Abstract
本发明属于生物工程和生物医药技术领域,涉及rAAV2/Retro作为针对视网膜感光细胞的递送系统及其在制备治疗视网膜疾病药物中的应用。以rAAV2/Retro为载体,构建携带外源性目的基因或shRNA的表达系统;以rAAV2/Retro作为递送系统,有效感染视网膜感光细胞,并特异性地表达所述目的基因或敲除内源性基因。本发明借助rAAV2/Retro独特的逆向回溯感染方式,特异性地作用于感光细胞,避免了传统腺相关病毒递送系统的非特异性;而且本发明用于治疗视网膜疾病的药物采用安全、全面的玻璃体腔注射,感染范围大于传统的腺相关病毒,具有更好的治疗效果。因此,利用本发明rAAV2/Retro病毒载体制得的用于治疗视网膜疾病的基因药物具有特异性强,安全性高,治疗效果好的优势。
Description
技术领域
本发明属于生物工程和生物医药技术领域,具体涉及新型rAAV2/Retro作为针对视网膜感光细胞的递送系统及其在制备治疗视网膜疾病药物中的应用。
技术背景
视网膜包括多种细胞,其中外核层主要由感光细胞组成(如图1)。感光细胞是视网膜内一类具有光信号转化功能的特殊神经细胞,是正常视觉产生的基础;视网膜疾病中普遍存在感光细胞病变,基因表达异常,从而严重影响视功能损。此外,感光细胞的基因突变或基因表达异常导致的视网膜疾病(如视网膜劈裂、先天性全色盲、年龄相关性黄斑变性、糖尿病视网膜病变、青光眼及眼外伤引起的病变)在致盲性眼病中占有相当比例。因此,针对视网膜疾病,靶向感光细胞的基因治疗是一种重要策略。但基因治疗的顺利推进面临着重要难题:在眼内缺乏针对感光细胞递送高效率的表达系统。
向感光细胞递送表达系统通常要依赖病毒感染,其中腺相关病毒(AAV)应用最为广泛,目前实现病毒感染感光细胞通常有如下两大类感染策略:(1)基于广谱强启动子的感染策略:利用对眼组织亲和性较好的腺相关病毒(如AAV2、AAV5、AAV7和AAV8)携带包括CAG或CMV广谱强启动子的表达系统进入感光细胞实现表达目的;(2)基于感光细胞特异性启动子的感染策略:该策略在采用对眼组织亲和性较好的腺相关病毒(如AAV2、AAV5、AAV7和AAV8)的基础上,进一步携带含有NR2E3、IRBP等感光细胞特异性启动子的表达系统,该表达系统进入感光细胞实现表达目的,并在非目的细胞中表达系统不表达。
但以上感染方法都有如下明显的缺陷:
(1)视网膜损伤风险。视网膜主要包括3层节构:节细胞层、内核层及感光细胞的外核层(图1);其中有相当部分的遗传性视网膜疾病都是由于感光细胞的基因缺陷或基因表达异常引起外核层损伤,从而致盲。此外,感光细胞的基因突变或基因表达异常导致的视网膜疾病(如视网膜劈裂、先天性全色盲、年龄相关性黄斑变性、糖尿病视网膜病变、青光眼及眼外伤引起的病变)在致盲性眼病中占有相当比例。目前针对外核层的感光细胞的给药方式主要有两种:①玻璃体腔注射(图2A),该方法注射针头只需穿过外侧的巩膜,睫状体扁平部,便可进入玻璃体腔,因而不造成视网膜脱离,视网膜细胞损伤小,炎症反应较轻;但其缺陷是普通病毒很难到达外核层。②视网膜下腔注射:这种方法需跨过玻璃体并再次穿过视网膜,如图2B所示,视网膜下腔是视网膜神经上皮层和色素上皮层分离时存在的潜在腔隙,病毒的扩散十分受限;同时,视网膜下腔注射会引起视网膜的局部分离,可能会引起胶质细胞增生,视网膜脱离,光感受器退化及视功能损害。
(2)感染范围有限。由于现有的腺相关病毒药物无法通过玻璃体腔注射达到外核层,而只能选择视网膜下腔注射给药,视网膜下腔是一个潜在的腔隙,局部的视网膜下腔注射并不能使病毒充分感染整个视网膜(图2B),导致感染范围受限。
(3)广谱强启动子专一靶向性差。AAV2、AAV5、AAV7和AAV8腺相关病毒的感染对神经元的种类并没有特异性,会感染病毒注射局部的所有类型的神经元和胶质细胞,病毒对非目的细胞的感染会降低目的感光细胞的感染效率并产生潜在的干扰和风险。
(4)感光细胞特异性启动子表达强度差。携带NR2E3、IRBP感光细胞特异型启动子的表达系统只在感光细胞中表达目的蛋白,但是表达强度远远不如广谱强启动子。
发明内容
基于现有技术中存在的问题,本发明的目的在于提供利用rAAV2/Retro构建针对视网膜感光细胞的高效率的表达系统;借助rAAV2/Retro独特的逆向回溯感染方式(图3),特异性地作用于感光细胞,避免了传统腺相关病毒递送系统感染细胞的非特异性。另外,本发明还给出了将该高效率的表达系统用于制备治疗视网膜疾病的药物,相对于传统的腺相关病毒药物,本发明药物能够采用安全、全面的玻璃体腔注射,从而规避了现有视网膜下腔注射给药造成的视网膜损伤,具有特异性强、治疗效果好、安全性高的优点。
基于上述目的,本发明采用如下技术方案:
第一方面,本发明提供了rAAV2/Retro作为针对视网膜感光细胞的递送系统的应用,以rAAV2/Retro为载体,构建携带外源性目的基因或shRNA的表达系统;以rAAV2/Retro作为递送系统,将所述目的基因或shRNA递送于视网膜感光细胞内,于视网膜感光细胞内表达外源性目的基因或敲除内源性基因。
