2.2 As can be seen from the test results shown in FIG. 1, the activities of luciferase in the cell lysates of HaCaT-FLG-Luc cells (15×10 cells/mL) treated with the anti-aging skin repair liquid containing vitamin A alcohol at mass concentrations of 0.1%, 0.5% and 4% were 26%, 97% and 364%, respectively, which indicates that the vitamin A alcohol solution of the present invention can promote the expression of silk Fibroin (FILAGGRIN), decompose to form moisturizing factors, and improve the moisturizing property of skin and the integrity of sebum.
3. The following in vitro experiments were carried out with the anti-aging skin repair liquid containing tretinoin prepared in example 2:
3.1 The experimental method comprises the following steps: triplicate samples were cultured in 96-well microplates with 293-VR1 cells, the anti-aging skin repair solutions containing tretinoin at concentrations of 0%, 1% and 4% were added to the culture cell fluids of the three samples, respectively, and the culture was continued for 30 minutes, with 1. Mu.M capsaicin for 3 hours, 10. Mu.M capsicum Zhuo Ping for a positive control of TRPV-1 receptor, and the cell activity of each sample after 3 hours was detected with an IncuCyt living cell imaging analyzer, and the detection results are shown in FIG. 2.
3.2 from the test results shown in FIG. 2, it can be seen that: the cell viability of the test sample with the capsaicin concentration of 1 mu M and the capsaicin concentration of 4% is respectively improved by about 2 times and 3.5 times compared with that of the control sample with the capsaicin concentration of 1 mu M, which proves that the vitamin A alcohol solution can increase the antagonism capability of cells to TRPV1 receptors, reduce external stimulus and ease skin sensitivity.
It is worth noting that TRPV1 (transient receptor potential vanillic acid subtype 1 channel protein) is present in unmyelinated C-fibers and aδ fibers, mainly regulating the human perception of burning, pain and stinging. TRPV1 plays an important role in hyperalgesia caused by inflammation. When inflammation occurs, inflammatory factors bind to respective receptors on neurons, act on TRPV1 through respective intracellular signaling pathways, cause sensitization of central or peripheral nerves, and exhibit hyperalgesia.
4. The following rat skin penetration test was performed with the anti-aging skin repair liquid containing tretinoin prepared in example 3:
4.1 detection instrument: a percutaneous absorption tester;
4.2 test method: smearing the free rhodamine B aqueous solution display agent on a test part of the skin of the mouse; diluting the anti-aging skin repair liquid containing vitamin A alcohol prepared in the embodiment 3 into 10% aqueous solution to serve as a test sample, smearing the aqueous solution on the test part, and respectively detecting a blank sample and the test sample at 2h and 4h, wherein an image obtained by detection is shown in a figure 3;
4.3 from the image shown in fig. 3 it can be seen that: the free rhodamine B water solution is concentrated on the stratum corneum at 2h and 4h, and cannot effectively permeate the stratum corneum barrier; the rhodamine B marked 10% concentration test sample has penetrated the stratum corneum barrier in 2 hours, and the fluorescence intensity of the stratum corneum penetrated part of the 10% test sample in 4 hours is enhanced in detail, which shows that the active ingredients of the 10% test sample have penetrated into the deep skin layers, and the skin permeability experiment results show that the anti-aging skin repair liquid containing the vitamin A alcohol can effectively deliver the active substances to the deep skin tissues.
