgather and 32kg of L-arabinose into the mixed powder obtained in the step (3), mixing, granulating, drying, subpackaging and preparing into clinically usable granules.
Example 2
A health food with an anxiolytic and antidepressant function comprises the following components in parts by weight: 25 parts of first composite fermentation powder and 18 parts of second composite fermentation powder;
the preparation method comprises the following steps:
1. preparing the first compound fermentation powder: the same as in example 1.
2. Preparing a second compound fermentation powder: the same as in example 1.
3. Mixing materials:
taking 25kg of the first composite fermentation powder obtained by the preparation method and 18kg of the second composite fermentation powder, wherein the raw materials are undersize materials which are sieved by a 40-mesh sieve, and obtaining mixed powder.
4. Preparing granules: the same as in example 1.
Experiment 1: animal experiments
The beneficial effects of the present invention are further illustrated by animal experiments as follows.
1. Sample preparation
Sample 1: the mixed powder prepared in example 1 (obtained in step 3);
sample 2: the first composite fermentation powder prepared in the example 1;
sample 3: the second composite baking powder prepared in example 1;
2. experimental materials and methods
2.1 test animals
C57BL/6 mouse, male, body weight 20-24 g. Free diet drinking water, feeding environment temperature (25 +/-1) DEG C, humidity (60 +/-5)%, 12h light and shade alternation, and random grouping after 5 days of adaptive feeding, wherein each group contains 10 animals. All animal behavioral experiments were performed at 13:00-15:00 and gavage administered at 10ml/kg body weight.
2.2 Tail suspension experiment
Mice were modeled as 5 groups of 10 mice each by subcutaneous injection of corticosterone (10 mg/kg). The experimental drug groups are 1-3 groups of samples, all the samples are administrated according to 2.1g/kg, the positive drug fluoxetine is administrated according to 10mg/kg, the control group is administrated with the same amount of normal saline, the intragastric administration is carried out once a day, the drug administration is continuously carried out for 14d, the 2cm part of the tail end of the animal is attached to a horizontal support after the last administration for 1h, the animal is in an inverted hanging state, the support is placed in an open box, and the head part of the support is 5cm away from the bottom surface. The immobility time of the mice was recorded within 6 min. Each group of mice was operated in parallel and statistically analyzed using animal behavioristics experimental analysis software.
2.3 forced swimming test
Mice were grouped and dosed as in tail suspension experiments. The test tool was a transparent plastic cylinder (20 cm in height, 12cm in diameter), 12cm in water level, and temperature (22 + -3) deg.C. The mice were placed in water 1h after the last administration for a test time of 6min, the whole process was recorded by video, and the immobility time of each mouse within 4min after the test was counted. Each group of mice was operated in parallel and statistically analyzed using animal behavioristics experimental analysis software.
3. Results and analysis of the experiments
The tail suspension experiment and the forced swimming experiment are classical ethological methods for researching animal depression behaviors, the state of different degrees of motion stopping of a mouse in a tail suspension and forced swimming model reflects the behavior of the animal, and the depression state of a human can be simulated.
As shown in Table 1, the composition of the invention can obviously shorten the immobility time of mice in both mouse tail suspension experiments and forced swimming experiments, and the difference has significance (P is less than 0.05). The immobility time of the 1 group of mixed powder composition samples prepared in the example 1 of the invention is equivalent to that of the positive medicine fluoxetine group, which shows that the composition can relieve depression symptoms of mice. In addition, the immobility time of the mixed powder composition sample 1 prepared in example 1 of the present invention is significantly shorter than that of the composite fermented powder sample 2 prepared in example 1 and the composite fermented powder sample 3 prepared in example 1, which indicates that the components of the present invention generate a synergistic effect therebetween, and the depression symptoms of the mice can be significantly alleviated.

Experiment 2: trial experiment of population
The crowd trial experiment adopts self-control design, and subjects meeting the conditions are screened, wherein each group comprises 25 persons. The granule obtained in step 4 of example 1 is administered 3 times a day, 2.0g each time, for 60 days. In the trial process, the subject stops taking other medicines or health care products with the function of regulating emotion, and the experimental results are shown in table 2.

The results in table 2 show that the composition of the invention has good improvement effect on various emotional symptoms without any adverse reaction when being taken.

