Abstract
本发明涉及一种利用混合物4F的用于增加干细胞生物学活性的组合物。处理根据本发明的混合物4F的干细胞不仅可以获得未分化特性(stemness),还具有提高细胞增殖能力及迁移能力的效果,从而在将干细胞移植到体内后,可以提高细胞存活及植入存活率,且可以进一步提高血管及组织再生能力,并以多种方式应用于干细胞分化、预防或治疗缺血性疾病的领域。

技术领域
本发明涉及利用混合物4F来增加干细胞生物学活性的组合物。
背景技术
心脏疾病、脑血管疾病、末梢血管性疾病等的缺血性血管疾病具有40%的高患病率,是世界上死亡率最高的疾病之一。对此,现有的主要治疗方法有以药物给药为主的内科治疗法和对血管的机械性中继性治疗及外科性迂回手术治疗等三种,但实际情况是有不少即使实施所有这些治疗,也因无法顺畅地供给氧气及养分的缺血组织的有限的再生而无法进行令人满意的治疗的情况。因此,对以目前医学技术无法治疗的广泛血管疾病而言,已经提出了通过促进缺血区域周围的组织中的血管生成来确保缺血组织的血流并减少组织损伤的血管生成治疗法作为治疗对策。
为了形成由细胞组成的血管组织,需要供应生成血管的细胞。因此,作为治疗策略,已经进行了通过直接施用血管生成促进因子或施用血管生成促进因子的基因来促进血管内皮细胞的增殖及发育的多种方法,但是实际情况是其效果不显著,并且提出了各种副作用和安全性问题,因此需要进行更多的研究以实际引入血管生成促进因子。
此外,已经发现血管内皮前驱细胞对受损血管的修复及缺血性组织再生是有效的细胞,并且目前为止正在尝试各种研究以通过直接施用生成血管的细胞来促进血管新生。目前,在使用干细胞的血管生成和再生领域中,成体干细胞用于临床应用,并且其优点在于它们可以从患者的身体中提取并自体移植到所需组织中,并且与胚胎干细胞相比没有伦理性问题。然而,由于由西方化的饮食生活和运动减少、老龄化等引起的源自患者细胞的衰老及功能降低,难以进入实用化阶段。因此,用于扩增干细胞和增强生物活性的研究成为细胞治疗实用化的核心因素,为此,需要开发对人体安全的功能增强因子并分析每种因子的作用机制以提供具有更优异的功能的干细胞的系统,并且由于这种需要,最近,积极地进行用于干细胞治疗剂实用化的细胞功能增强研究正成为趋势。使用基因导入的研究是通过扩增及缺失特定基因来改善细胞功能的方法,需要进一步研究其稳定性。
发明内容
技术问题
由此,为了解决如上述的现有技术的问题,本发明人开发了包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子的混合物4F,从而完成了本发明。
因此,本发明的一个目的是提供一种用于抑制干细胞衰老、促进干细胞增殖或诱导干细胞分化的组合物,所述组合物包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子作为活性成分。
本发明的另一个目的是提供一种用于预防或治疗缺血性疾病的药学组合物,所述药学组合物包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子作为活性成分。
本发明的另一个目的是提供一种干细胞培养用培养基组合物,所述干细胞培养用培养基组合物包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子作为活性成分。
本发明的又一个目的是提供一种无异源培养方法,所述无异源培养方法包括利用包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子作为活性成分的干细胞培养用培养基组合物处理干细胞的步骤。
本发明的又一个目的是提供一种干细胞治疗助剂,所述干细胞治疗助剂包含岩藻聚糖、牛磺熊去氧胆酸、橄榄油素和血管内皮生长因子作为活性成分。
本发明的又一目的是提供通过所述无异源培养方法培养的干细胞。
本发明的又一目的是提供包含所述干细胞的用于预防或治疗缺血性疾病的药学组合物。
技术方案
为了实现所述目的,本发明提供一种用于抑制干细胞的衰老、促进干细胞的增殖或诱导干细胞的分化的组合物,所述组合物包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子作为活性成分。
此外,本发明提供了用于预防或治疗缺血性疾病的药学组合物,所述药学组合物包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子作为活性成分。
此外,本发明提供了一种干细胞培养用培养基组合物,所述干细胞培养用培养基组合物包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子作为活性成分。
此外,本发明提供了一种无异源培养方法,所述无异源培养方法包括利用包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子作为活性成分的干细胞培养用培养基组合物处理干细胞的步骤。

技术效果
利用根据本发明的混合物4F处理的干细胞不仅会获得未分化特性(stemness),还具有提高细胞增殖能力及移动能力的效果,因此,将干细胞移植到体内后,可提高细胞存活及植入存活率,并可进一步提高血管及组织再生能力,从而可以在干细胞分化、预防或治疗缺血性疾病领域中广泛应用。

