Abstract

背景技术

发明内容
本发明的目的是提供一种适合作为具有合适血管化特性的骨替代物的新型生物材料。
本发明的目的还在于提供一种新的混合骨替代物,其包含矿物质组分和聚合物组分,所述骨替代物对于血管生成因子的持续释放是有效的。
本发明的另一个目的是提供一种包括纳米储层的新型生物材料,其允许生长因子的细胞接触依赖性释放,防止植入后被动释放并避免由活性分子的局部剂量递送引起的副作用。
本发明的另一个目的还在于提供一种适于治疗大骨缺损的新型生物材料。
因此,本发明涉及一种包括支架的生物材料,该支架含有矿物质组分,
其中所述矿物质组分包含至少一种磷酸钙化合物,并且
其中所述支架具有涂覆有由多层液滴制成的间断涂层的表面,所述多层液滴是由至少一个层对组成的液滴,该层对由一层聚阴离子和一层聚阳离子组成。
根据一种实施方式,本发明的生物材料的支架还包含聚合物组分。
“生物材料”是指适合在哺乳动物体内使用的任何材料,特别是在人类患者体内使用的任何材料。更具体地,根据本发明的生物材料适合用作植入物。
如上所述,根据本发明的生物材料的支架包含矿物质组分,并且任选地还包含聚合物组分。
矿物质组分
矿物质组分包含至少一种磷酸钙化合物,所述磷酸钙化合物优选地选自由以下组成的组:羟基磷灰石(HA)、无定形磷酸钙(ACP)、无水磷酸一钙(MCPA)、磷酸一钙一水合物(MCPM)、磷酸二钙二水合物(DCPD)、无水磷酸二钙(DCPA)、沉淀或缺钙磷灰石(CDA)、β-磷酸三钙(β-TCP)、磷酸四钙(TTCP)及其混合物。
根据一种实施方式,矿物质组分包括羟基磷灰石。
根据一种实施方式,矿物质组分包括β-磷酸三钙。
根据一种优选的实施方式,矿物质组分包括羟基磷灰石和β-磷酸三钙的混合物。
聚合物组分
根据一种实施方式,支架还包含聚合物组分。根据本发明,聚合物组分由聚合物制成。
优选地,聚合物组分由选自由以下组成的组中的聚合物制成:聚(ε-己内酯)、胶原蛋白、纤维蛋白、聚(乳酸)、聚(乙醇酸)、聚(乙二醇)-对苯二甲酸酯、聚(对苯二甲酸丁二醇酯)或其共聚物及其混合物。
当支架包含矿物质组分和聚合物组分时,涂覆在支架表面的如上定义的多层液滴存在于矿物质组分的表面,但也存在于聚合物部分的表面。
在支架的两个部分上都存在多层液滴是本发明的生物材料的有利特征,因此提供了具有有利特性的混合生物材料。
多层液滴
如上所述,本发明的支架具有涂覆有多层液滴的间断涂层的表面。这些液滴也可以命名为“纳米储层”或“纳米容器”。
本发明人惊奇地发现,可以用至少一个由以下组成的层对来涂覆支架(其矿物质组分和其聚合物组分):
-一层聚阴离子;和
-一层聚阳离子。
根据一些实施方式,生物材料支架是多层液滴涂覆的。
根据本发明的涂层优选地不规则地散布在支架表面上。
更具体地,根据本发明的生物材料支架逐层涂覆有交替带负电或带正电的层。
该涂层允许用治疗分子以产生治疗分子的纳米储层的方式将生物材料支架功能化,如下文所述。
术语“多层液滴”是指由至少一个层对组成的液滴或斑块(patches),该层对由一层聚阴离子和一层聚阳离子组成。所述液滴可以呈现不同的形状:圆形、椭圆形或鳞片形。优选地,所述液滴具有10至150nm、更优选15至100nm、甚至更优选25至50nm的尺寸。
根据本发明,术语“多层液滴涂层”是指设置在支架表面并通过带相反电荷的分子的多层液滴逐层(LbL)沉积获得的液滴或斑块的涂层。
术语“多层液滴涂层”还指支架的间断涂层,即沿着生物材料支架表面的涂层不是连续膜的形式。多层液滴涂层的特征可以在于其不规则的形状和/或这样的事实,即它不覆盖支架表面的全部,使得支架表面的至少一部分未被涂覆。本发明的多层液滴涂层可以与具有光滑表面并覆盖整个支架表面的薄膜涂层形成对比。
涂层的构建基于带相反电荷分子的逐层(LbL)沉积。也就是说,生物材料支架的涂层与制备聚电解质多层膜的方法相同。因此,根据本发明的生物材料在生物材料支架的表面上包含呈许多多层液滴形式的聚电解质多层。
与覆盖整个支架表面的薄膜涂层相反,根据本发明的多层液滴涂层优选仅部分地覆盖支架表面。根据本发明的涂层逐层施加(LbL),在连续吸附步骤之间通过冲洗步骤在每个步骤中去除过量的聚阴离子或聚阳离子。由于表面电荷的重新分配,聚阴离子或聚阳离子的第一层形成沿支架表面吸附的小液滴或斑块。在聚阴离子或聚阳离子涂覆的每个步骤中,每个液滴都被新的聚阴离子或聚阳离子层覆盖。当观察到多层液滴涂层时并在薄膜涂层之前停止涂覆过程。多层液滴涂层为支架提供了有利的特性,这是薄膜涂层无法观察到的。当获得薄膜涂层时,沿涂层支架表面无法再获得多层液滴。
与薄膜涂层或未涂覆支架相比,多层液滴涂层的第一个优点是其不规则的表面。这种不规则的形状提高了细胞对支架的粘附。此外,这种不规则的形状增加了涂层和细胞之间的接触表面,优化了涂层和细胞之间的交换。因此,需要较小浓度的治疗分子(如果存在)来观察对细胞生长的更好刺激。
此外,本发明的涂层比薄膜涂层使用更少的聚阴离子和聚阳离子层。因此需要减少数量的层来获得多层液滴涂层。
如本文进一步使用的,术语“聚电解质多层”特别包括涂覆根据本发明的生物材料支架的多层液滴。
在本说明书的框架中,术语“聚电解质”表示带有数个电解质基团的化合物,特别是其重复单元带有电解质基团的聚合物。这些基团将在水溶液中解离,产生聚阴离子或聚阳离子,视情况而定,并使聚合物带电。
涂覆纳米纤维支架的聚电解质多层由至少一个层对组成,该层对由一层聚阴离子和一层聚阳离子组成。它们可以例如包括1、2、3、4、5、6、7、8、9、10、11、12、13、14、15或更多个层对。优选地,它包括3至12个层对。
通过将生物材料支架交替浸入聚阴离子和聚阳离子溶液中,可以容易地获得聚电解质多层,特别是如本文所述的多层液滴。
对本领域技术人员显而易见的是,选择聚阴离子和聚阳离子的唯一要求是分子的电荷,即聚阴离子应带负电而聚阳离子应带正电。根据本发明的聚阴离子和聚阳离子可以对应于任何类型的分子,例如多肽(可选地化学修饰)或多糖(包括环糊精、壳聚糖等)。
