Abstract
本发明公开了一种针对抗炎性肠病的鸡枞菌与人参小分子肽营养补充剂组合物及其制备方法,包括润水、高压脉冲破壁处理、高压均质提取、离心除杂、酶解、纤维膜超滤、减压浓缩和活性炭脱色八个步骤。本发明制备的鸡枞菌和人参小分子肽营养补充剂组合物具有防御和修复肠道黏膜、提高肠病人群免疫力和提供均衡膳食营养的作用,并且能够与其他的有益成分组合应用于制备不同形式的制剂。
Description
技术领域
本发明属于营养食品生产领域,尤其涉及一种针对抗炎性肠病的鸡枞菌与人参小分子肽营养补充剂组合物及其制备方法。
背景技术
慢性肠炎在临床上较为常见,近些年来该类疾病的发病率呈现逐年递增的趋势,分析慢性肠炎的主要临床症状包括持续性的腹部疼痛感、肠鸣音、腹胀、增多大便次数、大便性状为稀状等,且带有黏液。中医理论认为,慢性肠炎属于读写、泄泻的范畴,以大小肠、脾胃等作为主要的病变部位,同时与肾、肝等脏器的联系十分紧密。当人体情志失调、精神紧张、肝气郁滞等情况时将容易引发脾虚湿盛、代谢失常等,并引发慢性肠炎病变。该类疾病具有病情复杂、病程漫长等特点,应及时进行缓解与治疗,帮助患者促进疾病的早日康复,改善生存质量。
小分子活性肽是介于氨基酸与蛋白质之间一种生化物质,它比蛋白质分子量小,又比氨基酸分子量大,是一个蛋白质的片段,对于缓解和治疗肠道炎症具有积极的作用。小分子多肽进入人体后,能够被肠道细胞直接吸收,从而大大减轻肠胃等负担,对消化系统的组织器官有很好的保护作用;小分子多肽能促进钙、锌、镁等矿物质的吸收,提高其生物利用率,还具有促进微生物生长发育和活跃其代谢的作用,能促进乳酸杆菌、双歧杆菌、酵母等菌类的生长,对胃炎有很好的调理作用;小分子多肽营养价值高,容易消化吸收,在胃里可覆盖在胃粘膜表面,防止胃腔内的H+扩散入胃黏膜上皮细胞内,免受侵蚀,可以促进胃炎恢复;小分子多肽能够调节胃肠道的运动,降低胃酸分泌,调节肠胃蠕动的速度,促进食物在肠胃道中的消化吸收,对防治胃炎有良好的作用。鸡枞菌是一种即可被用来食用又可以入药的大型真菌,肉质肥厚,营养丰富,尤其蛋白质含量较高,且含有20多种氨基酸,其中人体必需8种氨基酸种类齐全。人参在我国的中药领域中具有很重要的地位,长期以来都被奉为非常名贵的中药植物,含有皂苷、蛋白质和多糖等生物活性物质。基于鸡枞菌和人参的多种功能成分,可以利用生物技术开发功能性鸡枞菌和人参多肽食品。
中国发明专利CN 105154509 B公布了一种人参肽的提取方法,所述方法包括人参微波预提取、膨胀溶剂提取、复合酶解、两相萃取和分离纯化,最终得人参肽。本方法使用复合有机溶剂,具有成本高、污染大的缺点。中国发明专利CN 108034685 A 公布了一种人参多肽的制备方法,所述方法包括切片、水提、酶解、膜过滤、浓缩和喷雾干燥六个步骤,得到人参肽提取液。此方法虽然没有用到有机溶剂,但未将人参原料实现破壁处理,导致有效成分不能最大程度释放,萃取率低。中国发明专利CN 108653690 A一种治疗胃病的中药组合物和中国发明专利CN 108272961 A一种治疗胃病的中药组合物及制备方法及其在健胃方面的应用,以上两个专利都是通过使用附子、肉桂、干姜、苍术、白芍、川贝母和三七粉等为原料制成养胃中药组合物,中药方剂普遍存在成分复杂、治疗效果较慢的不足,并且只是起到治疗的作用,并不能为患者提供营养所需。目前鸡枞菌小分子肽在缓解与治疗肠道方面应用欠缺,且与人参分子肽组合能够解决“人参滥用”的问题。
发明内容
本发明的目的是为了增强慢性肠炎患者机体免疫力并提供均衡膳食营养,提出了一种针对抗炎性肠病的鸡枞菌与人参小分子肽营养补充剂组合物及其制备方法。