以下,对本发明进行详细说明。
根据本发明的一示例,本发明提供了一种用于抑制干细胞衰老、促进干细胞增殖或诱导干细胞分化的组合物,所述组合物包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子作为活性成分。
在本发明中,“混合物4F”是指岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子的混合物。
在本发明中,“干细胞”是指具有自我复制能力且具有能够分化为两个以上的细胞的能力的细胞,可分为万能干细胞(totipotent stem cell)、全能干细胞(pluripotentstem cell)、多能干细胞(multipotent stem cell)。所述干细胞可以无限制地根据目的适当地选择,并且可以来自包括人的哺乳动物,优选为来自人的已知的所有组织、细胞等的成体细胞,例如骨髓、脐带血、胎盘(或胎盘组织细胞)、脂肪(或脂肪组织细胞)等。例如,所述干细胞可以是从骨髓、脂肪组织、肌肉组织、离体(ex vivo)培养的自组织间充质干细胞、同种异体间充质干细胞、脐带血、卵黄囊、胎盘、脐带、骨膜、胎儿及青春期皮肤以及血液获得的干细胞而不限于此,并且可以是来自胎儿或出生后不久或成人的干细胞。
在本发明的具体例中,所述干细胞选自由血管内皮祖细胞(EndothelialProgenitor Cell)、血管干细胞(Angiogenic Stem Cell)、间充质干细胞(MesenchymalStem Cell)、胚胎干细胞(Embryonic Stem Cell)、成肌细胞(Myoblast)以及心脏干细胞(Cardiac Stem Cell)组成的组中,更优选为血管内皮祖细胞(Endothelial ProgenitorCell)或血管干细胞(Angiogenic Stem Cell),最优选为血管内皮祖细胞(EndothelialProgenitor Cell)。
在本发明中,“衰老(senescence)”是指干细胞抵抗从内部或外部受到的各种应激(例如,连续传代培养及体外培养时的高浓度的氧气条件等)而使细胞生长(cell growth)及细胞分裂停止或有意延迟。
在本发明中,“增殖(proliferation)”是指来自特征细胞类型或可能相同或不同的细胞的初始细胞群的细胞类型的数量的增加。用于增殖的初始细胞可以与在增殖中生成的细胞不同。
在本发明中,“分化诱导(differentiation induction)”不仅包括诱导从干细胞到特定细胞的完全分化的情况,还包括在干细胞完全分化为特定细胞之前在中间步骤实现的类胚体(embryonic body)的形成。
在本发明的具体例中,分化可以为血管生成。
在本发明中,“血管生成(angiogenesis)”是指新血管形成的过程,即,新血管在细胞、组织或器官内产生及分化。
本发明的血管生成包括诸如内皮细胞的引发、迁移、增殖、基质重建以及细胞稳定在内的与血管形成过程相关的血管再生、血管恢复以及血管分化。
新血管生成作用对于由血管损伤引起的缺血性组织的恢复是必不可少的。由于新血管生成和恢复仅通过现有血管内皮细胞的增殖是不够的,因此在血管生成过程中最重要的是将以骨髓为来源的血管干细胞动员到缺血区域并用于血管修复。
根据本发明,所述组合物通过增强血管干细胞从骨髓到缺血组织的动员,从而改善与血管内皮细胞之间的结合能力,并改善到血管的分化能力。
在本发明中,“岩藻聚糖(Fucoidan)”是由作为基本糖的岩藻糖(fucose)及硫酸基相结合的多糖类,大量包含于褐藻类中。已知所述岩藻聚糖具有以下作用:血液凝固防止作用、抗肿瘤作用、胃溃疡治疗促进作用、抗菌作用、血压上升抑制作用、肝细胞增殖因子(HGF)产生诱导、血糖上升抑制作用、免疫细胞调节作用、抗过敏作用、抗病毒作用。

在本发明中,术语“牛磺熊去氧胆酸(TUDCA:Tauroursodeoxycholic acid)”是指熊去氧胆酸(UDCA)的牛磺酸结合体,并且已知其对治疗胆结石、肝硬化、亨廷顿病、帕金森病以及脑中风等是有益的。

根据本发明的又一示例,本发明提供一种干细胞培养用培养基组合物,所述干细胞培养用培养基组合物包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷和血管内皮生长因子作为活性成分。
在本发明中,“培养基(media)”是指在体外培养条件下能够支持干细胞的生长及生存的培养液,包括适合于干细胞的培养的本领域中使用的所有通常的培养基。此外,可以根据细胞种类选择培养基和培养条件。用于培养的培养基优选为细胞培养最低培养基(CCMM:cell culture minimum medium),并且通常包含碳源、氮源以及微量元素成分。这种细胞培养最低培养基例如,包括达尔伯克氏改良伊格尔氏培养基(DMEM:Dulbecco’sModified Eagle’s Medium)、基本必需培养基(MEM:Minimal essential Medium)、基础伊格尔基础培养基(BME:Basal Medium Eagle)、RPMI1640、F-10、F-12、基本必需培养基(MEM:Minimal essential Medium)、Glasgow最小必需培养基(GMEM:Glasgow’s Minimalessential Medium)、Iscove改良的Dulbec co培养基等,但不一定局限于这些。

