Abstract
本发明公开了一种具有祛痘功效的护肤品组合物及其制备方法,其是由下述重量百分比的原料制得:卡波姆0.2‑0.3%,丁二醇2‑4%,甜菜碱水杨酸盐1‑3%,精氨酸1‑2%,季铵盐‑73,2‑4%,海藻糖0.5‑1.5%,烟酰胺0.5‑1.5%,生育酚乙酸酯1.5‑2.5%,寡肽‑1,4‑6%,金银花复配提取物3‑7%,纤连蛋白4‑6ppm,余量为水。本发明提供了一种含有纤连蛋白同时含有寡肽‑1的护肤品配方,对皮肤刺激性小,修复表皮损伤,祛痘的同时也不易留下痘印。

技术领域
本发明涉及护肤品技术领域,确切地说是一种具有祛痘功效的护肤品组合物及其制备方法。
背景技术
痘,是痤疮的俗称,又叫“面疱”“粉刺”“酒刺”“暗疮”等,是由于皮脂分泌过多、毛囊皮脂腺导管堵塞、细菌感染和炎症反应等因素所引发的一种慢性炎症性皮肤病。青春痘、粉刺、暗疮在医学上统称为痤疮,是常见的一种皮脂腺慢性病,其成因是青春期时,体内的荷尔蒙会刺激毛发生长,皮脂腺分泌旺盛,导致毛孔堵塞。大多数祛痘产品效果平平,容易留下痘印。因此,开发出一种具有显著祛痘功效,对皮肤刺激性小且不易留下痘印的产品是很有意义的。
目前现有的祛痘物质抗衰对皮肤刺激性大、效果不理想、易留下痘印,非常需要具有效果明确、作用安全、无不良反应且不易留下痘印的具有祛痘作用的产品。
发明内容
本发明的目的在于提供一种更易于被皮肤吸收,且具有显著的皮肤祛痘功效的护肤品组合物及其制备方法。
上述目的通过以下方案实现:
一种具有祛痘功效的护肤品组合物,其特征在于,其是由下述重量百分比的原料制得:卡波姆0.2-0.3%,丁二醇2-4%,甜菜碱水杨酸盐1-3%,精氨酸1-2%,季铵盐-732-4%,海藻糖0.5-1.5%,烟酰胺0.5-1.5%,生育酚乙酸酯1.5-2.5%,寡肽-14-6%,金银花复配提取物3-7%,纤连蛋白4-6ppm,余量为水;
一种具有祛痘功效的护肤品组合物,其特征在于,所述的金银花复配提取物是由下述重量份的原料制得:金银花10-15、薄荷叶4-6、葡萄糖酸锌0.1-0.3、视黄醇棕榈酸酯0.2-0.3,水适量。
所述的具有祛痘功效的护肤品的制备方法,其特征在于:包括以下步骤:
(1)在金银花复配提取物中加入适量水,加热至30-45℃搅拌5-10min后,得金银花复配提取液备用;
(2)卡波姆、丁二醇、甜菜碱水杨酸盐、精氨酸以及适量水投入搅拌锅中,搅拌均匀,加热至40-50℃,再加入酵母菌溶胞物提取物、干细胞外泌体,均质机均质3-5Min;
(3)在步骤(2)均质后的物料中加入金银花复配提取液以及其余原料,于30-45℃搅拌均匀,冷却,干燥,出料,即得。

(1)金银花、薄荷叶混匀后,用3-5%的氯化钠溶液浸泡3-5Min,洗净,捣碎,再研磨,得复配研磨粉备用;
(2)将复配研磨粉加入适量水中,加热至40-50℃搅拌5-10min后,加入葡萄糖酸锌、视黄醇棕榈酸酯,离心,去上清液后,干燥,即得。
本发明的有益效果为:
(1)本发明提供了一种含有纤连蛋白同时含有寡肽-1的护肤品配方,对皮肤刺激性小,修复表皮损伤,祛痘的同时也不易留下痘印。