rAAV2/Retro是经过定向进化改造的一种腺相关病毒次要衣壳蛋白,决定腺相关病毒的侵染特性,本发明将rAAV2/Retro作为次要衣壳蛋白组装腺相关病毒用于眼部侵染,这种rAAV2/Retro病毒在侵染眼部时具有以下特性:定点、高效以及创伤小,即具有逆向回溯感染能力,仅感染感光细胞,并高效率地表达外源基因,可以实现针对视网膜感光细胞递送高效表达系统的目的。
进一步地,上述外源性目的基因或shRNA包括与视网膜疾病相关基因或shRNA。
本发明以rAAV2/Retro为载体,充分利用rAAV2/Retro的逆向回溯感染能力,特异性地感染靶细胞如视网膜感光细胞,进而于视网膜感光细胞内高强度表达rAAV2/Retro上重组的外源目的基因或shRNA,从而达到高效、特异表达的目的。
第二方面,本发明提供了rAAV2/Retro在制备视网膜疾病治疗药物中的应用,以rAAV2/Retro为载体,构建与视网膜疾病相关基因或shRNA表达系统,制得用于治疗视网膜疾病的药物。
进一步地,在上述应用中,与视网膜疾病相关的基因包括但不限于RS1、CRX、CNGB3、CNGA3、GNAT2、PDE6C、NGF、BDNF、Neurotrophin-3、Neurotrophin-4基因,上述与视网膜疾病相关shRNA包括但不限于shNRL、shPTEN。
进一步地,上述视网膜疾病包括但不限于视网膜感光细胞、神经节细胞、双极细胞、水平细胞、无长突细胞或色素上皮细胞病变的疾病。
进一步地,在上述应用中,视网膜疾病包括但不限于年龄相关性黄斑变性、糖尿病视网膜病变、视网膜色素变性、先天性全色盲、先天性视网膜劈裂、GUCY2D-Leber先天性黑蒙以及青光眼和眼外伤引起的病变。
第三方面,基于上述应用,本发明提供一种用于治疗视网膜疾病的药物,该药物包含以rAAV2/Retro为载体,构建携带与视网膜疾病相关基因或shRNA的腺相关病毒。
进一步地,在上述药物中,与视网膜疾病相关基因包括RS1、CRX、CNGB3、CNGA3、GNAT2、PDE6C、NGF、BDNF、Neurotrophin-3、Neurotrophin-4;所述与视网膜疾病相关shRNA包括shNRL、shPTEN。
进一步地,在上述药物中,视网膜疾病包括视网膜感光细胞、神经节细胞、双极细胞、水平细胞、无长突细胞或色素上皮细胞病变的疾病。

进一步地,本发明提供了一种通过过表达目的基因治疗视网膜疾病的药物,用于治疗CNGA3基因突变导致的先天性全色盲,该药物中包含了rAAV2/Retro-Cnga3病毒;其中,rAAV2/Retro-Cnga3病毒中包含rAAV2/Retro载体和Cnga3的基因(ORF)。rAAV2/Retro-Cnga3病毒过表达Cnga3蛋白,Cnga3基因的核苷酸序列如SEQ ID NO.1所示。
Cnga3基因表达产生的Cnga3蛋白,主要表达于视网膜外核层的视锥细胞,当Cnga3基因突变致使Cnga3蛋白缺失,会导致视锥细胞逐渐变性死亡,视锥系功能逐渐丧失,引起全色盲及明视力的受损,主要表现为明适应后b波振幅的进行性下降,导致先天性全色盲的发生。
由于Cnga3基因作为视锥细胞的关键基因,当在rAAV2/Retro腺相关病毒上负载Cnga3基因(ORF),有望用于治疗Cnga3蛋白缺失引起的全色盲及明视力受损。
本发明通过在rAAV2/Retro载体上重组Cnga3基因制得rAAV2/Retro-CAG-Cnga3病毒,作为治疗由Cnga3蛋白缺失引起的全色盲及明视力受损的药物,借助rAAV2/Retro独特的逆向回溯感染方式,将Cnga3通过长轴突末端侵入神经元,特异性地作用于感光细胞,避免非特异性感染造成的潜在风险,相对于传统的AAV2和AAV8,本发明以rAAV2/Retro为载体制得的药物具有特异性强,治疗效果好,安全性高的优势。
进一步地,本发明提供了一种通过敲低目的基因治疗视网膜疾病的药物,该药物中包含rAAV2/Retro-shNrl病毒;其中,rAAV2/Retro-shNrl病毒中包含rAAV2/Retro载体和针对Nrl的shRNA序列。
Nrl基因表达产生Nrl蛋白,在视网膜色素变性中高表达于视网膜外核层的感光细胞,抑制Nrl基因的表达可以显著减少视网膜色素变性引起的感光细胞的凋亡。当在rAAV2/Retro腺相关病毒上负载Nrl的shRNA序列,有望用于治疗网膜色素变性引起的视网膜损伤。
本发明通过在rAAV2/Retro载体上重组针对Nrl基因的shRNA序列制得rAAV2/Retro-U6-shNrl病毒,抑制由RD1基因突变引起的感光细胞凋亡,借助rAAV2/Retro独特的逆向回溯感染方式,将Nrl的特异性shRNA通过长轴突末端侵入神经元,特异性地敲低感光细胞中的Nrl,避免非特异性感染造成的潜在风险,相对于传统的AAV2和AAV8,本发明以rAAV2/Retro为载体制得的药物具有特异性强,治疗效果好,安全性高的优势。
