동결건조가 끝난 발효물 0.5g를 보리새싹으로부터 폴리코사놀 추출을 위한 용매로 n-Hexane 추출 방법을 이용하여 보리새싹 유산균 발효물을 추출한 후 시험에 사용하였다. 25 mesh의 sieve에 거른 새싹보리 분말 100 g을 둥근 바닥 플라스크에 넣고, n-Hexane 2 L를 가한 뒤, 70 ℃로 맞춘 항온수조에 고정시켜 Hexane추출을 수행하였다. 6시간 후 추출액을 감압 여과(여과지 ADVANTEC Filter Paper No. 2)한 뒤 n-Hexane 1 L를 추가로 가해 3시간을 더 추출하고 다시 감압농축을 한 후 동결건조를 수행하였다. 보리새싹을로부터 폴리코사놀 추출을 위한 용매로 핵산만 국한하지 않고 물, 알코올 등 모두 유기용매 추출이 가능 하다.  0.5 g of the lyophilized fermented product was used as a solvent for policosanol extraction from barley sprouts, and the fermented product of barley sprout lactobacillus was extracted using n-Hexane extraction method. 100 g of the dried barley powder was poured into a round-bottomed flask, and 2 L of n-hexane was added thereto. 표 1 및 도 2를 살펴보면 비발효 보리새싹의 폴리코사놀 함량보다 유산균 발효를 시킨 보리새싹의 함량이 더 높다는 것을 확인할 수 있었다. 또한 S. thermophilus로 발효시킨 보리새싹보다 L. bulgaricus로 발효시킨 보리새싹이 폴리코사놀 함량이 더 높다는 것을 확인할 수 있었다. L. bulgaricus 발효물이 비발효물인 control(15 cm 보리새싹, 391.33 mg/100 g; 20 cm 보리 새싹, 465.50 mg/100 g)보다 폴리코사놀 함량이 약 2배 정도 높게 검출되었고, 15 cm 내외 보리새싹 L. bulgaricus 발효물 (BS-15-L)(826.02 mg/100 g)보다 20 cm 내외 보리새싹 L. bulgaricus 발효물 (BS-20-L)의 함량(935.82 mg/100 g)이 약 1.1배 정도 높게 나왔음을 확인할 수 있었다. 폴리코사놀이 가장 많이 분석되어진 L. bulgaricus 로 발효되어진 20 cm 보리새싹으로부터 폴리코사놀의 각 성분을 발효전과 후로 비교해볼 때, docosanol은 발효전에 비해 약 2.5배, tetracosanol은 약 2.3배, hexacosanol은 약 2.0배, heptacosanol은 약 2.5배, octacosanol은 약 2.4배 그리고 triacontanol은 약 2배가 증가하였다.When policosanol was inoculated from 20 cm barley sprouts fermented with L. bulgaricus , the content of docosanol was about 2.5 times higher than before fermentation, tetracosanol was about 2.3 times higher than before fermentation, hexacosanol was about 2.0 times higher than that before fermentation, heptacosanol was about 2.5 times, octacosanol was about 2.4 times, and triacontanol was about 2 times.

Cyclodextrin 무첨가구는 입자가 곱지 않고 뭉쳐있으며 만지면 달라붙는 emulsion 형태를 기준으로 cyclodextrin을 첨가하면 분말화가 진행됨을 알 수 있었고, 10% cyclodextrin 첨가 했을 때 완전히 분말화가 이루어짐을 확인할 수 있었다. 보리새싹 발효 유산균 스타터를 이용하여 40 ℃에서 72시간동안 발효시킨 후 동결건조 하여 헥산 추출 농축 전처리 과정 없이 시중에 판매되는 제품과 비교 분석한 결과를 하기 표 2에 나타내었다. 각각의 유산균을 사용하여 발효 했을 때와 같이 유산균 스타터를 사용하여 발효시켰을 때도 비발효 보리새싹보다 폴리코사놀 함량이 증가함을 확인할 수 있었다. 각각의 유산균을 사용하여 발효된 보리새싹의 폴리코사놀 함량 보다 적게 분석된 것은 헥산 추출을 따로 하지 않았기 때문에 함량이 상대적으로 적게 검출되었다. YC-X11로 발효한 보리새싹은 control(C)과 비교했을 때 폴리코사놀 함량이 높아 졌으며, 기능성을 가지고 있는 hexacosanol과 octacosanol이 증가된 것을 확인 할 수 있었다.  The fermented product was fermented at 40 ° C for 72 hours using barley germicide fermented lactic acid bacteria starter, followed by lyophilization and hexane extract concentration without pretreatment.