(2)与其他的产品相比,本发明通过受损表皮的修复与再生,重组人纤连蛋白能修复敏感肌、祛除红血丝、祛除色斑、晒斑、改善皮肤弹性;金银花复配提取物具有良好的抗菌效果与消炎效果,可以改善皮肤的敏感问题与长痘问题。甜菜碱水杨酸盐是水杨酸的衍生物,完美的结合了水杨酸祛痘紧肤和甜菜碱滋润保湿的功效,温和不刺激,促进皮肤新成代谢,能有效改善痘痘肌,防止毛孔堵塞;季铵盐-73作为一款强效但安全性极高的抗菌剂,对丙酸痤疮杆菌有极好的效果,还可抑制黑色素形成;烟酰胺作为一种维生素B3的衍生物不仅具有美白、抗糖化的作用,还具有一定的控油作用,可以缩小毛孔,抑制皮肤中分泌的脂肪酸,从而达到祛痘的效果;寡肽-1可以促进皮肤细胞增长,修复受损细胞,调理敏感肌,有很好的祛痘美白效果,在一定程度上还可以预防粉刺;生育酚乙酸酯不仅有锁水保湿抗氧化消除自由基的作用,对皮肤过敏也有一定的抑制作用,安全系数很高。

所述的金银花复配提取物是由下述重量份的原料制得:金银花12、薄荷叶4、葡萄糖酸锌0.3、视黄醇棕榈酸酯0.3,水适量。
所述的具有祛痘功效的护肤品的制备方法,其特征在于:包括以下步骤:
(1)在金银花复配提取物中加入适量水,加热至45℃搅拌5min后,得金银花复配提取液备用;
(2)卡波姆、丁二醇、甜菜碱水杨酸盐、精氨酸以及适量水投入搅拌锅中,搅拌均匀,加热至50℃,再加入酵母菌溶胞物提取物、干细胞外泌体,均质机均质5Min;
(3)在步骤(2)均质后的物料中加入金银花复配提取液以及其余原料,于45℃搅拌均匀,冷却,干燥,出料,即得。
所述的金银花复配提取物的制备方法包括以下步骤:
(1)金银花、薄荷叶混匀后,用5%的氯化钠溶液浸泡3-5Min,洗净,捣碎,再研磨,得复配研磨粉备用;
(2)将复配研磨粉加入适量水中,加热至40℃搅拌5min后,加入葡萄糖酸锌、视黄醇棕榈酸酯,离心,去上清液后,干燥,即得。
根据如下表格来对本发明的实施例进行进一步的详细说明。
1.促细胞粘附活性测定
试验原理:重组人纤连蛋白重要作用是促进细胞的黏连和粘附,可提高细胞的贴壁率、汇合率,缩短细胞汇合时间,使细胞形态结构良好,代谢率增强及蛋白质合成速度显著提高。而细胞的黏连和粘附是细胞修复、细胞生长完成的必要条件,因此重组人纤连蛋白具有促细胞修复的功效。本实验通过重组人纤连蛋白促MDBK细胞的粘附试验,统计不同重组人纤连蛋白浓度下各孔内细胞数,经四参数拟合得出重组人纤连蛋白的半数有效浓度(ED50)。
试验方法:将重组人纤连蛋白在96孔板中进行2倍梯度稀释,共做10个稀释度,每孔50μl重组人纤连蛋白稀释液,并设立阴性对照,加入50μlPBS作为对照,4℃过夜孵育。
孵育完成后弃去板中PBS,每孔加入100μl30g/LBSA,置于37℃孵育1h后,取出96孔板,弃去板中液体,加入MDBK重悬细胞(用无血清培养基重悬),细胞接种密度为1.0×105个/ml,每孔接种100μl,温箱孵育3-5h后PBS洗涤3次,镜下观察细胞贴壁情况,选取200倍镜下除边缘处五个点计贴壁细胞数,根据计数结果经四参数拟合计算后得出效价。
结果计算:以稀释倍数为横坐标,贴壁细胞数为纵坐标,在ELISACalc回归/拟合计算程序软件中设定Logistic曲线拟合2(四参数)为回归/拟合模型,输入记录测定的数据进行回归/拟合,可以得到回归曲线及回归方程。
试验数据采用四参数回归计算法进行处理后,按下式计算结果:
试供品生物学活性(U/ml)=Pr*Ds*Es/Dr*Er
式中Pr为标准品生物学活性,U/ml;
Ds为试供品预稀释倍数;