进一步地,rAAV2/Retro-CNGA3病毒、rAAV2/Retro-shNRL病毒、rAAV2/Retro-shPTEN病毒、rAAV2/Retro-RS1病毒、rAAV2/Retro-CRX病毒、rAAV2/Retro-CNGB3病毒、rAAV2/Retro-GNAT2病毒、rAAV2/Retro-PDE6C病毒、rAAV2/Retro-NGF病毒、rAAV2/Retro-BDNF病毒、rAAV2/Retro-Neurotrophin-3病毒、rAAV2/Retro-Neurotrophin-4病毒上重组的启动子包括但不限于CMV、CAG、U6、SYN、GFAP、EF1α。
进一步地,CAG启动子的核苷酸序列如SEQ ID NO.2所示。
与现有技术相比,本发明的有益效果如下:
(一)与传统AAV2、AAV5、AAV7和AAV8为载体的递送系统相比,本发明以rAAV2/Retro为载体,具有如下显著的优势:
(1)rAAV2/Retro具有特异性感染感光细胞的能力。
具体分析如下:rAAV2/Retro血清型腺相关病毒对神经元的感染形式为逆向回溯方式,如图3所示,rAAV2/Retro血清型腺相关病毒通过神经轴突末端侵入神经元,沿轴突逆向回溯至神经元胞体,进入细胞核从而发挥表达外源性基因的作用,而对胞体附近的神经元无感染能力,因而能够特异性地感染Recoverin阳性的感光细胞(如图4B、图5A所示),达到特异性治疗目的;而传统的AAV2、AAV5、AAV7和AAV8血清型腺相关病毒不具备逆向回溯感染能力和特异性感染感光细胞的能力,需要注射至胞体附近才能对神经元进行有效的感染,并且还会对胞体附近的神经元进行感染(图5B、图6所示),具有非特异性感染的风险(图5B白色箭头所示);因此,本申请以rAAV2/Retro血清型腺相关病毒为载体,能够有效避免基于广谱启动子的传统的AAV2、AAV5、AAV7和AAV8血清型腺相关病毒的非特异性感染造成的潜在风险。与此同时,由于rAAV2/Retro具有良好的特异性,可以使用高表达效率的广谱启动子,克服了基于细胞特异性启动子的腺相关病毒递送系统表达效率不足的缺点。
(2)rAAV2/Retro可以高效的表达有功能的外源蛋白。
申请者前期研究表明,表达外源性Gata3蛋白的rAAV2/Retro-CAG-mGata3-GFP病毒和对照rAAV2/Retro-CAG-GFP病毒高效、特异性感染视网膜外核层,Gata3蛋白(红色)作为转录因子特异的定位于细胞核内(图7B所示),而对照病毒只有GFP绿色荧光表达(图7A所示)。同时已有研究表明rAAV2/Retro血清型腺相关病毒对外源基因的表达效率比传统的AAV2、AAV5、AAV7和AAV8高,以rAAV2/Retro血清型腺相关病毒为载体的药物,能够通过低的用量达到高效治疗的目的,且能有效避免高滴度的病毒可能引发的免疫反应。
(3)建立简单有效的使用途径:玻璃体腔给药。
本发明以rAAV2/Retro为载体制得的递送载体,能够通过玻璃体腔注射,借助rAAV2/Retro的逆向回溯感染能力(图3),达到了只对感光细胞感染的目的,而不感染其他细胞或组织;而现有的AAV2和AAV8血清型腺相关病毒,由于不具备特异性,必须注射到视网膜下腔才能达到感染并表达的目的,具体分析如下:
视网膜组织为多层神经元结构,如图1所示,由最内层的节细胞、内核层的中间神经元和最外层的感光细胞组成;当需要对外核层的感光细胞进行感染治疗时,将rAAV2/Retro腺相关病毒注射到玻璃体腔即可,rAAV2/Retro腺相关病毒可以铺满整个视网膜表面,如图4A所示,具有逆向回溯功能的rAAV2/Retro腺相关病毒可通过轴突对外核层的神经元进行有效的感染,而且该rAAV2/Retro腺相关病毒定向感染外核层的细胞,对内核层及节细胞没有影响,因此,当携带有外源基因的rAAV2/Retro腺相关病毒被注射至玻璃体腔时,能够使得外源基因在外核层细胞高效表达,而不会在内核层细胞和节细胞中表达,达到特异性感染目的细胞的目的(如图4和图5A所示)。玻璃体腔给药的具有操作简单、损伤小、病毒作用范围广的优势。玻璃体腔注射时,注射针头只需穿过外侧的巩膜,色素上皮层和睫状体扁平部,如图2A所示,便可进入玻璃体腔;而且玻璃体腔内的病毒可充分的接触和感染整个视网膜。
而传统血清型腺相关病毒(AAV2,AAV4、AAV7和AAV8)必须注射到视网膜下腔。视网膜下腔注射还需跨过玻璃体并再次穿过视网膜,如图2B及图6所示,这种注射会引起视网膜的局部分离,可能会引起胶质细胞增生,视网膜脱离,光感受器退化及视功能损害;而且视网膜下腔是视网膜神经上皮层和色素上皮层分离时存在的潜在腔隙,病毒的扩散十分受限,病毒只能接触局部组织,然后感染外核层,如图5B、图6所示,而且内核层及节细胞层也有细胞感染(如图5B白色箭头、图6B绿色箭头)。
由上述可知,本发明以rAAV2/Retro腺相关病毒为载体时,能够实现将递送系统通过玻璃体腔注射即可,具有操作简单,且递送效果优异的优点。
(二)相对于现有传统的AAV2及AAV8载体,本发明以rAAV2/Retro为载体,能够在靶细胞中更加地高效表达其携带的外源基因,使得以rAAV2/Retro为载体的病毒在作为治疗药物时,在低剂量下便能够高效表达外源基因,从而降低药物剂量,避免高滴度的病毒引发机体的免疫反应。
