Third, vagina mucosa stimulation experiment
The detection method comprises the following steps: the test is carried out according to the disinfection technical specification (2002 edition) 2.3.5 vagina mucosa stimulation test. New Zealand rabbits (weight 2092.4g-2456.4 g) of common grade, supplied by the Guangdong provincial medical laboratory animal center (Sanshui base), were selected for testing, and had no secretion, congestion, edema and other injury conditions in the vaginal orifice.
The contamination method comprises the following steps: connecting a blunt hose with the length of 8cm with a 2mL syringe, and filling the syringe and the catheter with the antibacterial gel prepared by the invention; fixing the New Zealand rabbits on the back to expose perineum and vaginal opening, wetting the catheter with sodium chloride injection, gently inserting into vagina (4cm-5cm), slowly injecting 2mL of antibacterial gel (that is, the application amount of each New Zealand rabbit is 2mL) with a syringe, and withdrawing the catheter to complete contamination.
24 hours after the infection, the animals are killed by a carbon dioxide method, the whole vagina is taken out after the laparotomy, the longitudinal incision is made, and whether congestion, edema and other manifestations exist is observed by naked eyes.
The test result shows that the irritation of the antibacterial gel to the vaginal mucosa is extremely mild, and the standard of measurement refers to 'disinfection technical specification' (2002 edition) 2.3.13.1 (5).
Four or more complete skin irritation experiments
The detection method comprises the following steps: the test was carried out according to the skin irritation test "Disinfection Specification" (2002 edition) 2.3.3. New Zealand rabbits of the general grade (supplied by the medical laboratory animal center of Guangdong province (Sanshui)) weighing 2011.8g-2227.2g were selected for the test.
In the specific test, the hair at two sides of the spine of the rabbit is removed by using a shaver and depilatory cream 24 hours before the test, the left part and the right part are respectively 3cm by 3cm, and the rabbit is used as a test part and a control part without damaging the animal epidermis. 0.5mL of the antibacterial gel is smeared on a test area for skin contamination, the contamination is carried out for 1 time per day, and the continuous contamination is carried out for 14 days. The control zone was used as a blank control without contamination. Residual test article was removed with water 4h after each exposure, and the results were observed 24h after each exposure and scored for irritation response according to Table 2-11 of Disinfection Specification (2002).
The detection result shows that the antibacterial gel of the invention grades the irritation reaction strength of the complete skin of the New Zealand rabbit for many times into nonirritant, and meets the requirements of 'Disinfection technical Specification' (2002 edition) 2.3.13.1 (3).
The above embodiments are merely examples of the present invention, and it is obvious that the present invention is not limited to the above embodiments, but may be modified. All modifications directly or indirectly derivable by a person skilled in the art from the present disclosure are to be considered within the scope of the present invention.
