Detailed Description
The present invention is further illustrated by the following specific examples, but the scope of the present invention is not limited to the following examples.
Firstly, extracting flavonoid active ingredient from Chinese redbud
The preparation process of the bauhinia extract comprises the following steps:
1. weighing 10 parts by weight of Chinese redbud, and adding 20 parts by weight of 75vol.% ethanol;
2. reflux extracting at 70-80 deg.C;
3. concentrating the filtrate under reduced pressure at 50 deg.C or lower to obtain extract, and vacuum drying.
The content of the total flavone of the prepared Chinese redbud extract is 10-20%, and the Chinese redbud extract is added into a product as an active ingredient with the effects of clearing heat and cooling blood in the process of preparing an oral care product.
Second, the application of the Chinese redbud extract in oral care products (taking the preparation in toothpaste as an example)
The toothpaste in examples 1 to 4 comprises the following raw materials in percentage by weight as shown in table 1:
TABLE 1 toothpaste containing the extract of bauhinia flowers
Third, bacteriostatic effect of toothpaste containing Chinese redbud extract
Staphylococcus aureus widely lives in nature, is carried in air, water and dust, is a pyogenic microorganism with wide transmission and strong pathogenicity, can cause oral pyogenic infection, and is a main pathogenic bacterium of oral diseases. Coli can cause various urinary infections, acute diarrhea and the like outside the intestinal tract. Porphyromonas gingivalis is a gram-negative anaerobic coccobacillus which is not glycolytic and is one of pathogenic bacteria of periodontal diseases. Modern medical experiments prove that streptococcus mutans in dental plaque can generate acid in metabolism, corrode hard tissues of teeth, generate caries and are main pathogenic microorganisms causing caries.
The test method comprises the following steps:
the test is carried out according to the method for testing the bacteriostatic performance of the dissolution type antibacterial (bacteriostatic) product of the disinfection technical specification 2002 edition-2.1.11.3.2.
The 24-hour slant culture of the test bacteria was washed with PBS to prepare a bacterial suspension (the required concentration was 100. mu.l of the test bacteria was dropped on a control sample or 5ml of the sample, and the number of recovered bacteria was 1X 104 cfu/plate or ml-9X 104 cfu/plate or ml).
10g of the toothpaste sample of example 2 was added to 100mL of physiological saline, and mixed well to prepare a stock solution as a sample solution, and the sample solution (5mL) was taken in 3 tubes.
Taking the bacterial suspension, respectively dripping 100 mu l of the bacterial suspension into each sample liquid, uniformly coating/mixing, starting timing, acting for 3min, respectively putting a sample slice or sample liquid (0.5ml) into a test tube containing 5ml of PBS by using sterile forceps, fully and uniformly mixing, properly diluting, then taking 2-3 dilutions, respectively sucking 0.5ml, placing in 2 plates, pouring by using 15ml of nutrient agar medium (bacteria) or Sabouraud's agar medium (yeast) cooled to 40-45 ℃, rotating the plates to fully and uniformly, turning over the plates after agar is solidified, culturing for 48h (bacteria) or 72h (yeast) at 35 +/-2 ℃, and counting viable bacterial colonies.
Each test bacterium experiment was repeated 3 times, and the inhibition rate was calculated as follows:
in the formula: x is the bacteriostasis rate,%; n is a radical ofCMean colony number, cfu/plate, for control samples; n is a radical ofSThe average colony count of the samples to be tested, cfu/plate.
Table 2 antibacterial effect test results of toothpaste containing Chinese redbud extract
The antibacterial tests show that the toothpaste containing the Chinese redbud extract has antibacterial effect on escherichia coli, staphylococcus aureus, candida albicans and streptococcus mutans.
Fourth, the toothpaste containing the Chinese redbud extract has the hemostatic effect on the rabbit liver local wound injury bleeding
120 healthy New Zealand rabbits (30 rabbits each group) are taken, the weight is 2.0-2.5 kg, and the rabbits can be used for both males and females. Slowly injecting into ear vein of 30mg/kg of 3% sodium pentobarbital, anesthetizing, supining, fixing on operating table, making the middle incision of abdomen about 6-8 cm long, exposing and fixing liver, cutting liver left lobe with surgical scissors to obtain liver tissue with length of about 1cm, width of about 1cm, and height of about 1cm,
the wound surface is bleeding like spraying. After the sprayed blood is wiped off, 1g of a sample to be tested is immediately coated on the part of the wound surface, gauze is covered, a weight of 200g is used for compressing for 1min, the weight is removed after 1min, the hemostasis effect is observed, bleeding does not occur, if bleeding still occurs, the sample to be tested is coated and then is continuously pressurized, the observation is carried out once every 30s, the hemostasis time is recorded, if bleeding still occurs after exceeding 3min, the hemostasis time is recorded as the hemostasis failure, and the hemostasis time is recorded as more than 3 min. After each sample test, the residual test substance is washed away by physiological saline at 37 ℃ so that the wound surface bleeds again, and then the next test substance sample is tested. Four tests of hemostasis test of liver local wound injury are carried out on each rabbit, and each test sample is applied according to the Latin prescription procedure, so that the influence of incision difference and medication sequence of liver local wound injury is reduced. The hemostatic experiment of the rabbit liver local wound injury bleeding is carried out for 20 times in total for each test object sample. The hemostatic effect is marked as F by the reciprocal score method (10/T), hemostasis is not achieved for more than 3min, and no F score is marked as 0.
TABLE 3 hemostatic effect (X + -S) of toothpaste containing Cercis chinensis extract on rabbit liver local wound bleeding
Note: comparison with starch group: # # P < 0.001, P > 0.05.
Compared to a toothpaste blank matrix containing bauhinia flowers toothpaste: p < 0.001, Δ P > 0.05.
The experimental result shows that the toothpaste containing the Chinese redbud extract and the Yunnan white drug powder both have obvious hemostatic effect on the bleeding of the local wound surface of the rabbit liver and have obvious hemostatic effect;