Claims (10)
1. A health food with an anxiolytic and antidepressant function is characterized by comprising the following components in parts by weight: 20-30 parts of first composite fermentation powder and 10-20 parts of second composite fermentation powder;
the first compound fermentation powder is prepared by mixing and solid-state fermenting radix astragali, cortex Magnoliae officinalis, Atractylodis rhizoma, Poria, radix Angelicae sinensis, radix aucklandiae, cortex et radix Polygalae, arillus longan, semen Ziziphi Spinosae, fructus Jujubae, fructus Citri Sarcodactylis, and fructus Citri;
the second compound fermentation powder is prepared by mixing and fermenting rosemary, sage, saffron, basil, valerian root, passion flower, chamomile and balm.
2. The health food of claim 1, wherein the raw materials of the first composite fermentation powder comprise radix astragali, cortex Magnoliae officinalis, rhizoma Atractylodis Macrocephalae, Poria, radix Angelicae sinensis, radix aucklandiae, radix Polygalae, arillus longan, semen Ziziphi Spinosae, fructus Jujubae, fructus Citri Sarcodactylis, and fructus Citri in a mass ratio of 3: (1-1.5): (0.5-1.5): (0.5-1.5): (0.5-1.5): (0.5-1.5): (0.5-1.5): (0.5-1.5): (0.5-1.5): (0.5-1.5): (1-1.5): (1-1.5); in the raw materials of the second compound fermentation powder, the mass ratio of rosemary, sage, saffron, basil, valerian root, passion flower, chamomile and balm is 1: (1-2): (1-2): (1-2): (1-2): (1-2): (1-2): (1-2).
3. The health food as claimed in claim 1 or 2, wherein the first compound fermentation powder is obtained by mixed fermentation of bacillus subtilis, saccharomyces cerevisiae and lactobacillus acidophilus; the second compound fermentation powder is obtained by mixing and fermenting lactobacillus acidophilus, lactobacillus reuteri and lactobacillus plantarum.
4. The health food as claimed in claim 3, wherein the preparation method of the first composite baking powder comprises the following steps:
(1) pulverizing accurately weighed radix astragali, cortex Magnolia officinalis, Atractylodis rhizoma, Poria, radix Angelicae sinensis, radix aucklandiae, cortex et radix Polygalae, arillus longan, semen Ziziphi Spinosae, fructus Jujubae, fructus Citri Sarcodactylis, and fructus Citri, sieving with 40 mesh sieve, putting into fermentation tank at a certain proportion, adding water, mixing, and sterilizing with high pressure steam;
(2) after the product obtained in the step (1) is cooled, inoculating a zymocyte powder solution, wherein the ratio of bacillus subtilis to saccharomyces cerevisiae to lactobacillus acidophilus is 1: (1.5-2.5): (1.5-2.5), the inoculation amount is 5% -8%, and the fermentation product is obtained after the uniform mixing and the fermentation for 5-8 days at the temperature of 32-37 ℃;
(3) and (3) drying and crushing the fermentation product obtained in the step (2) at 45-55 ℃, and sieving the dried and crushed fermentation product with a 40-mesh sieve to obtain the first compound fermentation powder.
5. The health food as claimed in claim 3, wherein the preparation method of the second composite fermented powder comprises the following steps:
(a) pulverizing herba Rosmarini officinalis, herba Salvia officinalis, stigma croci Sativi, herba Ocimi, rhizoma et radix Valerianae, herba Passiflorae Caeruleae, flos Matricariae Chamomillae, and herba Melissae axillaris, sieving with 30 mesh sieve, adding into fermentation tank, adding water, mixing, sterilizing with high pressure steam, wherein the mass ratio of total dry matter raw materials to water is 1: (20-30);
(b) adjusting the pH value of the product obtained in the step (a) to 5-7, adding lactobacillus acidophilus, lactobacillus reuteri and lactobacillus plantarum into the product, and performing composite fermentation at the temperature of 28-38 ℃ for 48-96 hours to obtain a fermentation product;
(c) sterilizing the fermentation product obtained in the step (b) at the temperature of 130-135 ℃ for 5-10s, filtering, taking the filtrate, and drying to obtain the second composite fermentation powder.
6. The health food as claimed in claim 5, wherein in the step (b), the Lactobacillus acidophilus colony content in the fermentation system is 9 x 106CFU/mL, the colony content of Lactobacillus reuteri is 6.5X 106CFU/mL, the bacterial colony content of the lactobacillus plantarum is 8 multiplied by 106CFU/mL。
7. The health food of claim 3, wherein the health food composition is formulated in a clinically acceptable form in combination with an adjuvant.
8. The health food of claim 7, wherein the clinically acceptable dosage form is a powder, a tablet, a granule or a hard capsule.
9. The health food according to claim 7, wherein the auxiliary materials are pharmaceutically commonly used auxiliary materials and/or excipients, and comprise: one or more of saccharides, starch, cellulose and its derivatives, gelatin, talcum powder, solid lubricant, calcium sulfate, vegetable oil, polyol, alginic acid, emulsifier, wetting agent, colorant, flavoring agent, stabilizer, antioxidant and antiseptic.
10. The health food according to claim 3, wherein the daily dose of the health food composition is 2.1 to 3.3 g.
