Abstract
本发明涉及胰岛移植领域,尤其涉及ISL1在制备抑制移植胰岛早期铁死亡的药物中的用途和细胞。利用腺病毒构建过表达ISL1的胰岛细胞,得到ISL1过表达的胰岛细胞。利用慢病毒在大鼠β细胞INS‑1中过表达及敲减ISL1检测铁死亡相关蛋白(GPX4和ACSL3)和凋亡相关蛋白的表达变化情况,确定ISL1对STZ诱导INS‑1的铁死亡具有保护作用;利用EcoRI MCS‑ISL1的腺病毒构建过表达ISL1的大鼠和人源胰岛细胞,在胰岛细胞中明确ISL1通过促进ACSL3的表达抑制移植胰岛铁死亡。为基因修饰人类胰岛细胞改善移植胰岛存活及胰岛移植广泛应用于临床打下坚实的基础。构建的细胞能够用于实验和胰岛移植的可能性。

技术领域
本发明涉及胰岛移植领域,尤其涉及ISL1在制备抑制移植胰岛早期铁死亡的药物中的用途和细胞。
背景技术
移植胰岛的存活问题是影响移植胰岛早期存活的主要因素之一
成人胰岛移植一直是糖尿病研究领域的热点,2014年据国际胰岛移植注册处的资料表明,在约40个国家临床中心已有超过1500名患者接受了胰岛移植,移植后的疗效与胰腺器官移植的疗效相当,并有50-70%的患者在5年内达到不依赖胰岛素。虽然胰岛移植取得了令人振奋的成绩,但是胰岛移植在实施过程中仍存在许多不足,这限制了胰岛移植的进一步开展。最近的研究表明,移植胰岛的存活问题是影响移植胰岛早期存活的主要因素之一。
移植早期在缺血缺氧及炎症等复杂因素的影响下,超过25%的胰岛细胞在输注后立即丢失。移植胰岛的生存率是最好的移植物功能及胰岛素独立的预测指标,强调了在移植早期预防移植胰岛丢失的重要性。胰岛移植早期移植存活率主要取决两大因素:即时血液介导的炎症反应(IBMIR)和早期缺血缺氧环境。
IBMIR是早期移植胰岛丢失的主要原因之一。IBMIR是一种先天免疫系统的复杂反应,由移植胰岛与受者血液中的组织因子接触而启动。IBMIR并不局限于同种异体移植或者异种移植的环境,也存在于自体胰岛移植中,其特征主要包括补体系统激活、凝血级联反应、免疫调节和细胞因子的分泌。IBMIR在移植后几分钟后开始,约120min达到高峰。早期的IBMIR导致大量的胰岛细胞丢失,造成胰岛移植后5年胰岛素不依赖率低至10%,且可能需要从不同的供体多次输注胰岛,故有效控制IBMIR将克服临床胰岛移植的重要局限性。预防控制IBMIR可通过干预相关机制的激活达到。在临床胰岛移植中常规使用抗凝、抗血小板治疗和针对细胞因子反应的抗炎药预防IBMIR的发生。加入适量的抗凝剂(肝素)已被证实可显著改善移植疗效。最新研究表明,在小鼠模型中广泛表达于跨膜糖蛋白上的CD47在胰岛表面缓慢释放可减轻IBMIR,也建立了体内IBMIR监测模型。然而IBMIR未来将要面临的两大问题是:第一,在临床胰岛移植中探究使用人源胰岛细胞和猪源胰岛细胞而诱导的IBMIR的潜在相似性和差异性;第二,涉及胰岛移植的移植部位,IBMIR是只发生于门静脉移植,IBMIR与肝外移植部位之间的关系也有待进一步研究。
