Abstract
本发明涉及含5‑氨基乙酰丙酸的猪饲料、制备方法及在仔猪的应用。所述猪饲料,按重量份由以下组分组成:玉米粉50~60份,玉米蛋白粉2.5~5份,麸皮2.5~5份,酒糟20~25份,豆粕2.5~5份。制备方法,包括:⑴制备酵母菌种子液,将其转接入酵母菌液体发酵培养基中,制备成酵母菌发酵液,将酵母菌发酵液加入水中,并加入葡萄糖、甘氨酸、丁二酸后,制备成固体发酵用菌液;⑵固体发酵原料与固体发酵用菌液按照设定比例混合均匀,固体发酵36~72h后,发酵完成,烘干至水分12份以下;⑶营养添加剂复合包与固体发酵饲料按照设定比例混匀,即获得固体发酵含5‑氨基乙酰丙酸的猪饲料。
Description
技术领域
本发明属于饲料技术领域,特别涉及一种含5-氨基乙酰丙酸的猪饲料、制备方法及在仔猪的应用。
背景技术
5-氨基乙酰丙酸是生物体内四氢吡咯合成的前体,在各种生物中广泛存在,是合成叶绿素、血红素、维生素B12的必需因子,广泛存在于细菌、真菌、动物及植物等生物机体活细胞中的非蛋白质氨基酸,是动植物生命活动必需的、代谢活跃的生理活性物质,可以通过生物途径合成,也可以人工化学合成,没有毒副作用,易降解无残留。添加到饲料当中进行饲喂,可以明显改善动物的生长性能及血液指标。
目前ALA的生产菌株主要是原核细菌,如大肠杆菌、谷氨酸棒状杆菌等,通过酿酒酵母菌进行固体发酵的研究在国内外鲜有报道。其中大肠杆菌、谷氨酸棒状杆菌这两种菌均不在《饲料添加剂品种目录》中,而酵母菌是人类最早利用、公认安全且应用最广泛的微生物,是全球目前唯一年产量超过百万吨的微生物。酵母类饲料是公认的优质饲料。利用酿酒酵母菌进行固体发酵来生产富含5-氨基乙酰丙酸的饲料具有广阔的发展前景。
发明内容
本发明的目的是提供一种含5-氨基乙酰丙酸的猪饲料、制备方法及在仔猪的应用,该猪饲料不仅能够有效改善断奶仔猪的生长性能和血液指标,其浸出液还对金黄色葡萄球菌、大肠杆菌、肠炎沙门氏菌等致病菌有很强的抑制效果。
本发明的技术解决方案是所述固体发酵含5-氨基乙酰丙酸的猪饲料,其特殊之处在于,按重量份由以下组分组成:玉米粉50~60份,玉米蛋白粉2.5~5份,麸皮2.5~5份,酒糟20~25份,豆粕2.5~5份。
作为优选:还包括营养添加剂复合包,所述营养添加剂复合包按重量份由以下组分组成:黄芪提取物10~15份,单宁酸0.2~1份,纳米壳聚糖1~3份,丁酸钠3~5份,二氧化硅2~8份,余量为沸石粉载体。
本发明的另一技术解决方案是所述固体发酵含5-氨基乙酰丙酸的猪饲料的制备方法,其特殊之处在于,包括如下步骤:
⑴将各原料通过粉碎机粉碎后混匀;
⑵制备酵母菌种子液后,将其转接入酵母菌液体发酵培养基中,制备成酵母菌发酵液,将设定量的酵母菌发酵液加入水中,并加入设定量的葡萄糖、甘氨酸、丁二酸后,制备成固体发酵用菌液;
⑶固体发酵原料与固体发酵用菌液按照设定比例混合均匀,固体发酵36~72h后,发酵完成,烘干至水分12%以下;
⑷营养添加剂复合包与固体发酵饲料按照设定比例混匀,即获得固体发酵含5-氨基乙酰丙酸的猪饲料。
作为优选:所述步骤⑵进一步包括:
(2.1)制备酵母菌种子液时,将菌种在固体培养基中活化处理;固体培养基成分,按重量份由以下组分组成:葡萄糖1~3份,硫酸铵0.1~3份,无氨基酵母氮源0.1~1份,琼脂粉1~3份,30℃培养48~72h;
(2.2)活化后,菌种接入液体培养基,其培养基成分,按重量份由以下组分组成:葡萄糖1~3份,胰蛋白胨1~3份,酵母浸粉1~3份,氯化钠0.2~0.5份,30℃,220r/min培养24~48h。
作为优选:所述步骤(2.2)接续:
