技术领域和背景技术
在本发明的一些实施例中,本发明涉及多种培养γδ(Gammadelta)T细胞群的方法、多种培养的γδT细胞群的治疗用途以及多种包含培养的γδT细胞的试剂盒。更具体地,但并非特定地,本发明涉及多种培养的γδT细胞单独或与其他细胞组合用于移植的用途。
伽玛德尔塔(Gammadelta,γδ)T细胞是一种保护的先天性淋巴细胞群体,在组织内环境稳定、感染性疾病、自身免疫性疾病、炎症、移植和肿瘤监测期间参与多种免疫反应。γδT细胞可以共享适应性或先天性免疫系统的属性,或两者的属性,包含识别应激相关的抗原的胸腺和外周组织亚群(Vδ1细胞)、由磷酸化抗原激活的循环亚群(Vδ2细胞)和主要存在于肝脏中的亚群(Vδ3细胞),常见于病毒感染和白血病。

具体实施方式
在本发明的一些实施例中,本发明涉及多种培养γδ(Gammadelta)T细胞群的方法、多种培养的γδT细胞群的治疗用途以及多种包含培养的γδT细胞的试剂盒。更具体地,但并非特定地,本发明涉及多种培养的γδT细胞单独或与其他细胞组合用于移植的用途。
烟酰胺(NAM),来自烟酸的酰胺形式(烟酰胺,维生素B3)是一种碱基交换基质及NAD(+)依赖酶的有效抑制剂,赋予单ADP核糖转移酶和多ADP核糖转移酶的活性。作为一种微量的微营养素,烟酰胺需要以微摩尔的量来保证体外培养的哺乳动物细胞的活力和增殖,并且通常与其他维生素一起被纳入细胞培养基的配方中。与其他淋巴细胞组分一样,γδT细胞通常在含有烟酰胺的培养基中进行体外培养,浓度范围为约8μM烟酰胺(MEMα,RPMI)至约33μM(DMEM),以促进γδT细胞的强健生长(例如,参见Jakobovits等人美国专利申请案公开第2016/0175358号)。
已发现较高浓度的烟酰胺可有效增强CD34+和CD133+造血干细胞和祖细胞(例如,参见美国专利第7,955,852号和第8,846,393号)以及CD56+自然杀伤细胞(例如,参见Frei等人,Blood期刊,2011年,118:4035)的扩增和功能,但也有报道称,烟酰胺可抑制对激活信号的反应性及诱导T细胞(例如,参见Liu等人,J Immunol期刊,2001年,167:4942-4947)和中性粒细胞(Fernandes等人,Am J Physical Lung Cell Mol Phys期刊,2011年,300:L354-361)的细胞凋亡。
本申请发明人出人意料地示出,如本文进一步详述的,在γδT细胞增殖条件下体外培养的人类γδT细胞的富集群体中添加毫摩尔浓度的烟酰胺可增强功能性,例如,当注入SCID小鼠宿主时,γδT细胞的归巢和组织保留能力更强。
由于γδT细胞在组织内环境稳定、感染性和自身免疫性疾病、炎症、移植和肿瘤监测期间参与免疫反应,对感染细胞和肿瘤细胞表现出自发的非MHC限制的细胞毒性活性,并在体内介导对病毒感染和癌症发展的抵抗,有效增加γδT细胞功能性的方法可用于肿瘤治疗和清除感染细胞,具有更强的反应性和更少的不良反应。
因此,开发有效增强培养的γδT细胞功能以及在注入后其在宿主组织体内归巢和保留的可能性的方案,可以提高治疗的成功率,例如过继免疫治疗,γδT细胞用于治疗实体瘤、恶性肿瘤、病毒性和自身免疫性疾病等。
因此,根据本发明实施例的一个方面,提供了一种增强γδT细胞归巢和/或保留潜能的方法,所述方法包含:获得富集多个γδT细胞的一选定细胞群,体外为所述选定细胞群提供γδT细胞扩增的多个条件,及提供0.5至50mM范围内的烟酰胺持续一时间段,以显着增强γδT细胞归巢和/或保留潜能,从而增强所述选定细胞群中多个γδT细胞的归巢和/或保留潜能。
如本文所用,术语“γδT细胞”在本文中也可称为γδT细胞(γδT-cell,gammadelta T cell),或进一步称为gd T-细胞或gd T细胞。
γδT细胞是通过表达由γ(gamma)链和δ(delta)链组成的异二聚体T细胞受体(TCR)来定义的。这使它们区别于典型和更为人熟知的表达αβTCR的CD4+辅助T细胞和CD8+细胞毒性T细胞。γδT细胞选择(胸腺的)的机制仍不清楚。
γδT细胞通常显示共享相同TCR链的寡克隆亚群的组织特异性定位。例如,人类外周血γδ-T细胞主要是Vγ9/Vδ2+,而小鼠皮肤γδT细胞,即所谓的树突状表皮T细胞(DECT细胞),主要是Vγ5/Vδ1+。一般来说,γδT细胞在上皮和粘膜组织中富集,被认为是抵抗致病的挑战的第一道防线。
如本文所用,“γδT细胞激活”是指与γδT细胞相关联的任何可测量的生物现象,其代表被激活的此类T细胞。此类生物现象的非限制性示例包含细胞因子产生的增加、细胞表面蛋白质定性或定量组成的变化、T细胞增殖的增加,和/或T细胞效应物功能的增加,如此杀死标靶细胞或协助另一效应物细胞以杀死标靶细胞。
根据本发明的一些实施例,本发明的方法增强γδT细胞的归巢和/或保留潜能。
如本文所用,术语“功能”或“γδT细胞功能”是指归属于γδT细胞的任何生物功能。γδT细胞功能的非限制性列表包含,例如,细胞毒性、诱导凋亡、细胞运动、定向迁移、细胞因子和其他细胞信号反应、细胞因子/趋化因子的产生和分泌、试管中激活和抑制细胞表面分子的表达、细胞归巢和移植宿主体内保留,以及体内疾病或疾病过程的改变。在一些实施例中,通过暴露于烟酰胺和/或其他烟酰胺部分而增强的γδT细胞功能包含CD62L表面标记物的高表达、高迁移反应和γδT细胞的更高的细胞毒性活性中的至少一种,以及增强注入的γδT细胞的归巢和体内保留。在具体实施例中,通过暴露于烟酰胺和/或其他烟酰胺部分而增强的γδT细胞功能包含γδT细胞的CD62L表面标记物的提高的表达和注入的γδT细胞的提高的归巢和体内保留中的至少一种。在具体实施例中,通过将γδT细胞暴露于烟酰胺和/或其他烟酰胺部分而增强γδT细胞的CD62L表面标记物的表达和注入的γδT细胞的归巢和体内保留。
在本领域是众所周知,对粘附和迁移分子(例如CD62L、CXCR-4、CD49e等)的分析对于移植中细胞的归巢和保留是非常重要的。例如,可以通过流式细胞术、免疫检测、定量cDNA扩增、杂交等来分析细胞中的CD62L表达。在一个实施方案中,通过将细胞暴露于荧光标记的特异性人抗体(anti-human)的CD62L单克隆抗体(例如,来自BioLegend公司(加利福尼亚州圣地亚哥市,美国)的CD62L PE,目录号304806)并通过荧光激活细胞分选(FACS)分选细胞,在不同的γδT细胞群中检测CD62L表达。
细胞迁移的分析在本领域是众所周知的。例如,可以通过迁移分析或间隙闭合(gap closure)分析来分析细胞的迁移。在迁移分析中,如双室(two-chamber)技术,细胞通过屏障(如过滤器)与刺激物分离,并通过以特定间隔计数来源细胞的损失、穿过屏障的细胞累积,或其两者来检测细胞的迁移。在间隙闭合分析中,细胞被放置在可见间隙(刻痕琼脂板、围绕一圈等)的外围,并与刺激物一起培养。利用细胞仪、免疫检测、显微术/形态测定法等可视化通过细胞运动对刺激的反应应用的细胞之间空间的闭合。在一个实施例中,通过TranswellTM迁移分析确定不同细胞群的迁移潜能。
输血或移植细胞的归巢和体内保留的分析在本领域是众所周知的。如本文所用,术语“归巢”指注入或移植细胞到达宿主靶器官并在宿主靶器官中存活的能力。例如,γδT细胞靶器官可以是淋巴组织,肝细胞靶器官可以是肝实质,肺泡细胞靶器官可以是肺实质等。如本文所用,术语“体内保留”是指注入或移植细胞的留在于其中的能力,可选地增殖并在靶器官中保持存活。用于检测移植的γδT细胞归巢和体内保留的动物模型包含但不限于免疫缺陷的小型哺乳动物(例如SCID和IL2Rγ空(IL2Rγnull)小鼠等)。SCID-Hu小鼠模型采用C.B-17scid/scid(SCID)小鼠移植人类胎儿胸腺和肝组织或胎儿BM组织,以为评估移植的人类“γδT细胞保留和治疗潜能提供了合适的模型。移植细胞的归巢和体内保留也可以在人类宿主对象中进行评估。在一个实施方案中,在经辐照的NOD/SCID小鼠中分析归巢和体内保留,所述小鼠经输入例如约15X104、约15X105、约15X106、约15X107或更多的人类γδT细胞富集的多个细胞,所述人类γδT细胞富集的多个细胞根据本发明使用有效浓度的烟酰胺培养,并在输入后一预定时间(例如,输入后约5小时、10小时、12小时、1、2、3、4、5、6、7天、1、2、3、4、5周、2、3、4、4个月或更长时间)处死。处死小鼠后,通过FACS评估脾脏、骨髓、外周血和其他器官样本中是否存在人类γδT细胞。
此外,短语“归巢(homing)和/或保留潜能(retention potential)”是指细胞(例如γδT细胞)在注入到宿主生物体(例如对象)中时(最常见的是作为静脉注入进入循环),以离开循环系统并留在宿主器官或组织的能力。具体而言,如本文所使用的术语“保留(retention)”是指注入的细胞在“归巢”和该组织或器官的群体之后保留在宿主组织或器官中的能力。如本文所使用,短语“增强归巢和/或保留潜能”是指细胞移植的效率、质量或快速的改进,其可由对标靶组织或器官改进的归巢和/或保留、改进的粘附、减少排斥等的结果。
细胞毒性(“细胞杀伤”)的分析在本领域是众所周知的。用于重定向杀伤分析的合适靶细胞的示例为癌细胞系、原发癌细胞、实体瘤细胞、白血病细胞或病毒感染细胞。特别地,可以使用K562、BL-2、colo250和原发性白血病细胞,但是可以使用许多其他细胞类型中的任何一种,并且在本领域是众所周知的(参见,例如,Sivori等人(1997)J.Exp.Med.期刊186:1129-1136;Vitale等人(1998)J.Exp.Med.期刊187:2065-2072;Pessino等人(1998)J.Exp.Med.期刊188:953-960;Neri等人(2001)Clin.Diag.Lab.Immun.期刊8:1131-1135)。通过细胞活力分析(例如染料排除、铬释放、CFSE)、代谢分析(例如四氮唑盐)和直接观察来评估细胞杀伤。
细胞归巢和/或保留潜能可通过测量细胞功能性标记物(例如粘附分子,如CD62L、选择素配体等)或通过SCID-Hu小鼠模型中的体内注入和移植来确定。SCID-Hu小鼠模型采用C.B-17scid/scid(SCID)小鼠移植人类胎儿胸腺和肝组织或胎儿BM组织,以为评估可移植的推定人类淋巴组织和其他细胞提供了合适的模型。由于SCID小鼠与人类胎儿组织的重组,所述模型提供了人类细胞的归巢和保留,并在人类来源的微环境中发挥功能。通常对小鼠进行照射,然后将淋巴细胞送入移植物中,并通过任何数量的方法测量归巢/保留,包含FACS和重新填入器官的免疫组织化学(例如,参见下文的材料和实验方法)。
如本文所用,术语“体外”(ex-vivo)是指从活生物体中移除细胞并在生物体外繁殖(例如,在试管中)的过程。
如本文所用,术语“在试管中(in-vitro)”是指源自实验室维持的一个或多个细胞系(例如NTera2神经细胞、胚胎细胞系等)的细胞在生物体外被使用的过程。这种细胞系通常是永生化细胞。