根据优选的实施方式,在本发明的生物材料中,聚阳离子选自由以下组成的组:聚(赖氨酸)多肽(PLL)、共价偶联的环糊精-聚(赖氨酸)(PLL-CD)、聚(精氨酸)多肽、聚(组氨酸)多肽、聚(鸟氨酸)多肽、树枝状(Dendri-Graft)聚赖氨酸(例如树枝状聚-L-赖氨酸)、壳聚糖及其混合物。
更优选地,聚阳离子是壳聚糖。
根据优选的实施方式,在本发明的生物材料中,聚阴离子选自由以下组成的组:聚(谷氨酸)多肽(PGA)、聚(天冬氨酸)多肽及其混合物。
治疗分子
根据一种实施方式,本发明的生物材料还包含在至少一个多层液滴内的治疗分子或在所述治疗分子带电时形成至少一个多层液滴。
根据一种实施方式,如上所述,多层液滴的至少一个层对结合了治疗分子。
涂覆生物材料支架的聚电解质多层可以结合治疗分子,或聚电解质多层之一可以是治疗分子。
当结合到根据本发明的生物材料中的治疗分子带电时,当构建聚电解质多层时,所述治疗分子可用作聚阴离子或聚阳离子。当治疗分子不带电或带电不足时,它可以与聚阴离子或聚阳离子(例如上面列出的那些之一)共价连接以构建聚电解质多层。
根据一种优选的实施方式,在本发明的生物材料中,聚阴离子是治疗分子,特别是VEGF。
优选地,治疗分子是生长因子,该生长因子选自由以下组成的组:血管内皮生长因子(VEGF)、骨形态发生蛋白(BMP)、转化生长因子(TGF)、成纤维细胞生长因子(FGF)、编码其的核酸及其混合物。
治疗分子可结合到聚电解质多层中,如在例如WO 02/085423、WO2006/079928、Lynn(2006Soft Matter 2:269-273)、Decher(1997Science277:1232-1237)和Jessel等人,(2003Advanced Materials 15:692-695)中所述。
在本发明的框架内,生物材料支架可以用治疗分子功能化,允许所述治疗分子在根据本发明的生物材料的植入部位持续释放。
如本说明书通篇所用,术语“治疗分子”是指旨在治疗或预防疾病的任何分子。例如,它可以对应于已发布上市许可的药物(例如,由欧洲药品管理局(EMA)或美国食品和药物管理局(FDA)),或正在进行临床或临床前试验的候选药物。治疗分子可以例如对应于多肽(包括重组蛋白、抗体和肽)、核酸(包括RNA和DNA分子)、化学分子(例如小分子)或糖(例如脂多糖)。
当根据本发明的生物材料用于骨和/或软骨再生时,所述生长因子最优选地选自由以下组成的组:骨形态发生蛋白2(BMP2)、骨形态发生蛋白4(BMP4)、骨形态发生蛋白7(BMP7)、成纤维细胞生长因子1(FGF1)、成纤维细胞生长因子2(FGF2)、成纤维细胞生长因子4(FGF4)、成纤维细胞生长因子8(FGF8)、成纤维细胞生长因子9(FGF9)和成纤维细胞生长因子18(FGF18)。
在一具体实施方式中,聚电解质多层包括或由至少1、2、3、4、5、6、7、8、9、10、11、12、13、14、15或更多个层对组成,每个层对由以下组成:
-包含或由治疗分子(例如多肽,特别是生长因子)组成的聚阴离子层;和
-包含或由壳聚糖或赖氨酸聚合物(例如聚(赖氨酸)多肽(PLL)或树枝状聚赖氨酸(DGL))组成的聚阳离子层。
活细胞
根据一种实施方式,本发明的生物材料还包括活细胞。
在本发明的上下文中,生物材料支架可以进一步用活细胞功能化。事实上,植入活细胞是组织或器官修复的一个有前景的解决方案。
在骨和/或软骨再生的背景下,所述活细胞可以例如包含或由成骨细胞、软骨细胞、干细胞(例如间充质干细胞)、骨髓基质细胞或其混合物组成。优选地,所述活细胞包含或由成骨细胞、软骨细胞或其混合物组成。在一具体实施方式中,胚胎干细胞可以从根据本发明的活细胞中排除。
所述活细胞优选为人类细胞,最优选为自体细胞(即从待治疗患者获得的细胞)。
所述活细胞优选通过诱导型多能干细胞(iPSC)技术获得。
在一具体实施方式中,所述活细胞包含在沉积在所述涂覆支架上的水凝胶(例如藻酸盐水凝胶或胶原蛋白水凝胶)中。换言之,根据本发明的生物材料除了涂覆的支架之外还可以包括含有活细胞的水凝胶。
水凝胶是本领域技术人员众所周知的。胶原蛋白水凝胶可以例如通过将胶原蛋白(例如3mL的大鼠尾I型胶原蛋白)与含有10%FBS(例如5.5mL)的培养基和0.1M NaOH溶液(例如0.5mL)混合来制备。藻酸盐水凝胶可以例如是藻酸盐和透明质酸的混合物(例如藻酸盐:透明质酸溶液(4:1),其可以在pH7.4的0.15M NaCl溶液中制备)。
在根据本发明的一优选实施方式中,根据本发明的生物材料包括或由以下组成:
-涂覆有至少一个由一层聚阴离子和一层聚阳离子组成的层对的支架;和
-可选地包含在胶原蛋白水凝胶中的成骨细胞(沉积在所述涂覆的支架上)。
在另一优选实施方式中,根据本发明的生物材料包含或由以下组成:
-涂覆有至少一个由一层聚阴离子和一层聚阳离子组成的层对的支架;
-可选地包含在胶原蛋白水凝胶中的成骨细胞(沉积在所述涂覆的支架上);和
-包含在藻酸盐水凝胶中的软骨细胞(沉积在所述涂覆的支架上)。
在根据本发明的又一优选实施方式中,根据本发明的生物材料包括或由以下组成:
-涂覆有至少一个由一层聚阴离子和一层聚阳离子组成的层对的支架;
-包含在藻酸盐水凝胶中的软骨细胞(沉积在所述涂覆的纳米纤维支架上)。
在根据本发明的又一优选实施方式中,根据本发明的生物材料不包含活细胞。更具体地,它可以简单地由上面定义的支架组成,该支架涂覆有至少一个由一层聚阴离子和一层聚阳离子组成的层对。
制备生物材料的方法
本发明还涉及一种制备如上定义的生物材料的方法,所述方法包括用至少一个由一层聚阴离子和一层聚阳离子组成的层对涂覆含有矿物质组分和任选的聚合物组分的支架的步骤。
优选地,上述用至少一个层对涂覆的步骤包括以下步骤:
i.将支架浸入包含聚阳离子的溶液中(例如在约5至60分钟期间,优选在约15分钟期间);
ii.冲洗在步骤(i)结束时获得的支架(例如在约5至60分钟期间,优选在约15分钟期间);
iii.将在步骤(ii)结束时获得的支架浸入包含聚阴离子的溶液中(例如在约5至60分钟期间,优选在约15分钟期间);
iv.冲洗在步骤(iii)结束时获得的支架(例如在约5至60分钟期间,优选在约15分钟期间);和可选地;
