Abstract

技术领域
本发明涉及包含细胞或器官的类器官的基质组合物。更详细而言,本发明涉及由多种包含细胞等的基质形成的、具有特定结构的基质组合物。本发明还涉及这样的基质组合物的制造方法和用途。
背景技术
以针对肝脏、胰腺等各种器官的疾病的医药品开发、实现再生医疗为目标,已开展了研究开发以制作使培养细胞自组织化、重现了各个器官的三维结构、血管、胆管、其他脉管的复杂结构的细胞结构体、即器官的类器官。作为这样的器官的类器官的发展性的细胞结构体,已能够通过将包含未分化细胞的特定种类的细胞组合进行培养来制作形成于发育初期的器官原基、即器官芽(例如,作为肝脏器官芽的肝芽)。对于这样的被称为“迷你器官”的类器官等而言,较之传统的单独培养的器官的细胞等,可以用作药效或毒性的评价更贴近生物体的评价体系,还可以用于移植手术、血浆蛋白的生产等,利用价值极高。
专利文献1记载了可通过将血管内皮细胞、间充质细胞(例如间充质干细胞)和器官细胞(例如肝脏内胚层细胞、或胰腺β细胞)按规定的比例混合的细胞混合物“在被胞外基质成分支承的特殊环境下”(具体而言,例如接种至固相化于培养容器上的基质胶)进行培养来制作被称为“器官芽”(例如肝芽、或胰芽)的三维结构体。另一方面,专利文献1(尤其是实施例1和2)记载了在将细胞混合物“包埋于基质胶中”的情况下,“并未形成立体结构”。
非专利文献1记载了具有如下形态(三层梯度系统)的精巢类器官(基质组合物),其用不含细胞的第一基质包裹混合有精巢细胞的第二基质(Corning公司“基质胶”),并用不含细胞的第三基质封闭第一基质的开口部。记载于该文献中的系统由于第一和第三基质不含细胞而产生了细胞密度的梯度,促进了基于包含于第二基质中的精巢细胞的类器官的形成。然而,非专利文献1并未记载在所述系统中培养精巢细胞以外的细胞(类器官),也未记载第二基质以外的基质含有细胞。
非专利文献2记载了下述情况:在涂布于载玻片上的基质胶上(以二维方式)分别培养了(1)从人微血管内皮细胞(Microvessel endothelial cells;MVEC)和肝脏细胞制作的类器官;(2)从人脐带静脉内皮细胞(Human umbilical-vein endothelial cells;HUVEC)和肝脏细胞制作的类器官;(3)从大鼠肝窦内皮细胞(Liver sinusoidalendothelial cells;LSEC)和肝脏细胞制作的类器官;和(4)从人脐带静脉内皮细胞(HUVEC)、人皮肤成纤维细胞(Normal human dermal fibroblasts;NHDF)和肝脏细胞制作的类器官,结果,血管内皮细胞的网络在上述(2)和(3)中最终崩塌,在上述(1)和(4)中持续存在,最终形成成纤维细胞的囊并从载玻片上浮起,等等。然而,非专利文献2并未记载在(具有多层结构的)基质胶中培养如前文所述的细胞或类器官。
非专利文献3记载了在培养下层凝胶包含神经前体细胞(NPC)、上层凝胶包含NPC、星状细胞或神经元的结构体的情况下,尤其是在上层凝胶包含星状细胞时,下层凝胶的NPC向上层迁移,促进NPC分化为神经细胞。然而,非专利文献3并未公开或暗示下层凝胶“包裹”上层凝胶、下层凝胶“具有至少一个开口部”。

发明所要解决的课题
本发明的课题是提供用于制作接近于生物体内的器官、可进行血浆蛋白的分泌活动、免疫应答的器官的类器官的手段。
用于解决课题的手段
本申请的发明人发现,通过将不同种类的细胞等包埋于具有多层结构的基质的各层中,在气液边界面培养该基质多层结构体中所含的细胞等,可形成较之传统更接近于生物体内器官的类器官。具体而言,发现:制作具备下述第二基质及第一基质的基质多层结构体,所述第二基质包含构成器官的细胞(优选能形成器官的类器官的细胞)或器官的类器官自身,所述第一基质对该第二基质排除其一部分(开口部)而进行包裹,包含脉管细胞(例如血管内皮细胞、平滑肌细胞等)、神经细胞、血液细胞等,以在从其开口部向包含于第二基质中的细胞等供给氧气等的同时将第一基质和第二基质浸渍在液体培养基中的方式进行培养,由此使得包含于第一基质中的细胞形成层级式细胞网络(hierarchical cellnetwork),同时与包含于第二基质中的细胞等成为一体而形成器官的类器官。并且,发现了这样的基质多层结构体(也可以称为包含细胞或者器官的类器官和基质等的基质组合物)能够用于生产血浆蛋白和其他器官的类器官的预期目的,从而完成了本发明。
即,本发明为了解决上述课题,提供了下述[1]~[10]。
[1]基质组合物,其包含:
(1)第一基质,其包含选自由脉管细胞、神经细胞和血液细胞组成的组中的一个以上的细胞;和
(2)第二基质,其包含构成器官的细胞及/或器官的类器官,
其中,第二基质被第一基质包裹,
第一基质具有至少一个开口部。
[2]项1所述的基质组合物,其中,在基质组合物中具有层级式细胞网络。
[3]项1所述的基质组合物,其中,第一基质中的脉管细胞为造血性血管内皮细胞。

步骤(1)对项1所述的基质组合物进行培养;和
步骤(2)从所述步骤(1)培养的培养物中回收培养上清液。
发明效果
本发明提供的基质组合物具有接近于生物体内器官的层级式细胞网络(脉管结构等),也可以包含支承细胞或类器官的基质而整体视为新型的器官的类器官。通过使用这样的基质组合物,优选通过制作移植有所述基质组合物的非人嵌合动物,能够提高人的药物代谢曲线的预测、药效评价、毒性评价、药物相互作用评价的可信度等。

·血管内皮细胞
在本申请说明书中,“血管内皮细胞”为包括造血性血管内皮细胞(hemogenicendothelial cell;HEC)和非造血性血管内皮细胞(non-hemogenic endothelial cell;non-HEC)这两个概念的术语。HEC为能够产生造血干细胞(具有造血能力)的血管内皮细胞,也被称为血细胞产生型血管内皮细胞。另一方面,non-HEC为不具有这样的造血能力的血管内皮细胞。
用于本发明的血管内皮细胞可以是从生物体采集的血管内皮细胞(例如微血管内皮细胞(microvessel endothelial cells:MVEC)、肝窦内皮细胞(liver sinusoidalendothelial cells:LSEC)、脐带静脉内皮细胞(umbilical-vein endothelial cells:UVEC)等)的高纯度的细胞群,也可以是使ES细胞、iPS细胞等多能干细胞、其他具有分化为血管内皮细胞的能力的细胞分化而得的血管内皮细胞的高纯度的细胞群。上述血管内皮细胞的高纯度的细胞群包含相对于细胞群中的细胞总数而言为70%以上、80%以上、90%以上、95%以上、或99%以上的血管内皮细胞。