通过根据本发明的无异源培养方法培养的干细胞不仅会获得未分化特性(stemness)的效果,还具有生成血管、提高细胞增殖能力以及迁移能力的效果,因此,在将血管内皮祖细胞移植到体内后,可以提高细胞存活及植入存活率,并且可以进一步提高血管及组织再生能力。

在本发明中,“个体”指需要治疗疾病的对象,更具体地,指人类或非人类的灵长类、小鼠(mouse)、大鼠(rat)、狗、猫、马以及牛等的哺乳类。
本发明的药学组合物可以根据通常的方法以各种形态剂型化使用。例如,可以剂型化为粉剂、颗粒剂、片剂、胶囊剂、悬浮剂、乳剂、糖浆等的口服型制剂,也可以剂型化为外用制剂、栓剂以及无菌注射溶液的形态使用。
本发明的组合物可以与所述活性成分一同含有对缺血性疾病具有预防或治疗效果的一种以上的公知的活性成分。

本发明的组合物可以在实际临床给药时以口服或非口服的多种剂型给药,在进行制剂化的情况下,可以使用通常使用的填充剂、增量剂、结合剂、湿润剂、崩解剂、表面活性剂等的稀释剂或赋形剂来制剂,本技术领域中公知的适合的制剂优选为利用在莱明顿的文献中公开的制剂。
用于所述口服给药的固体制剂包括片剂、丸剂、粉剂、颗粒剂、胶囊剂等,这种固体制剂混合至少一种的赋形剂(例如淀粉、碳酸钙(Calcium car bonate)、蔗糖(Sucrose)或乳糖(lactose)、明胶等)来制备。此外,除了简单的赋形剂之外,还使用润滑剂(如硬脂酸镁、滑石)。此外,用于口服给药的所述液体制剂包括悬浮剂、溶液剂、乳剂、糖浆剂等,并且除了通常使用的简单稀释剂的水、液体石蜡之外,还可以包括各种赋形剂,例如润湿剂、甜味剂、芳香剂、防腐剂等。

本发明的岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子可以以药学上允许的盐的形态使用,并且包括通过常规方法制备的所有盐、水合物以及溶剂化物。
作为盐,由药学上可接受的游离酸(free acid)形成的酸加成盐较有用。酸加成盐通过常规方法制备,例如,将化合物溶解在过量的酸性溶液中,并使用混溶性有机溶剂(例如甲醇、乙醇、丙酮或乙腈)沉淀该盐。可以对等摩尔量的化合物和水中的酸或醇(例如,乙二醇单甲醚)进行加热,然后可以蒸发并干燥所述混合物,或者抽滤析出的盐。此时,可以将有机酸和无机酸用作游离酸,可以使用盐酸、磷酸、硫酸、硝酸、酒石酸等用作无机酸,使用甲磺酸、对甲苯磺酸、乙酸、三氟乙酸、马来酸(maleic acid)、琥珀酸、草酸、苯甲酸、酒石酸、富马酸(fumaric acid)、扁桃酸、丙酸(propionic acid)、柠檬酸(citric acid)、乳酸(lactic acid)、乙醇酸(glycollic acid)、葡萄糖酸(gluconic acid)、半乳糖醛酸、谷氨酸、戊二酸(glutaric acid)、葡萄糖醛酸(glucuronic acid)、天冬氨酸、抗坏血酸、碳酸、香草酸、氢碘酸等用作有机酸,但不限于此。
此外,可以使用碱性基团制备药学上可接受的金属盐。碱金属盐或碱土金属盐例如可以通过将化合物溶解在过量的碱金属氢氧化物溶液或碱土金属氢氧化物溶液中,然后过滤不溶性化合物盐,并蒸发并干燥滤液而获得。在这种情况下,在金属盐中,尤其钠盐、钾盐或钙盐的制备是药学上可接受的,但不限于此。此外,与此对应的银盐可以通过使碱金属盐或碱土金属盐与适量的银盐(例如,硝酸银)反应来获得。

本发明的药学组合物的给药量可根据所述药学组合物的制剂化方法、给药方式、给药时间和/或给药途径等而不同,可以根据通过所述药学组合物的给药来实现的反应的种类和程度、作为给药对象的个体的种类、年龄、体重、一般健康状态、疾病的症状或程度、性别、饮食、排泄、对相应个体同时或不同时间一起使用的药物的其他组合物的成分等在内的多种因素及医药领域中已知的类似因素而不同,在本技术领域中具备普通知识的技术人员可以容易地确定对目标治疗有效的给药量并开出处方。
本发明的药学组合物的给药途径及给药方式可以各自独立,其方式不受特别限制,只要所述药学组合物能够到达目标的相应部位,则可以根据任意的给药途径及给药方式来进行。
所述药学组合物可以以口服或非口服的给药方式给药。所述非口服给药方式包括例如静脉内给药、腹腔内给药、肌肉内给药、经皮给药或皮下给药等。
本发明的药学组合物为了预防或治疗缺血性疾病可以单独使用或与手术、放射治疗、激素治疗、化学治疗以及使用生物反应调节剂的使用方法组合而使用。
根据本发明的又一示例,本发明提供了一种干细胞治疗助剂,所述干细胞治疗助剂包含岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子作为活性成分。