v.重复步骤(i)至(iv)至少第二次;和可选地;
vi.对在步骤(iv)或(v)结束时获得的支架进行灭菌(例如通过暴露于紫外线)。
在步骤(i)和(iii)中,包含聚阳离子或聚阴离子的溶液可以例如包含约20μM至约500μM、优选约50μM至约200μM浓度范围内的聚阳离子或聚阴离子。除了聚阴离子或聚阳离子之外,所述溶液可以例如包含或由0.02M 2-(N-吗啉代)乙磺酸(MES)和0.15M NaCl组成。溶液的pH值优选为中性(例如pH值为7.4)。
在步骤(ii)和(iv)中,支架可以例如用具有中性pH(例如pH7.4)的溶液冲洗。所述溶液可以例如包含或由0.02M MES和0.15M NaCl组成。
步骤(v)可以重复任意次数,取决于应涂覆支架的层对的数量。
步骤(vi)可以例如通过暴露于紫外光(例如在254nm、30W,在20cm的照射距离下,约15分钟至约1小时,优选约30分钟)来进行。
在使用之前,可以平衡根据本发明的生物材料(例如通过使其与无血清培养基接触)。
对本领域技术人员显而易见的是,将纳米纤维支架浸入包含聚阳离子或聚阴离子的溶液中的步骤可以替换为将所述溶液喷涂到支架上的步骤。
制备根据本发明的生物材料的上述方法还可以包括以下步骤:
a)提供或获得活细胞(例如从患有骨和/或软骨缺损的患者身上分离的成骨细胞或软骨细胞);
b)将所述活细胞与水凝胶(例如胶原蛋白水凝胶或藻酸盐水凝胶)混合;和
c)将步骤(d)中获得的混合物沉积在步骤(b)中获得的生物材料上。
制备水凝胶的方法是本领域技术人员公知的。胶原蛋白水凝胶可以例如通过将胶原蛋白(例如3mL大鼠尾I型胶原蛋白)与含有10%FBS(例如5.5mL)的培养基和0.1M NaOH溶液(例如0.5mL)混合来制备。藻酸盐水凝胶可以例如是藻酸盐和透明质酸的混合物(例如藻酸盐:透明质酸溶液(4:1),其可以在pH7.4的0.15M NaCl溶液中制备)。
在一具体实施方式中,活细胞是成骨细胞,并且水凝胶是胶原水凝胶。在该实施方式的框架中,步骤(d)可以通过将成骨细胞悬浮液(例如以2ⅹ105个细胞mL-1)与胶原蛋白水凝胶(例如与9mL水凝胶混合的1mL成骨细胞悬浮液)混合来进行。在步骤(e)中,可以将胶原蛋白制剂倒在步骤(b)中获得的生物材料的顶部,然后可以培养以允许聚合(例如在37℃下约30分钟)。
在另一具体实施方式中,活细胞是软骨细胞,并且水凝胶是藻酸盐水凝胶。在本实施方式的框架中,步骤(d)可以通过将软骨细胞悬浮液(例如1×105个细胞mL-1)与藻酸盐水凝胶混合来进行。在步骤(e)中,可以将该制剂倒在步骤(b)中获得的生物材料的顶部。
使用前,可以切割圆柱体(例如使用无菌活检穿孔器),并在约37℃下培养,例如在5%CO2的潮湿环境中过夜。
或者,活细胞也可直接沉积在步骤(b)或(e)中获得的涂覆的支架上,无需预先与水凝胶混合。
当成骨细胞和软骨细胞都应沉积在本发明的涂覆的支架上时,上述制备根据本发明的生物材料的方法在步骤(a)和(b)之后还可以包括以下步骤:
c)提供或获得成骨细胞(例如从患有骨和/或软骨缺损的患者分离);
d)可选地将所述成骨细胞与胶原蛋白水凝胶混合;
e)将步骤(c)中获得的成骨细胞或步骤(d)中获得的混合物沉积在步骤(b)中获得的生物材料上;
f)提供或获得软骨细胞(例如从患有骨和/或软骨缺损的患者分离);
g)将所述软骨细胞与藻酸盐水凝胶混合;和
h)将步骤(g)中获得的混合物沉积在步骤(e)中获得的生物材料上。
这些步骤(c)至(h)例如可以如上文详细描述的那样进行。
本发明进一步提供了可通过本文所述的方法获得的生物材料。
用途
本发明还涉及如上定义的生物材料作为骨替代物的用途。
本发明还涉及如上定义的生物材料作为骨和/或软骨缺损填充材料的用途,或在骨和/或软骨再生中的用途。
本发明还涉及如上定义的生物材料在治疗骨和/或软骨缺损中的用途。
骨和/或软骨缺损可影响骨或软骨,或两者。它可以是例如软骨缺损、骨软骨缺损或软骨下骨缺损。
在根据本发明的具体实施方式中,骨和/或软骨缺损是软骨下骨缺损。因此,本发明提供了在以上段落中描述的生物材料在软骨下骨再生中的用途和/或在治疗软骨下骨缺损中的用途。
本发明还提供了在以上段落中描述的生物材料在骨软骨的骨再生中的用途和/或在治疗骨软骨的骨缺损中的用途。
特别地,根据本发明的生物材料可用于治疗患有剥离性骨软骨炎、骨坏死、骨软骨骨折、脊柱融合、由于受伤(例如运动伤害或事故造成的伤害)而导致的骨和/或软骨缺损、由于衰老而导致的骨和/或软骨缺损、需要颌面重建的骨和/或软骨缺损、需要鼻窦提升的骨和/或软骨缺损、需要牙槽嵴增强的骨和/或软骨缺损,或由于肿瘤(包括良性和癌性肿瘤)而导致的骨和/或软骨损失的患者的骨和/或软骨缺损。

材料和方法
VEGF/HEP复合物的沉积
将HEP(20mM/0.15mM Tris/NaCl中500μg ml-1,pH 6.8)、VEGF(20mM/0.15mM Tris/NaCl中200μg ml-1,pH 6.8)或HEP/VEGF(500μg ml-1/200μg ml-1)溶液(Sigma-Aldrich,Saint-Quentin-Fallavier,France)的液滴滴在盖玻片上并干燥。通过2次冲洗步骤,每次5分钟,缓冲液中的盐晶体溶解在去离子水中。该沉积过程重复6次以增加材料的量。
扫描电子显微镜(SEM)和原子力显微镜(AFM)研究
为了分析骨替代物上形成的HEP/VEGF-NR,样品在4℃下用4%多聚甲醛(PFA)固定10分钟。脱水后,通过SEM(使用Hitachi TM1000或FEG Sirion XL;FEI)在常规高真空模式下使用二次电子检测器观察样品。采用商用独立AFM显微镜Solver Pro(Nt-Mdt Inc.,Moscow,Russia)获取AFM图像。使用轻敲成像模式,NSG10悬臂梁设有105kHz的典型共振频率和2Nm-1的弹簧常数。图像分辨率设置为512ⅹ512,扫描速率为1Hz。使用开源软件Gwyddion 2.245(KlapetekP.Gwyddion:用于SPM数据分析的开源软件(an open-source software for SPM data analysis)。In:Open Physics(ed^(eds)(2012))分析图像。
