Description
Technical Field
The invention belongs to the technical field of edible fungus strains, and particularly relates to an application of a hypsizigus marmoreus strain in preparation of folic acid.
Background
Hypsizigus marmoreus (Hypsizygusguisammoreus), the scientific name Hypsizigus marmoreus, and wild Hypsizigus marmoreus are mainly distributed in Japan, Europe, North America, and the like. Artificial cultivation of hypsizigus marmoreus has been successfully achieved in japan, korea, china, etc., and artificially cultivated hypsizigus marmoreus products have been commercialized. The hypsizigus marmoreus is widely popular with Asian people because of the fleshy fruiting body, crisp and tender stipe, and sweet and delicious taste.
The common white beech mushroom (white Hypsizygus giganteus) on the market is a white line of the hypsizigus marmoreus, called as 'golden branch jade leaves' in the hypsizigus marmoreus, and the mushroom body is crisp, tender, fresh and smooth, fresh, sweet and delicious; hypsizygus marmoreus (Xieweiguu), also known as Hypsizygus marmoreus, Lyophyllum decastes, is a famous rare Hypsizygus marmoreus, is a gray strain of Hypsizygus marmoreus, has a gray pileus, has a long stipe, has a gray upper portion of the stipe, is introduced from Japan, and has a unique crab flavor; hypsizygus marmoreus (Haixiangu) is a pure white strain of Hypsizygus marmoreus, and has longer stipe than Hypsizygus marmoreus, delicate taste, fragrant smell and delicious taste.
The strains used by the domestic hypsizigus marmoreus manufacturers at present are basically from Japan, and the dominant strains of the hypsizigus marmoreus currently used are numbered wz-29, wz-65 and wz-77; the fruiting performance of the strain wz-29 is good in quality and relatively low in yield per unit; wz-65 and wz-77 have relatively high yield per unit, and the mushroom has a cap with a tumor, short shelf life and relatively poor quality.
Folic acid refers to tetrahydrofolic acid and derivatives thereof, is a water-soluble B vitamin, and is an essential component of organisms. Folic acid is involved in nucleotide biosynthesis, amino acid metabolism and methylation cycle. Folate and the synthetic pathway are critical for cellular metabolism. The deficiency of folic acid causes a number of health problems, such as neural tube malformations in newborns, cancer, cardiovascular disease, megaloblastic anemia, arteriosclerosis, acute myocardial infarction, and stroke. The human body has no ability to synthesize folic acid by itself and can only take it by diet. To meet the needs of normal life activities, adults should be supplemented with 400 μ g of folic acid per day, while pregnant women should be supplemented with 600 μ g per day.
Recently, as consumers attach importance to health, preference for substances extracted from natural materials rather than chemical synthesis has increased.
Disclosure of Invention
The invention aims to provide the application of a specific hypsizigus marmoreus strain in preparing folic acid, folic acid can be conveniently and quickly extracted from the hypsizigus marmoreus strain, and the extraction purity of the folic acid is high.
In order to achieve the technical purpose, the technical scheme provided by the invention is as follows:
an application of hypsizigus marmoreus strain in preparing folic acid, wherein the hypsizigus marmoreus strain is hypsizigus marmoreus XCXH-2 with the preservation number of CCTCC NO: m2020068, the preservation date is 2020, 4 and 17 months, the preservation address is Wuchang Lojia mountain in Wuhan city, Hubei province, and the preservation unit is the China center for type culture Collection.
According to the detection result, the folic acid content of the hypsizigus marmoreus strain is not less than 56.8 mug per 100g hypsizigus marmoreus strain.
In another aspect, the present invention also provides a method for preparing folic acid, comprising the steps of:
(1) drying and pulverizing Hypsizigus marmoreus, wherein the strain of Hypsizigus marmoreus is Hypsizigus marmoreus XCXH-2, and the preservation number is CCTCC NO: m2020068, wherein the preservation date is 2020, 4 and 17 months, the preservation address is Wuchang Lojia mountain in Wuhan city, Hubei province, and the preservation unit is China center for type culture Collection;
(2) adding water and organic acid into Hypsizygus marmoreus, heating under high pressure for hydrolysis to obtain extract;
(3) concentrating and drying the extract.