·造血性血管内皮细胞(HEC)
在本申请说明书中,“造血性血管内皮细胞(HEC)”意指作为HEC细胞标记物的CD34为阳性且CD73为阴性的、具有造血能力的血管内皮细胞。此外,用于本发明的HEC还可以包含其前体细胞。作为这样的前体细胞,典型地可举出存在于从细胞标记物Flk-1(CD309、KDR)阳性的血管内皮细胞的前体细胞(例如,侧板中胚层谱系细胞)分化为HEC细胞的过程中的细胞(参见Cell Reports 2,553-567,2012)。需要说明的是,Flk-1(CD309、KDR)阳性这样的分化早期阶段的前体细胞是HEC细胞和non-HEC细胞中共通的前体细胞,除非另外说明,“HEC前体细胞”也包括HEC细胞和non-HEC细胞中共通的前体细胞。

在本申请说明书中,“非造血性血管内皮细胞(non-HEC)”意指作为non-HEC的细胞标记物的CD31、CD73和CD144为阳性的、不具有造血能力的血管内皮细胞。此外,用于本发明的non-HEC可以包含其前体细胞。作为这样的前体细胞,典型地可举出存在于从细胞标记物Flk-1(CD309、KDR)阳性的血管内皮细胞的前体细胞(例如,侧板中胚层谱系细胞)分化为non-HEC细胞的过程中的细胞(参见Cell Reports 2,553-567,2012)。需要说明的是,Flk-1(CD309、KDR)阳性这样的分化早期阶段的前体细胞是HEC细胞和non-HEC细胞中共通的前体细胞,除非另外说明,“non-HEC前体细胞”也包含HEC细胞和non-HEC细胞中共通的前体细胞。
用于本发明的非造血性血管内皮细胞(non-HEC)可以是从生物体采集的non-HEC的高纯度的细胞群,也可以是使ES细胞、iPS细胞等多能干细胞、其他具有分化为血管内皮细胞的能力的细胞(例如,侧板中胚层谱系细胞)分化而获得的non-HEC的高纯度的细胞群。上述non-HEC的高纯度的细胞群包含相对于细胞群中的细胞总数而言为70%以上、80%以上、90%以上、95%以上、或99%以上的non-HEC及/或non-HEC前体细胞。在一个实施方案中,上述non-HEC的高纯度的细胞群包含相对于细胞群中的细胞总数而言为70%以上、80%以上、90%以上、95%以上、或99%以上的non-HEC。
使ES细胞、iPS细胞等多能干细胞、其他具有分化为血管内皮细胞的能力的细胞分化为非造血性血管内皮细胞(non-HEC)的方法是已知的,例如,可以基于iPS细胞,根据NatCell Biol.2015;17(8):994-1003、Cell Rep.2017;21(10):2661-2670等中记载的方法实施。
·间充质细胞
在本申请说明书中,“间充质细胞”是指主要存在于源自中胚层的结缔组织中的、形成在组织中发挥功能的细胞的支承结构的结缔组织细胞,是包括已分化的细胞(分化间充质细胞)和虽然确定了向间充质细胞分化的命运但尚未分化为间充质细胞的细胞(未分化间充质细胞)、即所谓间充质干细胞这两个概念的术语。但是,“血管内皮细胞”虽然是从未分化间充质细胞分化的一种细胞,其被排除在本申请说明书中的“间充质细胞”的定义之外。
某一细胞是未分化间充质细胞还是分化间充质细胞可以通过Stro-1、CD29、CD44、CD73、CD90、CD105、CD133、CD271、Nestin等中的1种或2种以上是否为阳性(阳性则为未分化间充质细胞、阴性则为分化间充质细胞)来判别。
间充质细胞还可以根据本发明中作为目标的器官的类器官、组合使用的“构成器官的细胞”而表达对于特定的器官(组织)具有特异性的细胞标记物。作为这样的细胞标记物,可举出例如作为横隔间充质(septum transversum mesenchyme;STM)的细胞标记物的FOXF1、COL4A和ALCAM。
肝细胞
在本申请说明书中,“肝细胞”(肝脏细胞)为肝脏的实质细胞,是包括已分化的肝细胞(分化肝细胞)和虽然确定了向肝细胞分化的命运但尚未分化为肝细胞的细胞(未分化肝细胞)、即所谓肝前体细胞(例如,肝脏内胚层细胞)这两个概念的术语。分化肝细胞可以是自生物体采集的(从生物体内的肝脏分离的)细胞,也可以是使ES细胞、iPS细胞等多能干细胞、肝前体细胞、其他具有分化为肝细胞的能力的细胞分化而获得的细胞。未分化肝细胞可以是从生物体采集的细胞,也可以是使ES细胞、iPS细胞等多能干细胞、其他的干细胞或前体细胞分化而获得的细胞。能够分化为肝细胞的细胞可以根据例如K.Si-Taiyeb等,Hepatology,51(1):297-305(2010)、T.Touboul等,Hepatology.51(5):1754-65.(2010)来制作。使ES细胞、iPS细胞等多能干细胞、肝前体细胞、其他具有分化为肝脏细胞的能力的细胞分化为肝脏细胞的方法是已知的,例如,可以基于iPS细胞,根据Hepatology,2010;51(1):297-305、Cell Rep.2017;21(10):2661-2670等中记载的方法实施。对于从生物体采集而得的细胞群、通过ES细胞、iPS细胞等的分化诱导而制作的细胞群(尤其对于后者)而言,均可以使用分化肝细胞的高纯度的细胞群或未分化肝细胞的高纯度的细胞群,也可以使用包含任意比例的分化肝细胞和未分化肝细胞的细胞混合物。
某一细胞是否为分化肝脏细胞可以通过成熟肝细胞标记物、例如去唾液酸糖蛋白受体1(ASGR1)、作为未成熟肝细胞标记物(初期肝分化标记物)的甲胎蛋白(AFP)、作为初期肝分化标记物的白蛋白(ALB)、视黄醇结合蛋白(RBP4)、转甲状腺素蛋白(TTR)、葡萄糖-6-磷酸酶(G6PC)等中的1种或2种以上的表达是否为阳性来判别。另一方面,某一细胞是否为未分化肝脏细胞可以通过HHEX、SOX2、HNF4α、AFP、ALB等中的1种或2种以上细胞标记物的表达是否为阳性来判别。
·细胞标记物
在本申请说明书中,“细胞标记物”为在规定的细胞型中特异性表达(阳性标记物)或不表达(阴性标记物)的基因,具体是指作为由基因组中的所述基因的转录而得的mRNA,或作为由其mRNA的翻译而得的蛋白质生成(阳性标记物)或不生成(阴性标记物)的物质。细胞标记物优选为能够通过荧光物质进行标记(染色),能容易地进行表达有所述细胞标记物的细胞的检测、浓缩、分离等的、表达于细胞表面的蛋白质(细胞表面标记物)。