骨替代物上的纳米储层沉积
将海绵(10%胶原蛋白I和III,90%陶瓷;Medical Biomat,Vaulx-en-Velin,France)切成5mm宽的碎片并置于96孔板中。将它们用紫外线(254nm,30W,距离20cm,暴露30分钟)进行消毒。壳聚糖-HEP-VEGF-NR是通过逐层沉积应用的,如前所述[29]。简而言之,将骨替代物交替浸入500μg ml-1壳聚糖溶液(Protasan UP CL 113,Novamatrix,Sandvika,Norway)和20mM/0.15mM Tris/NaCl中的500μg ml-1/200ng ml-1HEP/VEGF复合溶液中,pH 6.8。每次水浴后,将骨替代物在Tris/NaCl缓冲液中冲洗3次,每次5分钟。使用前,将骨替代物在无血清杜氏改良伊格尔培养基(D-MEM)中进行平衡。
HEP-VEGF复合物的分子建模
为了准备模拟的起始结构,从PDB结构文件(Fairbrother WJ,Champe MA,Christinger HW,Keyt BA,Starovasnik MA,血管内皮生长因子肝素结合域的溶液结构(Solution structure of the heparin-binding domain of vascular endothelialgrowth factor),Structure 6,637-648(1998))中提取了HEP钠盐(CAS9041-08-1)和VEGF165肝素结合域坐标(pdb id:2VGH,55个氨基酸)。使用AmberTools的前室程序,基于AM1-BCC方法分配肝素钠盐分子的部分原子电荷。肝素的范德华力和键合参数取自一般琥珀力场(GAFF)(Wang J,Wang W,Kollman PA,Case DA,分子力学计算中的自动原子类型和键类型感知(Automatic atom type and bond type perception in molecularmechanical calculations),J Mol Graph Model 25,247-260(2006))。然后使用acpypepython脚本将这些分子的AMBER格式文件转换为GROMACS格式。使用Cluspro和Patchdock对接程序,生成了肝素和VEGF的复合物。选择6个最佳复合物的坐标来为每个复合物生成拓扑文件以运行分子动力学(MD)模拟。
使用GROMACS-4.6、显式水模型进行30纳秒的MD模拟。将复合物置于立方周期盒的中心,并通过添加SPC水分子进行溶剂化。然后根据需要通过添加反离子来中和系统上的净电荷。使用最速下降算法完成能量最小化。使用v-重放缩(rescale)温度和Parrinello-Rahman压力耦合方法将温度和压力保持在300K和1个大气压。生产模拟进行30纳秒,时间步长为2飞秒。为了计算肝素和VEGF复合物的相互作用自由能,我们使用了MM/PBSA方案。使用g_mmpbsa工具82进行计算,该工具使用GROMACS软件包实施MM-PBSA方法。从100个快照(对于每个复合物)的样品中获得MM/PBSA能量,这些快照是从MD轨迹中提取的。所有计算均使用BWUniCluster Karlsruhe上的计算时间完成。
细胞培养
将表达绿色荧光蛋白的HUVEC(EssenBiotech,France)和来自骨髓的hMSC(PromoCell,Heidelberg,Germany)在各自的完全培养基(内皮生长培养基;间充质干细胞生长培养基,PromoCell)中于37℃、5%CO2的潮湿环境中培养。将1.5×104hMSC接种在每个沉积或未沉积HEP/VEGF-NR的海绵片段上,并在间充质干细胞生长培养基中培养7天。7天后,将5.5×104GFP-HUVEC接种在相同的样品上,并在由一半hMSC生长培养基和一半内皮生长培养基组成的培养基中培养。接种细胞的骨替代碎片共培养21天,然后根据下游应用进行处理。
细胞生物相容性分析
Alamar(Thermo Fischer Scientific, Waltham, MA, USA)用于评估细胞代谢活动随时间的变化(n=4)。在GFP-HUVEC接种3、14和21天后,将细胞在不含酚红(Lonza, Levallois-Perret, France)的10% AlamarD-MEM中于37℃和5%CO2下培养。4小时后,将上清液转移到96孔板中,并在Multiskan FC 读板器 (plate reader)(Thermo Fischer Scientific,Illkirch-Graffenstaden, France)中测量570和595nm处的吸光度, 以确定还原的Alamar的百分比。
免疫荧光
用细胞接种的骨替代物在4℃下用4% PFA固定10分钟,然后用PBS冲洗,然后脱矿物质并嵌入石蜡中,用于4μm连续切片。兔抗人PECAM1/CD31
(Microm,Brignais,France)和非物种特异性抗PECAM1/CD31(Abcam,Pairs,France),然后针对兔抗体产生二抗并与Alexa 594荧光染料(Molecular Probes,LifeTechnologies,Thermo Fisher Scientific)偶联,在PBS 1% BSA中以1:200 稀释。在PBS中多次冲洗后,将样品在200 nM 4',6-二脒基-2-苯基吲哚(DAPI;Sigma-Aldrich)中孵育10分钟,用于核复染。用LEICA DM4000B落射荧光显微镜(Leica Microsystems, Nanterre,France)观察样品。
在裸鼠中植入骨替代物
