Abstract

The invention discloses a preparation method of tea powder fermentation liquor and application of the tea powder fermentation liquor in cosmetics. The preparation method comprises the following steps: pulverizing tea, mixing with deionized water, pasteurizing, and fermenting with Lactobacillus plantarum. The probiotic inoculated tea powder is fermented, so that the biological activity of the tea powder is improved, the loss of active ingredients and the emission of harmful substances caused by a chemical extraction method are avoided, and the environment-friendly effect is achieved. The fermentation method adopted by the invention does not add any chemical organic reagent, has mild fermentation conditions and ensures the safety of the fermentation product. The tea powder fermentation liquor prepared by the preparation method disclosed by the invention is high in DPPH free radical clearance rate and can be used as an effective active ingredient in a skin care product.

Technical Field

The zinc-selenium organic tea belongs to the ecological health-care tea with pure nature, high quality and no public nuisance. The organic tea is produced without gene engineering, chemical fertilizer, pesticide, growth promoter and other synthetic preparation and radiation technology. Completely follows the natural and ecological principles, and depends on crop straws, waste materials, animal manure, natural green manure and mineral raw materials as much as possible to maintain nutrients. And biological and physical measures are utilized to prevent and control plant diseases and insect pests. Product pollution is prohibited in the packaging and transportation process.
The active substances in the tea mainly comprise tea polysaccharide, tea polyphenol and the like, wherein the tea polyphenol is the most extensive substance in the research of the tea. Tea polyphenol is a general term of polyphenol compounds, accounts for 18-36% of the dry weight of tea leaves, and mainly comprises catechins, flavonoids, anthocyanins and phenolic acids. Wherein the catechin compound accounts for 70-80% of the total amount and is the main component of the secondary metabolites of the tea trees. Catechin is further classified into catechin, epigallocatechin, gallate and epicatechin gallate. Theanine is a non-protein amino acid, accounts for more than 50% of the total amount of amino acids in the tea leaves and accounts for 1% -2% of the dry weight of the tea leaves, and the active ingredients not only provide the biological activity of antioxidation for the tea leaves, but also add the flavor of the tea leaves.
The lactobacillus plantarum is an important probiotic, and can be used for inoculating fermented tea powder to improve the bioactivity potential of the tea powder without destroying nutritional ingredients. Meanwhile, harmful substance residues caused by a chemical extraction residue process are avoided, the utilization rate of active ingredients of tea powder is improved, and the method is environment-friendly.
At present, tea has related applications in the field of cosmetics, and is mainly focused on the extraction and application of tea polyphenol, and the defects that the utilization rate of active ingredients is not high due to the fact that various active ingredients cannot be simultaneously and effectively extracted, and in addition, organic reagent residues are easily generated in the chemical extraction process, and the environmental pollution is easily generated.

The technical problem solved by the invention is as follows: the prior extraction and application of tea polyphenol have the problems that the utilization rate of active ingredients is not high due to the fact that a plurality of active ingredients cannot be simultaneously and effectively extracted, organic reagent residues are easily generated in the chemical extraction process, and environmental pollution is easily caused.
In order to solve the technical problem, the invention provides a preparation method of tea powder fermentation liquor, which comprises the following steps:
step 1: pulverizing dried folium Camelliae sinensis and sieving to obtain folium Camelliae sinensis powder;
step 2: according to the mass ratio of 1: 30-50, mixing the tea powder obtained in the step 1 with water, and sterilizing at the temperature of 115 ℃ and 121 ℃ for 10-15min to prepare tea powder liquid;
and step 3: pretreatment of zymophyte: streaking lactobacillus plantarum stored at low temperature to obtain single colony, selecting well-grown single colony, inoculating to MRS broth culture medium, culturing at 30-37 deg.C in incubator for 16-20 hr, and inoculating to the tea powder solution obtained in step 2, with inoculation concentration of 106-109CFU/mL mixed solution;
and 4, step 4: culturing the mixed liquor obtained in the step 3 in a constant-temperature incubator for 24-72 hours under a sealed anaerobic condition to obtain fermentation liquor, wherein the fermentation temperature is 30-37 ℃;
and 5: and (4) sterilizing the fermentation liquor obtained in the step (4) by high-pressure steam, centrifuging and taking supernatant fluid to obtain the tea powder fermentation liquor.
Preferably, the tea powder in the step 1 is crushed and then passes through a 80-200 mesh sieve.
Preferably, the rotation speed of the centrifugation in the step 5 is 3000-8000r/min, and the centrifugation time is 10-30 min.
Preferably, the pH value of the tea powder fermentation liquor in the step 5 is 4.0-6.0.
The invention also provides a skin-care product, which comprises the tea powder fermentation liquor prepared by the preparation method of the tea powder fermentation liquor as claimed in any one of claims 1 to 4.
Preferably, the tea powder fermentation liquor is applied as an effective active ingredient in a skin care product.
Compared with the prior art, the invention has the following beneficial effects:

Example 1

The tea powder fermentation liquor prepared by the embodiment can be used for skin care cosmetics such as astringent and essence.
The tea powder fermentation liquor prepared by the embodiment can also be directly used as toning lotion.
Example 2
Taking 150g of dried tea leaves, crushing, sieving with a 200-mesh sieve, weighing 5g, adding 150mL of water, mixing, and sterilizing at 115 ℃ for 20min to obtain initial tea powder fermentation liquor; streaking lactobacillus plantarum stored at low temperature to obtain single colony, selecting well-grown single colony, inoculating to MRS broth culture medium, culturing at 37 deg.C in incubator for 16 hr, inoculating to the obtained initial tea powder solution with inoculation concentration of 108CFU/mL mixed solution; culturing the inoculated strain in a constant temperature incubator for 24h under a sealed anaerobic condition, wherein the fermentation temperature is 37 ℃; sterilizing the fermentation liquor with high pressure steam, centrifuging, and collecting supernatant to obtain the desired tea powder fermentation liquor.
The tea powder fermentation liquor prepared by the embodiment can be used for skin care cosmetics such as astringent and essence.

Example 3

Comparative example

Analytical experiments:

The results of comparing the relevant material components of examples 1-3 with the comparative examples show that the total phenol content after fermentation is obviously improved compared with that before fermentation.
Measurement of DPPH radical scavenging Rate
A tube was charged with 2mL of ethanol DPPH solution (2mM, 75% ethanol) and 2mL of the test solution. Mixing, standing at 25 deg.C in dark for 30min, zeroing with anhydrous ethanol at 517nm wavelength, and measuring absorbance A1. The blank control was prepared by replacing the sample solution with absolute ethanol to determine the absorbance A0, and by replacing the ethanol solution DPPH with an equal volume of absolute ethanol to determine the absorbance A2. The results of comparing relevant substance components of examples 1-3 with comparative examples show that the antioxidant activity of the fermented tea powder liquid is obviously improved compared with that before fermentation.
In conclusion, the tea powder fermentation liquor provided by the invention has good antioxidant activity and can be added into cosmetics as an anti-aging additive.

Claims (6)