标记物基因为“阳性”意指该基因的mRNA或蛋白质的表达量能够通过对本领域技术人员而言普通或已知的方法进行检测、或高于规定的阈值(背景水平等)。标记物基因为“阴性”意指该基因的mRNA或蛋白质的表达量不能通过对本领域技术人员而言普通或已知的方法进行检测、或低于规定的阈值(背景水平等)。
细胞标记物是阳性还是阴性可以通过对本领域技术人员而言普通或已知的方法,基于定性或定量的结果进行判定。作为蛋白质的细胞标记物可以利用使用特异于所述蛋白质的抗体的免疫学分析,例如ELISA、免疫染色、流式细胞术等进行检测或测定表达量。作为mRNA的细胞标记物可以利用使用特异于所述mRNA的核酸的分析,例如RT-PCR(包括定量PCR)、微阵列、生物芯片等进行检测或测定表达量。
-基质组合物-
本发明的基质组合物为具有如下结构的组合物:其包含(1)第一基质,其包含选自由脉管细胞、神经细胞和血液细胞组成的组中的一个以上的细胞;和(2)第二基质,其包含构成器官的细胞及/或器官的类器官,其中,第二基质被第一基质包裹,第一基质具有至少一个开口部(参见图1)。
第一基质具有的“开口部”是指没有包裹第二基质的部分。即,第二基质在“开口部”中,能够由第二基质自身直接地、或介由“替代第一基质的其他物质”而间接地自外部获取氧、营养、包含在第二基质中的细胞所必需的其他物质。如果第二基质被第一基质完全包裹,则存在由于包含于第一基质中的脉管细胞等或由其形成的网络使得第二基质中所含的构成器官的细胞等无法自外部获取足够量的氧、营养等的情况。在第一基质设置“开口部”是用于避免这样的问题的手段,只要能够实现这一目的,也可以用“替代第一基质的其他物质”填充“开口部”。
作为上述“替代第一基质的其他物质”的代表示例,可举出“不含细胞的基质”(在本申请说明书中,称为“第三基质”)。即,在一个实施方案中,本发明的基质组合物为包含含有脉管细胞等的第一基质、含有构成器官的细胞等的第二基质、和不含细胞的第三基质的基质组合物,其中,第二基质被第一基质和第三基质包裹,第一基质具有至少一个开口部,该开口部的一部分或全部被第三基质填充。
本发明中的“基质”指在用于培养构成器官的细胞的、及用于形成器官的类器官的三维细胞培养中使用的含有胞外基质(ECM)、和根据需要而含有的生长因子等添加成分的制品。已知多种这样的基质,可以根据本发明的目的选择合适的基质使用。
作为基质成分包含的ECM基本上以纤维状蛋白质和蛋白多糖作为主要成分,作为这样的成分可举出例如弹性蛋白、巢蛋白、骨粘连蛋白、胶原蛋白(IV型胶原蛋白等)、腱蛋白、血小板反应蛋白(thrombospondin)、串珠素(perlecan)、波连蛋白、原纤维蛋白、纤连蛋白、肝素(硫酸盐)蛋白多糖、层粘连蛋白等。
作为可根据需要向基质添加的生长因子,可举出例如EGF、PDGF、IGF-1、TGF-β、VEGF等。
上述的含有ECM和生长因子的基质已有市售,可举出例如“BD基质胶(BDMatrigel)”(BD Biosciences)、基质胶(Matrigel)(Corning)等。
本领域技术人员可以根据发明的目的适当调整基质的组成、浓度(原液、或稀释物)。例如,基质胶可根据需要与液体培养基混合使用,使其操作性提高,并且在固化时具有适当的硬度。作为这样的实施方案中的用于与基质胶混合的液体培养基,可以使用本说明书中另外记载的与基质组合物的制造方法中的、用于浸渍基质组合物并进行培养的液体培养基相同的培养基。
需要说明的是,基质胶通常在低温(例如4℃以下)下为流体,而在常温以上(例如37℃以上)固化成为凝胶。包含规定的细胞等的基质通常是在通过将流体状态的基质胶与规定的细胞等混合后升温并静置以使其固化而获得的。
进一步地,作为水凝胶而为人所知的自组织化肽、例如含有精氨酸、甘氨酸和天冬酰胺的水凝胶等也可以作为本发明中的基质成分使用。在使用包含这样的水凝胶的基质的情况下,添加使其固化时所必需的离子型溶液或离子型分子即可。
本发明中的第一、第二(以及任选使用的第三)基质可以是相同的制品,也可以是彼此不同的制品。例如,根据添加到基质中的生长因子、基质的硬度,包含在基质中的细胞的分化、器官的类器官(例如器官芽)的形成可能发生变化,因此可以根据发明的实施方案使用合适的基质。
·第一基质
第一基质包含选自由脉管细胞、神经细胞和血液细胞组成的组中的1种以上的细胞。这些细胞来源的物种没有特别限定,可以是人,也可以是人以外的动物,例如小鼠、大鼠、狗、猪、猴等哺乳动物。
作为脉管细胞,可举出例如血管内皮细胞、血管平滑肌细胞和周细胞。
例如,基于能够赋予包含于第二基质中的肝脏、胰腺、其他器官的类器官层级式微血管结构、适合作为血管类疾病(例:窦壁阻塞综合征)的模型的观点,血管内皮细胞作为包含于第一基质中的细胞(脉管细胞)是优选的。此外,由于在形成层级式微血管结构的同时,还能够从其产生血液细胞(例如巨噬细胞、嗜中性细胞等免疫活性细胞),因此血管内皮细胞中的造血性血管内皮细胞是更优选的。
本发明中使用的HEC可以是从生物体采集的HEC的高纯度的细胞群,也可以是使ES细胞、iPS细胞等多能干细胞、其他具有分化为血管内皮细胞的能力的细胞(例如,侧板中胚层谱系细胞)分化而获得的HEC的高纯度的细胞群。需要说明的是,使ES细胞、iPS细胞等多能干细胞、其他具有分化为血管内皮细胞的能力的细胞分化为造血性血管内皮细胞(HEC)的方法是已知的,例如,可以基于iPS细胞,根据PLoS One,2013;8(4):e59243、NatBiotechnol.2014;32(6):554-61、Sci Rep.2016;6:35680等中记载的方法实施。HEC的“高纯度的”细胞群包含相对于细胞群中的细胞总数而言为70%以上、80%以上、90%以上、95%以上、或99%以上的HEC及/或HEC前体细胞。
作为神经细胞,可举出例如GABA能神经元、多巴胺能神经元和运动神经元。
血液细胞(血细胞谱系细胞)可以大致分为血小板、红细胞和白细胞。作为白细胞,可举出淋巴细胞(NK细胞、T淋巴细胞、B淋巴细胞、巨噬细胞、树突状细胞)、单核细胞、嗜酸性细胞、嗜碱性细胞、嗜中性细胞。例如,基于能够制造可用作炎性疾病等病况模型的基质组合物的观点,巨噬细胞、嗜中性细胞等免疫活性细胞作为包含于第一基质中的细胞(血细胞)是优选的。