缺血缺氧是影响胰岛存活和移植的另一个重要因素。胰岛细胞是直径在50-400微米的高度血管化的多细胞构成的球状细胞团块,由分泌胰岛素的β细胞、分泌胰高血糖素的α细胞、分泌生长抑素的δ细胞、分泌生长素的ε细胞和胰腺多肽细胞组成。胰岛微结构及其内部广泛的微血管网络传递葡萄糖(和氧气)、相关激素和信号分子支持胰岛功能,而胰岛功能又受到旁分泌、自分泌和内分泌信号的严格调控。胰岛细胞中的β细胞由于抗氧化剂的低表达尤其对缺氧特别敏感,它们是通过在厌氧条件下在细胞内表达低水平的乳酸脱氢酶,催化丙酮酸转化为乳酸来提供能量三羧酸腺苷(ATP)。在移植胰岛早期的缺氧环境中不能产生ATP严重影响胰岛细胞的存活率。缺氧除了对胰岛细胞的存活有影响外,在缺氧环境中存活下来的胰岛细胞再次复氧后也会表现为一个持续功能受损的状态。即使成功地使用抗氧化应激或凋亡等其他药物或者方式来最小化缺氧对移植胰岛活力的影响,也不可以改变缺氧对胰岛细胞产生严重损害后遗症的现状。这也就表明即使短暂的暴露于缺氧环境也可以对胰岛素释放通路产生长期损害。大量的封装材料涌入胰岛移植领域,其免疫隔离、提供活性氧和预血管化的特征为改善胰岛移植存活提供了新思路及方案,也吸引了大批研究者。在研究过程中发现单纯补充氧气或者ATP,虽然可以很大程度上提高移植胰岛的生存能力,但是无法改善胰岛细胞的功能,同时出现了封装装置中后期纤维化等不良反应。这一研究结果使我们意识到封装及移植后的生物材料特性分析的重要性。细胞外基质、包裹封装装置周围的功能性血管系统和将细胞有效地暴露于循环刺激物中是封装细胞治疗的必备因素,任何一个因素的缺乏均会导致封装设备故障,正如迄今为止有限的胰岛移植临床试验所证明。综上所述,为了进一步提高移植胰岛的存活率并且保证胰岛具有完整的功能,减少早期移植胰岛丢失是必要的。
铁死亡是最近发现的受管制细胞死亡的途径,其特征是氧化应激和过氧化脂依赖性增加。铁死亡的主要机制是,通过二价铁或酯氧合酶,促进细胞膜上不饱和脂肪酸的高表达,导致脂质过氧化,诱导细胞死亡;还可降低抗氧化体系(谷胱甘肽系统)的调控核心酶GPX4,导致细胞死亡。这种基因决定和编程的过程被认为是导致胰岛功能受损,并可促进胰岛坏死,增强胰岛免疫原性。在胰岛细胞分离、培养和移植过程中,铁死亡在移植胰岛丢失中起着重要作用,并最终影响移植疗效。
ISL1在胰腺内分泌的四种主要细胞、中枢和周围神经系统的神经元以及心脏前体中均有表达。作为一种关键转录因子,在胚胎发生期间心血管细胞的发育中起着重要作用。ISL1在胰岛中的主要作用为促进胰岛内分泌细胞的分化、发育和增殖,也可调控内分泌细胞的激素分泌。然而,ISL1在胰岛移植铁死亡的研究国内外尚未见报道,有待进一步研究。
发明内容
发明的目的:为了提供效果更好的ISL1在制备抑制移植胰岛早期铁死亡的药物中的用途和细胞,具体目的见具体实施部分的多个实质技术效果。
为了达到如上目的,本发明采取如下技术方案:
方案一:
ISL1在制备抑制移植胰岛早期铁死亡的药物中的用途。
方案二:
一种用于实验和移植的ISL1过表达胰岛细胞,其特征在于,利用腺病毒构建过表达ISL1的胰岛细胞,得到ISL1过表达的胰岛细胞。
本发明进一步技术方案在于,包含如下步骤,
在大鼠β细胞(INS-1)中,明确ISL1抑制胰岛细胞铁死亡的作用;在分子水平上实现ISL1调节INS-1中的ACSL3的表达及作用;
在Lewis近交系大鼠胰岛细胞中,明确ISL1通过ACSL3抑制胰岛细胞铁死亡的作用;
在人源胰岛细胞中过表达ISL1,明确ISL1通过ACSL3抑制人源胰岛细胞铁死亡的作用;
本发明进一步技术方案在于,在大鼠β细胞(INS-1)中明确ISL1抑制铁死亡的作用;
