The amplification culture method adopts a variable temperature culture mode, the seed liquid with the volume percentage of 10 percent is inoculated into a fermentation amplification culture medium, the fermentation culture is carried out for 48 hours under the conditions of 33 ℃ and 200r/min, then the temperature is increased to 35 ℃, the culture is continued for 48 hours, and the thallus density is more than or equal to 30g wet thallus/L. After the expanded culture, the cells were recovered by filtration and stored at a low temperature of 4 ℃ for further use. In the step (2), the lactobacillus is subjected to amplification culture, an ultrasonic biological stimulation growth instrument is used for stimulation culture, the processing parameters of the ultrasonic biological stimulation growth instrument are ultrasonic frequency of 30kHz, power of 10W, ultrasonic time of 10s and interval time of 20s, and ultrasonic stimulation is carried out for 30 minutes in total.
Example 2: dried Ginseng radix raw material
This embodiment 2 specifically includes the following steps:
(1) pretreatment of raw materials: selecting dried ginseng rhizome planted for more than 5 years 0.1 kg, and pulverizing to 200-300 mesh. Adding 6 times of purified water for soaking, and adjusting pH to 7.0.

(3) Homogenizing: a micro-jet high-pressure homogenizer is adopted, and the feeding is pumped by a booster pump or a homogenizer plunger pump; material pressurization: pressurizing the material in a plunger booster pump to a set pressure value of 38 Mpa; the pressurized material is in ultramicro homogeneous state in the micro-jet high-pressure homogenizer, and the homogenized ginseng material is collected in a discharge container. In the process, the material subjected to high-pressure micro-jet treatment is cooled by a heat exchanger, and in the embodiment, the real-time temperature control temperature is set to be 40 ℃;
4) fermentation treatment:
adding the crushed other raw materials and water into the homogenized material obtained in the step, and adding lactobacillus for fermentation treatment. The other raw materials comprise 0.4 kg of astragalus, 0.1 kg of dendrobium, 0.4 kg of semen cuscutae, 0.4 kg of schisandra chinensis and 0.3 kg of radix ophiopogonis, and the raw materials are crushed into 80-120 meshes; the volume ratio of the homogenized material to other raw materials to water is 1: 10-20; the addition amount of lactobacillus is 90 g; the lactobacillus is lactobacillus acidophilus;
fermentation culture: the culture process needs a 10L fermentation tank, and adopts an inoculation mode of feeding liquid seeds into the tank. The pH value can be adjusted to 6.5-8.0 by 0.5N-1NHCl and 0.5N-1NNaOH or 10% -20% ammonia water in the whole fermentation process. Larger aeration volumes were used after inoculation.
Example 3: fresh ginseng raw material
This embodiment 3 specifically includes the following steps:
(1) pretreatment of raw materials: selecting 0.3 kg of fresh ginseng roots planted for more than 5 years, adding 5 times of purified water, and adjusting the pH value to 7.5; homogenizing, pulverizing, and sieving with 200-300 mesh sieve.
(2) An enzymolysis step: adding endoprotease 0.5 wt% of Ginseng radix and cellulase 0.1 wt% of Ginseng radix for enzymolysis for 3 hr, and stirring for 5 min every 1 hr during enzymolysis to allow enzymolysis to proceed uniformly and sufficiently.
(3) Homogenizing: a micro-jet high-pressure homogenizer is adopted, and the feeding is pumped by a booster pump or a homogenizer plunger pump; character material pressurization: pressurizing the material in a plunger booster pump to a set pressure value of 35.8 Mpa; the pressurized material is in ultramicro homogeneous state in the micro-jet high-pressure homogenizer, and the homogenized ginseng material is collected in a discharge container. In the process, the material subjected to high-pressure micro-jet treatment is cooled by a heat exchanger, and in the embodiment, the real-time temperature control temperature is set to be 40 ℃;
4) fermentation treatment:
adding the crushed other raw materials and water into the homogenized material obtained in the step, and adding lactobacillus for fermentation treatment. The other raw materials comprise 0.2 kg of astragalus, 0.05 kg of dendrobium, 0.2 kg of semen cuscutae, 0.2 kg of schisandra chinensis and 0.15 kg of radix ophiopogonis, and the raw materials are crushed into 80-120 meshes; the volume ratio of the homogenized material to other raw materials to water is 1: 10-20; the addition amount of lactobacillus is 80 g; the lactobacillus is lactobacillus thermophilus;
fermentation culture: the culture process needs a 10L fermentation tank, and adopts an inoculation mode of feeding liquid seeds into the tank. The pH value can be adjusted to 6.5-8.0 by 0.5N-1NHCl and 0.5N-1NNaOH or 10% -20% ammonia water in the whole fermentation process. Larger aeration volumes were used after inoculation.
Referring to FIGS. 1-2, FIG. 1 is a schematic flow diagram of a method of the present invention; FIG. 2 is the total saponin absorption spectrum of the fermentation broth ginsenoside and the reference solution. The control was prepared as in example 2 without the fermentation step.