有关动物和组织的所有程序的设计均符合欧盟(2010/63/EU)的建议,并由Préfecture du Bas-Rhin 授权的调查人员执行。所有实验均在来自表示法国农业部兽医科学部的Préfecture du Bas-Rhin兽医公共卫生服务中心的批准号为C-67-482-35的斯特拉斯堡医学中心动物研究所进行。6周大的裸雄性小鼠(Crl:NIH-Foxn1nu;Charles River,L'arbresle,France)通过腹腔注射混合有10 mg kg-1甲苯噻嗪(2%, VIRBACSantéAnimale; Centravet, Nancy,France) 的100 mg kg-1氯胺酮(VIRBAC SantéAnimale;Centravet,Nancy,France)进行麻醉并放置在保持在37℃的加热板上。植入前5天,每个骨替代物接种1.0ⅹ105 hMSC。对于皮下植入物,进行背部皮肤切口并将骨替代物(直径=5mm)放置在皮肤和下方肌肉之间。用可吸收材料缝合切口,并在整个实验时间内观察小鼠。在4(n=6)或12(n=12)dpi后,通过腹膜内注射致死剂量的氯胺酮处死植入的小鼠。对于骨临界尺寸缺损的植入,进行皮肤切口,使用无
菌圆钻(500μm深,直径5mm)在颅骨(颅骨顶区)钻孔,并将骨替代物(直径=5mm)放入缺损处。用可吸收材料缝合切口,并在整个实验时间内观察小鼠。小鼠(n=12)在12dpi处死,外植体接受组织学、TEM和/或微CT评估。
苏木精-伊红(HE)组织学染色
对于HE染色,皮下外植体用4%PFA固定,脱矿物质并嵌入石蜡中,用于7μm连续切片。然后对样品进行HE染色并在Leica DM4000B显微镜(Leica Microsystems)上观察。对于血管化的定量分析,使用了imageJ软件。分析了在外植体中发现的血管的数量、平均大小和总面积。至少,每个切片5个图像,每个样本4个切片用于分析。
脉管系统的微血管造影和定量分析
皮下植入后12天,小鼠接受深度全身麻醉(戊巴比妥钠120mg Kg-1)。打开胸廓后,将输液针置于左心室。小鼠依次灌注(2ml min-1的速率)肝素(50U ml-1;以清除心血管系统)、4%PFA、PBS和不透射线硅橡胶(MV-122,Flow Tech Inc.)。灌注后,夹住心脏以避免造影剂泄漏;然后将小鼠置于4℃下过夜以允许造影剂聚合。然后将外植体在4%PFA中后固定48小时,然后在37℃下在恒定缓慢搅拌下用乙二胺四乙酸(EDTA,15%,PH=7.4)脱矿物质1周。将外植体放置在1%琼脂中,以避免在微CT采集过程中样品发生任何移动。使用配备有180kV-15W高功率纳米聚焦管和钨传输靶的微型CT X射线系统(GE Sensing&Inspection Technologies GmbH,Wunstorf,Germany)进行断层扫描实验。以5mm2的各向同性像素尺寸和约15.4×12.0mm2的视野进行X射线显微CT。对于每次测量,用60kV加速电压和310mA束电流的X射线照射样品。在每个旋转角度位置,采集六幅图像并将其平均为一个投影。360°上的1700次投影导致总扫描持续时间约为100分钟。
使用phoenix datos|x 2.0软件(phoenix|x-ray,GE Sensing&InspectionTechnologies GmbH,Wunstorf,Germany)进行数据采集和重建。
从显微CT扫描获得的灰度图像被分为两类,以区分血管内的体素和外部的体素。对于每个图像,将手动调整阈值应用于强度水平。将10μm宽的结构化组件应用于每个图像,以对分割进行去噪。然后,擦除连接的小于20μm3的体素组。分割后,生成的血管被绘制为骨架,其中骨架被定义为定义中心线和血管形状的局部半径的有序点集,如前面描述的算法。
透射电子显微镜(TEM)
微CT扫描后,将12dpi的外植体固定在2.5%戊二醛和2.5%PFA的0.1M甲次砷酸盐缓冲液中,pH7.4。将样品后固定在1%四氧化锇中,脱水,在环氧丙烷中调节并嵌入Spurr的环氧树脂Epon 812(Electron Microscopy Sciences,Ft.Washington,PA)。半薄切片(2μm)和超薄切片(70nm)在Leica Ultracut UCT超薄切片机(Leica Microsystem)上制备,与醋酸铀酰(Laurylab,Brindas,France)和柠檬酸铅(Euromedex EMS,Souffelweyersheim,France)进行对比,并用Morgagni 268D电子显微镜(FEI-Phillips,ThermoFisherScientific)在70kv下检查。使用Mega View III相机(Soft Imaging System,Münster,Germany)捕获数字图像。
统计分析
使用BioStatGV(Sentiweb,France)和Prism5(GraphPad,La Jolla,CA,USA)进行统计分析。Alamar Blue代谢测定、组织学数据和微血管造影数据使用未配对的双尾学生t检验进行分析。当发现数据集之间的方差不同时,则应用Welch校正。
实施例1:用血管生成复合分子纳米功能化的骨替代物的设计和建模
为了促进移植的骨替代物的血管化,发明人使用NR技术(图1A)(Mendoza-Palomares C等人,配备治疗剂纳米储层的智能混合材料(Smart hybrid materialsequipped by nanoreservoirs of therapeutics),ACS Nano 6,483-490(2012))对FDA批准的海绵进行了功能化;Eap S等人,用于骨再生的配备“鱼鳞”状生长因子纳米储层的胶原植入物(Collagen implants equipped with'fish scale'-likenanoreservoirs of growth factors for bone regeneration),Nanomedicine(Lond)9,1253-1261(2014);Schiavi J等人,用于骨再生纳米医学的结合人体干细胞微组织和生长因子的活性植入物(Active implant combining human stem cell microtissues andgrowth factors for bone-regenerative nanomedicine),Nanomedicine(Lond)10,753-763(2015);和Eap S等人,用于骨再生的用活性生长因子和干细胞双功能化的活体厚纳米纤维植入物(A living thick nanofibrous implant bifunctionalized with activegrowth factor and stem cells for bone regeneration),Int J Nanomedicine 10,1061-1075(2015)。