(1)慢病毒构建过表达或敲减ISL1的INS-1细胞系,利用Western blot及RT-PCR分别在蛋白及mRNA水平检测INS-1细胞中ISL1及GPX4的表达水平变化情况;实验分组:取对数生长期的INS-1计数后分为以下6组:1)INS-Scrambled:将阴性对照慢病毒加入含有2×105INS-1细胞的六孔板中,作为过表达ISL1的阴性对照组;2)INS-1-ISL1组:利用ISL1慢病毒转染INS-1,构建过表达ISL1的INS-1细胞系,作为过表达ISL1组;3)Scramble-shRNA组:将阴性对照慢病毒加入含有2×105INS-1细胞的六孔板中,作为敲减ISL1的阴性对照组;4)shISL1-1组:利用ISL1慢病毒转染INS-1,构建敲减ISL1的INS-1细胞系,作为shISL1-1组;5)shISL1-2组:利用ISL1慢病毒转染INS-1,构建敲减ISL1的INS-1细胞系,作为shISL1-2组;6)shISL1-3组:利用ISL1慢病毒转染INS-1,构建敲减ISL1的INS-1细胞系,作为shISL1-3组;在分组标记好的六孔板内加入含10%胎牛血清的RPMI-1640,放入37℃,恒温恒湿培养箱内培养(5%CO2);培养后进行RT-PCR和Western blot分析;并利用链脲霉素STZ构建体外糖尿病环境,发现ISL1可以降低STZ诱导INS-1产生的铁死亡;
(2)利用免疫共沉淀-质谱(IPMS)进一步探究ISL1在铁死亡中的作用;取3.5cm2的小皿的1×107个INS-1-ISL1细胞,利用ISL1抗体做免疫共沉淀,同型IgG作为对照;样品进行凝胶电泳后进行考马斯亮蓝染色,并保留凝胶进行进一步的质谱分析;质谱分析结果表明在与ISL1相互作用的蛋白中铁死亡相关蛋白长链酰基辅酶A合成酶3(ACSL3),进一步证实ISL1可抑制铁死亡的结论;在过表达ISL1和干扰ISL1的INS-1细胞中进一步证实了ISL1与ACSL3之间的结合关系。
本发明进一步技术方案在于,ISL1对大鼠胰岛细胞铁死亡的抑制作用;
(1)Lewis近交系大鼠胰岛细胞分离纯化:SPF级别、8-12周龄、200g Lewis大鼠胆总管内逆行注入8ml的胶原酶进行原位灌注,胰腺取下后用眼科镊把胰腺撕碎至1mm3左右,移入含汉克斯平衡盐Hank’s溶液的50ml离心管37℃消化12分钟,当肉眼可见胰腺组织消化成泥沙状时,视为到达消化终点;后利用Ficol l400连续密度梯度离心法纯化大鼠胰岛细胞,并用DTZ鉴定胰岛细胞的纯度;
(2)糖尿病大鼠模型构建:Lewis SPF级近交系大鼠,禁食不禁水3小时后,分别称重,按每只50mg/kg快速腹腔注射STZ,然后再每2天测定1次随机血糖和体重;当随机血糖>16.7mol/L并维持15天以上,出现多饮、多食、多尿和消瘦症状为糖尿病大鼠造模成功;
(3)构建过表达ISL1的体系:我们利用腺病毒构建过表达ISL1的大鼠胰岛细胞,得到ISL1过表达的大鼠胰岛细胞;
(4)在体外利用STZ构建糖尿病环境,将过表达ISL1的大鼠胰岛细胞置于糖尿病环境中培养48h,检测细胞内GPX4和ACSL3的蛋白及mRNA水平变化情况;并将处理后的过表达ISL1的大鼠胰岛细胞将上述修饰后胰岛细胞向糖尿病大鼠模型的肾包膜下定向移植;将上述各组活性良好的1000IEQ的新鲜胰岛向糖尿病大鼠的肾包膜下定向移植;实验分组分为5组,每组6只大鼠:空白对照组、糖尿病大鼠模型组、单纯胰岛组、转染阴性对照胰岛组、过表达ISL1的胰岛治疗组;