step (1), raw material pretreatment: comprises processing dried Ginseng radix or fresh Ginseng radix material into appropriate size to reach suitable sample size. Soaking the pulverized ginseng material in an extraction solvent, wherein the extraction solvent is water, and preferably the extraction solvent is purified water or deionized water;
step (2), enzymolysis step: adding endoprotease and cellulase into the ginseng material, performing enzymolysis for 3-4h, stirring regularly during the enzymolysis process, and inactivating the enzyme of the obtained enzymolysis liquid after the enzymolysis is finished;
step (3), homogenizing: a micro-jet high-pressure homogenizer is adopted, and the feeding is pumped by a booster pump or a homogenizer plunger pump;
step (4), fermentation treatment;
adding the crushed other raw materials and water into the homogenized material obtained in the step, and adding lactobacillus for fermentation treatment; the other raw materials comprise astragalus, dendrobium, dodder, schisandra and ophiopogon root, and the raw materials are crushed into 80-120 meshes.
2. The preparation method of the anti-aging traditional Chinese medicine formula by utilizing probiotic fermentation according to claim 1, characterized in that the raw materials comprise, by weight, 18-22 parts of astragalus membranaceus, 4-6 parts of ginseng, 4-6 parts of dendrobe, 18-22 parts of semen cuscutae, 18-22 parts of schisandra chinensis and 14-16 parts of radix ophiopogonis.

The preparation method of the anti-aging traditional Chinese medicine formula by utilizing probiotic fermentation according to claim 2, characterized in that the raw materials comprise, by weight, 19-21 parts of radix astragali, 4.5-5.5 parts of ginseng, 4.5-5.5 parts of dendrobe, 19-21 parts of semen cuscutae, 19-21 parts of schisandra chinensis and 14.5-15.5 parts of radix ophiopogonis.
4. The preparation method of the anti-aging traditional Chinese medicine formula by utilizing probiotic fermentation according to claim 3, characterized in that the raw materials comprise, by weight, 20 parts of radix astragali, 5 parts of ginseng, 5 parts of dendrobe, 20 parts of semen cuscutae, 20 parts of schisandra chinensis and 15 parts of radix ophiopogonis.

The preparation method of the anti-aging traditional Chinese medicine formula by utilizing probiotic fermentation according to any one of claims 1 to 4, wherein in the step (1) of the method, the weight ratio of the ginseng to the purified water is 1: 6-10 times, and adjusting the pH value to 7-7.5.

The method for preparing anti-aging traditional Chinese medicine fermentation liquor by using probiotics according to any one of claims 1 to 4, wherein in the step (2), the enzymolysis is carried out by adding endoprotease 0.5 to 0.7 percent of the weight of the ginseng raw material and cellulase 0.1 to 0.2 percent of the weight of the ginseng raw material. In the enzymolysis process, stirring is carried out for 5-10 minutes every 1 hour, so that the enzymolysis is uniformly and fully carried out. The cell wall of the ginseng is further destroyed by the action of cellulase and endoprotease.

The method according to any one of claims 1 to 4, wherein in the step (1), the lactobacillus is enlarged and cultured by using an ultrasonic biostimulation growth instrument, and the ultrasonic biostimulation growth instrument is used for treating parameters of 30kHz, 10W of power, 10s of ultrasonic time and 20s of interval time for 30 minutes of ultrasonic stimulation.

The method for preparing the anti-aging traditional Chinese medicine formula by utilizing probiotic fermentation according to any one of claims 1 to 4, characterized in that the step of homogenizing treatment comprises the following steps of: pressurizing the material in a plunger booster pump to a set pressure value of 30-40 MPa, preferably 35-38 MPa, more preferably 35.8 MPa; collecting the homogenized ginseng material into a discharging container; the material treated by the high-pressure micro-jet flow has the phenomenon of instantaneous temperature rise, and needs to be cooled by a heat exchanger, and the real-time temperature control range is set to be 38-42 ℃, and is preferably 38.8-40 ℃.

The method for preparing an anti-aging traditional Chinese medicine formula by utilizing probiotic fermentation according to any one of claims 1 to 4, characterized in that in the fermentation treatment step, the volume ratio of the homogenized material and other raw materials to water is 1: 10-20; the addition amount of lactobacillus is 5-8% of the dry matter weight of all raw materials.

The method according to any one of claims 1 to 4, wherein the lactobacillus is cultured in an expanded medium according to the following formula in step (1): 60g/L glucose, 2.0g/L urea, KH2PO40.6g/L, 30g/L ammonium chloride, MgSO47H20.5g/L of O, 0.5g/L of yeast extract, 20g/L of liquid paraffin and water as a solvent; the expansion culture method adopts a variable temperature culture mode, and preferably comprises the following steps: inoculating the seed liquid into a fermentation amplification culture medium according to the volume percentage of 10%, performing fermentation culture for 48 hours at the temperature of 33 ℃ and at the speed of 200r/min, then raising the temperature to 35 ℃, and continuing to culture for 48 hours, wherein the thallus density is more than or equal to 30g wet thallus/L; after the expanded culture, filtering and recovering thalli, and preserving at a low temperature of 4 ℃ for later use; when used, a lactic acid bacterium suspension was prepared with 10mmol/L phosphate buffer solution (pH 8.0).