根据本发明方法的一些实施例,向γδT细胞提供γδT细胞扩增的多个条件。
在具体实施例中,γδT细胞扩增的多个条件包含提供多种营养素和多个细胞因子。
能够支持γδT细胞的合适培养基包含HEM、DMEM、RPMI、F-12等。如果需要,培养基可以包含细胞代谢所需的多种补充剂,例如谷氨酰胺和其他氨基酸、维生素、矿物质和有用的蛋白质,例如转铁蛋白等。培养基可能包含也可能不包含添加的血清。培养基还可含有抗生素以防止酵母、细菌和真菌污染,例如青霉素、链霉素、庆大霉素等。如果要培养细胞,多个条件应接近多个生理条件(最好是pH值约为6至8,温度约为30℃至40℃)。在一些实施例中,培养基可任选地补充以下至少一种:多种增殖诱导生长因子、多个细胞因子和/或多个趋化因子,例如IL-2、IL-4、IL-7、IL-15、IL-12、IL-21、IL-23或IL-33及其组合。在具体实施例中,培养基补充有IL-2和/或IL-15。除增殖诱导生长因子外,还可向培养基中添加其他生长因子。在一些示例性实施方案中,γδT细胞在含细胞因子(IL-2、IL-4、IL-7、IL-15、IL-12、IL-21、IL-23或IL-33)、生长因子(胰岛素和转铁蛋白、胰岛素样生长因子)、白蛋白、脂质(胆固醇、脂质溶液、脂质前体)、维生素、铜、铁、硒、蛋白质水解物、必需氨基酸、非必需氨基酸和剪切保护剂(Pluronic F-68)的无血清培养基(例如,Ex-Vivo 10培养基、Ex-Vivo15培养基、Ex-Vivo 20培养基、AIMV培养基、Optimizer CTS培养基)中被刺激和扩增。
多个细胞因子和其他生长因子通常以0.5-100纳克/毫升或1.0-80纳克/毫升、更通常的5-750纳克/毫升、更通常的5.0-50纳克/毫升的浓度提供(可考量最高达这样的浓度的10倍),并可从美国新泽西州洛基山的Perpo Tech公司处购买。在一个实施方案中,允许细胞增殖的多个条件包含提供细胞因子白细胞介素2或白细胞介素15。在具体实施方案中,γδT细胞与20纳克/毫升的IL-15和/或IL-2一起培养。
此外,在这方面应理解,不断发现新的细胞因子,其中一些可用于本发明的γδT细胞增殖方法。对于将细胞引入(或重新引入)人体的应用,通常优选的使用无血清制剂,例如用于淋巴细胞培养的AIM VRTM无血清培养基或MARROWMAX.RTM骨髓培养基。此类培养基配方和补充剂可从商业来源获得,如Invitrogen(GIBCO)(加利福尼亚州卡尔斯巴德)。多种培养物可以补充多种氨基酸、多种抗生素和/或多个细胞因子,以促进最佳生存能力、增殖、功能和/或存活。
此类无血清培养基可进一步补充多种添加剂,以支持悬浮培养(例如,WAVE生物反应器)中的高细胞密度γδT细胞生长,同时保持γδT细胞的生物功能性。
根据本发明的一个方面,可通过为γδT细胞或γδT细胞群提供体外细胞增殖的多个条件和使用烟酰胺部分体外培养γδT细胞,来实现γδT细胞的体外培养,从而体外扩增和/或体外增强γδT细胞群的归巢和/或保留潜能。