(三)rAAV2/Retro载体具有独特的逆向回溯感染方式,使得基于rAAV2/Retro载体构建的病毒药物仅通过长轴突末端侵入神经元,而对胞体在附近的神经元无感染能力,特异性地作用于感光细胞,从而避免传统AAV2及AAV8载体的非特异性感染造成的潜在风险。
(四)本发明构建的用于治疗视网膜疾病的药物具有操作简单、损伤小的优点,将药物进行玻璃体腔注射,注射针头只需穿过外侧的巩膜,色素上皮层和睫状体扁平部便可进入玻璃体腔,不造成视网膜脱离,视网膜细胞损伤小,炎症反应较轻。避免了现有药物需要通过视网膜下腔注射的方式,该注射方式还需跨过玻璃体并再次穿过视网膜,会引起视网膜的局部分离,可能会引起胶质细胞增生,视网膜脱离,光感受器退化及视功能损害的副作用。
(五)本发明构建的用于治疗视网膜疾病的药物还具有作用范围更大的优点,由于本发明构建的药物逆向回溯感染,可以通过玻璃体腔注射感染外核层。玻璃体腔为视网膜前腔隙,玻璃体腔内的病毒药物可充分地接触和感染整个视网膜,解决了现有药物仅能通过视网膜下腔注射,视网膜下腔是视网膜神经上皮层和色素上皮层分离时存在的潜在腔隙,病毒药物的扩散十分受限的问题。

具体实施方式
为更好地说明本发明的目的、技术方案和优点,下面将结合附图和具体实施例对本发明作进一步说明。以下实施例所涉及各原料如无特别说明,均为市售通用产品。
实施例1.rAAV2/Retro用于装配腺相关病毒
用三质粒系统共转HEK293细胞系生产具有感染能力的病毒颗粒。HEK293细胞是用修饰过的5型腺病毒DNA转化人胚肾细胞得到的细胞系,表达腺病毒E1蛋白。三质粒包括rAAV2/Retro辅助质粒(从Addgene购得,货号为81070)、pAdDeltaF6辅助病毒质粒(从Addgene购得,货号为112867)和pAAV表达质粒(购自Addgene,货号为37825)。
rAAV2/Retro辅助质粒如图8所示,在HEK293细胞系中产生病毒装配所需的次要衣壳蛋白rAAV2/Retro。其中,43至145为ITR;413至2152为病毒主要衣壳蛋白1(VP1);2169至4406为病毒次要衣壳蛋白retro;5185至5698为M13 ori;6449至6549为ampicillin启动子;6550至7410为ampicillin;7581至8169为pUC ori;病毒次要衣壳蛋白retro是腺相关病毒感染特异性的关键。
pAAV如图9所示,经改造后作为表达质粒。其中,182至770为pUC ori;941至1801为ampicillin;1932至2387为f1 ori;2459至2588为AAV2 ITR;3095至3336为多克隆位点(MCS);3343至3931为WPRE;3974至4095为SV40 polyA;4257至4386为AAV2 ITR。在多克隆位点内可以插入多种启动子,包括但不限于CMV、CAG、U6、SYN、GFAP、EF1α。其后可以插入各类目的表达片段,包括但不限于各类普通基因片段、视网膜遗传性疾病相关基因片段、shRNA。
实施例2制备rAAV2/Retro病毒用于在视网膜感光细胞递送pAAV-CAG-Cnga3-GFP表达系统
当Cnga3基因突变致使Cnga3蛋白缺失,会导致视锥细胞逐渐变性死亡,视锥系功能逐渐丧失,引起全色盲及明视力的受损,主要表现为明适应后b波振幅的进行性下降。过表达外源性的Cnga3蛋白可以逆转这一过程。
rAAV2/Retro-CAG-Cnga3-GFP病毒药物由pAAV-CAG-Cnga3-GFP、pAdDeltaF6、rAAV2/Retro helper质粒经病毒包装工艺得到,包含了rAAV2/Retro病毒载体和Cnga3基因(ORF)及报告基因GFP,其中GFP在正式的疾病治疗基因药物中可以去掉。
rAAV2/Retro-CAG-Cnga3-GFP病毒药物的具体制备过程如下:
1)pAAV-CAG-Cnga3-GFP质粒的构建
将CAG启动子、全长小鼠Cnga3 ORF、GFP串联克隆到pAAV表达质粒中。以pAAV多克隆位点的BamHI和XbaI位点作为克隆靶点,如图9所示,主要待克隆的片段为全长小鼠Cnga3ORF,通过PCR扩增反应从小鼠661W细胞系中获得,通过重叠PCR技术将CAG启动子、全长小鼠Cnga3 ORF、GFP串联扩增并引入BamHI和XbaI位点。pAAV表达质粒与重叠PCR获得的CAG-Cnga3-GFP串联产物经BamHI和XbaI酶切后进行质粒重组,重组质粒在E.coli Stbl3中繁殖,并在赛默飞公司测序设施中进行序列验证。其中,Cnga3的核苷酸序列如SEQ ID NO.1所示;CAG启动子的核苷酸序列如SEQ ID NO.2所示。
在产生rAAV2/Retro-CAG-Cnga3-GFP病毒药物之前,确认了pAAV-CAG-Cnga3-GFP质粒DNA序列的准确性。其中,182至770为pUC ori;941至1801为ampicillin;1932至2387为f1 ori;2459至2588为AAV2 ITR;2899至3176为CAG promoter;3490至5499为m-Cnga3;5512至6231为GFP;6356至6844为WPRE;6887至7008为SV40 polyA;7170至7299为AAV2 ITR。