在本发明中,“细胞治疗剂”是指为了恢复细胞和组织的功能,通过在体外增殖、筛选存活的自体(autologous)、同种(allogenic)、异种(xenogenic)细胞或通过其他方法改变细胞的生物学特性等一系列行为而以治疗、诊断、预防为目的使用的医药品,尤其,可以分为“干细胞治疗剂”、胚胎干细胞治疗剂以及成体干细胞治疗剂。
在本发明中,“干细胞治疗助剂”是指可以用于辅助增强本领域中通常使用的干细胞治疗剂的效果的制剂,通过使用本发明的组合物,可以促进干细胞治疗剂的干细胞的衰老抑制和增殖来改善治疗剂的效果。在本发明的优选实施例中,所述干细胞治疗为缺血性疾病治疗。
只要所述干细胞治疗助剂可以到达靶组织,则可以通过任何常规的途径施用于人体。
非口服给药可以是例如腹腔内给药、静脉内给药、肌肉内给药、皮下给药,但不限于此。
所述干细胞治疗助剂也可以借由能够将活性物质转移至靶细胞的任意装置而给药。可以与通常用于干细胞治疗的药学载体一起施用,生理盐水可以作为这种载体的一例。
以下,将通过实施例更详细地说明本发明。然而,应当解释为这些实施例仅用作本发明的示例,本发明的范围不受这些实施例的限制,这对于本领域具备普通知识的技术人员应该是显而易见的。
实施例
实施例1.混合物4F的制备
将岩藻聚糖(150nM)、牛磺熊去氧胆酸(25μM)、橄榄苦苷(0.5μM)以及血管内皮生长因子(2.5nM)(重组人VEGF165)混合以制备混合物4F。在后述的实施例中,不使用添加到现有培养基(EndoGo XF培养基,Biological Industries公司)中的b-FGF、IGF、EGF、抗坏血酸,仅将4F追加到基础培养基中以用于实验。
实施例2.根据经过混合物4F处理的血管内皮祖细胞的形态学特征的确认
为了研究4F处理对细胞表现型的影响,将主要细胞标记物大致分为未分化标记物(stem cell marker)和内皮细胞系标记物(Endothelial lineage marker),并利用流式细胞分析仪进行分析。具体地,将混合物4F(岩藻聚糖(Fucoidan),牛磺熊去氧胆酸(TUDCA),橄榄苦苷(Oleuropein),血管内皮生长因子(VEGF))与5%FBS和1%青霉素/链霉素一起添加至用于培养血管内皮细胞的基础培养基(EBM2,龙沙(Lonza))中,然后以各种期间条件进行处理,然后使用流式细胞分析仪(flowcytometry)分析未分化标记物(CD34、CD90、c-kit、CXCR4)、内皮细胞系标记物(Tie2、CD31)的表达。为了验证结果的再现性,从源自5名孕妇的脐带血分离并培养L-EPC(n=5),以确认各个标记物的表达。图1示出了根据经过混合物4F处理的血管内皮祖细胞的形态学特征的确认结果。
如图1所示,确认了表达未分化标志物的细胞群以及在具有未分化性质的同时表达内皮细胞系的明显的分化标志物的细胞群会以依赖于混合物4F处理天数的方式显著增加。由此,确认了通过混合物4F的处理进一步获得了血管干细胞的以下特征:在保持原本细胞所具有的内皮细胞系的文化特征的同时,通过仅增加未分化特性(stemness),从而具有与内皮细胞相似的表现型和功能,并且作为更高级别而具有单分化为内皮细胞的能力。
实施例3.根据混合物4F处理的细胞迁移能力的确认
已知CXCR4是以依赖于4F处理天数的方式调节干细胞的迁移能力,并且确认了所述CXCR4的表达增加,基于此,使用作为CXCR4的配体的SDF-1α分析了作为血管形成及再生所必需过程的细胞迁移能力。具体地,为了分析因趋化因子刺激引起的细胞迁移能力而实施了Transwell迁移试验(transw ell migration assay)。将接种各实验组细胞(5×104cells)的8.0μm的网状膜嵌入件插入于培养板,并在下部添加作为刺激细胞移动的趋化因子的基质细胞衍生因子(SDF-1α)(100ng/ml)后进行了培养。6小时后,利用结晶紫对借由下部的基质细胞衍生因子(SDF-1α)刺激而脱离网状膜的孔隙(pore)的细胞进行染色,并对移动的细胞进行了计数。SDF-1α是从缺血组织内分泌而出,并重塑与血管和组织再生相关的细胞的主要细胞因子。图2示出了根据经过混合物4F处理的细胞迁移能力的确认结果。
如图2所示,细胞的迁移能力通过混合物4F的引发而相比于对照组有显著的增加。此外,虽然观察到在没有特定刺激的情况下进行的阴性对照组具有与阳性对照组类似水平的移动能力,然而确认了根据经过混合物4F处理的基质细胞衍生因子(SDF-1α)刺激的细胞移动能力增加了三倍以上。所述结果意味着通过混合物4F的引发来增进细胞对基质细胞衍生因子(SDF-1α)的反应性及向缺血组织的特异性移动能力。
实施例4.根据本发明的混合物4F在下肢缺血动物模型中的血管再生效果的确认
制备作为重症血管疾病模型的下肢缺血动物模型以评价借由混合物4F处理而使得功能增强的血管干细胞的治疗效果。具体地,使用激光多普勒流速计,从血流动力学分析细胞移植后血流的改善程度。具体地,为了在重症缺血性动物模型中确认用混合物4F引发的细胞的治疗效果,利用7-8周龄的裸鼠(雄性)制作了下肢缺血动物模型。下肢缺血动物模型以通过股动脉结扎术(femoral artery ligation)阻断下肢血流的方式制作,并且在血管切除后,肌内注射细胞(5×105cells/PBS50μl),并使用激光多普勒流速计按天(第3天、第7天、第14天)对阻断血流的恢复程度进行了血流动力学分析。此外,利用活细胞成像的方式通过组织学分析确认了移植细胞的植入和存活以及血管生成作用,其结果图示于图3a和3b。