分别观察以下指标:1)观察胰岛的特征以及移植后大鼠的生理指标;包括:动物的摄食量、饮水量及体重变化;空腹血糖水平、葡萄糖耐量试验、血C2肽及胰岛素含量;胰岛细胞存活(排斥反应)时间;2)通过病理切片观察移植后大鼠胰岛的情况:手术获取人工生物胰岛移植物,用4%的多聚甲醛固定过夜并用石蜡包埋,利用荧光染色技术观察胰岛细胞中GPX4、ISL1和ACSL3的表达变化情况。本发明进一步技术方案在于,ISL1对人源胰岛细胞铁死亡的抑制作用;
(1)人源胰岛细胞分离纯化:在取得供者知情同意的情况下,获得小块胰腺组织用眼科镊把胰腺撕碎至1mm3左右,移入含汉克斯平衡盐Hank’s溶液和胶原酶的50ml离心管37℃消化32分钟,当肉眼可见胰腺组织消化成泥沙状时,视为到达消化终点;后利用Ficoll400连续密度梯度离心法纯化人源胰岛细胞,并用DTZ鉴定胰岛细胞的纯度;
(2)构建过表达ISL1的体系:我们利用腺病毒构建过表达ISL1的人源胰岛细胞,得到ISL1过表达的人源胰岛细胞;
(3)在体外利用STZ构建糖尿病环境,将过表达ISL1的人源胰岛细胞置于糖尿病环境中培养48h,检测细胞内GPX4和ACSL3的蛋白及mRNA水平变化情况;并将处理后的过表达ISL1的人源胰岛细胞用4%的多聚甲醛固定过夜并用石蜡包埋,利用荧光染色技术观察胰岛细胞中GPX4、ISL1和ACSL3的表达变化情况。
采用如上技术方案的本发明,相对于现有技术有如下有益效果:在体外明确ISL1减少胰岛铁死亡的作用,利用INS-1过表达检测GPX4和ACSL3的表达情况,确定ISL1是否可影响INS-1的铁死亡,并利用大鼠胰岛细胞islets验证细胞系结果;其次,利用慢病毒构建敲减ISL1的INS-1,检测GPX4和ACSL3表达变化情况,且利用pull-down等现代分子生物学技术手段,进一步阐明ISL1与ACSL3的具体结合位点;进一步构建过表达ISL1的大鼠及人源胰岛细胞,在体外糖尿病的环境中,检测人源及大鼠过表达ISL1的胰岛细胞中GPX4和ACSL3的表达水平,验证ISL1在改善胰岛细胞铁死亡方面的作用。最终,明确ISL1通过促进ACSL3的表达抑制移植胰岛铁死亡的作用。构建的细胞能够用于实验,提供了胰岛移植的可能性。

具体实施方式
下面结合附图和具体实施方式,进一步阐明本发明,应理解下述具体实施方式仅用于说明本发明而不用于限制本发明的范围。
本专利选用的腺病毒为腺病毒pAdEasy-EF1-MCS-CMV-EGFP(EcoRI MCS)载体。其中腺病毒购买于汉恒生物。
本专利的重现性为百分之百。
胰岛素基因增强子结合蛋白-1(ISL1)可与ACSL3相互作用,进而减少胰岛细胞铁死亡
胰岛素基因增强子结合蛋白-1(ISL1),ISL1属于LIM同源域转录因子,是LIM同源域(LHX)蛋白家族的成员之一,ISL1在其N端附近包含两个排列整齐的LIM结构域和一个C端DNA结合同源域(HD)。LIM结构域为大小50-60个氨基酸,共有两个特异性高度保守的锌指片段,其各含有八个保守的残基,主要由半胱氨酸和组氨酸,它们与两个锌原子协调发挥结合其他蛋白作用,中心同源结构域(HD)包含60个负责特异性DNA结合的保守氨基酸残基。同时,ISL1-HD是真核DNA结合基序的重要家族之一,为DNA识别提供了重要的模型系统,它可在靶基因启动子中识别并结合共有的八聚体结合位点5’-ATAATTA-3’。此外,ISL1中的C端区域还包含LIM同源框3(LHX3)的结合域(LBD),它与LHX3的LIM域相互作用。本专利利用Raptor X web server预测ISL1结构模型及蛋白序列如下图5所示。