本发明的技术方案为:
一、制备鸡枞菌与人参小分子肽
(1)取新鲜鸡枞菌和人参,用水洗涤干净以除去土壤杂质;
(2)将(1)中洗净的鸡枞菌和人参原料用纯净水复水0.1~1h后放入高压脉冲破壁设备中,压力为0.2~2.0 MPa,维持时间0.5~10 min后快速泄压,收集从卸料口排出的鸡枞菌和人参;
(3)将(2)中处理后的鸡枞菌和人参原料进行高压均质处理,以料水比1:10,压力40~100Mpa,时间10~100 min,得到浸提液;
(4)将(3)中得到的浸提液以5000r/min 的转速离心5~12min,取上清液备用;
(5)将(4)中得到的上清液温度调节为 30~37℃,pH 为 6.5~8.0,按上清液重量0.01~5%加入胰蛋白酶,酶解时间30~90min;
(6)将(5)中得到的酶解液灭活,酶活温度为80~100℃,酶活时间5~10min,得到小分子肽粗液;
(7)将(6)灭活液通过分子量为7000~20000Da的中空纤维膜超滤,除去大分子蛋白,得到小分子肽溶液;
(8)将(7)中小分子肽溶液减压浓缩和活性炭脱色得到比重在1.05~1.20的浓缩液,即为鸡枞菌和人参小分子肽。
二、利用得到的鸡枞菌与人参小分子肽制备鸡枞菌与人参小分子肽营养补充剂液体组合物,方法如下:
(1)分别将约2~20%的鸡枞菌与人参小分子肽与其他成分混合,生成混合物;
(2)对混合物灭菌;
(3)将灭菌的混合物包装在合适的容器中。
三、利用得到的液体组合物制备鸡枞菌与人参小分子肽营养补充剂固体组合物,方法如下:
将鸡枞菌与人参小分子肽营养补充剂液体组合物通过冷冻干燥或喷雾干燥的方式得到固体物质,即为鸡枞菌与人参小分子肽营养补充剂固体组合物。
本发明的有益效果是:
(1)本发明采用高压脉冲破壁技术处理鸡枞菌和人参原料,不仅能够使原料破壁,有利于胞内功能性成分的溶出,还能够使淀粉、半纤维素等部分水解生成可溶性糖,多糖能够提供人体能量,增强机体免疫力;
(2)本发明采用高压脉冲破壁和高压均质技术,具有灭菌的效果,能够将处理过程中滋生的微生物杀死,减少霉变的几率,能够延长物质的储存期;
(3)本发明采用高压脉冲破壁和高压均质技术的连用,高压脉冲破壁技术能够促进原料活性成分的提高,高压均质技术能够将处理过的原料进一步打碎,提高提取率;
(4)本发明采用高压均质工艺进行提取,使用温度低,避免发生物料高温氧化、变质和有效成分的损失;
(5)本发明抗炎性肠病的鸡枞菌与人参小分子肽营养补充剂组合物具有防御和修复肠道黏膜、提高肠病人群免疫力和提供均衡膳食营养的作用。
具体实施方式
下面将结合实施例对本发明的实施方案进行详细描述,但是本领域技术人员将会理解,下列实施例仅用于说明本发明,而不应视限定本发明的范围。
实施例中未标注具体条件者,按照常规条件或制造商建议的条件进行。所用试剂或仪器未标注生产厂商者,均为通过市购获得的常规产品。
制备鸡枞菌和人参提取液
(1)分别取新鲜鸡枞菌和人参5 kg,用水洗涤干净以除去土壤杂质;
(2)将(1)中洗净的鸡枞菌和人参原料用纯净水复水0.5 h后放入高压均质设备中,压力为0.8 MPa,维持时间5 min后快速泄压,收集从卸料口排出的鸡枞菌和人参;
(3)将(2)中处理后的鸡枞菌和人参原料进行高压均质处理,以料水比1:10,压力70Mpa,时间30min,得到浸提液;
(4)将(3)中得到的浸提液以5000r/min 的转速离心8min,取上清液备用;
(5)将(4)中得到的上清液温度调节为35℃,pH为7.0,按上清液重量2%加入胰蛋白酶,酶解时间60min;