如本文所用,短语“烟酰胺类似物”是指已知在上述或类似分析中类似于烟酰胺作用的任何分子。烟酰胺类似物的代表性示例可包含但不限于苯甲酰胺、烟碱乙酰胺(烟酰胺的硫醇类似物)、烟酸和α-氨基-3-吲哚丙酸。
短语“烟酰胺衍生物”进一步是指烟酰胺本身或烟酰胺类似物的任何结构衍生物。此类衍生物的示例包含但不限于取代的苯甲酰胺、取代的烟酰胺和烟碱乙酰胺以及N-取代的烟酰胺和烟碱甲酰胺、3-乙酰吡咯烷和烟酸钠。在本发明的一个具体实施例中,烟酰胺部分为烟酰胺。
适于在本发明的一些实施例中使用的烟酰胺或烟酰胺部分浓度通常在约0.5mM(毫莫尔浓度)至约50mM、约1.0mM至约25mM、约1.0mM至约25mM、约2.5mM至约10mM、约5.0mM至约10mM、约0.5mM至约20mM的范围内。基于这些浓度的烟酰胺对γδT细胞归巢和/或保留的影响,烟酰胺的示例性有效浓度可为约0.5至约15mM、1.0-10.0mM、通常为2.5或5.0mM。根据本发明的一些实施例,以约0.5、约0.75、约1.0、约1.25、约1.5、约1.75、约2.0、约2.25、约2.5、约2.75、约3.0、约3.25、约3.5、约3.75、约4.0、约4.5、约4.75、约5.0、约5.25、约5.5、约5.75、约6.0、约6.25、约6.5、约6.75、约7.0、约7.25、约7.5、约7.75、约8.0、约8.25、约8.5、约8.75、约9.0、约9.25、约9.5、约9.75、约10.0、约11.0、约12.0、约13.0、约14.0、约15.0、约16.0、约17.0、约18.0、约20.0mM、约23.0mM、约25.0mM、约30.0mM、约35.0mM、约40.0mM、约45.0mM或约50.0mM范围内的浓度提供烟酰胺。所有有效的中间浓度皆被考虑在内。在具体实施例中,允许增殖的条件包含0.5至50mM、1.0至10.0mM的烟酰胺。在又一其它实施方案中,增强γδT细胞归巢和/或保留的条件包含5.0mM的烟酰胺。
可根据γδT细胞归巢和/或保留或CD62L表达的任何分析来确定烟酰胺和/或烟酰胺部分的适当浓度。与烟酰胺含量小于0.1mM、小于0.2mM或小于0.4mM及来自相同γδT细胞来源(例如脐带血、骨髓或外周血制备)测试的对照培养物相比,在相同分析和类似培养条件下(暴露于烟酰胺的持续时间、暴露于烟酰胺的时间、扩张条件),烟酰胺的适当浓度是在培养物中使用增强或导致γδT细胞归巢和/或保留功能净增加的浓度。
应注意的是,根据本发明的方法用于扩增和增强γδT细胞的条件也可能有利于培养在与γδT细胞混合的细胞群中发现的其他类型的细胞。因此,在一些实施例中,根据本文公开的方法提供γδT细胞扩增和烟酰胺的条件也增强了其他淋巴细胞(例如自然杀伤(NK)细胞)的归巢和/或保留潜能,与没有添加烟酰胺培养和/或扩增的类似细胞群相比,提供了扩增细胞群比类似细胞群具有潜在更大治疗功效。
在具体实施例中,本发明的方法可以扩增和增强各种γδT细胞群的功能,例如Vγ1+、Vγ2+、Vγ3+、γδT细胞群。在某些情况下,γδT细胞群可以被离体培养少于36天、少于35天、少于34天、少于33天、少于32天、少于31天、少于30天、少于29天、少于28天、少于27天、少于26天、少于25天、少于24天、少于2天3天、少于22天、少于21天、少于20天、少于19天、少于18天、少于17天、少于16天、少于15天、少于14天、少于13天、少于12天、少于11天、少于10天、少于9天、少于8天、少于7天、少于6天、少于5天、少于4天、少于3天。在某些情况下,γδT细胞群培养1至8周、1至5周、1至4周、1至3周、1至2周、1至14天、2至13天、1至10天、2至8天、1至7天、3至12天以及5至14天。在一些实施例中,还设想到将γδT细胞的富集细胞短期离体暴露于烟酰胺和/或其他烟酰胺部分,时间为几分钟、几小时、1天等。
在一些研究中,通过使用多种营养素、血清、多个细胞因子和烟酰胺培养体外扩增γδT细胞不需要在培养期间补充培养基或进行操作,而其他研究则主张在γδT细胞培养期间以不同的间隔补充培养基(“再饲养(re-feeding)”)。在本发明的某些实施例中,γδT细胞部分在培养期间“再饲养”。因此,在具体实施方案中,培养γδT细胞群包含在体外培养开始后8-10天用新鲜的多种营养素、血清、多个细胞因子和烟酰胺补充γδT细胞的富集细胞。在一些实施方案中,在体外培养开始后的8-9天之间、体外培养开始后的9-10天之间或在培养γδT细胞的富集细胞开始后的8-10天之间提供补充。在一些实施方案中,所述补充(或“再饲养”)包含移除约30-80%、约40-70%或约45-55%的培养基,并当培养基移除时,用具有相同成分和水平的多种营养素、血清、多个细胞因子(例如IL-2和/或IL-15)的类似(例如等效)体积的新鲜培养基替换。在一些实施方案中,所述补充(或“再饲养”)包含移除培养的约50%的培养基,并用具有相同成分和水平的多种营养素、血清、多个细胞因子(例如IL-2和/或IL-15)和烟酰胺的类似(例如等效)体积的新鲜培养基替换移除的培养基。在其他实施方案中,再饲养后的培养体积达到γδT细胞富集的多个细胞培养(“接种”)开始时原始培养体积的大约两倍。
γδ细胞群可以使用多种方法和装置进行培养。培养设备的选择通常基于培养的规模和目的。细胞培养的放大优选地包含使用专用装置。在一些实施方案中,在多个烧瓶中以每烧瓶100-4000X106个细胞的细胞密度培养γδT细胞富集的多个部分。在具体实施方案中,在多个烧瓶中以200-300X106个细胞/烧瓶的细胞密度培养γδT细胞富集的多个部分(例如,体外培养开始和/或“再饲养”)进行。在某些实施例中,所述多个烧瓶是包含气体可渗透膜的多个烧瓶,例如G-Rex培养装置(G-Rex 100M或封闭系统G-Rex MCS,WolfWilson公司,明尼苏达州圣保罗)。
培养γδT细胞富集的多个细胞可以在具有或没有饲养细胞或饲养细胞层的情况下进行。无饲养层离体培养对于培养细胞的临床应用非常有利。因此,根据一个实施例,在没有饲养层或饲养细胞的情况下培养γδT细胞群富集的多个细胞。
在一些方面,提供了通过用激活剂接触γδT细胞来扩增各种γδT细胞或γδT细胞群的方法。在某些情况下,激活剂与γδT细胞的细胞表面受体上的特定表位结合,例如单克隆抗体。激活剂可特异性地激活一种或多种类型的γδT细胞的生长,例如δ1、δ2或δ3细胞群。在一些实施例中,激活剂特异性激活δ1细胞群的生长。在其他情况下,激活剂特异性地激活δ2细胞群的生长。活化剂可刺激γδT细胞快速生长。