肝素对VEGF的血管生成活性至关重要,其通过位于其羧基端结构域的肝素结合结构域结合生长因子(Fairbrother WJ,Champe MA,Christinger HW,Keyt BA,Starovasnik MA,血管内皮生长因子的肝素结合域的溶液结构(Solution structure ofthe heparin-binding domain of vascular endothelial growth factor),Structure6,637-648(1998);Krilleke D,NgY-Shan E,Shima David T,肝素结合域赋予VEGF-A在发育和疾病中的多种功能:结构-功能研究(The heparin-binding domain confers diversefunctions of VEGF-A in development and disease:a structure–function study),Biochemical Society Transactions 37,1201-1206(2009))。为了通过NR介导的细胞接触依赖性递送使血管生成作用最大化,发明人首先研究了HEP/VEGF165复合物对骨替代物功能化的适用性。将肝素、VEGF或其复合物逐滴沉积在玻璃盖玻片上,并通过扫描电子显微镜可视化(图1B-D)。复合的HEP/VEGF165形成大团聚体(60.0±15.4nm),如AFM成像所证实的(图1E)。
为了理解为什么复合的HEP/VEGF聚集体具有如此大的尺寸,发明人使用分子动力学模拟对HEP和VEGF165之间的分子相互作用进行了建模。肝素是一种线性硫酸化多糖,带高负电荷,而VEGF165的HEP结合域的表面静电势带正电荷(Ono K,Hattori H,Takeshita S,Kurita A,shihara M,肝素中与VEGF165相互作用并调节其生物活性的结构特征(Structural features in heparin that interact with VEGF165 and modulate itsbiological activity),Glycobiology 9,705-711(1999))。因此,使用HEP的硫酸根阴离子和VEGF的精氨酸(Arg)残基作为模拟分子相互作用的指导,进行了HEP模型与VEGF165的NMR模型的对接模拟。大多数阳性Arg残基要么聚集在VEGF165的中央结构域(Arg35、Arg39、Arg46、Arg49)中,要么聚集在其N端结构域的环内(Arg13、Arg14)。根据计算出的溶剂可及表面的静电势,这些区域代表高负电荷分子(如HEP)的结合位点。使用分子对接生成的HEP/VEGF复合模型经过最陡峭的体面能量最小化,然后是平衡和30纳秒的分子动力学生产运行。然后通过分子力学泊松-玻尔兹曼表面积(MM/PBSA)方法,根据复杂模型的能量对复杂模型进行排序。由此产生的具有最低能量的HEP/VEGF复合结构(图1F)清楚地表明,净电荷在HEP(图1F中的棒模型)与VEGF165(图1F中的同源模型)的亲和力中起着重要作用。HEP的硫酸盐基团(图1F中的黄色)与上述Arg残基和VEGF165的亮氨酸17和苏氨酸47残基的侧链强烈相互作用,稳定了HEP/VEGF复合物并可能促进大团聚体的形成。
实施例2:用HEP/VEGF-NR纳米功能化的骨替代物改善体外内皮细胞的组织
肝素/VEGF复合物被整合到壳聚糖NR中并沉积在海绵上。在SEM上观察到功能化的骨替代物,并与仅壳聚糖-NR(非功能化NR:NF-NR)进行比较。在沉积6个循环后,我们观察到HEP/VEGF-或NF-NR在骨替代物的矿物质(图2A、C)和胶原蛋白(图2B、D)部分均匀分布。为了研究纳米功能化骨替代物的促血管生成作用,将HEP/VEGF-NR、HEP/BSA-NR或NF-NR沉积在骨替代物的碎片上,这些骨替代物又接种了hMSC和表达绿色荧光蛋白(GFP)的人脐静脉内皮细胞(GFP-HUVEC)。培养21天后监测内皮细胞的组织。在HEP/VEGF-NR存在的情况下,GFP-HUVEC组织成一系列细胞,类似于血管状结构(图2E中的红色星号),在许多情况下显示管腔。相反,在存在NF-NR或HEP/BSA-NR的情况下(图2E),内皮细胞主要以单细胞形式分布在移植物上。血管样结构通过连续拉伸排列的GFP-HUVEC的数量进行评估。在纳米活性支架上,这些平均为3.4±0.5个细胞长度,而在非功能化支架或用非血管生成HEP/BSA功能化的支架上,这些分别为1.2±0.1和1.4±0.2个细胞长度(p≤0.001)(图2F)。由于HEP/BSA-NR的存在产生了与NF-NR相似的结果,我们得出结论,纳米活性骨替代物在体外促进内皮细胞组织的能力取决于HEP/VEGF复合物的存在和细胞可用性。我们还评估了接种在非功能化支架上的细胞的代谢活性,并通过阿拉玛蓝测定将其与在HEP/VEGF-NR存在下接种的细胞的代谢活性进行比较。在这两种情况下,阿拉玛(alamar)蓝的减少从第0天到第21天增加(图2G),表明没有细胞毒性线索影响细胞。在所考虑的培养期间,NF-NR和HEP/VEGF-NR之间没有观察到显著差异。然而,虽然用NF-NR培养的细胞在第3天和第14天之间显示出突然的代谢增加(p≤0.1),但在存在HEP/VEGF-NR的情况下培养的细胞显示出更稳定的代谢率(对于第3-14天和第14-21天,分别为p≤0.05和p≤0.01)。这些数据表明,在非功能化支架上,细胞可能比在HEP/VEGF-NR存在下增殖更多。