(6)将(5)中得到的酶解液灭活,酶活温度为90℃,酶活时间8min,得到小分子肽粗液;
(7)将(6)灭活液通过分子量为7000~20000Da的中空纤维膜超滤,除去大分子蛋白,得到小分子肽溶液;
(8)将(7)中小分子肽溶液减压浓缩和活性炭脱色得到比重在1.10的浓缩液,即为鸡枞菌和人参小分子肽。

将原料按照实施例1、2、3的配比彻底混合,然后根据表2中列出的任一种条件灭菌并且通过无菌操作和包装方法装瓶

Claims (8)
1.一种针对抗炎性肠病的鸡枞菌与人参小分子肽营养补充剂组合物及其制备方法,其特征在于所述鸡枞菌和人参小分子肽营养补充剂组合物具有防御和修复肠道黏膜、提高肠病人群免疫力和提供均衡膳食营养的作用,制备方法如下:润水、高压脉冲破壁处理、高压均质提取、离心除杂、酶解、纤维膜超滤、减压浓缩和活性炭脱色
(1)取新鲜鸡枞菌和人参,用水洗涤干净以除去土壤杂质;
(2)将(1)中洗净的鸡枞菌和人参原料用纯净水复水0.1~1h后放入高压脉冲破壁设备中,压力为0.2~2.0 MPa,维持时间0.5~10 min后快速泄压,收集从卸料口排出的鸡枞菌和人参;
(3)将(2)中处理后的鸡枞菌和人参原料进行高压均质处理,以料水比1:10,压力40~100Mpa,时间10~100 min,得到浸提液;
(4)将(3)中得到的浸提液以5000r/min 的转速离心5~12min,取上清液备用;
(5)将(4)中得到的上清液温度调节为 30~37℃,pH 为 6.5~8.0,按上清液重量0.01~5%加入胰蛋白酶,酶解时间30~90min;
(6)将(5)中得到的酶解液灭活,酶活温度为80~100℃,酶活时间5~10min,得到小分子肽粗液;
(7)将(6)灭活液通过分子量为7000~20000Da的中空纤维膜超滤,除去大分子蛋白,得到小分子肽溶液;
(8)将(7)中小分子肽溶液减压浓缩和活性炭脱色得到比重在1.05~1.20的浓缩液,即为鸡枞菌和人参小分子肽。
2.根据权利要求1所述,鸡枞菌与人参小分子肽营养补充剂液体组合物的制备方法如下:
(1)分别将2.0~20%鸡枞菌与人参小分子肽与其他成分混合,生成混合物;
(2)对混合物灭菌;
(3)将灭菌的混合物包装在合适的容器中。
3.根据权利要求2所述,其中脂肪包括中长链脂肪酸食用油;益生元包括低聚半乳糖、低聚木糖和低聚果糖的一种或多种;益生菌包括乳酸双歧杆菌、长双歧杆菌、保加利亚乳酸菌、嗜热链球菌和干酪乳杆菌中的一种或多种;膳食纤维包括菊粉、麦麸膳食纤维的一种或两种;维生素包括维生素A、B1、B2、B6、B12、C、D、E、K1、烟酸、泛酸、叶酸中的一种或多种;矿物质包括钠、钾、钙、镁、铁、锌、铜、锰、氯、碘。
4.根据权利要求2和3所述,鸡枞菌与人参小分子肽营养补充剂液体组合物的配比如下:2.0~20%鸡枞菌小分子肽;2.0~20%人参小分子肽;4~10%脂肪酸;50~70%低聚糖;2~10%膳食纤维;1~5%益生元;0.01~1%维生素、0.01~1%矿物质以及105~107cfu/ml的益生菌。
5.根据权利要求2所述,将鸡枞菌与人参小分子肽营养补充剂液体组合物通过冷冻干燥或喷雾干燥的方式得到固体物质,即为鸡枞菌与人参小分子肽营养补充剂固体组合物。
6.根据权利要求2所述,人参小分子肽营养补充剂固体组合物包括片剂、胶囊或者粉末。
7.根据权利要求2所述,混合物灭菌包括巴氏灭菌、高温短时灭菌和超高温灭菌。
8.根据权利要求2所述,包装灭菌的混合物包括热灌装工艺和冷罐装工艺。