第一基质的硬度可以考虑例如由气相提供的氧、由液体培养基提供的成分等物质的透过性,对其他包含于第一基质中的细胞的生长产生的作用等而进行适当调节,例如,可以在0.05~50kPa的范围内。需要说明的是,这样的“硬度”指标可以适当转换为用培养基稀释基质胶时的倍率、或基质中的ECM的浓度等。此外,也可以考虑上述相同的事项而适当调节第一基质的厚度、与第二基质的接触面的面积等。
·第二基质
第二基质包含构成器官的细胞及/或器官的类器官。这些细胞或类器官来源的物种没有特别限定,可以是人,也可以是人以外的动物,例如小鼠、大鼠、狗、猪、猴等哺乳动物。
第二基质包括例如下述(i)~(iii)的实施方案:
(i)构成器官的细胞、特别是能够形成器官的类器官的1种或2种以上细胞,以尚未形成器官的类器官的状态(例如通过本发明的基质组合物的制造方法的步骤(a))包含于第二基质中;
(ii)已预先另行形成的器官的类器官(例如通过本发明的基质组合物的制造方法的步骤(a))包含于第二基质中;
(iii)能够形成器官的类器官的1种或(至少包括“构成器官的细胞”的)2种以上细胞(例如通过本发明的基质组合物的制造方法的步骤(a))包含于第二基质中,对其(例如通过本发明的基质组合物的制造方法的步骤(c))进行培养而形成的器官的类器官包含于第二基质中。
·构成器官的细胞
本发明中的“构成器官的细胞”包含(I)构成器官的实质细胞和(II)构成器官的非实质细胞。此外,(I)实质细胞和(II)非实质细胞分别包含(i)已分化成熟的、或已达到终末分化的、具有作为实质细胞或非实质细胞的规定功能性的细胞(本说明书中简称为“分化细胞”),以及(ii)具有分化为实质细胞或非实质细胞的能力的、或者已确定分化命运(已定向)但尚未分化的、或者处于干细胞或前体细胞的阶段而尚未充分具有作为实质细胞或非实质细胞的规定功能性的细胞(本说明书中简称为“未分化细胞”)。
作为构成器官的细胞可使用选自由实质细胞的分化细胞、实质细胞的未分化细胞、非实质细胞的分化细胞和非实质细胞的未分化细胞组成的组中的至少1种细胞,优选能够形成“器官的类器官”(例如器官芽)的2种以上细胞的组合。各种器官的类器官(例如器官芽)的形成所需的细胞的种类、培养条件是已知的,在本发明(第二基质)中也是同样。
作为“器官”可举出例如肝脏、胰腺、肾脏、心脏、肺、脾脏、食道、胃、甲状腺、甲状旁腺、胸腺、生殖腺、脑、脊髓等。
作为构成器官的“实质细胞”可举出例如肝脏的肝细胞、胰腺的内分泌细胞(例如α细胞、β细胞、δ细胞、ε细胞、PP细胞)和胰管上皮细胞、肾脏的肾小管上皮细胞和肾小球上皮细胞、肺的肺胞上皮细胞、心脏的心肌细胞、肠道的上皮细胞、脑的神经细胞和神经胶质细胞、脊髓的神经细胞和施旺氏细胞等。
作为构成器官的“非实质细胞”可举出例如肝脏的肝窦内皮细胞、肝星状细胞和枯否细胞、胰腺的胰星状细胞和胰微血管内皮细胞、肾脏的肾小球内皮细胞、肺的肺动脉内皮细胞和肺成纤维细胞、心脏的心微血管内皮细胞、大动脉内皮细胞、冠状动脉内皮细胞和心成纤维细胞、肠道的肠道微血管内皮细胞、脑的脑微血管内皮细胞、血管周细胞、脉络丛内皮细胞和脑血管外膜成纤维细胞等。
作为具有分化为构成器官的实质细胞或非实质细胞的能力的“未分化器官细胞”可举出例如能分化为脑、脊髓、肾上腺髓质、表皮、毛发·指(趾)甲·皮肤腺、感觉器官、外周神经、晶状体等外胚层器官的细胞;能分化为肾脏、输尿管、心脏、血液、生殖腺、肾上腺皮质、肌肉、骨骼、真皮、结缔组织、中皮等中胚层器官的细胞;能分化为肝脏、胰腺、肠道、肺、甲状腺、甲状旁腺、尿路等内胚层器官的细胞;等。
·器官的类器官
“器官的类器官”是人为创造的与器官类似的组织体(三维结构体),已知各种器官的类器官,例如肝脏、胰腺、肾脏、心脏、肺、脾脏、食道、胃、甲状腺、甲状旁腺、胸腺、生殖腺、脑、脊髓等的类器官(参见例如https://www.nejm.org/doi/pdf/10.1056/NEJMra1806175、https://www.nature.com/articles/s41568-018-0007-6、http://www.amsbio.com/brochures/organoid-culture-handbook.pdf)。本发明中,任意器官的类器官均可包含于第二基质中进行使用。需要说明的是,“器官的类器官”还包含作为最终会达到具有复杂构成的器官的初期阶段结构体的“器官芽”(例如肝芽、胰芽)。
作为本发明中的器官的类器官的代表示例,可举出“肝脏类器官”。肝脏类器官的制作方法是已知的,作为本发明中的肝脏类器官,可使用通过已知的各种制作方法获得的肝脏类器官。
肝脏类器官优选为“肝芽”。肝芽的制作方法也是已知的,例如前述专利文献1所记载的,可以基于肝脏内胚层细胞、血管内皮细胞和间充质干细胞的组合制作肝芽。需要说明的是,肝脏内胚层细胞相当于本说明书中的实质细胞的未分化细胞,血管内皮细胞和间充质干细胞相当于本说明书中的非实质细胞的未分化细胞。作为用于制作肝脏类器官(例如肝芽)的血管内皮细胞,优选非造血性血管内皮细胞(non-HEC)。
在本发明的一个实施方案中,选择肝脏类器官或肝芽作为器官的类器官时,或者选择肝脏内胚层细胞、血管内皮细胞和间充质干细胞的组合、或其他用于制作肝脏类器官或肝芽的细胞的组合作为包含于第二基质中的细胞时,作为包含于第一基质中的细胞,可以选择例如脉管细胞,优选血管内皮细胞,更优选造血性血管内皮细胞(HEC)。
作为本发明中的器官的类器官的代表示例之一,可举出“胰腺类器官”。胰腺类器官的制作方法是已知的,作为本发明中的胰腺类器官,可使用通过已知的各种制作方法获得的胰腺类器官。
胰腺类器官优选为“胰芽”。胰芽的制作方法也是已知的,例如前述专利文献1所记载的,可以基于胰腺β细胞、血管内皮细胞和间充质干细胞的组合制作胰芽。需要说明的是,胰腺β细胞相当于本说明书中的实质细胞的分化细胞,血管内皮细胞和间充质干细胞相当于本说明书中的非实质细胞的未分化细胞。
本发明中,可以基于与各种器官的类器官的制作方法相关的已知信息(参见例如http://www.amsbio.com/brochures/organoid-culture-handbook.pdf),制作可埋入第二基质中的类器官。需要说明的是,这样的制作方法也可以用作从埋入第二基质中的细胞形成各种器官的类器官时的、本发明特有的构成以外的基本方法。