ISL1可通过诱导胰岛内ACSL3表达,减少移植胰岛铁死亡的发生,为提高早期移植胰岛存活的防治探索新的策略和途径。
鉴于早期移植胰岛处于缺血缺氧的环境中,氧化应激损伤在这个过程中也发挥重要作用。在胰岛细胞分离、培养和移植过程中,铁死亡在移植胰岛丢失中起着重要作用,并最终影响移植疗效。ISL1在胰岛中的主要作用为促进胰岛内分泌细胞的分化、发育和增殖,也可调控内分泌细胞的激素分泌。但其是否参与移植胰岛铁死亡目前缺乏报道。我们前期研究结果发现在大鼠β细胞系(INS-1)中过表达ISL1可提高胰岛存活率。进一步研究显示,在利用过表达ISL1的INS-1细胞进行IPMS时,发现与铁死亡相关的ACSL3可与ISL1相互作用,提示ISL1可能参与调控胰岛移植铁死亡的过程。以及移植胰岛的存活问题是影响移植胰岛疗效的主要因素之一,为探究ISL1是否为抑制移植胰岛早期铁死亡的机制之一,本项目确定如下的技术方案:本项目的实施为抑制移植胰岛铁死亡,改善胰岛低氧状态,提高移植胰岛存活率具有重要意义,为下一步应用于胰岛移植改善移植胰岛的缺血缺氧的微环境奠定理论基础。
基于本专利发现,ISL1可与ACSL3(长链乙酰-CoA合成酶3)相互作用。由此推测,
ISL1促进ACSL3的表达抑制移植胰岛铁死亡的发生。
1、在大鼠β细胞(INS-1)中,明确ISL1抑制胰岛细胞铁死亡的作用
(1)明确胰岛细胞中ISL1的表达变化伴随着细胞中的铁死亡发生;
(2)利用IPMS探究ISL1在铁死亡中的作用机制;
(3)在分子水平上研究ISL1调节INS-1中的ACSL3的表达及作用机制。
2、ISL1对胰岛细胞铁死亡的抑制作用
(1)利用密度梯度离心法提取大鼠及人源胰岛细胞;
(2)在STZ构建的体外糖尿病模型中,检测过表达ISL1的胰岛细胞中GPX4和ACSL3的表达水平,揭示ISL1抑制胰岛细胞铁死亡的作用机制。
目前,国内外关于移植胰岛铁死亡的这些过程知之甚少,是胰岛移植研究的热点,也是胰岛移植广泛应用于临床的主要阻碍之一。针对此问题我们利用基因修饰技术在INS-1细胞中改变ISL1的表达,实验结果表明ISL1可明显改善移植胰岛的功能、提高胰岛移植的存活率,其机制可能与抑制胰岛细胞铁死亡等作用相关。但是,ISL1对移植胰岛铁死亡的影响机制尚不清楚。进一步IP-MS,我们发现胰岛细胞中ISL1可与ACSL3相互作用。但国际关于ISL1抑制铁死亡尚未见报道。本项目首次提出了研究ISL1在抑制胰岛细胞铁死亡方面的重要作用及机制。本项目的实施对于填补ISL1在铁死亡领域的空缺,也为降低胰岛铁死亡率、改善胰岛低氧状态,提高移植胰岛存活率具有重要意义,为下一步应用于胰岛移植改善移植胰岛的缺血缺氧的微环境奠定理论基础,同时对于其他领域铁死亡的抑制也具有深远意义。
1、在大鼠β细胞(INS-1)中明确ISL1抑制铁死亡的作用;
(1)慢病毒构建过表达或敲减ISL1的INS-1细胞系,利用Western blot及RT-PCR分别在蛋白及mRNA水平检测INS-1细胞中ISL1及GPX4的表达水平变化情况;实验分组:取对数生长期的INS-1计数后分为以下6组:1)INS-Scrambled:将转染阴性对照慢病毒加入含有2×105INS-1细胞的六孔板中,作为过表达ISL1的阴性对照组;