在一些方面,本发明提供了用于培养已从一对象分离的多个γδT细胞的方法。γδT细胞可以从一对象的一复合样本中分离出来。复合样本可以是外周血样本、脐带血样本、肿瘤、干细胞前体、肿瘤活检、组织、淋巴,或者来自直接接触外部社会环境的一对象的多个上皮部位,或者衍生自干细胞前体细胞。在具体实施例中,样本衍生自一器官,所述器官选自由以下的群组:肌肉、皮肤、骨骼、淋巴器官、胰腺、肝脏、胆囊、肾脏、消化道器官、呼吸道器官、生殖器官、泌尿道器官、血液相关器官、胸腺、脾脏和神经系统器官。在具体实施例中,多个γδ细胞或γδ细胞群衍生自一来源,所述来源选自由以下组成的群组:多个造血细胞、多个脐带细胞、多个外周血细胞(可动的或不可动的)和多个骨髓细胞。在又一实施例中,从骨髓或外周血样本、新生儿脐带血或单核细胞部分分离γδ细胞或群体。
例如,通过使用流式细胞术技术对表达一个或多个细胞表面标记物的(多个)γδT细胞进行分选,一γδT细胞可直接从一对象的一复合样本中分离。野生型γδT细胞表现出与(多个)γδT细胞相关联的许多抗原识别、抗原呈递、共刺激和粘附分子。一个或多个细胞表面标记物,例如特异性γδTCR、抗原识别、抗原呈递、配体、粘附分子或共刺激分子,可用于从复合样本中分离野生型γδT细胞。与γδT细胞相关联或通过γδT细胞表达的各种分子可用于从复合样本中分离γδT细胞。在一些实施例中,本发明提供了用于Vδ1+、Vδ2+、Vδ3+细胞或其任何组合的混合群体分离的方法。
外周血单个核细胞可以从对象身上采集,例如,使用单采机(apheresismachine),所述单采机包含Ficoll-PaqueTMPLUS(GE Healthcare公司)系统或其他合适的装置/系统。(多个)γδT细胞或(多个)γδT细胞的所需亚群可通过例如流式细胞术技术从收集的样本中纯化。脐带血细胞也可以在对象出生时从脐带血中获得。在具体实施例中,γδT细胞或γδT细胞群来自单采样本(apheresis sample)或衍生自单采样本。
在收集的(多个)γδT细胞上表达的细胞表面标记物的阳性和/或阴性选择可用于直接从外周血样本、脐血样本、肿瘤、肿瘤活检、组织、淋巴组织,或者从对象的上皮样本中分离γδT细胞或表达类似细胞表面标记物的(多个)γδT细胞的群体。例如,可以基于CD2、CD3、CD4、CD8、CD24、CD25、CD44、套件组(Kit)、TCRα、TCRβ、TCRδ、NKG2D、CD70、CD27、CD30、CD16、CD337(NKp30)、CD336(NKp46)、OX40、CD46、CCR7和其他合适的细胞表面标记物的阳性或阴性表达,从复合样本中分离γδT细胞。具体实施例中,所述选定细胞群是通过αβT细胞损耗来富集γδT细胞的一淋巴细胞群。应了解,这种γδT细胞富集的细胞群可包含其他细胞类型的多个显着部分(significant fraction),例如自然杀伤(NK)细胞。
在其它实施例中,所述选定细胞群是通过阳性TCRγδT细胞选择来富集γδT细胞的一淋巴细胞群。应了解,在这种γδT细胞富集的细胞群中,非γδT细胞将是稀缺的。在一些实施例中,γδT细胞阳性选定的富集的细胞群不含自然杀伤(NK)细胞。
可以从体外培养的复合样本中分离γδT细胞。在具体实施例中,多个富集的γδT细胞群可在其特定激活和扩增之前产生。在一些实施例中,例如单核细胞、T细胞、B细胞和自然杀伤细胞等附加细胞群包含在富集的γδT细胞群中,并且在某些情况下可与γδT细胞一起被激活和扩增。在某些方面,γδT细胞的激活和扩增在不存在天然或设计的多种别藻蓝蛋白(APCs)的情况下进行。在一些方面,可以使用固定的γδT细胞有丝分裂原(包含γδTCR特异性抗体)和其他γδTCR激活剂(包含凝集素),从肿瘤样本中分离和扩增γδT细胞。
在某些实施方案中,在γδT细胞富集的细胞培养开始的14-16天后从培养物中收获γδT细胞富集的多个细胞。可以手动地,通过释放附着的细胞(例如,“刮”培养容器表面)或通过设计用于有效地将细胞从其培养容器中冲洗出来并自动收集细胞的细胞采集装置进行细胞采集。在具体实施例中,通过细胞收获装置(例如,WolfWilson公司(明尼苏达州圣保罗市)的收获装置:g-Rex MCS)从培养容器中收获扩增的细胞部分。
在一些实施方案中,从培养物收获扩增的γδT细胞富集的多个细胞为从培养容器中移除大部分或几乎所有细胞。在其他实施例中,可分两个或多个步骤进行收获,允许未收获的细胞保留在培养物中,直到稍后收获。在收获第一部分和第二部分之间,可以以几小时、几天或更长的间隔来收获这两部分。
为了制备用于移植的扩增的γδT细胞富集的多个细胞,收获的细胞需要洗去培养基,评估多个关键参数,并将体积调整到适合在临床上合理时间内注入的浓度。
收获后,扩增的γδT细胞富集的多个细胞群可手动清洗,或优选地使用用于临床应用的封闭系统的自动装置。洗涤后的细胞可以用输液重新构成(例如,一种示例性输液包含8%w/v人血清白蛋白(HSA)和6.8%w/v葡聚糖-40(Dextran-40))。在一些实施例中,在封闭系统中执行重新构成。在一些实施例中,对输液进行筛选以适合与本发明的方法和组合物一起使用。选择合适输液的示例性标准包含安全性测试,表明无细菌、酵母或霉菌生长、内毒素含量低于0.5Eu/毫升、外观清晰、无异物颗粒。
上文描述的用于体外培养γδT细胞和γδT细胞的多个富集群体的方法,除其他外,可产生多个γδT细胞和γδT细胞富集的多个细胞的一培养群体。
因此,进一步根据本发明的一个方面,提供了多个γδT细胞的一群体,其特征在于以下的至少一个:与在其他相同培养条件下使用烟酰胺和/或其他烟酰胺部分小于0.1mM培养的γδT细胞和/或γδT细胞富集的多个细胞的一群体相比,CD62L的表达提高、迁移反应提高、归巢和体内保留提高、细胞毒性活性增强。在一些实施例中,多个γδT细胞和/或γδT细胞富集的多个细胞的群体的特征在于以下的至少任何两个、至少任何三个、至少任何四个或全部五个:与在其他相同培养条件下使用烟酰胺和/或其他烟酰胺部分小于0.1mM培养的γδT细胞富集的多个细胞的一群体相比,CD62L的表达提高、迁移反应提高、归巢和/或体内保留提高、细胞毒性活性增强。
在实施例1中,发明人已示出根据本发明方法制备的多个γδT细胞群增加了细胞表面标记物(parker)CD62L(L-选择素)的表达,这对细胞粘附和“滚动(rolling)”很重要。在实施例2中,发明人已示出,根据本发明方法制备的多个富集γδT细胞群具有增加的体内功能潜能,如通过在靶器官(例如脾脏、骨髓)中的定位和体内保留所证明。因此,在本发明的一些实施例的特定方面中,提供了一种γδT细胞富集的多个细胞的群体,其特征在于在移植时具有以下的至少一种:增强的CD62L表达和增强的归巢和/或体内保留。