The hypsizigus marmoreus XCXH-2 selected as a natural product has higher folic acid content and extremely low contents of other vitamins, particularly water-soluble vitamin C and vitamin B1, so the hypsizigus marmoreus can be extracted by a simpler extraction process and has high extraction purity.
As a further improvement of the technical proposal, the organic acid in the step (2) is formic acid or acetic acid. Formic acid or acetic acid can stabilize the structure of folic acid in the extraction process, prevent the structure of folic acid from being damaged by heating influence, and improve the extraction yield of folic acid.
As a further improvement of the technical scheme, the weight ratio of the hypsizigus marmoreus and the water in the step (2) is 1: 10-20. If the amount of water used is too small, folic acid cannot be fully extracted; if the amount of water used is too much, folic acid is diluted in water and has low concentration, and the subsequent concentration and drying take longer, so that the production efficiency is reduced.
As a further improvement of the technical scheme, the weight ratio of the water to the organic acid in the step (2) is 100: 1-5. If the content of the organic acid is too low, the organic acid does not function sufficiently; if the organic acid content is too high, the acidity of the whole extract becomes strong and folic acid may be destroyed.
As a further improvement of the technical scheme, in the step (2), the heating pressure is 0.1-0.12 Mpa, the heating temperature is 80-100 ℃, and the heating time is 2-5 hours. If the heating temperature is lower than 80 ℃, folic acid is not fully extracted; if the heating temperature exceeds 90 c or the heating time is too long, the structure of folic acid may be broken.
According to the invention, through detection research on specific hypsizigus marmoreus XCXH-2, the content of folic acid contained in the hypsizigus marmoreus is high, the content of other water-soluble vitamins is low, and when the hypsizigus marmoreus is applied to folic acid extraction, the extraction process is simple and the purity is high.

The detection methods of the substances were carried out according to the detection standards specified in the industry, and the detection results are shown in table 1 below.
Table 1: hypsizigus marmoreus XCXH-2 folate and other vitamin content
As can be seen from the detection results, the content of folic acid in the hypsizigus marmoreus XCXH-2 is higher, but the content of other vitamins, particularly water-soluble vitamin C and vitamin B1 is extremely low, so that the hypsizigus marmoreus XCXH-2 is very suitable to be used as a natural product source for extracting folic acid.
Example 2: extraction of Hypsizigus marmoreus XCXH-2 folic acid
(1) Weighing and cutting dried hypsizigus marmoreus XCXH-2(100g), wherein the preservation number of the hypsizigus marmoreus XCXH-2 is CCTCC NO: m2020068, the preservation date is 2020, 4 and 17 months, the preservation address is Wuchang Lojia mountain in Wuhan city, Hubei province, and the preservation unit is the China center for type culture Collection.
(2) Adding 1500ml distilled water into Hypsizigus marmoreus, and immersing Hypsizigus marmoreus XCXH-2. If the amount of water used is too small, folic acid cannot be fully extracted; if the amount of water used is too much, folic acid is diluted in water and has low concentration, and the subsequent concentration and drying take longer, so that the production efficiency is reduced.
(3) Further adding 7.5ml acetic acid, and mixing well. The acetic acid can stabilize the folic acid structure in the extraction process, prevent the folic acid structure from being damaged by heating influence, and improve the extraction yield of folic acid. If the content of the organic acid is too low, the organic acid does not function sufficiently; if the organic acid content is too high, the acidity of the whole extract becomes strong and folic acid may be destroyed.
(4) The mixture was heated to 80 ℃ under 0.1MPa for 2 hours. If the heating temperature is lower than 80 ℃, folic acid is not fully extracted; if the heating temperature exceeds 90 c or the heating time is too long, the structure of folic acid may be broken.
(5) Concentrating and drying the extract. The concentration and drying means may be vacuum concentration, freeze-drying concentration, etc. which are commonly used in the art.