第二基质的硬度可以考虑例如由气相提供的氧、由液体培养基提供的成分等物质的透过性,对其他包含于第二基质中的细胞或类器官的生长产生的作用等而进行适当调节,例如,可以在0.05~50kPa的范围内。需要说明的是,这样的“硬度”指标可以适当转换为用培养基稀释基质胶时的倍率、或基质中的ECM的浓度等。此外,也可以考虑上述相同的事项而适当调节第二基质的厚度、与第一基质(以及根据需要设置的第三基质)的接触面的面积等。
·第三基质
为了使包含在第二基质中的细胞有效获取透过第三基质的氧等,根据需要使用的第三基质不包含细胞、类器官等。但是,在能够实现上述功能的范围内,允许含有例如影响包含于第一基质中的细胞或包含于第二基质中的细胞及/或器官的类器官的培养的物质。作为一个示例,可举出下述情况:当第一基质中包含脉管细胞(血管内皮细胞等)时,使处于夹持第二基质而与第一基质彼此相对的位置的第三基质含有VEGF,使得从脉管细胞形成的血管更易于向第二基质(所包含的细胞)的方向伸长。
第三基质的硬度可以考虑例如氧和其他物质的透过性等而进行适当调节,例如,可以在0.05~50kPa的范围内。需要说明的是,这样的“硬度”指标可以适当转换为用培养基稀释基质胶时的倍率、或基质中的ECM的浓度等。此外,也可以考虑上述相同的事项而适当调节第三基质的厚度、与第二基质的接触面的面积等。
本发明的基质组合物优选在其中具有“层级式细胞网络”。“层级式细胞网络”意指例如如下状态(参见图2):针对从血管内皮细胞构建的血管网来看,第二基质内的器官的类器官中包含的血管内皮细胞组成的缺乏方向性(无序地延伸)的血管网,并另行形成了由第一基质中包含的血管内皮细胞组成的具有方向性的血管网,就基质组合物整体而言,形成了前者与后者的“层级式”的细胞网络(血管网)。
如上所述的本发明的基质组合物可以是通过任何制造方法获得的组合物。作为优选的制造方法,可举出以下记载的本发明的基质组合物的制造方法。
-基质组合物的制造方法-
本发明的基质组合物的制造方法至少包括下述步骤(a)和(b),根据需要还包括下述步骤(c):
步骤(a),将构成器官的细胞及/或器官的类器官悬浮于流体形态的第二基质中,然后使第二基质固化;
步骤(b),将选自由脉管细胞、神经细胞、和血液细胞组成的组中的细胞悬浮于流体形态的第一基质中,通过第一基质以第一基质具有至少一个开口部的方式包覆第二基质,然后使第一基质固化;
步骤(c),将通过所述步骤(a)和(b)获得的基质组合物在悬架于透气性膜的状态下,在确保通过该透气性膜的气体交换的同时,浸渍在液体培养基中进行培养,由此使第二基质中的构成器官的细胞形成立体结构。
关于步骤(a),在流体形态的第二基质中悬浮构成器官的细胞等的方法没有特别限定。例如,在使用基质胶与培养基的混合物作为基质的情况下,通过在基质胶成为流体的低温(例如4℃)下混合基质胶与培养基,并将该混合液与构成器官的细胞等混合,获得了用于步骤(a)的悬浮液。
将用于步骤(a)的悬浮液中包含的第二基质进行固化的方法没有特别限定。例如,使用基质胶与培养基的混合物作为基质制备用于步骤(a)的悬浮液时,通过升温至所述基质胶进行固化(凝胶化)的温度(例如37℃),可获得所述悬浮液的固化物。
关于步骤(b),将脉管细胞等悬浮在流体形态的第一基质中的方法、和使第一基质固化的方法都没有特别限定,可以分别与上述步骤(a)中的悬浮方法和固化方法同样地实施。
作为“利用第一基质以第一基质具有至少一个开口部的方式包覆第二基质,然后使第一基质固化”的方法,可举出例如首先将包含规定的细胞的第二基质滴至透气性膜上使其固化,然后将包含规定的细胞等的第一基质滴至该已固化的第二基质上使其固化的方法。上述第二基质与透气性膜的接触部分未被上述第一基质覆盖,相当于开口部。
此外,在前述使用第三基质的实施方案中,可举出例如首先将不含细胞的第三基质直接滴至透气性膜上使其固化,然后将包含规定的细胞的第二基质滴至该已固化的第三基质上使其固化,最后将包含规定的细胞等的第一基质滴至该已固化的第二基质上使其固化的方法。上述第三基质填充了上述第一基质的开口部。
关于步骤(c),“透气性膜”为至少具有透氧性、根据需要还具有透过二氧化碳、其他期望的气体的性质的膜,各种透气性膜都是已知的。作为透气性膜,可举出例如用聚对苯二甲酸乙二醇酯(PET)、聚二甲基硅氧烷(PDMS)、碳氟化合物、聚四氟乙烯(PTFE)、聚氨酯等纤维制作的膜。透气性膜根据需要可实施提高或降低细胞附着性的表面处理,例如用胶原蛋白等ECM进行包被的处理。此外,透气性膜根据需要可以是将用不同于透气性膜的纤维制作而得的多孔膜(网)层叠得到的膜(杂化膜)。
处于“悬架于透气性膜的状态”的基质组合物可以通过下述方式制得:如前所述,经由包括将包含规定的细胞等的第二基质滴至透气性膜上(或预先滴加第三基质)等的步骤(a)和(b)获得附着于透气性膜的状态的固化的基质组合物,并使基质组合物朝向下方(上下翻转)。将处于该状态的基质组合物使用保持件等合适的构件浸渍在容纳于培养容器中的液体培养基中,另一方面,透气性膜不进行浸渍而置于空气中(或所期望的培养气氛中),由此能够实现“在确保通过透气性膜的气体交换的同时,浸渍在液体培养基中进行培养”。培养时间可以是足以“使第二基质中的构成器官的细胞形成立体结构”的时间,优选足以形成基于第一基质中的脉管细胞等的层级式结构的时间。
用于步骤(c)的液体培养基只要适合培养包含在第一基质中的脉管细胞等和包含在第二基质中的构成器官的细胞及/或器官的类器官即可。本领域技术人员可根据进行培养的细胞或器官的类器官选择合适的种类和量的基础培养基以及根据需要使用的添加物,制备将它们混合而得的培养基。此外,由于第一基质和第二基质包含多种(至少2种)细胞,因此,可以使用将用于培养各细胞的基础培养基和必需的添加物以合适比例混合而得的培养基。尤其是在第二基质中包含用于形成器官的类器官(例如肝芽)的多种细胞的情况下、或者包含已形成的器官的类器官的情况下,使用适合该器官的类器官的形成及/或生长的组成的培养基是恰当的。
