Abstract

A method for extracting corn protein concentrate by gluten water comprises the following steps: the method comprises the following steps: adjusting pH of the gluten water to 4-8, adding alpha amylase, and reacting at 70-100 deg.C for 30-150min to obtain enzymatic hydrolysate; step two: adding alkali liquor into the enzymolysis liquid obtained in the step one to adjust the pH value to 8-12, and reacting at the temperature of 40-70 ℃ for 30-60min to obtain a corn concentrated protein liquid; step three: and centrifugally separating the concentrated corn protein liquid obtained in the step two, washing with water, and drying to obtain the concentrated corn protein. The invention uses alpha-amylase to enzymolyze starch in corn gluten water, and reduces the dry fat content from 2.5% to about 0.5-1% after degreasing by an alkaline method. After water washing and centrifugal treatment, the color of the corn protein powder is obviously reduced, the corn protein powder can reach faint yellow to creamy yellow after spray drying, and the dry basis protein content can reach more than 90 percent after enzymolysis and starch removal and alkali degreasing. The sulfide content of the corn concentrated protein powder product can be reduced to below the national standard limit.

Description

Method for extracting corn protein concentrate by gluten water
Technical Field
The invention belongs to the field of recycling of gluten water in corn starch production, and particularly relates to a method for extracting corn concentrated protein from gluten water.
Background
In 2018, the consumption of Chinese protein products reaches 8829.6 ten thousand tons, and the consumption of protein products in European Union and America respectively reaches 4426.7 ten thousand tons and 4134.9 ten thousand tons. The consumption of protein products will further increase in the future, and the supply gap of Chinese protein products in 2030 is expected to reach more than 3800 ten thousand tons. The vegetable protein has the health advantages of zero cholesterol, low fat and the like, reduces the risk of diseases caused by meat products, and meets the concept of green and healthy consumption. The edible corn protein powder is produced by taking the corn gluten meal as a raw material, so that the corn resource is comprehensively utilized, the problem of lack of protein resource is solved, and the demand of people on healthy plant protein is met. Corn is one of main crops which have been used for producing starch, a large amount of gluten water is generated when the corn is used for producing the starch by wet grinding at present, the dry-based starch content in the gluten water is about 15 percent, the dry-based protein content is about 65 percent and the dry-based fat content is about 2.5 percent according to the detection of the formula, so the gluten water has great recycling value, and a gluten water recycling process is lacked at present.
Disclosure of Invention
The invention provides a method for extracting corn protein concentrate by gluten water, which is used for overcoming the defects in the prior art.
The invention is realized by the following technical scheme:
a method for extracting corn protein concentrate by gluten water comprises the following steps:
the method comprises the following steps: adjusting pH of the gluten water to 4-8, adding alpha amylase, and reacting at 70-100 deg.C for 30-150min to obtain enzymatic hydrolysate;
step two: adding alkali liquor into the enzymolysis liquid obtained in the step one to adjust the pH value to 8-12, and reacting at the temperature of 40-70 ℃ for 30-60min to obtain a corn concentrated protein liquid;
step three: and centrifugally separating the concentrated corn protein liquid obtained in the step two, washing with water, and drying to obtain the concentrated corn protein.
The method for extracting the corn protein concentrate by the gluten water comprises the step one, wherein the addition amount of the alpha amylase is 100-1000U/kg.
The method for extracting the corn protein concentrate from the gluten water comprises the step three, wherein the washing times are 1-5 times, and the ratio of the washing materials to the washing liquids is 1: 1-6.
The invention has the advantages that: the invention uses the gluten water of the starch produced by corn wet milling as the raw material to produce the corn concentrated protein. The invention uses alpha-amylase to carry out enzymolysis on starch in corn gluten water, carries out enzymolysis on starch components into soluble sugar such as dextrin and the like, and removes the components through water washing so as to improve the dry basis protein content in the corn protein powder. After the starch is enzymolyzed, the dry starch content of the corn protein powder can be reduced to about 0.5-1.5%, and the dry protein content can be increased to about 88-91%. After degreasing by an alkaline method, the content of dry fat is reduced from 2.5 percent to about 0.5 to 1 percent.