基因治疗:对于成功的长期基因治疗来说,高频率的转基因细胞和稳定整合在其基因组中的转基因是一项必须的要求。以病毒为基础(如逆转录病毒)的载体需要活跃的细胞分裂,以便将转基因整合到宿主基因组中。因此,将基因转移到一些未受刺激的新鲜细胞群体中是非常低效的。体外储存和处理所选定γδT细胞群,以及增强其归巢和保留潜能的能力将提供增加成功使用转基因细胞移植的可能性。
过继免疫疗法:体外扩增、定义的淋巴亚群已被研究并用于各种恶性肿瘤、免疫缺陷、病毒和遗传疾病的过继免疫治疗(Freedman,Nature Medicine期刊2:46,(1996);Heslop,Nature Medicine期刊2:551,(1996);Protti,Cancer Res期刊56:1210,(1996))。
所述治疗可增强所需的免疫反应或取代功能缺陷。Rosenberg等人(Rosenberg,JNatl Cancer Inst.期刊85:622,1993)在临床上率先使用了大量自体和同种异体体外扩增的非特异性杀伤性T细胞,以及随后体外扩增的特异性肿瘤浸润淋巴细胞。

在一些实施例中,所述治疗性细胞组合物是一药物组合物,所述药物组合物包含一扩增的γδT细胞富集的群体和药学上可接受的一载体。如上文详细讨论的,γδT细胞富集的多个细胞的体外培养可有利地用于γδT细胞移植或植入。因此,根据本发明的另一方面,提供了一种将多个γδT细胞或γδT细胞富集的多个细胞或群体移植或植入到一接受者的方法。根据本发明此方面的方法通过以下来执行:(a)根据本发明的多种方法,通过用于γδT细胞扩增的多个条件和烟酰胺来培养细胞群,体外扩增一选定γδT细胞群,其中所述扩增的选定γδT细胞群具有增强的γδT细胞归巢和/或保留潜能,其与一类似的选定γδT细胞群相比,所述类似的选定γδT细胞群不使用0.5-50nM范围的烟酰胺来扩增,以及(b)将扩增的多个γδT细胞注入需要的一对象。

“癌症相关联的”是指在一对象细胞中,核酸及其表达或核酸缺失,或蛋白质及其水平或活性或蛋白质缺失与恶性肿瘤发病之间的关系。例如,癌症可能与正常健康细胞中未表达或表达水平较低的特定基因的表达相关联。相反地,癌症相关联的基因可以是在恶性细胞(或正在转化的细胞)中不表达的基因,或在恶性细胞中的表达水平低于在正常健康细胞中的表达水平的基因。
“过度增殖性疾病”是指细胞增殖快于正常组织生长的任何疾病(disease或disorder)。因此,过度增殖细胞是比正常细胞增殖更快的细胞。
“晚期癌症”是指不再局限于原发肿瘤部位的癌症,或者根据美国癌症联合委员会(AJCC)的说法是III或IV期癌症。
“良好耐受性”是指在治疗过程中不会出现健康状况的不利变化,并会影响治疗决定。