如图3a所示,确认了相比于对照组,当移植经过4F处理的细胞时,体内血管再生显著增强。
如图3b所示,应用生物体内成像技术,在被GFP标记的细胞移植后的第3天以存活状态对血管缝合部位进行成像,结果发现在缝合的动脉血管周围能够观察到许多移植的GFP+细胞。所述结果意味着,通过混合物4F处理存活能力得到改善的血管干细胞被重塑以用于血管再生。
实施例5.在重症心肌梗塞动物模型中根据经过混合物4F处理的心脏功能效果增强的确认
利用重症心肌梗塞动物模型,确认了由混合物4F引发的细胞的治疗功效。具体地,使用7-8周龄的裸鼠制作重症心肌梗塞模型。所述重症心肌梗塞模型以切除左冠状动脉的方式而制作,每个个体分别直接移植5×105个细胞到心脏的缺血部位。针对诱发心肌梗塞后经过28天时间点的心脏功能,利用超声心动图来测量了左心室的质量、容积[左心室舒张末期直径(LVEDD:Leftventricle end diastolic diameter)、左心室收缩末期直径(LVESD:Left ventricleend systolic diameter)]、左心室射血分数[Left ventricularejectionfraction(LVEF)]等。此外,评价动物模型的心输出量、左心室舒张功能。图4a示出了超声心动图、心输出量以及左心室舒张功能的分析结果。
如图4a所示,根据在诱发心肌梗塞后经过28天时间点的心脏功能评价结果,确认了相对于对照组,混合物4F处理组的射血分数(EF:Ejection Fraction)、缩短分数(FS:Fractional Shortening)显著增加。所述结果意味着,混合物4F进一步提高了现有血管干细胞的功能。
此外,在诱发心肌梗塞28天后,通过进行能够将阿胶纤维特征性地染色的马松三色(Masson’s trichrome)染色,分析了心肌纤维化的程度(蓝色染色部分),其结果图示于图4b中。
如图4b所示,确认了通过混合物4F处理,左心室的纤维化程度显著降低。
此外,在向重症心肌梗塞模型移植被绿色荧光蛋白(GFP)标记的细胞起经过3天后,通过对心肌内细胞的植入细胞及增殖率细胞的增殖标记物(即,增殖细胞核抗原(PCNA))进行免疫染色而进行了确认,其结果图示于图4c。
在图4c中的混合物4F处理组中观察到多个与绿色荧光蛋白(GFP)合并(merge)的增殖细胞核抗原(PCNA)的表达。
上述的结果意味着,在重症心肌梗塞动物模型中,用移植的混合物4F引发的血管干细胞通过增殖来增进心脏内心肌恢复及心脏功能。
实施例6.根据混合物4F处理的增殖能力、细胞周期变化、细胞衰老及集落形成能力的分析
为了分析细胞增殖能力,在96孔板(96well plate)中接种10000个细胞后,根据混合物4F处理天数,通过MTS分析确认了细胞增殖能力。此外,为了确认作为细胞增殖调节因子的CDK抑制剂(p21、p16)的蛋白质表达,利用蛋白质溶解缓冲液来分离、提取及定量对照组的细胞内蛋白质后,使用等量的蛋白质执行了蛋白质印迹。将蛋白质经过SDS-PAGE之后转移到PVDF膜,并在5%脱脂乳中封闭(blocking)了30分钟。封闭后,将以1:1000的比例稀释的各一次抗体p53(abcam)、p27(abcam)、p21(abcam)、p16(abcam)以及b-actin(santacruz)与膜在4℃下反应过夜。洗涤膜后,在常温下与结合有HRP的二次抗体反应1小时,利用LAS3000(Fujifilm公司)使蛋白质的表达可视化。此外,为了确认细胞增殖能力的降低是因未分化特性的增加所诱导的细胞周期的停止而引起的,利用PI染色法和Hst/PY染色法分析了细胞周期。具体地,对60%-70%铺满培养的细胞进行了PI(SantaCruz公司)、hoechst33342(SantaCruz公司)、PY(SantaCruz公司)染色,并利用流式细胞术分析了细胞周期。图5a和5b示出了根据经过混合物4F处理的细胞增殖率和细胞周期变化的确认结果。
如图5a和5b所示,确认了细胞增殖能力以依赖于混合物4F处理天数的方式显著减少。此外,确认了在用混合物4F处理的细胞中,作为细胞增殖调节因子的CDK抑制剂(p21、p16)的蛋白质表达显著增加。此外,根据细胞周期分析结果,确认了当用混合物4F处理时(4天),相比于对照组,发生DNA复制和细胞质分裂的S基团和G2/M基团显著降低,但是G0/G1基团显著增加。不仅如此,还确认了相比于对照组,在用混合物4F处理时(4天),处于休眠状态的G0基团的细胞群显著增加。
分析了细胞周期停止的细胞和通过重复传代而衰老的细胞(old)的衰老程度。使用SA-β-gal检测试剂盒(Cell Signaling公司)进行细胞衰老分析,并根据制造商手册进行了实验。通过显微镜成像测量每组细胞中衰老细胞(β-阳性-绿色)的数量并进行了图表化,其结果图示于图5c。