Es为试供品相当于标准品半效数的稀释倍数;
Dr为标准品预稀释倍数;
Er为标准品半效量的稀释倍数;

本次实验结果表明本发明所涉及的组合物具有促细胞粘附活性。该化合物可以通过促进细胞粘附、迁移来起到皮肤修复效果,可做到祛痘除印。
2.促细胞增殖活性检验
试剂配制
完全培养液 量取小牛血清100ml,加培养液定容至1000ml。
维持培养液 量取小牛血清4ml,加培养液定容至1000ml。
消化液:称取乙二胺四乙酸二钠0.2g、氯化钠8g、氯化钾0.2g、磷酸氢二钠1.152g、磷酸二氢钾0.2g、胰蛋白酶2.5g,加纯化水溶解并定容至1000ml,过滤除菌。
噻唑蓝(MTT)溶液:称取MTT粉末0.10g,加PBS20ml使溶解,经0.22μm滤膜过滤除菌。4℃避光保存。
PBS:称取氯化钠8.0g、氯化钾0.20g、磷酸氢二钠1.44g、磷酸二氢钾0.24g,加纯化水溶解并定容至1000ml,经121℃15分钟灭菌。
细胞株:BALB/C3T3细胞。
rhEGF国家标准品:6800IU/支,研磨粉;
试验操作
标准品制备:取50μl标准品于450μl完全培养液中进行10倍稀释,重复操作直至稀释100倍。取735μl100倍稀释标准品于265μl完全培养液中进行1.36倍稀释。取136倍稀释液在96孔细胞培养板中,从1︰4开始依次做4倍递增梯度稀释(具体操作步骤为首先于96孔细胞培养板中加入150μl完全培养液,取50μl1000倍稀释的供试品溶液加入96孔板中第1列,吹打稀释。充分稀释后于第1列孔中抽取50μl于第2列孔,吹打稀释。充分稀释后于第2列孔中抽取50μl于第3列孔,吹打稀释。重复上述操作,直至第10列),共做10个稀释度(1~10孔),每个稀释度同时设置一个复孔。
重组人表皮生长因子供试品制备:取100μl供试品于900μl完全培养液中进行10倍稀释,重复操作直至稀释1000倍。取1000倍稀释液在96孔细胞培养板中,从1︰4开始依次做4倍递增梯度稀释(具体操作步骤为首先于96孔细胞培养板中加入150μl完全培养液,取50μl1000倍稀释的供试品溶液加入96孔板中第1列,吹打稀释。充分稀释后于第1列孔中抽取50μl于第2列孔,吹打稀释。充分稀释后于第2列孔中抽取50μl于第3列孔,吹打稀释。重复上述操作,直至第10列),共做10个稀释度(1~10孔),每个稀释度同时设置一个复孔。
注:样品预稀释倍数根据其活性值进行调整。
测定方法
①BALB/C3T3细胞株用完全培养液于37℃、5%二氧化碳培养,控制细胞浓度为每1ml含1.0×105-5.0×105个细胞,传代后24~36小时用于生物学活性测定。
②弃去培养瓶中的培养液,消化和收集细胞用完全培养液配成每1ml含5.0×104~8.0×104个细胞的细胞悬液,接种于96孔板中,接种过程中不停摇匀,保持每孔接种数相同,每孔100μl,于37℃、5%二氧化碳条件下培养。
③24小时后换成维持培养液。置37℃、5%二氧化碳培养24小时。
④制备的细胞培养板弃去维持液,加入标准品溶液和供试品溶液,每孔100μl。置37℃、5%二氧化碳培养64~72小时。
⑤MTT比色法:每孔加入MTT溶液20μl,于37℃、5%二氧化碳培养5小时。以上操作在无菌条件下进行。弃去培养板中的液体后,向每孔中加入DMSO100μl,混匀后在酶标仪上,以630nm为参比波长,570nm为试验波长测定吸光度,记录测定结果。