2)rAAV2/Retro-CAG-Cnga3-GFP病毒制备
①rAAV2/Retro-CAG-Cnga3-GFP病毒的出毒包装
a.准备HEK-293细胞:在每一个100mm细胞培养盘10mL DMEM生长培养基中加入3×106个HEK-293细胞,48小时后用于转染。为了获得较高的滴度,HEK-293细胞的健康和传代的密度是非常重要的因素。当细胞汇合率达到50%时须传代。并在细胞处于低代并生长健康时进行大量冻存。当传代和铺板用于转染时要避免细胞成团。在用pAAV-CAG-Cnga3-GFP,pAdDeltaF6,rAAV2/Retro helper转染之前细胞会生长到较高的汇合率。
b.HEK-293细胞的转染:尽管大量的转染实验方案可以成功用于这些载体的转染,但是推荐使用下述的PEI MAX 40K(购自Polysciences公司,货号49553-93-7)转染方法,本实验方案用于转染HEK-293细胞时,可稳定获得>107病毒颗粒/毫升的产品滴度。具体步骤如下:
*转染前检查两天前传代的HEK293细胞,它们应当达到70%~80%的汇合度;
*从-20℃冰箱中取出将进行共转染的质粒,pAAV-CAG-Cnga3-GFP,pAdDeltaF6,rAAV2/Retro helper用pH7.5 TE缓冲液将质粒的浓度调整到1mg/mL;
*根据包装盘数计算所需的转染体系和质粒用量,如果包装一盘则吸取pAAV-CAG-Cnga3-GFP、pAdDeltaF6、rAAV2/Retro helper质粒各5μL(或5μg)到一个含150ul无血清DMEM的1.5mL EP离心管中,混合均匀后室温静置2分钟,然后加入60ul 1mg/mL的PEI MAX,涡旋混合后室温静置10分钟,加入7mL DMEM生长培养基制得质粒DNA/PEI MAX/DMEM混合液。
*将细胞培养盘从37℃培养箱取出,吸弃原有培养基,加入全部质粒DNA/PEI MAX/DMEM混合液;
*将培养盘送回到孵箱中,另外培养66~72小时。
②观察出毒
通过观察HEK-293细胞的形态变化可以得知rAAV2/Retro-CAG-Cnga3-GFP病毒颗粒的包装进行程度。为了比较方便,做一盘病毒包装阴性对照,以不加前述质粒的转染组作为阴性对照。病毒包装成功的最明显的征兆是与与阴性对照相比,培养基的颜色由红色变为橙色或黄色,且随着出毒的进行,有些细胞会变圆并从盘上脱落,可以看到它们漂浮在培养基中。转染后三天是收集rAAV2/Retro-CAG-Cnga3-GFP病毒颗粒的最佳时机。
③收集rAAV2/Retro-CAG-Cnga3-GFP病毒颗粒
rAAV2/Retro-CAG-Cnga3-GFP病毒颗粒同时存在于包装细胞和培养上清中,因此,将细胞和培养上清都收集下来以获得最好的回收率。具体步骤如下:
*准备一个干冰乙醇浴和37℃水浴;
*将产毒的细胞连同培养基一同收集到一个15mL的离心管中,收集细胞时,将培养盘倾斜一定角度将细胞刮到培养基中;
*将上述收集有产毒细胞和培养基的离心管于200g、3分钟的离心条件下,离心分离产毒细胞和培养上清,将培养上清另外存放,细胞用1mL PBS重悬得到细胞重悬液;
*将细胞重悬液在干冰乙醇浴和37℃水浴中反复转移,冻融裂解四次,每次融解后稍加震荡每次凝固和解冻大概需要十分钟的时间;
*冻融裂解后的细胞重悬液于10000g、5分钟的离心条件下,离心去除细胞碎片,离心后的细胞冻融裂解上清转移到一个新离心管中。
④rAAV2/Retro-CAG-Cnga3-GFP病毒的浓缩
*在新离心管内的上清中加入40%PEG8000直到其终浓度为8%,在冰上放置2小时,其中每隔15分钟来回混匀一次,随后于2500xg的离心条件下,离心30分钟,去掉上清液,收集的沉淀用PBS重悬后与前述细胞冻融裂解上清合并;
*将PBS重悬的沉淀与细胞裂解上清的混合液于3,000xg的离心条件下,离心30分钟,将上清转移到另一个干净的管中。如果上清仍有部分碎片,则再次离心收集上清;
*向收集的上清中加入Benzonase核酸酶(终浓度为50U/mL)消化去除残留的质粒DNA,合上管盖,颠倒几次以充分混合,在37℃孵育30分钟;
*用0.45um过滤头过滤,取滤出液,即得rAAV2/Retro-CAG-Cnga3-GFP病毒浓缩液。
⑤rAAV2/Retro-CAG-Cnga3-GFP病毒的初步纯化
*向rAAV2/Retro-CAG-Cnga3-GFP病毒浓缩液中添加固体氯化铯(CsCl)直到密度为1.41g/mL,大概是10mL rAAV2/Retro-CAG-Cnga3-GFP病毒浓缩液中加6.5g CsCl,振荡使之溶解;
*将溶解于CsCl的rAAV2/Retro-CAG-Cnga3-GFP病毒浓缩液加入到超速离心管中,用预先配好的1.41g/mL CsCl溶液将离心管剩余空间填满;
*在175,000g下离心24小时,以形成密度梯度。按顺序分步收集不同密度的样品,取样进行滴度测定,收集富集有rAAV2/Retro-CAG-Cnga3-GFP病毒颗粒的组分;