如图5c所示,确认了由混合物4F处理而引起的细胞周期停止不是因细胞损伤或衰老引起的,而是由未分化特性的增加而引起的。
此外,为了确认细胞的集落形成能力,在96孔板涂覆1%明胶后,接种细胞,并培养了14天至20天。通过显微镜成像,针对培养细胞测量形成集落(colony)的孔(well)数并进行了图表化,并且将其图示于图5d。
如图5d所示,通过分析单一细胞集落,对单一细胞的集落形成能力分析的结果,确认了血管干细胞因混合物4F的引发而陷于休眠状态,并且作为干细胞的未分化特性大大增加。
实施例7.根据经过混合物4F处理的有关干细胞分化基因表达的分析
测量根据混合物4F处理的血管内皮细胞的未分化或有关分化的基因的表达。具体地,从利用混合物4F引发的血管内皮细胞中提取总RNA。准备2μg的总RNA,并委托Teragen公司进行了RNA测序。差异表达基因(DEG:Differentially expressed gene)分析是基于Cuffdiff方法提取了小于0.05的q值阈值(q-value threshold)并进行了分析。有关干细胞分化的基因表达的分析结果图示于图6a和6b。
如图6a和6b所示,通过RNA测序分析转录体的结果,确认了用混合物4F引发的细胞与完全分化的血管内皮细胞(HUVEC)具有明显的遗传学差异,并且相比于多度生长内皮细胞(OEC:outgrowing EC,L-EPC),一部分有关未分化干细胞的标志物(CD34、KIT、HEY1、PODXL等)的表达显著增加。确认了分化的血管内皮细胞特异性基因的表达水平也与过度生长内皮细胞(OEC:outgrowing EC,L-EPC)的表达水平相似或略有减少。所述结果意味着,用混合物4F引发的细胞会获得未成熟(immature)特性。
实施例8.根据混合物4F处理的未分化干细胞的凋亡率分析
未分化干细胞由于具有对如缺血刺激的病态生理学环境的防御能力而生存能力变高,从而移植及植入存活率高于分化细胞。为了确认这一点,通过体外实现缺血状态(无养分及低氧状态)分析了细胞的凋亡率。通过在FBS、生长因子以及细胞因子被去除的无营养培养基中在1%O2的缺氧条件下培养细胞组24小时来进行细胞凋亡率分析,从而在体外实现了体内缺血状态。之后,使用膜联蛋白V/PI染色试剂盒(BD Biosciences公司)进行流式细胞分析,其结果图示于图7。
如图7所示,确认了相比于对照组,用混合物4F引发的细胞的活细胞群显著增加。上述结果意味着,混合物4F处理使细胞的未分化特性增加,其意味着它会增强对缺血刺激的防御机制,从而有助于提高细胞生存能力。
实施例9.根据混合物4F处理的未分化干细胞的增殖率及标记物表达分析
分析了在将通过混合物4F处理而细胞周期停止的细胞向用于现有细胞培养的营养培养基(添加b-FGF、EGF、IGF-1、抗坏血酸)进行再灌注时的细胞增殖能力和标记物表达的恢复。具体地,针对通过用混合物4F处理4天而细胞周期停止的细胞,在营养培养基(添加b-FGF、EGF、IGF-1、抗坏血酸)条件下培养(reperfusion)了5天。在更换为营养培养基后的第3天和第5天,使用MTS分析试剂盒确认了细胞增殖能力的恢复。根据制造商的手册的说明书进行了MTS分析。细胞增殖率图示于图8a,标记物表达分析结果图示于图8b和图8c。
如图8a至图8c所示,确认了用混合物4F引发的细胞不仅恢复了因营养培养基再灌注而减少了的增殖能力,而且标记物表达也会恢复到与对照组相似的水平。从所述结果确认,根据混合物4F的处理而引起的细胞周期停止及由此而引起的细胞增殖能力的降低不是细胞损伤或衰老之类的不可逆的现象,而是与细胞的未分化性质的增加一同出现的休止期细胞的一般特征,是可以通过必要刺激充分恢复的可逆的现象。
实施例10.根据混合物4F或单一成分处理的标志物表达及血管形成能力比较
比较了根据混合物4F或单一成分(岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子)的处理的标志物表达和血管形成能力。本实验的实验组经过了混合物4F处理,对照组利用了未处理组(con)、岩藻聚糖处理组(Fu,0.1μg/ml)、牛磺熊去氧胆酸处理组(TD,25μM)、橄榄苦苷处理组(OLP,0.5μM)以及血管内皮生长因子处理组(VEGF,100ng/ml)。
首先,通过实施例1的方法分析了用混合物4F引发的细胞的标记物,即CD90、c-kit、CXCR4、Tie2以及CD144的表达,其结果分别图示于图9a至图9e。
如图9a至图9e所示,相比于各成分的单一处理条件,混合物4F处理组中增加了较多表达未分化标记物(CD34、CD90、c-kit、CXCR4)的细胞群。
为了分析血管形成能力,将60μl基质胶GFR(BD Biosciences公司)添加到96孔板中以固化,然后接种10000个细胞并培养。培养6小时后,用显微镜观察管状网络的形成程度。使用图像处理软件(Image J)通过分支数计数(branch number counting)对显微镜观察结果绘制了细胞的血管形成能力图,其结果图示于图10。