2)INS-1-ISL1组:利用ISL1慢病毒转染INS-1,构建过表达ISL1的INS-1细胞系,作为过表达ISL1组;3)Scramble-shRNA组:将阴性对照慢病毒转染加入含有2×105INS-1细胞的六孔板中,作为敲减ISL1的阴性对照组;4)shISL1-1组:利用ISL1慢病毒转染INS-1,构建敲减ISL1的INS-1细胞系,作为shISL1-1组;5)shISL1-2组:利用ISL1慢病毒转染INS-1,构建敲减ISL1的INS-1细胞系,作为shISL1-2组;6)shISL1-3组:利用ISL1慢病毒转染INS-1,构建敲减ISL1的INS-1细胞系,作为shISL1-3组;在分组标记好的六孔板内加入含10%胎牛血清的RPMI-1640,放入37℃,恒温恒湿培养箱内培养(5%CO2);培养后进行RT-PCR和Western blot分析;并利用链脲霉素STZ构建体外糖尿病环境,发现ISL1可以降低STZ诱导INS-1产生的氧化应激损伤;
(2)利用免疫共沉淀-质谱(IPMS)进一步探究ISL1在铁死亡中的作用;取3.5cm2的小皿的1×107个INS-1-ISL1细胞,利用ISL1抗体做免疫共沉淀,同型IgG作为对照;样品进行凝胶电泳后进行考马斯亮蓝染色,并保留凝胶进行进一步的质谱分析;质谱分析结果表明在与ISL1相互作用的蛋白中铁死亡相关蛋白长链酰基辅酶A合成酶3(ACSL3),进一步证实ISL1可抑制铁死亡的结论;在过表达ISL1和干扰ISL1的INS-1细胞中进一步证实了ISL1与ACSL3之间的结合关系。(如全部附图所示)。
2、ISL1对大鼠胰岛细胞铁死亡的抑制作用
(1)Lewis近交系大鼠胰岛细胞分离纯化:SPF级别、8-12周龄、200g Lewis大鼠胆总管内逆行注入8ml的胶原酶进行原位灌注,胰腺取下后用眼科镊把胰腺撕碎至1mm3左右,移入含汉克斯平衡盐Hank’s溶液的50ml离心管37℃消化12分钟,当肉眼可见胰腺组织消化成泥沙状时,视为到达消化终点;后利用Ficol l400连续密度梯度离心法纯化大鼠胰岛细胞,并用DTZ鉴定胰岛细胞的纯度;
(2)糖尿病大鼠模型构建:Lewis SPF级近交系大鼠,禁食不禁水3小时后,分别称重,按每只50mg/kg快速腹腔注射STZ,然后再每2天测定1次随机血糖和体重;当随机血糖>16.7mol/L并维持15天以上,出现多饮、多食、多尿和消瘦症状为糖尿病大鼠造模成功;
(3)构建过表达ISL1的体系:我们利用腺病毒构建过表达ISL1的大鼠胰岛细胞,得到ISL1过表达的大鼠胰岛细胞;
(4)在体外利用STZ构建糖尿病环境,将过表达ISL1的大鼠胰岛细胞置于糖尿病环境中培养48h,检测细胞内GPX4和ACSL3的蛋白及mRNA水平变化情况;并将处理后的过表达ISL1的大鼠胰岛细胞将上述修饰后胰岛细胞向糖尿病大鼠模型的肾包膜下定向移植;将上述各组活性良好的1000IEQ的新鲜胰岛向糖尿病大鼠的肾包膜下定向移植;实验分组分为5组,每组6只大鼠:空白对照组、糖尿病大鼠模型组、单纯胰岛组、转染阴性对照胰岛组、过表达ISL1的胰岛治疗组;