*重复上述过程一次,对收集的rAAV2/Retro-CAG-Cnga3-GFP病毒颗粒再次进行纯化。
⑥超滤脱盐纯化
*淋洗:在Amicon-15超滤装置中加入4mL去离子水;一旦在超滤装置浸湿后,则不能任由膜再次晾干。如果不是淋洗后立即使用,则将水留在膜上直到开始实验;
*将密度梯度离心得到的rAAV2/Retro-CAG-Cnga3-GFP病毒液加入到超滤装置中,加PBS至总体积为4mL,盖上盖子;随后,于1,500xg下离心10分钟,每5分钟检查一次剩余体积数,直到最终体积为200~250μL,不要将体积减少到200μl以下,因为这会导致病毒克隆的聚集。如果滴度较高,则要相应地增加最小体积;
*将滤除液收集到一起以便做消毒处理,将滤膜放回到装置中;加1×PBS稀释浓缩后的病毒使体积为4mL;
*重复上述过程3次;离心超滤管,使最终体积大约为0.5mL;在病毒浓缩液中加入甘油使得终浓度为5%。分装后保存在-80℃。
⑦rAAV2/Retro-CAG-Cnga3-GFP病毒滴度的测定
我们采用定量PCR方法检测rAAV2/Retro-CAG-Cnga3-GFP病毒载体的基因组拷贝数来测定rAAV2/Retro-CAG-Cnga3-GFP的病毒颗粒数。标准曲线绝对定量PCR检测滴度的准确性和可靠性是rAAV2/Retro-CAG-Cnga3-GFP质量控制的最关键要素,其结果会影响下游实验的精确度。因此,在我们的质控步骤中,对rAAV2/Retro-CAG-Cnga3-GFP病毒滴度做了完善的设计以保证其准确性和稳定性,如在检测设计一系列的标准品、验证样品(病毒和质粒)和对照(为了除去背景和DNA污染)。质控流程中所使用的滴定引物是常备试剂,因此可缩短病毒生产周期以适应快节奏的载体构建需求。
使用上述方法产生rAAV2/Retro-CAG-Cnga3-GFP病毒,滴定病毒颗粒并将其重悬于含有0.014%Tween-20的平衡盐溶液中,浓度为1.5×1012病毒载体基因组/mL(vg/mL),在最终产品中确认了无菌性和缺乏内毒素。
作为对比,使用上述方法,仅将前述三质粒转染系统中的prAAV2/Retro替换为传统pAAV2/5(addgene货号104964)产生rAAV2/5-CAG-Cnga3-GFP病毒,滴定病毒颗粒并将其重悬于含有0.014%Tween-20的平衡盐溶液中,浓度为1.5×1012病毒载体基因组/mL(vg/mL),在最终产品中确认了无菌性和缺乏内毒素。
实施例3制备rAAV2/Retro病毒用于在视网膜感光细胞递送pAAV-U6-shNrl表达系统
Nrl基因表达产生Nrl蛋白,在视网膜色素变性中高表达于视网膜外核层的感光细胞,抑制Nrl基因的表达可以显著减少视网膜色素变性引起的感光细胞的凋亡。
rAAV2/Retro-U6-shNrl病毒药物由pAAV-U6-shNrl、pAdDeltaF6、rAAV2/Retrohelper质粒经病毒包装工艺得到,包含了rAAV2/Retro病毒载体和针对Nrl基因的shRNA。
rAAV2/Retro-U6-shNrl病毒药物的具体制备过程如下:
1)pAAV-U6-shNrl质粒的构建
将U6启动子、针对小鼠Nrl基因的shRNA(shNrl)串联克隆到pAAV表达质粒中。以pAAV多克隆位点的BamHI和XbaI位点作为克隆靶点,如图9所示,通过重叠PCR技术将U6启动子、shNrl串联扩增并引入BamHI和XbaI位点。pAAV表达质粒与重叠PCR获得的U6-shNrl串联产物经BamHI和XbaI酶切后进行质粒重组,重组质粒在E.coli Stbl3中繁殖,并在赛默飞公司测序设施中进行序列验证。其中,将shNrl的序列打乱得到shControl作为对照,构建pAAV-U6-shControl。shNrl的核苷酸序列如SEQ ID NO.3所示;U6启动子的核苷酸序列如SEQ ID NO.4所示;shControl的核苷酸序列如SEQ ID NO.5所示。
在产生rAAV2/Retro-U6-shNrl病毒药物之前,确认了pAAV-U6-shNrl质粒DNA序列的准确性。其中,182至770为pUC ori;941至1801为ampicillin;1932至2387为f1 ori;2459至2588为AAV2 ITR;3206至3341为U6 promoter;3452至3500为shNrl;3529至4117为WPRE;4160至4281为SV40 polyA;4443至4542为AAV2 ITR。
2)rAAV2/Retro-U6-shNrl和rAAV2/Retro-U6-shControl病毒制备
参照实施例2中rAAV2/Retro-CAG-Cnga3-GFP病毒的出毒包装步骤,将步骤中使用pAAV-CAG-Cnga3-GFP质粒全部等量替换为pAAV-U6-shNrl质粒,即在最终产物中获得rAAV2/Retro-U6-shNrl病毒。
参照实施例2中rAAV2/Retro-CAG-Cnga3-GFP病毒的出毒包装步骤,将步骤中使用pAAV-CAG-Cnga3-GFP质粒全部等量替换为pAAV-U6-shControl质粒,即在最终产物中获得rAAV2/Retro-U6-shControl病毒。