作为用于血管内皮细胞的基础培养基,可举出例如DMEM/F-12(Gibco)、Stempro-34SFM(Gibco)、Essential 6培养基(Gibco)、Essential 8培养基(Gibco)、EGM(Lonza)、BulletKit(Lonza)、EGM-2(Lonza)、BulletKit(Lonza)、EGM-2MV(Lonza)、VascuLife EnGSComp Kit(LCT)、Human Endothelial-SFM Basal Growth Medium(Invitrogen)、人微血管内皮细胞生长培养基(TOYOBO)等。作为用于血管内皮细胞的添加物,可举出例如由B27Supplements(GIBCO)、BMP4(骨形成因子4)、GSKβ抑制剂(例如CHIR99021)、VEGF(血管内皮细胞生长因子)、FGF2(成纤维细胞生长因子(也称为bFGF(碱性成纤维细胞生长因子))、Folskolin、SCF(干细胞因子,Stem Cell Factor)、TGFβ受体抑制剂(例如SB431542)、Flt-3L(Fms相关酪氨酸激酶3配体)、IL-3(白介素3)、IL-6(白介素6)、TPO(血小板生成素)、hEGF(重组人上皮细胞生长因子)、氢化可的松、抗坏血酸、IGF1、FBS(胎牛血清)、抗生素(例如,庆大霉素、两性霉素B)、肝素、L-谷氨酰胺、酚红和BBE组成的组中的1种以上。
作为用于肝细胞的基础培养基,可举出例如RPMI(Fujifilm)、HCM(Lonza)等。作为用于肝细胞的添加物,可举出例如选自由Wnt3a、激活素A、BMP4、FGF2、FBS、HGF(肝细胞生长因子)、抑瘤素M(OSM)和地塞米松(Dex)组成的组中的1种以上。用于肝细胞的培养基可根据需要添加选自抗坏血酸、BSA-FAF、胰岛素、氢化可的松和GA-1000中的至少1种。更具体而言,作为用于肝细胞的培养基,可举出例如从HCM BulletKit(Lonza)中去除hEGF(重组人上皮细胞生长因子)而得的培养基;向RPMI1640(Sigma-Aldrich)中添加1%B27 Supplements(GIBCO)和10ng/mL hHGF(Sigma-Aldrich)而得的培养基。尤其是在制作肝芽的情况下,还可以使用例如向将EGM BulletKit(Lonza)与去除了hEGF(重组人上皮细胞生长因子)的HCMBulletKit(Lonza)按1:1混合而得的培养基中添加地塞米松、抑瘤素M和HGF而得的培养基。
作为用于胰腺β细胞的基础培养基,可举出例如CMRL1066(Corning)、MCDB 131(GIBCO)等。作为用于胰腺β细胞的添加物,可举出例如Alk5抑制剂II、T3(L-3,30,5-三碘甲状腺原氨酸)、Trolox、N乙酰半胱氨酸、R428(AXL抑制剂)、肝素等。更具体而言,作为用于胰腺β细胞的培养基,可举出例如向CMRL基础培养基中添加10%FBS、10μM Alk5抑制剂II、1μMT3而得的培养基。尤其是在制作胰腺类器官的情况下,还可以使用例如向将EGM BulletKit(Lonza)与DMEM/F-12(Gibco)按1:1混合而得的培养基中添加1%B27 Supplements(GIBCO)、0.2%BSA和10μM烟酰胺获得的培养基。
收容液体培养基的培养容器可以使用与基质组合物的形状、大小等相对应的容器,例如具备多个孔的平板。培养容器优选用基质不会附着于表面的材料制作、或实施了表面处理的容器,例如,可以使用市售的低吸附培养容器。
可根据第二基质中的构成器官的细胞的组成适当调节步骤(c)的培养条件(气氛、培养温度、培养时间等),使立体结构(或器官芽)形成。例如,步骤(c)的培养可以于5%CO2、30~40℃(优选约37℃)实施1~10天(例如肝芽则为1~3天)。
可以通过肉眼或显微镜观察来确认立体结构(三次元结构)的形成。此外,对于类器官的制作而言,除了立体结构的形成以外,可以通过规定的细胞标记物、尤其是器官的实质细胞的标记物是否呈阳性、更优选通过这些标记物的蛋白质是否被分泌至培养上清液中来进行判别。例如,如果为肝的类器官或肝芽,可以将HHEX、SOX2、AFP、ALB、HNF4α等标记物的表达是否呈阳性、ALB(白蛋白)是否被分泌至培养上清液中作为判别基准。如果为胰腺的类器官或胰芽,可以将PDX1、SOX17、PSX9等标记物的表达是否呈阳性作为判别基准。
在本发明的其他方面,本发明的基质组合物的制造方法可以变形为在步骤(c)中使用“物质透过性膜”替代“透气性膜”的实施方案。即,可以是下述技术方案:第二基质通过与浸渍有凸状基质的液体培养基(在本说明书中称为“第一液体培养基”)不同的液体培养基(在本说明书中称为“第二液体培养基”)和“物质透过性膜”获取营养物、其他添加成分、以及溶解于第二液体培养基中的氧等气体,以此取代直接地或介由根据需要设置的第三基质而间接地通过暴露于气相中的“透气性膜”获取氧、二氧化碳、其他气体。在此情况下,合适的是使第二液体培养基介由物质透过性膜仅与第二基质或第三基质进行接触(物质交换),而非介由物质透过性膜与第一液体培养基进行接触(物质交换),换言之,使第二基质或第三基质覆盖“物质交换膜”的整个面。
“物质透过性膜”为在阻断细胞通过的同时确保了所期望的物质的透过性的膜。这样的物质透过性膜是已知的,可举出例如胶原蛋白制或聚二甲基硅氧烷(PDMS)制的膜。
在上述其他方面,本发明的基质组合物的制造方法的步骤(c)可以变更为例如下述步骤(c’):
步骤(c’),将通过所述步骤(a)和(b)获得的基质组合物在接触物质透过性膜的状态下,在确保与通过了物质透过性膜的第二液体培养基中的物质交换的同时,浸渍在第一液体培养基中进行培养,由此使第二基质中的构成器官的细胞形成立体结构。
进一步地,在下述的本发明的系统中,可以将步骤(c)涉及的记载更换为步骤(c’)涉及的记载。
-非人嵌合动物的制作方法-
本发明的非人嵌合动物的制作方法包括将本发明的基质组合物或通过本发明的基质组合物的制造方法获得的基质组合物移植至非人动物中,使其分化为组织和器官。
通过包括步骤(c)的本发明的基质组合物的制造方法获得的基质组合物形成于透气性膜上,在向非人动物移植时,剥去该透气性膜,仅移植基质组合物即可。或者,可以使用在生物体内分解的物质作为透气性膜。
作为非人动物可举出例如小鼠、兔、猪、狗、猴等。
本发明的非人嵌合动物为通过上述制造方法获得的非人嵌合动物,即,具有从所移植的本发明的基质组合物或通过本发明的基质组合物的制造方法获得的基质组合物分化而得的组织和器官的非人动物。
本发明的非人嵌合动物可以用于例如人的药物代谢曲线的预测、药效评价、毒性评价、药物相互作用评价等评价药剂的方法。
-培养系统-
本发明的系统为用于实施本发明的基质组合物的制造方法、尤其是用于实施步骤(c)的系统,其具备悬架于透气性膜的状态的基质组合物、和浸渍有该基质组合物的状态的液体培养基。