造血细胞移植已成为治疗各种遗传性或恶性疾病的首选方法。然而,造血细胞成分通常富集T淋巴细胞,这有助于移植物抗宿主病。由于血液恶性肿瘤患者通常缺乏γδT细胞数量和功能,目前正在研究外源性施用γδT细胞以及造血细胞移植,以增强长期植入和预防移植物抗宿主病。因此,在本发明的又一实施例中,提供了一种治疗或预防有需要的对象的移植物抗宿主病的方法。因此,在本发明的另一个实施例中,提供了一种治疗有需要的对象的自身免疫性疾病或病症的方法。根据本发明此方面的方法通过向所述对象施用治疗剂量的本发明γδT细胞群或包含相同γδT细胞群的组成物来实现。
治疗方案
根据本发明一些实施例的一些方面,提供了包含本发明的γδT细胞富集的细胞群的药物组合物,以用于治疗疾病,例如转移癌、实体瘤、自身免疫性疾病、过度增殖性疾病或病毒感染,与药学上可接受的载体一起制备。一些组合物包含本发明的多个(例如,两个或更多)γδT细胞富集的多个细胞群的组合。
在预防性应用中,向易罹患某病或具有疾病或病症(即,过度增殖性疾病或实体肿瘤)风险的患者施用药物组合物或药物以足以消除或降低过度增殖性疾病或实体肿瘤复发风险的剂量,以减轻疾病的严重程度或延迟疾病的开始,包含疾病的生化、组织学和/或行为症状、并发症以及疾病发展过程中出现的中间病理表型。在治疗应用中,向怀疑患有或已经患有这样的疾病的患者施用足够剂量的组合物或药物,以治愈或至少部分缓解所述疾病的症状(生化、组织学和/或行为),包含其并发症和疾病发展过程中的中间病理表型。足以完成治疗或预防治疗的剂量定义为治疗或预防有效剂量。在预防和治疗方案中,药物通常分为几个剂量施用,直到达到显着的抗增殖反应。通常,如果抗增殖反应开始减弱,则监测抗增殖反应并重复给药。

有效剂量
本文所述用于治疗疾病(例如,转移癌、实体瘤或过度增殖性疾病)的γδT细胞群的组合物的有效剂量取决于许多不同的因素,包含给药方式、标靶位置、患者的生理状态,患者是人或动物,施用的其他药物,以及治疗是预防性的还是治疗性的。通常,患者是人类,但非人类哺乳动物包含转基因哺乳动物在内的也可以被治疗。治疗剂量需要滴定测量以优化安全性和疗效。
对于使用治疗性γδT细胞或γδT细胞富集的细胞群施用,剂量范围为每名患者约1X106至约1X109个γδT细胞和/或γδT细胞富集的细胞。对于γδT细胞富集的细胞群的施用,剂量范围为每千克接受者重量约1X105至约1X109个γδT细胞和/或γδT细胞富集的细胞,或剂量范围为每千克接受者重量约5X105至约1x18个γδT细胞和/或γδT细胞的富集细胞。一种示例性的治疗方案需要每两周或每月或每3到6个月施用一次。在一些方法中,同时施用两种或多种γδT细胞富集的细胞群,在这种情况下,施用的每个γδT细胞富集的细胞群的剂量在指示的范围内。可以多次施用γδT细胞富集的多个细胞群。单一剂量之间的间隔可以是每周、每月或每年。间隔也可以是不规则的,如通过测量患者体内γδT细胞富集的细胞群的血液水平来指示。替代地,可以将γδT细胞富集的多个细胞群作为缓释制剂施用,在这种情况下,需要较少的施用频率。剂量和频率取决于患者体内γδT细胞富集的多个细胞群的半衰期。根据治疗是预防性的还是治疗性的,施用的剂量和频率可能会有所不同。在预防性应用中,在相当长的一段时间内相对频率较少的间隔施用相对较低的剂量。一些患者在其余生继续接受治疗。在治疗应用中,有时需要在相对较短的时间间隔内使用相对较高的剂量,直到疾病进展减少或终止,并且最好直到患者表现出疾病症状的部分或完全改善。据此,本专利可在预防性制度下实施。
施用途径
用于治疗疾病(例如,转移癌、实体瘤或过度增殖性疾病)的治疗性γδT细胞富集的细胞群的组合物可通过静脉内、囊内、鞘内、肠外、局部、皮下、口内、鼻内、动脉内、颅内、腹腔、或肌肉内的施用方式。作为预防/辅助或治疗疾病,治疗性γδT细胞富集的多个细胞群标靶过度增殖性疾病或实体瘤,例如泌尿生殖系统恶性肿瘤和/或治疗性治疗。免疫原的制剂最典型的施用途径是皮下注射,尽管其他途径也同样有效。其次最常见的途径是肌肉注射。这种类型的注射通常在手臂或腿部肌肉中进行。在一些方法中,药剂被直接注射到沉积物积累的特定组织中,例如颅内注射。静脉输液时肌肉内注射是施用γδT细胞富集的细胞群的优选方法。在某些方法中,将特定的治疗性细胞群直接注入膀胱。
配方
γδT细胞富集的细胞群的多种组合物,用于治疗疾病,例如转移癌、实体瘤、病毒或其他感染、炎症或过度增殖性疾病。
用于治疗疾病(例如,转移癌、实体瘤或过度增殖性疾病)的治疗性γδT细胞富集的细胞群的多种组合物通常以包含活性治疗剂(即和多种其他药学上可接受的组分)的药物组合物施用。例如,参见Alfonso R Gennaro(编辑),《雷明顿:药学科学与实践》(原雷明顿制药科学)第20版,利平科特,威廉姆斯·威尔金斯出版社,2003年,通过引用全部并入本文。优选的形式取决于预期的施用模式和治疗应用。根据所需的配方,所述组合物还可包含医药上可接受的无毒载体或稀释剂,所述载体或稀释剂被定义为通常用于配制供动物或人类施用的药物组合物的载体。稀释剂的选择是为了不影响组合物的生物活性。此类稀释剂的一个示例是X-vivo 20培养基(Cambrex Bio Science公司,马里兰州沃克斯维尔市)含有10%热失活的人类AB血清或10%自体血清。此类稀释剂的进一步示例为蒸馏水、生理磷酸盐缓冲盐水、林格氏溶液、葡萄糖溶液和汉克斯溶液。此外,药物组合物或制剂还可包含其他载体、佐剂或无毒、非治疗、非免疫原性的稳定剂等。
药物组合物还可包含大型的、缓慢代谢的大分子,例如蛋白质、多糖,例如壳聚糖、聚乳酸、聚乙醇酸和共聚物(例如乳液功能化的Sepharose.TM、琼脂糖、纤维素等)、聚合氨基酸、氨基酸共聚物和脂质聚集体(如油滴或脂质体)。此外,这些载体可以作为免疫刺激剂(即佐剂)发挥作用。
对于肠外给药,本发明的组合物可以作为在生理上可接受的稀释剂中物质的溶液或悬浮液的可注射剂量施用,药物载体可以是无菌液体,例如水油、盐水、甘油或乙醇。此外,组合物中还可以有辅助物质,如润湿剂或乳化剂、表面活性剂、pH缓冲物质等。药物组合物的其他成分为石油、动物、植物或合成成分,例如花生油、大豆油和矿物油。一般来说,乙二醇(如丙二醇或聚乙二醇)是优选的液体载体,尤其是用于注射溶液。治疗性的γδT细胞富集的多个细胞群可以以贮库型注射剂或植入制剂的形式施用,其配制方式可以允许活性成分的持续释放。一种示例性组合物包含5mg/mL的治疗性γδT细胞的富集细胞群,其在含有50mM的L-组氨酸、150mM的NaCl组成的水性缓冲液中配制,并用HCl调节至pH 6.0。
通常情况下,将组合物制备为可注射,或作为液体溶液或悬浮液;也可制备适于在注射前在液体载体中溶解或悬浮的固体形式。如上文所述,制剂也可乳化或封装在脂质体或微粒(例如聚乳酸、聚乙交酯或共聚物)中以增强佐剂效果。Langer,科学期刊,249:15271990;Hanes,Advanced Drug Delivery Reviews期刊,28:97-1191997,全部通过引用并入本文。本发明的制剂可以贮库型注射剂或植入制剂的形式施用,其配制方式可允许活性成分的持续或脉动释放。适用于其他施用方式的其他制剂包含口服、鼻内和肺部制剂、栓剂和透皮应用。