实施例4.rAAV2/Retro-CAG-Cnga3-GFP病毒的治疗效果
本实施例以先天性全色盲小鼠作为实验对象,探究由实施例2制得的rAAV2/Retro-CAG-Cnga3-GFP病毒的治疗效果,具体实验方案如下:
实验动物:正常10周龄C57bl/6j小鼠购自广东省实验动物中心;先天性全色盲转基因小鼠cpfl5 Mice购自维通利华公司。光照周期12h光照-12h黑暗,自由采食,自由饮水,所有的动物研究严格按照国家科学技术委员会颁发的《实验动物管理条例》进行。
主要试剂、耗材:生理盐水(浙江天瑞药业有限公司),一次性注射针头(美国Becton Dickinson and Company公司),山羊血清(Sigma),聚乙二醇辛基苯基醚(TritonX-100)(Sigma),OCT组织包埋剂(樱花),多聚甲醛(Sigma),DAPI(碧云天),复方托吡卡胺(沈阳兴齐眼药股份有限公司),三溴乙烷(Sigma),PBS溶液(浙江森瑞生物科技有限公司),妥布霉素眼膏(托百士)。
主要仪器:眼科实验手术显微镜(日本Nikon公司);眼科显微器械(苏州明仁医疗器械有限公司);微量进样器(美国Hamilton公司);荧光显微镜(德国蔡司公司),数码眼底彩照及荧光造影系统(Topcon),ERG系统(Roland Consult),冰冻切片机(徕卡)。
眼内注射:用三溴乙醇按照0.14mL/10g体重的剂量麻醉小鼠,0.5%的复方托比卡胺以扩张瞳孔,使用30G的破囊针头于巩膜角膜缘后无血管区倾斜45°避开晶体刺入玻璃体腔,然后再使用33G的显微注射器从破囊针破口部位45°进针避开晶体进入玻璃体腔,分别注射1μL浓度为1.5×1012vg/mL的rAAV2/Retro-CAG-Cnga3-GFP病毒药物和rAAV2/Retro-CAG-GFP对照病毒。留置10s后缓缓拔出显微注射器,确认无液体漏出。使用妥布霉素软膏涂布眼表预防感染,并将老鼠置于37℃复苏毯上直至老鼠苏醒。术后老鼠常规饲养。
ERG检查:在术后14天,28天,2个月,3个月,6个月行ERG检查,确认基因治疗对视功能的改善情况。用三溴乙醇(0.14mL/10g体重)麻醉小鼠,复方托比卡胺(0.5%)扩张瞳孔,纤维素钠滴眼液涂布小鼠眼表。ERG检测方法如文献[Yang S,Luo X,Xiong G,So KF,YangH,Xu Y.The electroretinogram of Mongolian gerbil(Meriones unguiculatus):comparison to mouse.Neurosci Lett.2015;589:7.]中所示,对小鼠进行基因药物(rAAV2/Retro-CAG-Cnga3-GFP)治疗6个月前后小鼠明适应视网膜电生理检测结果如图10所示,发现在对cpfl5小鼠进行基因药物治疗6个月后,cpfl5小鼠的b波振幅比眼内注射对照病毒组小鼠视功能明显改善,且经本发明基因药物治疗后cpfl5小鼠的b波振幅与正常同周龄小鼠无显著差异。
随后,通过对经本发明基因药物治疗后的cpfl5小鼠进行免疫荧光检测,具体步骤如下:
组织处理:术后6个月进行取材,动物首先经过PBS全身灌注去除血液,后使用4%多聚甲醛灌注固定,灌注速度为5mL/min,眼球取出后在巩膜角膜缘扎孔后浸泡于4%多聚甲醛4℃固定过夜,10%,20%,30%梯度蔗糖脱水后OCT包埋,置于液氮中快速冰冻。使用冰冻切片机制作厚度为10um的视网膜切片。
免疫荧光检测:取眼球冰冻切片,使用PBS洗涤3次洗去OCT;使用DAPI孵育5min标记细胞核。并用PBS洗涤3次去除多余的DAPI,使用抗荧光淬灭剂封片。使用荧光显微镜观察GFP表达情况以及视网膜形态结构并拍照,结果如图11所示,可见GFP荧光依然特异而高效地表达在视网膜外核层,同时也表明该基因药物能够在6个月内稳定表达。
因此,通过将实施例1制得的rAAV2/Retro-CAG-Cnga3-GFP病毒药物,注射于cpfl5小鼠玻璃体腔中,通过特异性感染外核层的感光细胞,使得有功能的Cnga3蛋白重新表达,从而有效抑制了cpfl5小鼠视锥细胞变性过程,使得治疗后的cpfl5小鼠的明适应的b波振幅达到正常小鼠水平,可见,由本发明制得的rAAV2/Retro-CAG-Cnga3-GFP病毒药物对先天性全色盲具有良好的疗效。
实施例5.rAAV2/Retro-U6-shNrl病毒的治疗效果
本实施例以视网膜色素变性小鼠为实验对象,探究由实施例3制得的rAAV2/Retro-U6-shNrl病毒的治疗效果,具体实验方案如下:
实验动物:正常10周龄C57bl/6j小鼠购自广东省实验动物中心;视网膜色素变性转基因小鼠rd1 Mice购自维通利华公司。光照周期12h光照-12h黑暗,自由采食,自由饮水,所有的动物研究严格按照国家科学技术委员会颁发的《实验动物管理条例》进行。