本发明的系统所规定的“悬架于透气性膜的状态的基质组合物”和“浸渍有基质组合物的状态的液体培养基”分别为本说明书中前述的物质,可以适用本发明的基质组合物的制造方法等的相关记载相同的技术事项。
本发明的系统除上述构成要素以外,还可以具备对本发明的实施而言必需或优选的构成要素,例如,用于保持悬架有基质组合物的状态的透气性膜的器具、用于收容液体培养基的器具(例如具备孔的平板)、用于在合适的气氛和温度下进行培养的培养装置等。
-血浆蛋白的制造方法-
本发明的血浆蛋白的制造方法包括下述步骤(1)和(2):
步骤(1),对本发明的基质组合物进行培养;和
步骤(2),从所述步骤(1)培养的培养物中回收培养上清液。

步骤(1)的培养方法中的其他实施方案、例如培养基的组成等可以以本发明的“基质组合物的制造方法”中的步骤(c)的培养方法中的其他实施方案为基准。
在本发明的一个实施方案中,步骤(1)中的培养基是血清浓度已调节为合适的范围的培养基,优选血清浓度为0%(即不添加血清的培养基)~5%的培养基。需要说明的是,作为“血清”,可以使用“血清替代物”,该情况下的血清替代物的浓度范围可对应上述血清的浓度范围。
在本发明的一个实施方案中,步骤(1)中的培养基为添加有Nrf2(核因子E2相关因子2,nuclear factor erythroid-2-related factor 2)激活剂的培养基。作为Nrf2激活剂,可举出例如R-α-硫辛酸、叔丁基氢醌、萝卜硫素、含有白藜芦醇的虎杖(Polygonumcuspidatum)提取物、BM(甲基巴多索隆(Bardoxolone methyl))等。Nrf2激活剂的添加量可以由本领域技术人员适当确定,优选为20nM以上。
步骤(1)的培养条件(气氛、培养温度、培养时间等)可以以能够在步骤(2)中回收所期望的培养上清液的方式适当调节,例如,可以是于5%CO2、30~40℃(优选约37℃)通常进行10~50天。需要说明的是,步骤(1)中的截止至回收培养上清液的培养时间通常长于步骤(c)中的截止至形成器官的类器官(例如器官芽)的培养时间,可以到血浆蛋白的产量(培养上清液中的浓度)达到所期望的水平为止。
凝血因子、补体、其他血浆蛋白的检测、定量可以通过已知的方法,例如ELISA法(酶联免疫吸附测定法)实施,用于此的试剂盒也已有市售[例如Human CoagulationFactor II(F2)ELISA Kit(Biomatik)、Human Coagulation Factor V(F5)ELISA Kit(Biomatik)、Human Coagulation Factor VII(F7)ELISA Kit(Biomatik)、HumanCoagulation Factor VIII(F8)ELISA Kit(Biomatik)、Human Coagulation Factor IX(F9)ELISA Kit(Biomatik)、Human Coagulation Factor X(F10)ELISA Kit(Biomatik)、Human Coagulation Factor XI(F11)ELISA Kit(Biomatik)、High Sensitive HumanAlbumin(ALB)ELISA Kit(Biomatik)、(C3):Complement C3 Human ELISA kit(Abcam)、(C5):Complement C5 Human ELISA kit(Abcam)、Human Factor H ELISA Kit(Abcam)、Human Factor B ELISA Kit(Abcam)]。
通过本发明的制造方法,可以制造可用于各种疾病的治疗等的血浆蛋白。更具体而言,通过本发明的血浆蛋白的制造方法的步骤(2),可以回收以如下浓度包含例如凝血因子、补体、α1抗胰蛋白酶(AAT)、溶酶体酶等血浆蛋白的培养上清液,所述浓度就用于制造针对起因于各血浆蛋白的缺损、缺乏、异常、活性降低等疾病的治疗用组合物而言是优选的。由此获得的培养上清液的毒性(例如急性毒性、慢性毒性、遗传毒性、生殖毒性、心毒性、癌原性)低,可以直接进行制剂化,或加入用于医药品的添加剂而进行制剂化。
作为与凝血因子相关的疾病(凝血病)的具体例子,可举出血友病(A型血友病(第VIII因子缺乏症)、B型血友病(第IX因子缺乏症))、血友病相关疾病(第II因子缺乏症、第V因子缺乏症、第VII因子缺乏症、第X因子缺乏症、第XI因子缺乏症)、血栓病(抗凝血酶缺乏症)。
作为与补体相关的疾病(补体异常疾病)的具体例子,可举出补体第3成分缺乏症、补体第5成分缺乏症、B因子缺乏症、H因子缺乏症。
作为与α1抗胰蛋白酶(AAT)相关的疾病的具体例子,可举出AAT缺乏症。

为了进行上述制剂化,本发明的血浆蛋白的制造方法可根据需要还包括步骤(3)浓缩前述步骤(2)中回收的培养上清液。经浓缩的培养上清液优选例如基于活化部分凝血酶原时间(activated partial thromboplastin time:APTT)的活性升高(缩短APTT)等作为凝血及/或补体异常疾病的治疗用组合物的用途,或者由于经浓缩而以高浓度含有多种酶,而优选作为溶酶体病(例如II型粘脂贮积病(mucolipidosis)和III型粘脂贮积病)的治疗用组合物的用途。
实施例
[实施例1]制作包含造血性血管内皮细胞和肝脏构成细胞的基质组合物
[实验方法]
[1]制作人非造血性血管内皮细胞
通过将人iPS细胞(1383D2;京都大学iPS研究所)在向DMEM/F-12(Gibco)(10ml)中添加1%B-27Supplements(GIBCO)、BMP4(25ng/ml)和CHIR99021(8μM)而得的培养基中于5%CO2、37℃培养3天,诱导中胚层谱系细胞。将获得的中胚层细胞进一步在向Stempro-34SFM(Gibco)(10ml)中添加VEGF(200ng/ml)和Folskolin(2μM)而得的培养基中于5%CO2、37℃培养7天,得到CD31阳性、CD73阳性和CD144阳性的人非造血性血管内皮细胞群。
[2]制作人造血性血管内皮细胞