所述药物组合物通常包含治疗性γδT细胞富集的细胞群的组合物,其形式适合给患者施用。药物组合物通常是被制备为无菌、基本等渗,并符合美国食品和药物管理局的所有良好生产规范(GMP)规定。
毒性
优选地,本文所述的γδT细胞富集的细胞群的组合物的治疗有效剂量将提供治疗益处而不会引起实质性毒性。
本文所述的治疗性γδT细胞的富集细胞群的毒性可通过细胞培养或实验动物中的标准制药程序来确定,例如,通过确定LD50(对50%的群体致死的剂量)或LD100(对100%的群体致死的剂量)。毒性与疗效之间的剂量比是治疗指标。从这些细胞培养分析和动物研究中获得的数据可用于制定对人体无毒的剂量范围。本文所述的治疗性γδT细胞富集的细胞群的剂量优选地位于循环浓度范围内,该循环浓度包含毒性很小或没有毒性的有效剂量。剂量可以在这个范围内变化,这取决于所采用的剂型和使用的施用途径。具体的配方、给药途径和剂量可由个别医生根据患者情况选择。(参见,例如,Fingl等人,《治疗学的药理学基础》,第1章,1975年),通过引用将其全部内容并入本文。

试剂盒

低温保存

实验程序
γδT细胞的体外培养
使用midiMACSTM柱和TCRα/β试剂盒(Miltenyi Biotec公司,马里兰州盖瑟斯堡市)去除健康献血者血液单位样本中TCRαβ表达的T细胞。在(Saint-Gobain公司,马里兰州盖瑟斯堡市)袋中,使用添加0(对照组)或5mM烟酰胺(NAM)和50纳克/毫升IL-2的培养基培养αβT细胞损耗群体12-13天。
T细胞受体特性
为了表征培养中CD3+细胞上表达的T细胞受体(TCR),使用CliniMACSTMCD3试剂纯化CD3阳性细胞(Miltenyi公司,273-01,马里兰州盖瑟斯堡市),然后对CD3+/γδ+和CD3+/α/β+细胞的百分比进行FACS分析。根据应用细胞表面特异性抗体的标准程序进行FACS染色和分析。
在扩增的γδT细胞中的CD62L表达
来自NAM处理和对照(-NAM)培养物的纯化γδT细胞对CD62L(L-选择素)进行染色,并使用抗CD62L抗体通过FACS进行分析。
扩增的γδT细胞的移植和体内功能
如上所述,从培养物中纯化CD3+/γδ+细胞,然后用CFSE(Invitrogen,ThermoFisher公司,加利福尼亚州卡尔斯巴德市)标记。NSG SCID小鼠以350cGy剂量辐射照射。辐射照射后第二天,将来自NAM和对照(-NAM)培养物每只小鼠以5-6x106个CFSE标记的CD3+/γδ阳性细胞静脉注射到小鼠体内。
细胞注入后4天处死小鼠,切除器官。通过对器官细胞悬液的FACS评估CFSE染色细胞的部分。
示例1:用NAM体外培养人类γδT细胞来增强T细胞功能
对用5mM烟酰胺(NAM)培养的αβ损耗T细胞的FACS分析表明,αβT细胞部分的损耗提供了一个扩增的CD3+细胞群,所述扩增的CD3+细胞群包含超过90%的γδT细胞。用烟酰胺培养细胞似乎不会影响培养的T细胞群中γδ阳性成分的扩增(图1)。
CD62L(L-选择素)是一种重要的淋巴细胞粘附分子,是淋巴细胞归巢和进入淋巴组织的“归巢受体”,也是一种T淋巴细胞共刺激信号。图2显示了与没有使用NAM培养的相同细胞相比,使用5mM NAM培养的γδT细胞中CD62L表达的显著增强(通过FACS检测)。
示例2:使用NAM培养γδT细胞的增强体内归巢和保留
注入淋巴细胞的有效组织归巢和保留的频率低,构成了T细胞治疗成功的严重阻碍,增加了达到最佳效果所需的细胞数量。NAM培养的γδT细胞功能的增加反映在注入到受辐射照射的scid NSG小鼠后,归巢和保留在组织中的发生率提高。
图3显示了在注入NAM培养的γδT细胞4天后,NAM对体内归巢和保留的影响程度。特别重要的是,用NAM培养后,留存在淋巴组织(脾脏、骨髓)中的γδT细胞得到了强力的增强,同时留存在血液和肺组织中的γδT细胞也增加了近三倍。