如图10所示,确认了相比于各成分的单一处理条件,混合物4F处理组的血管形成能力增加得较大。
上述的结果意味着,相比于处理单一成分的情况,处理混合物4F使细胞的未分化特性及血管形成能力显著增加。
实施例11.利用混合物4F的无异源培养方法
图11是简要示出利用混合物4F的无异源培养方法的图。具体地,对于血管内皮祖细胞(EPC)的分离和培养而言,通过获得将韩国梁山釜山大学医院妇产科就诊的孕妇的脐带血(IRB批准号05-2017-053,50cc以上)而以利用聚焦糖(Ficoll)的密度差离心分离的方式分离获取了单核细胞(MNCs)。进行红细胞溶解后,将细胞接种(7×106细胞/孔)在涂覆有纤连蛋白(1μg/cm2)的6孔板,并且在37℃、5%CO2的各培养基(包含混合物4F的无异源培养基或已知的现有血管内皮细胞分化培养基(EGM2培养基))条件下培养了5天。包含所述混合物4F的无异源培养基是排除了现有培养基内添加的细胞因子或生长因子及动物血清(FBS)并在基础培养基中只添加混合物4F和人血清(2%)的培养基。进行培养后,每天更换培养基,确认EPC集落附着情况,在70%以上的铺满状态的情况下,进行传代培养。针对培养的细胞,根据培养基的种类,将通过在现有的血管内皮细胞分化培养基中培养而获得的细胞命名为“L-EPC”,并且将通过在含有混合物4F的无异源培养基中培养而获得的细胞命名为“X-EPC”。
实施例12.对通过利用混合物4F的无异源培养方法培养的血管干细胞的形态学特性的确认
在细胞的形态学比较中,通过在70%至80%的铺满状态下进行显微镜成像来确认了细胞的形态学特征。具体地,在作为内皮细胞系特异性功能评估的Ac-LDL吸收和UEA-1结合分析中,将在10%-20%的铺满状态下培养的细胞以血清饥饿状态(serum starvation)培养了2小时。将培养的细胞用浓度为10μg/ml的Ac-LDL(Invitrogen公司)处理,然后在37℃下反应了4小时。将反应的细胞在室温下用4%PFA固定了10-15分钟,并在室温下用10μg/ml浓度的UEA-1(SigmaAldrich公司)反应了1小时。将反应的细胞用磷酸盐缓冲盐溶液(PBS)洗涤,然后用DAPI染色法染色后,用荧光显微镜进行了观察。图12示出了分析通过使用混合物4F的无异源培养方法培养的血管干细胞的形态学特征的结果。
图12确认了通过使用混合物4F的无异源培养方法培养的细胞(X-EPC)具有与通过现有培养方法培养的细胞(E-EPC、L-EPC)相同的表达型。此外,确认了应用混合物4F的无异源培养方法未引起细胞的特定形态学变化。
实施例13.针对通过利用混合物4F的无异源培养方法培养的血管干细胞中有关未分化干细胞的标记物的基因表达的分析
分析了在所述实施例11中培养的细胞中的造血系标志物(CD11b、CD14、CD45)、未分化标志物(CD34、c-kit、CXCR4)、内皮细胞系标志物(VEGFR2、PECAM、VE-钙粘蛋白)的表达。具体地,将实施例11中培养的细胞与胰蛋白酶-EDTA以1:100、4℃的条件下反应30分钟,并从培养板上剥离。通过流式细胞分析仪(flowcytometry)分析了所述细胞的有关未分化干细胞标记物基因表达,其结果图示于图13。
如图13所示,确认了相比于通过现有培养方法获得的细胞组(E-EPC、L-EPC、HUVEC),通过利用混合物4F的无异源培养方法培养的细胞(X-EPC)中几乎未确认到造血系标记物(CD11b、CD14、CD45)的表达,并且确认了未分化标记物和血管内皮细胞系标记物的表达保持较高的结果(按各个脐带血捐献者(donor)确保3批(lot),并且即使在连续传代培养后,代表性标记物CD34、CXCR4、VE-钙粘蛋白的表达也仍保持高于L-EPC)。由此,确认了通过利用混合物4F的无异源培养方法获得的细胞(X-EPC)是具有比通过现有培养方法培养的细胞更高的未分化特性的血管干细胞。
实施例14.针对通过利用混合物4F的无异源培养方法培养的血管干细胞的增殖能力、细胞周期变化以及集落形成能力的分析
将根据所述实施例11的方法通过利用混合物4F的无异源培养方法培养的细胞分离成单一细胞,并通过与实施例6相同的方法分析了增殖能力、细胞周期变化以及集落形成能力。本实施例的对照组利用了通过现有培养方法培养的细胞(L-EPC)以及在无异源培养基培养的包含各单一成分的培养基的细胞[岩藻聚糖处理组(Fu,0.1μg/ml)、牛磺熊去氧胆酸处理组(TD,25μM)、橄榄苦苷处理组(OLP,0.5μM)以及血管内皮生长因子处理组(VEGF,100ng/ml)]。集落形成能力的分析结果图示于图14a和14b。
如图14a和14b所示,确认了通过利用混合物4F的无异源培养方法培养的细胞集落形成能力优异并且分裂时间短。但是,在处理单一成分的对照组未形成EPC集落。只不过,在经过血管内皮生长因子处理的细胞中确认了EPC集落,但是VE-钙粘蛋白的表达低,并且细胞分裂时间延长,从而使细胞增殖能力降低。
实施例15.针对通过利用混合物4F的无异源培养方法培养的血管干细胞的有关干细胞分化的基因表达的分析