实施例3:皮下植入纳米活性骨替代物的募集的宿主脉管系统
用HEP/VEGF-NR功能化的骨替代物改善了体外内皮细胞的组织,引发了血管样结构的形成。因此,发明人评估了血管生成纳米活性骨替代物是否以及如何能够有效地触发体内血管生成。本发明人在裸鼠的背部皮下植入HEP/VEGF-NR或NF-NR骨替代物。在植入之前,所有骨替代物都接种hMSC并培养7天。在植入后4天或12天(dpi,即植入后天数)从小鼠中取出骨替代物,并在组织学水平上评估其血管化程度。一般来说,与NF相比,外植的纳米活性骨替代物显示出更好的宿主血管募集(图3A、B中的白色箭头)。仅在存在HEP/VEGF-NR的情况下,骨替代物核心中发现的血管的量化显示显著增加(在4dpi和21dpi时分别为14.8±1.7mm-1和61.0±12.9mm-1;p=0.002),与NF骨替代物相比(在4dpi和12dpi时分别为13.2±2.74mm-1和22.6±8.0mm-1)(图3C)。在4dpi时未观察到募集的血管直径有显著差异(在HEP/VEGF-NR和NF-NR骨替代物中分别为17±1μm与18±2μm)(图3E)和相对血管面积(在任一条件下均低于1%)(图3E)。当在HEP/VEGF-NR骨替代物中发现的宿主血管的大小和相对面积增加1.5-(p=0.002)和8倍(p=0.0003)时,照片在12dpi时发生了相当大的变化,而在NF-NR骨替代物中没有观察到差异(图3E)。
这些结果表明,由于NR技术,HEP/VEGF复合物的存在和延长的可用性如何增加了从骨替代物核心中的宿主募集的血管的数量和大小,这表明还可以预期移植物具有更好的功能。
实施例4:纳米活性骨替代物在临界尺寸颅骨骨缺损小鼠模型中促进血管生成
最后,我们测试了纳米活性HEP/VEGF骨替代物在临界尺寸骨缺损小鼠模型中的作用。针对小鼠钻出一部分颅顶(诱发颅骨缺损),并填充HEP/VEGF-NR或NF-NR骨替代物。在12dpi时,植入的动物被灌注放射性橡胶造影剂,然后移植骨替代物并进行显微CT扫描。与空NR相比,产生移植小鼠的3D微血管造影(显微CT扫描图像的3D描述),以显示纳米活性HEP/VEGF复合物的存在对宿主血管系统募集的影响(图4A)。新形成的血管紧密地穿透HEP/VEGF-NR骨替代物,几乎覆盖了植入物的整个体积(图4B,左图)。相反,宿主脉管系统稀疏地定植了NF-NR骨替代物(图4B,右图)。值得注意的是,当使用标准欧拉距离生成到分割图像中最近相邻血管的距离图时,可以看出曲线的趋势(图4C)和分布的模式(对于1NF、2NF和3NF,分别为0.32、0.25和0.05mm;对于1F、2F和3F,分别为0.06、0.02和0.05mm)清楚地表明,与NF相比,纳米活性骨替代物中任何点到最近相邻血管的平均距离更小(p=0.0423)。分析还显示,NF骨替代物的血管密度为0.75±0.99%,而HEP/VEGF-NR骨替代物的血管密度为2.8±0.76%(p=0.0167),几乎是NF骨替代物的血管密度的4倍。
总之,这些数据表明骨替代物的纳米功能化导致在治疗的颅骨骨缺损内侧形成更密集的新血管云,这是骨再生的必要前提。
在存在矿物质块的情况下,血管会停止或被切成碎片,从而产生许多非常小的血管,如图4所示。必须指出的是,为了使血管网络可见,该块预先脱矿物质,这意味着它的矿物质元素被消除了。
使用本发明的创新技术,如图4所示,血管不再遇到障碍物,因此能够沿着它们的路径前进。
实施例5:接种在骨替代物上的人类MSC有助于血管生成
在显微血管造影中观察到的骨替代物内的血管看起来与周围血管连接良好。此外,植入的动物没有出血,当灌注造影剂时,它们的周围骨骼中没有出现任何造影剂渗漏,这可能指向亚功能性脉管系统。为了更仔细地观察新形成的血管的形态,我们在超微结构水平上对其进行了分析。在骨替代物中发现的血管的特征是存在紧密连接的正常形态的内皮细胞,周围环绕着为血管提供结构稳定性的壁细胞(图5A和5B)。植入后骨替代物内发现的血管的功能以及它们形成的速度对于避免移植骨替代物的细胞成分坏死都至关重要。功能化和非功能化骨替代物在植入前都接种了hMSC,以再生缺失的骨骼。间充质干细胞是多能干细胞,已知能够在许多细胞类型中分化,如成骨细胞、脂肪细胞、软骨细胞甚至神经元。研究还表明,至少在体外,MSC可以有效地在内皮细胞中分化。因此,发明人研究了接种在骨替代物上的hMSC是否有助于在植入物中发现的血管的形成。通过免疫组织化学,发明人检测到抗人PECAM1抗体阳性的几个内皮细胞(图5C,上图),它们是骨替代物内新形成的血管内皮的一部分(图5C中的白色星号)。这些细胞中只有一部分源自移植的hMSC,如使用与人和小鼠PECAM1发生交叉反应的抗体的免疫组织化学所示(图5C,中图)。正如预期的那样,在对照小鼠骨骼上使用抗人PECAM1抗体未检测到信号(图5C,下图)。这些结果表明,植入的骨替代物内新血管的形成取决于NR和hMSC的同时存在。因此得出的结论是,血管生成生长因子在较长时间内的细胞接触依赖性释放与内皮细胞中hMSC的主动分化相结合,协同促进了渗入纳米功能化骨替代物的功能性血管化。
本发明基于骨替代材料的纳米功能化,其允许VEGF的细胞接触依赖性释放。为了最大限度地发挥血管生成作用,VEGF165与HEP复合。收集到的证据表明,HEP/VEGF复合物形成了比单独的VEGF更大的聚集体(图1B-E),这是由于与HEP的化学物理相互作用,正如计算机建模所预期的(图1F)。纳米储层介导的骨替代物的功能化非常有利,因为它有效地克服了与高局部剂量VEGF相关的副作用。此外,由于VEGF165的半衰期很短,约为90分钟,当暴露于细胞外环境时,在壳聚糖NR内,它不会发生降解,因此细胞可以长时间使用。在这项工作中,我们展示了HEP/VEGF-NR如何在体外诱导血管样结构中的内皮细胞的组织(图2E、F),并可以改善临床使用的皮下植入(图3A)-C)或在临界尺寸的颅骨骨缺损中植入(图4A-C)的双相骨替代物的血管化。尽管发现一种植入颅骨缺损的NF-NR骨替代物中的宿主血管的空间分布与HEP/VEGF-NR骨替代物相似,与纳米活性骨替代物(2.8%)相比,血管密度(1.9%)较低。这些结果可能归因于血管的平均直径,并且可以通过在一些组织学标本中观察到的血管成熟度的差异来解释。