Claims (34)
1.一种增强γδT细胞归巢和/或保留潜能的方法,其特征在于,所述方法包含:
(a)获得富集多个γδT细胞的一选定细胞群;
(b)体外为所述选定细胞群提供γδT细胞扩增的多个条件;及
(c)提供0.5至50mM范围内的烟酰胺持续一时间段,以显着增强γδT细胞归巢和/或保留潜能,从而增强所述选定细胞群中多个γδT细胞的归巢和/或保留潜能。
2.一种增强γδT细胞CD62L表达的方法,其特征在于,所述方法包含:
(a)获得富集多个γδT细胞的一选定细胞群;

5.如权利要求1所述的方法,其特征在于,所述烟酰胺选自由以下组成的群组:烟酰胺、一烟酰胺类似物、一烟酰胺代谢物、一烟酰胺类似物代谢物及其衍生物。
6.如权利要求1所述的方法,其特征在于,所述选定细胞群是通过αβT细胞损耗来富集γδT细胞的一淋巴细胞群。
7.如权利要求6所述的方法,其特征在于,所述选定细胞群包含多个自然杀伤细胞。
8.如权利要求7所述的方法,其特征在于,所述方法进一步包含为自然杀伤细胞扩增提供多个条件。
9.如权利要求7所述的方法,其特征在于,提供γδT细胞扩增的所述多个条件和所述烟酰胺来增强所述选定细胞群中所述多个自然杀伤细胞的归巢和/或保留潜能和/或CD62L表达。
10.如权利要求1所述的方法,其特征在于,所述选定细胞群是通过选择多个γδT细胞来富集的γδT细胞的一淋巴细胞群。
11.如权利要求10所述的方法,其特征在于,所述选定细胞群不含多个自然杀伤细胞。
12.如权利要求1所述的方法,其特征在于,所述多个γδT细胞的群体衍生自一器官,所述器官选自由以下组成的群组:一肌肉、皮肤、一骨骼、一淋巴器官、一胰腺、一肝脏、一胆囊、一肾脏、一消化道器官、一呼吸道器官、一生殖器官、一泌尿道器官、一血液相关器官、一胸腺、一脾脏、或一神经系统器官。
13.如权利要求1所述的方法,其特征在于,所述多个γδT细胞的群体来源于选自由以下组成的群组:多个造血细胞、多个脐带血细胞、多个可动的外周血细胞和多个骨髓细胞。
14.如权利要求1所述的方法,其特征在于,所述多个γδT细胞的群体来源于骨髓或外周血。
15.如权利要求1所述的方法,其特征在于,所述多个γδT细胞的群体来源于新生儿脐带血。
16.如权利要求1所述的方法,其特征在于,所述多个细胞的群体来源于一单核细胞部分。
17.如权利要求1所述的方法,其特征在于,所述多个γδT细胞的群体来自一单采样本。
18.如权利要求1所述的方法,其特征在于,所述步骤(c)的所述时间段为1到3周之间。
19.如权利要求1所述的方法,其特征在于,所述步骤(c)的所述时间段为1到7天之间。
20.如权利要求1所述的方法,其特征在于,所述烟酰胺的一浓度为在0.5-20mM的范围内。
21.如权利要求1所述的方法,其特征在于,所述烟酰胺被提供在一浓度为5mM。
22.如权利要求1所述的方法,其特征在于,所述方法进一步包含根据一细胞标记来选择一γδT细胞群,所述细胞标记选自由以下组成的群组:一肿瘤抗原、一病毒抗原和一细菌抗原。
23.一种治疗性细胞组合物,其特征在于,所述治疗的细胞组合物包含:一扩增的选定γδT细胞群,所述扩增的细胞群在体外使用γδT细胞扩增的多个条件,及烟酰胺在0.5-50mM范围内的一剂量来培养,其中所述扩增的选定γδT细胞群的特征在于以下至少一个:
(i)增强的γδT细胞归巢和/或保留潜能,以及
(ii)增强的CD62L表达,
其与一类似的选定γδT细胞群相比,所述类似的选定γδT细胞群使用相同的多个条件及烟酰胺不超过0.1mM来扩增。
24.如权利要求23所述的治疗性细胞组合物,其特征在于,所述治疗性细胞组合物包含根据一方法来培养的多个γδT细胞,所述方法包含:
(a)获得富集多个γδT细胞的一选定细胞群;
(b)体外为所述选定细胞群提供γδT细胞扩增的多个条件;及
(c)提供0.5至50mM范围内的烟酰胺持续一时间段,以显着增强γδT细胞归巢和/或保留潜能。
25.如权利要求23所述的治疗性细胞组合物,其特征在于,所述治疗性细胞组合物进一步包含多个自然杀伤细胞。
26.一种在一对象中移植多个细胞的方法,其特征在于,所述方法包含:
(a)体外扩增一选定γδT细胞群,通过使用γδT细胞扩增的多个条件以及0.5-50mM范围内的烟酰胺来培养所述细胞群持续一时间段,足以显着增强γδT细胞归巢和/或保留潜能和/或CD62L表达,其中所述扩增的选定γδT细胞群的特征在于以下至少一个:
(i)增强的γδT细胞归巢和/或保留潜能,以及
(ii)增强的CD62L表达,
其与一类似的选定γδT细胞群相比,所述类似的选定γδT细胞群不使用0.5-50mM范围的烟酰胺来扩增,及
(b)将扩增的多个γδT细胞注入需要的一对象。
27.如权利要求26所述的方法,其特征在于,所述步骤(a)根据一方法作用,所述方法包含:
(i)获得富集多个γδT细胞的一选定细胞群;
(ii)体外为所述选定细胞群提供γδT细胞扩增的多个条件;及
(iii)提供0.5至50mM范围内的烟酰胺持续一时间段,以显着增强γδT细胞归巢和/或保留潜能。

29.如权利要求26所述的方法,其特征在于,所述多个γδT细胞与所述对象为同种异体。

31.如权利要求26所述的方法,其特征在于,所述对象患有一疾病,所述疾病选自由以下组成的群组:一癌症、一细菌感染、一病毒感染、一自身免疫性疾病和一炎症性疾病。
32.如权利要求31所述的方法,其特征在于,所述多个细胞在所述对象中的移植包含一辅助疗法。
33.如权利要求32所述的方法,其特征在于,所述辅助疗法与一疗法组合,所述疗法选自由以下组成的群组:抗病毒疗法、抗炎疗法、抗生素疗法、杀菌疗法、化疗、外科手术、免疫疗法、免疫化疗、放疗、骨髓移植和造血干细胞移植。
34.如权利要求32所述的方法,其特征在于,所述对象在移植所述扩增的多个γδT细胞之前、同时或之后,使用在大于1.0mM的烟酰胺的培养物中扩增的脐带血造血干细胞进行治疗。