通过将人iPS细胞(625A4;京都大学iPS研究所)在AK02N(味之素)(8ml)中于5%CO2、37℃培养6~7天,使其形成直径500-700μm的iPS细胞集落。将得到的集落在向Essential 8培养基(Gibco)(8ml)中添加BMP4(80ng/ml)、VEGF(80ng/ml)和CHIR99021(2μM)而得的培养基中于5%CO2、37℃培养2天。然后,通过更换为向Essential 6培养基(Gibco)(8ml)中添加VEGF(80ng/ml)、FGF2(25ng/ml)、SCF(50ng/ml)和SB431542(2μM)而得的培养基于5%CO2、37℃再培养2天,诱导侧板中胚层谱系细胞。之后,更换为向Stempro-34SFM(Gibco)(8ml)中添加VEGF(80ng/ml)、SCF(50ng/ml)、Flt-3L(50ng/ml)、IL-3(50ng/ml)、IL-6(50ng/ml)和TPO(5ng/ml)而得的培养基,于5%CO2、37℃培养2天。进一步更换为自上述组成的培养基中去除VEGF而得的培养基,于5%CO2、37℃培养1天,由此获得CD34阳性、CD73阴性的造血性血管内皮细胞。
[3]制作人肝脏内胚层细胞
通过将人iPS细胞(1383D2;京都大学iPS研究所)在向基础培养基RPMI(Fujifilm)(2ml)中添加Wnt3a(50ng/mL)和激活素A(100ng/ml)而得的培养基中于5%CO2、37℃培养5天,诱导内胚层谱系细胞。向所述基础培养基中添加1%B27 Supplements(GIBCO)和FGF2(10ng/ml),通过将得到的内胚层谱系细胞于5%CO2、37℃再培养5天,获得AFP、ALB和HNF4α为阳性的人肝脏内胚层细胞群。
[4]制作人间充质细胞
通过将人iPS细胞(1383D2;京都大学iPS研究所)在向基础培养基DMEM/F-12(Gibco)(10ml)中添加1%B-27Supplements(GIBCO)、BMP4(25ng/ml)和CHIR99021(8μM)而得的培养基中于5%CO2、37℃培养3天,诱导中胚层谱系细胞。向相同培养基中添加PDGFBB和激活素A,将得到的中胚层谱系前体细胞于5%CO2、37℃再培养3天。回收培养后的细胞,再次接种至新的明胶包被平板上,向所述基础培养基中添加bFGF和BMP4,于5%CO2、37℃再培养4天,由此获得FOXF1、COL4A、ALCAM和CD73为阳性的人间充质细胞群。
[5]细胞向细胞外基质的包埋和基质组合物的制作
制备将向HCM(Lonza公司制)中加入FBS、HGF、OSM和Dex而得的肝诱导培养基及EGM(Lonza)按1:1的体积比例混合而得的培养基(本说明书中称为“类器官用培养基”)。进一步制备将类器官用培养基以及基质胶(BD Pharmingen)按1:1的体积比例混合而得的基质(本说明书中称为“类器官用培养基/基质胶混合基质”)。
将类器官用培养基/基质胶混合基质5μL滴至翻转的PET Millicell HangingCell Culture Inserts(Millipore公司)上,通过升高温度至37℃使其固化,形成本发明中的“第三基质”。
将如上所述制作的人肝脏内胚层细胞、人非造血性内皮细胞和人间充质细胞按10:7:1的比例在类器官用培养基/基质胶混合基质内于4℃混合。将得到的混合液3μL滴至如上所述地固化的第三基质上,通过升高温度至37℃使其固化,形成本发明中的“第二基质”。
进一步地,将如上所述地制作的人造血性血管内皮细胞在类器官培养基/基质胶混合基质内于4℃混合。将得到的混合液8μL滴至如上所述地固化的第二基质上,通过升高温度至37℃使其固化,形成本发明中的“第一基质”。
向低吸附24孔板中加入600μL添加有VEGF(80ng/ml)、SCF(50ng/ml)、Flt-3L(50ng/ml)、IL-3(50ng/ml)、IL-6(50ng/ml)和TPO(5ng/ml)的类器官培养基。朝向孔内的培养基方向插入如上所述地固化于悬滴插入物(hanging drop insert)的、包含第一~第三基质的基质组合物。截止至培养开始后的第四天,按每天更换一半量来实施培养基更换,之后,每2~3天实施一次更换。经时性地采集培养至约21~42天为止的培养液,测定凝血因子和补体因子的产量。
[结果]
培养第21天的培养液中所含的凝血因子(第VIII因子)的产量、以及补体因子(补体第3成分、补体第5成分、B因子、H因子)的产量如表1所示。

Claims (8)
1.基质组合物,其包含:
(1)第一基质,其包含选自由脉管细胞、神经细胞和血液细胞中的一种以上组成的组中的一个以上的细胞,所述一个以上的细胞没有形成具有方向性的细胞网络;和
(2)第二基质,其包含构成器官的细胞及/或器官的类器官,
其中,第二基质被第一基质包裹,
第一基质具有至少一个用于供给氧气的开口部,所述开口部为没有包裹第二基质、并且被不含细胞的基质填充的部分。
2.如权利要求1所述的基质组合物,其中,第一基质中的脉管细胞为造血性血管内皮细胞。
3.如权利要求1所述的基质组合物,其中,第二基质中的构成器官的细胞为肝脏内胚层细胞、血管内皮细胞和间充质干细胞的组合,或胰腺β细胞、血管内皮细胞和间充质干细胞的组合。
4.基质组合物的制造方法,其包括:
步骤(a),将构成器官的细胞及/或器官的类器官悬浮于流体形态的第二基质中,然后使第二基质固化;和
步骤(b),将选自由脉管细胞、神经细胞和血液细胞中的一种以上组成的组中的细胞悬浮于流体形态的第一基质中,利用第一基质以该第一基质具有至少一个用于供给氧气的开口部的方式包覆第二基质,所述开口部为没有包裹第二基质、并且被不含细胞的基质填充的部分,然后使第一基质固化。

7.培养系统,其具备:
悬架于透气性膜的状态的基质组合物;和
浸渍有该基质组合物的状态的液体培养基,其中,
该基质组合物包含:
(1)第一基质,其包含选自由脉管细胞、神经细胞和血液细胞中的一种以上组成的组中的一个以上的细胞,所述一个以上的细胞没有形成具有方向性的细胞网络;和
(2)第二基质,其包含构成器官的细胞及/或器官的类器官,
其中,所述第二基质被该第一基质包裹,该第一基质具有至少一个开口部,所述开口部为没有包裹第二基质、并且被不含细胞的基质填充的部分,
所述透气性膜不进行浸渍而置于空气中或其他所期望的培养气氛中,从而通过所述透气性膜进行气体交换,以通过所述开口部至少将氧提供至包含在所述第二基质中的细胞。
8.血浆蛋白的制造方法,其包括以下步骤:
步骤(1),对权利要求1所述的基质组合物进行培养;和
步骤(2),从所述步骤(1)培养的培养物中回收培养上清液。