主要试剂、耗材:生理盐水(浙江天瑞药业有限公司),一次性注射针头(美国Becton Dickinson and Company公司),山羊血清(Sigma),聚乙二醇辛基苯基醚(TritonX-100)(Sigma),OCT组织包埋剂(樱花),多聚甲醛(Sigma),DAPI(碧云天),复方托吡卡胺(沈阳兴齐眼药股份有限公司),三溴乙烷(Sigma),PBS溶液(浙江森瑞生物科技有限公司),妥布霉素眼膏(托百士)。
主要仪器:眼科实验手术显微镜(日本Nikon公司);眼科显微器械(苏州明仁医疗器械有限公司);微量进样器(美国Hamilton公司);荧光显微镜(德国蔡司公司),数码眼底彩照及荧光造影系统(Topcon),ERG系统(Roland Consult),冰冻切片机(徕卡)。
眼内注射:用三溴乙醇按照0.14mL/10g体重的剂量麻醉小鼠,0.5%的复方托比卡胺以扩张瞳孔,使用30G的破囊针头于巩膜角膜缘后无血管区倾斜45°避开晶体破入玻璃体腔,使用33G的显微注射器从破囊针破口部位45°进针避开晶体进入玻璃体腔,分别注射1μL浓度为1.5×1012vg/mL的rAAV2/Retro-U6-shNrl病毒药物和rAAV2/Retro-U6-shControl对照病毒。留置10s后缓缓拔出显微注射器,确认无液体漏出。使用妥布霉素软膏涂布眼表预防感染,并将老鼠置于37℃复苏毯上直至老鼠苏醒。术后老鼠常规饲养。
ERG检查:在术后14天,28天,2个月,3个月,6个月行ERG检查,确认基因药物治疗对视功能的改善情况。用三溴乙醇(0.14mL/10g体重)麻醉小鼠,复方托比卡胺(0.5%)扩张瞳孔,纤维素钠滴眼液涂布小鼠眼表。ERG检测方法如文献[Yang S,Luo X,Xiong G,So KF,Yang H,Xu Y.The electroretinogram of Mongolian gerbil(Meriones unguiculatus):comparison to mouse.Neurosci Lett.2015;589:7.]中所示,对小鼠进行基因药物(rAAV2/Retro-U6-shNrl)治疗6个月前后小鼠的明适应视网膜电生理检测结果如图12所示,发现在对Rd1小鼠进行基因药物治疗6个月后,Rd1小鼠的b波振幅比眼内注射对照病毒组小鼠视功能明显改善。
随后,通过对经本发明基因药物治疗后的rd1小鼠进行免疫荧光检测,具体步骤如下:
组织处理:术后6个月进行取材,动物首先经过PBS全身灌注去除血液,后使用4%多聚甲醛灌注固定,灌注速度为5mL/min,眼球取出后在巩膜角膜缘扎孔后浸泡于4%多聚甲醛4℃固定过夜,10%,20%,30%梯度蔗糖脱水后OCT包埋,置于液氮中快速冰冻。使用冰冻切片机制作厚度为10um的视网膜切片。
免疫荧光检测:取眼球冰冻切片,使用PBS洗涤3次洗去OCT;使用特异标记Nrl的抗体标记感染了基因药物rAAV2/Retro-U6-shNrl和对照病毒AAV2/Retro-U6-shControl的视网膜,并使用DAPI孵育5min标记细胞核。用PBS洗涤3次去除多余的DAPI,使用抗荧光淬灭剂封片。使用荧光显微镜观察Nrl表达情况以及视网膜形态结构并拍照,结果如图13所示,图13A为对照病毒处理后视网膜外核层中Nrl表达情况图,如图中红色三角形标识部分,可见视网膜外核层中Nrl表达非常强;图13B为基因药物处理后视网膜外核层中Nrl表达情况图,如图中粉色三角形标识部分,与对照组图13A中相比,基因药物处理后视网膜外核层中Nrl表达明显减弱,同时可以看出,如黄色箭头标记部分所示,基因药物处理后有助于促进外核层神经纤维的恢复。
因此,通过将实施例3制得的rAAV2/Retro-U6-shNrl病毒药物,注射于rd1小鼠玻璃体腔中,通过特异性感染外核层的感光细胞,敲低Nrl基因的表达,从而有效抑制了rd1小鼠感光细胞变性过程,使得治疗后的rd1小鼠ERG的b波振幅明显改善,可见,由本发明制得的rAAV2/Retro-U6-shNrl病毒药物对先天性视网膜色素变性具有良好的疗效。
最后所应当说明的是,以上实施例仅用以说明本发明的技术方案而非对本发明保护范围的限制,尽管参照较佳实施例对本发明作了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的实质和范围。

1.rAAV2/Retro在制备针对视网膜感光细胞的递送载体中的应用,其特征在于,以rAAV2/Retro为载体,构建携带外源性目的基因或shRNA的表达载体,以所述表达载体作为递送系统,将所述目的基因或shRNA递送于视网膜感光细胞内,于视网膜感光细胞内表达外源性目的基因或敲除内源性基因;
所述载体的43bp至145bp为ITR,413bp至2152bp为病毒主要衣壳蛋白1,2169bp至4406bp为病毒次要衣壳蛋白retro,5185bp至5698bp为M13 ori,6449bp至6549bp为ampicillin启动子,6550bp至7410bp为ampicillin,7581bp至8169bp为pUC ori;
所述外源性目的基因为Cnga3,所述Cnga3的核苷酸序列如SEQ ID NO.1所示;
所述shRNA为shNrl,所述shNrl的核苷酸序列如SEQ ID NO.3所示。