数据处理
将各个样品及标准品的OD值数据进行四参数拟合。
结果计算
试验数据采用计算机程序或四参数回归计算法进行处理。
供试品工作效价计算公式U=预稀释倍数*4C(C为四参数方程中C值)
供试品校正效价并按下式计算结果:
校正效价(IU/ml)=Pr*Ds*Es/Dr*Er
式中Pr为标准品生物学活性,IU/ml;
Ds为试供品预稀释倍数;
Es为试供品相当于标准品半效数的稀释倍数;
Dr为标准品预稀释倍数;
Er为标准品半效量的稀释倍数;
本次实验结果表明本发明所涉及的组合物具有促细胞增长活性。
3.超氧自由基清除率活性测定
实验原理:超氧阴离子自由基作为生物体代谢过程中产生的一种自由基,可攻击生物大分子,如脂质、蛋白质、核酸和聚不饱和脂肪酸等,使其交链或者断裂,引起细胞结构和功能的破坏,与机体衰老和病变有很密切的关系,清除超氧阴离子自由基的研究已经得到了广泛的关注。
AP-TEMED系统产生超氧阴离子,与盐酸羟胺反应生成NO2-,NO2-与对氨基苯磺酸和α-萘胺的作用生成红色的偶氮化合物,在530nm处有特征吸收峰,样品对超氧阴离子的清除能力与530nm的吸光值呈负相关。
操作步骤
将试剂盒中各试剂按以下顺序加入。
计算公式:超氧阴离子清除率I%=(A对照管-A测定管)/A对照管×100%
结果:
本次实验结果表明本发明所涉及的组合物具有超氧自由基清除活性,具有抗氧化作用。
另外需要说明的是,在上述具体实施方式中所描述的各个具体技术特征,在不矛盾的情况下,可以通过任何合适的方式进行组合,为了避免不必要的重复,本发明对各种可能的组合方式不再另行说明。
Claims (4)
1.一种具有祛痘功效的护肤品组合物,其特征在于,其是由下述重量百分比的原料制得:卡波姆0.2-0.3%,丁二醇2-4%,甜菜碱水杨酸盐1-3%,精氨酸1-2%,季铵盐-73 2-4%,海藻糖0.5-1.5%,烟酰胺0.5-1.5%,生育酚乙酸酯1.5-2.5%,寡肽-14-6%,金银花复配提取物3-7%,纤连蛋白4-6ppm,余量为水。
2.所述的一种具有祛痘功效的护肤品组合物,其特征在于,所述的金银花复配提取物是由下述重量份的原料制得:金银花10-15、薄荷叶4-6、葡萄糖酸锌0.1-0.3、视黄醇棕榈酸酯0.2-0.3,水适量。
3.一种如权利要求1所述的具有祛痘功效的护肤品的制备方法,其特征在于:包括以下步骤:
(1)在金银花复配提取物中加入适量水,加热至30-45℃搅拌5-10min后,得金银花复配提取液备用;
(2)卡波姆、丁二醇、甜菜碱水杨酸盐、精氨酸以及适量水投入搅拌锅中,搅拌均匀,加热至40-50℃,再加入酵母菌溶胞物提取物、干细胞外泌体,均质机均质3-5Min;
(3)在步骤(2)均质后的物料中加入金银花复配提取液以及其余原料,于30-45℃搅拌均匀,冷却,干燥,出料,即得。
4.根据权利要求3所述的具有祛痘功效的护肤品的制备方法,其特征在于:
所述的金银花复配提取物的制备方法包括以下步骤:
(1)金银花、薄荷叶混匀后,用3-5%的氯化钠溶液浸泡3-5Min,洗净,捣碎,再研磨,得复配研磨粉备用;
(2)将复配研磨粉加入适量水中,加热至40-50℃搅拌5-10min后,加入葡萄糖酸锌、视黄醇棕榈酸酯,离心,去上清液后,干燥,即得。