与HEP/VEGF-NR一起,双相海绵在植入前接种了hMSC。最近,随着第三代生物材料的出现,成体间充质干细胞作为治疗剂的使用引起了临床应用的极大兴趣。由于营养因子的分泌,这些细胞不仅能够分化为成熟组织,而且还能够调节免疫反应、防止细胞凋亡和促进血管生成。此外,间充质干细胞显示出通过在壁细胞和内皮细胞中分化来稳定新形成的血管。hMSC、内皮细胞和3D生物材料的联合使用显示出特别增加血管网络和新骨组织的形成。
因此,发明人将血管生成NR与hMSC的存在结合到临床使用的双相海绵上,并观察到这两种参与者如何协同促进骨替代物在体内的血管生成。我们发现,除了改善从宿主募集内皮细胞外,hMSC还积极促进这些内皮细胞形成血管,因为在定植骨替代物的脉管系统中发现了几种人类来源的内皮细胞(图5C)。
总之,由于血管生成因子和hMSC在骨替代物上同时存在细胞接触依赖性缓释,因此进行了以下观察:1)功能性脉管系统的形成,这通过在大移植物的核心处也存在壁细胞来证明(图5A、B);2)在存在VEGF/HEP-NR和hMSC的情况下发现的血管的数量、大小和相对面积方面,皮下植入的骨替代物的血管形成更好(图3A-C);3)在颅骨大骨缺损中植入的骨替代物中的血管网络更密集(图4A-C);和4)hMSC对血管生成的积极贡献(图5C)。
截至今天,自体骨移植(自体移植)是治疗大骨缺损的金标准。全世界每年有≥200万例手术,它是仅次于输血的第二大最常见的组织移植。但是,它也有缺点。就感染而言,骨自体移植物比使用合成骨替代物的并发症更多,而且成本更高。同种异体骨移植物比自体移植物更广泛,因为它们必须在临床使用前进行适当的治疗。然而,自体移植物必然会引入第二个手术部位、更长的手术室时间和更长的术后慢性疼痛,这增加了该技术的总体成本。因此,需要先进的功能化材料来克服目前的局限性(缺乏从宿主组织募集的血管和植入骨替代物的核心处诱导的坏死)并诱导植入骨替代物的血管化,这是功能性骨组织再生的基石。
因此,本发明涉及基于血管生成纳米储层和间充质干细胞的组合存在的纳米功能化骨替代物,其旨在改善临界尺寸骨缺损中的血管生成(vasculoneogenesis)。从生物医学和公共卫生的角度来看,这里介绍的结果都具有很大的相关性。这些结果表明,这种创新策略应用于已经在临床中使用的骨替代物,能够1)在体外在血管样结构中组织内皮细胞,2)有助于皮下植入物中新血管的形成和3)改善关键骨缺小鼠模型中的血管生成。本发明的纳米功能化骨替代物现在可以在治疗大骨缺损时替代自体和同种异体移植物,并且对活性成分(细胞和生长因子)的组成进行适当的修改,也可用于其他组织的再生。
Claims (14)
1.一种生物材料,包括含有矿物质组分的支架,其中,所述矿物质组分包含至少一种磷酸钙化合物,
其中,所述支架具有涂覆有由多层液滴制成的间断涂层的表面,所述多层液滴是由至少一个层对组成的液滴,所述层对由一层聚阴离子和一层聚阳离子组成。
2.根据权利要求1所述的生物材料,其中,所述支架还含有聚合物组分。
3.根据权利要求1或2所述的生物材料,还包括在至少一个多层液滴内的治疗分子或当所述治疗分子带电时形成至少一个多层液滴。
4.根据权利要求3所述的生物材料,其中,所述治疗分子是选自由以下组成的组中的生长因子:血管内皮生长因子(VEGF)、骨形态发生蛋白(BMP)、转化生长因子(TGF)、成纤维细胞生长因子(FGF)、编码其的核酸及其混合物。
5.根据权利要求1至4中任一项所述的生物材料,其中,所述磷酸钙化合物选自由以下组成的组:羟基磷灰石(HA)、无定形磷酸钙(ACP)、无水磷酸一钙(MCPA)、磷酸一钙一水合物(MCPM)、磷酸二钙二水合物(DCPD)、无水磷酸二钙(DCPA)、沉淀或缺钙磷灰石(CDA)、β-磷酸三钙(β-TCP)、磷酸四钙(TTCP)及其混合物。
6.根据权利要求2至5中任一项所述的生物材料,其中,所述聚合物组分由选自由以下组成的组中的聚合物制成:聚(ε-己内酯)、胶原蛋白、纤维蛋白、聚(乳酸)、聚(乙醇酸)、聚(乙二醇)-对苯二甲酸酯、聚(对苯二甲酸丁二醇酯)或其共聚物,及其混合物。
7.根据权利要求1至6中任一项所述的生物材料,其中,所述聚阳离子选自由以下组成的组:聚(赖氨酸)多肽(PLL)、共价偶联的环糊精-聚(赖氨酸)(PLL-CD)、聚(精氨酸)多肽、聚(组氨酸)多肽、聚(鸟氨酸)多肽、树枝状聚赖氨酸(例如树枝状聚-L-赖氨酸)、壳聚糖及其混合物。
8.根据权利要求1至7中任一项所述的生物材料,其中,所述聚阴离子选自由以下组成的组:聚(谷氨酸)多肽(PGA)、聚(天冬氨酸)多肽及其混合物。
9.根据权利要求1至8中任一项所述的生物材料,还包括活细胞。
10.一种制备根据权利要求1至9中任一项所述的生物材料的方法,所述方法包括以下步骤:用至少一个层对涂覆含有矿物质组分和任选的聚合物组分的支架,所述层对由一层聚阴离子和一层聚阳离子组成。
11.根据权利要求10所述的方法,其中,用至少一个层对涂覆的步骤包括以下步骤:
vii.将支架浸入包含聚阳离子的溶液中;
viii.冲洗在步骤(i)结束时获得的支架;
ix.将在步骤(ii)结束时获得的支架浸入包含聚阴离子的溶液中;
x.冲洗在步骤(iii)结束时获得的支架;和任选地;
xi.重复步骤(i)至(iv)至少第二次;和任选地;
xii.对在步骤(iv)或(v)结束时获得的支架进行灭菌。
12.根据权利要求1至9中任一项所述的生物材料作为骨替代物的用途。
13.根据权利要求1至9中任一项所述的生物材料作为骨和/或软骨缺损填充材料的用途,或在骨和/或软骨再生中的用途。
14.根据权利要求1至9中任一项所述的生物材料在治疗骨和/或软骨缺损中的用途。