分别观察以下指标:1)观察胰岛的特征以及移植后大鼠的生理指标;包括:动物的摄食量、饮水量及体重变化;空腹血糖水平、葡萄糖耐量试验、血C2肽及胰岛素含量;胰岛细胞存活(排斥反应)时间;2)通过病理切片观察移植后大鼠胰岛的情况:手术获取人工生物胰岛移植物,用4%的多聚甲醛固定过夜并用石蜡包埋,利用荧光染色技术观察胰岛细胞中GPX4、ISL1和ACSL3的表达变化情况。
3、ISL1对人源胰岛细胞铁死亡的抑制作用
(1)人源胰岛细胞分离纯化:在取得供者知情同意的情况下,获得小块胰腺组织用眼科镊把胰腺撕碎至1mm3左右,移入含汉克斯平衡盐Hank’s溶液和胶原酶的50ml离心管37℃消化32分钟,当肉眼可见胰腺组织消化成泥沙状时,视为到达消化终点;后利用Ficoll400连续密度梯度离心法纯化人源胰岛细胞,并用DTZ鉴定胰岛细胞的纯度;
(2)构建过表达ISL1的体系:我们利用腺病毒构建过表达ISL1的人源胰岛细胞,得到ISL1过表达的人源胰岛细胞;
(3)在体外利用STZ构建糖尿病环境,将过表达ISL1的人源胰岛细胞置于糖尿病环境中培养48h,检测细胞内GPX4和ACSL3的蛋白及mRNA水平变化情况;并将处理后的过表达ISL1的人源胰岛细胞用4%的多聚甲醛固定过夜并用石蜡包埋,利用荧光染色技术观察胰岛细胞中GPX4、ISL1和ACSL3的表达变化情况。
作为非必要的改进和进一步的方案和描述,本专利构建的过表达细胞,能够用于科学实验,用于制药甚至用于胰岛的防止铁死亡的移植。构建的细胞能够用于实验,提供了胰岛移植的可能性,ISL1过表达胰岛细胞能够用于体外器官生成,或者形成细胞群注入人体进行细胞级别的治疗。该ISL1过表达胰岛细胞和部分干细胞混合培养,利用混合培养中ISL1过表达胰岛细胞对部分干细胞的诱导,过表达细胞和母体干细胞混合培养利用胰岛细胞表面蛋白对干细胞进行分化诱导,生成生存能力和母体极其亲和的细胞群,作为实验或医疗用。或者该ISL1过表达胰岛细胞和干细胞进行融合,也可以用于组织修复或者是新的融合细胞,用于实验和医疗。

1.ISL1在制备抑制移植胰岛早期铁死亡的药物中的用途。
2.一种用于实验和移植的ISL1过表达胰岛细胞,其特征在于,利用腺病毒构建过表达ISL1的胰岛细胞,得到ISL1过表达的胰岛细胞。
3.如权利要求1所述的一种用于实验和移植的ISL1过表达胰岛细胞,其特征在于,包含如下步骤,
在大鼠β细胞INS-1中,明确ISL1抑制胰岛细胞铁死亡的作用在分子水平上实现ISL1调节INS-1中的ACSL3的表达及作用;
在Lewis近交系大鼠胰岛细胞中,明确ISL1通过ACSL3抑制胰岛细胞铁死亡的作用;
在人源胰岛细胞中过表达ISL1,明确ISL1通过ACSL3抑制人源胰岛细胞铁死亡的作用。
4.如权利要求1所述的一种用于实验和移植的胰岛细胞,其特征在于,在大鼠β细胞INS-1中明确ISL1抑制铁死亡的作用;

(4)在体外利用STZ构建糖尿病环境,将过表达ISL1的大鼠胰岛细胞置于糖尿病环境中培养48h,检测细胞内GPX4和ACSL3的蛋白及mRNA水平变化情况;并将处理后的过表达ISL1的大鼠胰岛细胞将上述修饰后胰岛细胞向糖尿病大鼠模型的肾包膜下定向移植;将上述各组活性良好的1000IEQ的新鲜胰岛向糖尿病大鼠的肾包膜下定向移植;实验分组分为5组,每组6只大鼠:空白对照组、糖尿病大鼠模型组、单纯胰岛组、转染阴性对照胰岛组、过表达ISL1的胰岛治疗组;