通过实施例7的方法,对通过根据所述实施例11的方法而利用混合物4F的无异源培养方法培养的细胞中有关干细胞分化的基因的表达进行了分析。基于RNA测序结果分析了基因本体、类别以及表达模式。有关干细胞分化的基因表达的分析结果图示于图15至图17。
如图15所示,通过利用混合物4F的无异源培养方法培养的细胞(X-EPC)与通过现有培养方法获得的L-EPC存在遗传学上的差异,并且通过PCA分析按各细胞组的相关性的结果,确认了与呈现出高相似性的HUVEC/L-EPC不同地,所述X-EPC与各细胞组具有低相似性。
如图16所示,以在X-EPC中与L-EPC相比具有2倍以上表达差异的基因为对象分析了基因本体及类别,结果确认了有关内皮细胞活性(endothelial cell activation)、血管生成(angiogenesis)、内皮细胞迁移(endothelial cell migration)等的基因的表达变化程度较大。
如图17a和17b所示,确认了X-EPC显著增加了一部分有关未分化干细胞基因(CD34、KIT、HEY1、PODXL等)的表达。
实施例16.确认利用混合物4F的无异源培养方法培养的血管干细胞的血管形成能力
通过实施例10的方法确认了根据实施例11的所述方法中利用混合物4F的无异源培养方法培养的细胞的血管形成能力。进一步地,3D微球(beads)发芽分析是将培养用微球和各细胞培养4小时,用细胞包被了微球。在被细胞包被的培养纤维蛋白原/凝血酶凝胶中接种100至200个细胞包被微球,并在37℃下进行了培养。在培养过程中观察微球的血管新生发芽程度。2D和3D血管形成能力分析的结果图示于图18。
如图18所示,确认了相比于L-EPC和HUVEC,通过利用混合物4F的无异源培养方法培养的细胞(X-EPC)具有改善的血管形成能力并持续更长时间。
实施例17.确认利用混合物4F的无异源培养方法培养的血管干细胞的周边分泌血管形成的效果
根据所述实施例11的方法,分析通过利用混合物4F的无异源培养方法培养的细胞的周围分泌对血管形成的影响。具体地,使用Trizol试剂从实施例11的细胞中提取了总RNA,并基于提取的总RNA合成了cDNA。此外,执行以合成的cDNA作为模板的实时荧光定量多聚核苷酸链式反应(qPCR),分析了主要血管生成刺激因子(VEGF-A、IL-8、b-FGF、Ang2)的mRNA表达。用于聚合酶链式反应(PCR)的引物图示于表1,并且实时荧光定量多聚核苷酸链式反应(qPCR)结果图示于图19。

如图19所示,确认了相比于对照组(L-EPC、E-EPC、HUVEC),通过利用混合物4F的无异源培养方法培养的细胞(X-EPC)呈现出显著更高的刺激血管生成的主要生长因子和细胞因子的表达。
实施例18.在下肢缺血动物模型中确认利用混合物4F的无异源培养方法培养的血管干细胞的血管再生效果
通过与实施例4相同的方法,确认了根据所述实施例11的方法而以利用混合物4F的无异源培养方法培养的细胞的血管再生效果,其结果图示于图20和图21。
如图20和图21所示,确认了相比于对照组,将通过利用混合物4F的无异源培养方法培养的细胞(X-EPC)移植到下肢缺血动物模型的体内血管再生显著增强。此外,在移植通过利用混合物4F的无异源培养方法培养的细胞(X-EPC)的下肢缺血动物模型中,在缝合的动脉血管周围观察到了许多移植的GFP+细胞,并且确认了GFP+细胞会进入现有血管的现象。移植L-EPC的下肢缺血动物模型在移植后7天仅观察到细胞碎片,相反,移植X-EPC的下肢缺血动物模型在移植后的第21天检测到了活细胞的GFP信号。所述GFP信号的检测意味着细胞的植入及生存率得到了提高。此外,还确认了相比于L-EPC移植组,在移植X-EPC的下肢缺血动物模型中有更多CD31(红色:小鼠容器)阳性血管。上述的结果意味着X-EPC在体内(invivo)血管再生效果优异。
总而言之,本发明人已经发现,当针对血管内皮祖细胞进行包含有岩藻聚糖、牛磺熊去氧胆酸、橄榄苦苷以及血管内皮生长因子的混合物4F的处理时,血管内皮祖细胞不仅会获得了未分化特征(stemness),而且还具有生成血管、改善细胞增殖能力以及迁移能力的效果,因此,在将血管内皮祖细胞移植到体内后,可以改善细胞存活和植入存活率,并且可以进一步改善血管再生能力和组织再生能力,从而可以以各种方式应用于干细胞分化、预防或治疗缺血性疾病的领域。
以下,将通过制剂例更详细地说明本发明。制剂例仅用于例示本发明,不应解释为本发明的范围受制剂例的限制。

产业上可利用性
根据本发明,混合物4F处理的干细胞不仅获得未分化特性(stemness),而且具有提高细胞增殖能力及移动能力的效果,由此,通过将所述干细胞移植到体内作为缺血性疾病的治疗剂,可提高细胞存活及植入、血管及组织的再生能力。
可以通过利用混合物4F的无异源培养方法培养干细胞,并且通过该无异源培养方法培养的干细胞比通过现有培养方法培养的细胞具有更高的未分化特性、集落形成能力以及增殖能力,用于刺激血管形成的主要生长因子和细胞因子的表达较高,并且可以改善生物体内的细胞存活和植入、血管和组织的再生能力,因此,根据本发明的混合物4F和所述干细胞可以有效地用于开发用于缺血性疾病的治疗剂的基础研究和临床领域。