(2.3)制备酵母菌发酵液时,将培养好的种子液接入发酵罐中的液体发酵培养基,其发酵培养基成分,按重量份由以下组分组成:黄豆饼粉0.5~1.5份,柠檬酸铵1~2份,无水硫酸镁0.1~0.5份,七水合硫酸锌0.01~0.05份,无水葡萄糖1~3份,30℃,150r/min培养24~48h。
作为优选:所述步骤(2.3)接续:
(2.4)制备固体发酵用菌液时,于混合罐中加入相对于固体发酵原料质量50份的水,加入葡萄糖0.1~1份,甘氨酸0.1~1份,丁二酸0.1~1份,混匀后加入1~3份的酵母发酵液。
作为优选:所述步骤⑶进一步包括:
(3.1)固体发酵原料与固体发酵用菌液为1:0.3~1:0.6的比例。
作为优选:所述步骤⑷进一步包括:
(4.1)营养添加剂复合包与固体发酵饲料为1:9~2:8的比例。
本发明的再一技术解决方案是所述含5-氨基乙酰丙酸的猪饲料在断奶仔猪上的应用,其特殊之处在于,包括如下步骤:
⑴含5-氨基乙酰丙酸的猪饲料对断奶仔猪生长性能的影响:
(1.1)选取90头断奶仔猪进行为期35天的饲喂实验,初始体重为7.05±0.46kg,实验开始,这90头猪随机被分成3个处理组;每个处理组有3个重复;所述3个处理组分别为对照组,对照组:基础日粮,不添加含5-氨基乙酰丙酸的猪饲料,试验1组:基础日粮+0.5%含5-氨基乙酰丙酸的猪饲料;试验2组:基础日粮+1%含5-氨基乙酰丙酸的猪饲料;
(1.2)将饲喂过程分为三个阶段,第一阶段:0~7天、第二阶段:8~21天、第三阶段:22~35天,分别测定其每个阶段中各处理组断奶仔猪的平均日增重、平均日采食量和肉料比;
实验三个阶段和整个阶段:试验1组断奶仔猪的生长性能:ADG、ADF1和G:F要优于对照组,但差异不显著,说明基础日粮中添加0.5%的含5-氨基乙酰丙酸的猪饲料对断奶仔猪的生长性能没有太大的影响;而试验2组断奶仔猪的生长性能明显要优于对照组和试验1组,说明基础日粮中添加1%的含5-氨基乙酰丙酸的猪饲料能显著提高断奶仔猪的生长性能;
⑵含5-氨基乙酰丙酸的猪饲料对断奶仔猪血液指标的影响:
对于血液学指标,饲喂试验结束后,将断奶仔猪的血液样本收集到非肝素化和K3EDTA中,分别用真空管获得血清和全血样,血样随后4℃,2000×g离心30min后放入4℃冰箱内保存备用;
红血球、白血球用全自动血细胞分析仪测定,血红蛋白和血细胞比容采用全自动生化分析仪测定;用流式细胞仪测定淋巴细胞的数量;对照组和试验1组血液中各指标差异不大,试验2组血液中各指标含量较对照组和试验1组相比,均有较明显的提高,说明在基础饲料中添加1份的含5-氨基乙酰丙酸的猪饲料可以显著提高动物血液中血红蛋白、血细胞比容、红血球、白血球和淋巴细胞的含量。
与现有技术相比,本发明的有益效果:
⑴本发明利用酿酒酵母菌固体发酵生产含5氨基乙酰丙酸的饲料,在之前的研究中没有报道。
⑵本发明生产的含5-氨基乙酰丙酸的猪饲料,其5-氨基乙酰丙酸的含量可以达到338.87mg/kg,为目前研究中固体发酵中含量最高。
⑶本发明生产的含5-氨基乙酰丙酸的猪饲料,选用菌株为酿酒酵母,绿色无污染,较为环保。

具体实施方式
本发明下面将结合实施例作进一步详述:
【实施例1】含5-氨基乙酰丙酸的猪饲料制备方法,包括步骤:
⑴制备酵母菌种子液
(1.1)将菌种在固体培养基中活化处理;固体培养基成分为葡萄糖2%,硫酸铵0.5%,无氨基酵母氮源(YNB)0.15%,琼脂粉2%,30℃培养72h;
(1.2)活化后,菌种接入液体培养基,其培养基成分为葡萄糖2%,胰蛋白胨2%,酵母浸粉1%,氯化钠0.2%;在摇床中30℃,220r/min培养36h;
⑵制备酵母菌发酵液:
(2.1)将培养好的种子液接入发酵罐中的液体发酵培养基,其发酵培养基成分为黄豆饼粉1%,柠檬酸铵2%,无水硫酸镁0.1%,七水合硫酸锌0.01%,无水葡萄糖2%;温度30℃,空气流量30m3/h,转速150r/min,培养24h。
⑶制备固体发酵原料:
按以下重量份称取玉米粉60份,玉米蛋白粉5份,麸皮5份,酒糟25份,豆粕5份,粉碎后混匀;
⑷制备固体发酵用菌液:
(4.1)于混合罐中加入相对于固体发酵原料重量份水50份,加入葡萄糖1份,甘氨酸1份,丁二酸1份,混匀后加入2份的酵母发酵液:
⑸制备固体发酵饲料:
(5.1)将固体发酵原料与固体发酵用菌液按1:0.5的比例混匀后,平铺于发酵床上,固体发酵72h;
发酵完成后,将固体发酵饲料烘干至水分12%以下,即获得含5-氨基乙酰丙酸的猪饲料。
【实施例2】猪饲料中5-氨基乙酰丙酸含量的测定:
⑴所需试剂
乙酸盐缓冲液(pH=4.6):于700ml水中加入57ml冰乙酸,82g无水乙酸钠,溶解后加水至1000ml;
显色剂:于30ml冰乙酸中加入1g对-二甲氨基苯甲醛,5ml高氯酸,5ml水,溶解后用冰乙酸稀释至50ml,混匀。
(2)5-氨基乙酰丙酸标准曲线的制备
(2.1)配制5-氨基乙酰丙酸标准溶液:准确称取0.01280g标准样品。用水溶解后,移入100ml容量瓶中,稀释至刻度,此溶液1ml=0.10mg5-氨基乙酰丙酸。再用水稀释成1ml=10μg的标准应用溶液。
(2.2)取11支10ml具塞比色管,按表1配制标准管。

(2.3)向各管中加入2ml乙酸盐缓冲液,混匀,再加入0.4ml乙酰乙酸乙酯,充分混匀。于沸水浴中加热12min,取出于冷水中冷却至室温。向各管加入4ml乙酸乙酯,充分混匀,4,000r/min离心5min。
(2.4)各取2ml乙酸乙酯提取液(上层)于另11支10ml具塞比色管中,各加2ml显色剂,混匀,静置10min,在554nm处,用10mm比色杯,以0号管为参比测量吸光度。
注:10min内测完。
(2.5)以5-氨基乙酰丙酸含量为横坐标,以吸光度为纵坐标,绘制标准曲线如图1。
(3)猪饲料中5-氨基乙酰丙酸含量的测定
准确称取1.00g猪饲料,加入5mL水,混匀,4000r/min离心10min后,取上清0.5mL,加入1.5mL水,2mL乙酸盐缓冲液后,混匀,后加入0.4mL乙酰乙酸乙酯,混匀后沸水浴12min,冷却至室温,加入4mL乙酸乙酯,混匀后4000r/min离心10min,取上层萃取液2mL与显色剂2mL混匀后,测定其554nm处的吸光度值。通过标准曲线上的方程,计算出猪饲料中5-氨基乙酰丙酸的含量为338.87mg/kg。
【实施例3】含5-氨基乙酰丙酸的猪饲料在断奶仔猪上的应用
⑴含5-氨基乙酰丙酸的猪饲料对断奶仔猪生长性能的影响
(1.1)选取90头断奶仔猪进行为期35天的饲喂实验,初始体重(BW)为7.05±0.46kg,实验开始,这90头猪随机被分成3个处理组。每个处理组有3个重复。基础日粮成分见表2。这3个处理组分别为对照组(基础日粮,不添加含5-氨基乙酰丙酸的猪饲料)、试验1组(基础日粮+0.5%含5-氨基乙酰丙酸的猪饲料)、试验2组(基础日粮+1%含5-氨基乙酰丙酸的猪饲料)。
(1.2)将饲喂过程分为三个阶段,第一阶段(0~7天)、第二阶段(8~21天)、第三阶段(22~35天),分别测定其每个阶段中各处理组断奶仔猪的平均日增重(ADG)、平均日采食量(ADF1)和肉料比(G:F)(表3)
结果显示,实验三个阶段和整个阶段:试验1组断奶仔猪的生长性能(ADG、ADF1和G:F)要优于对照组,但差异不显著,说明基础日粮中添加0.5%的含5-氨基乙酰丙酸的猪饲料对断奶仔猪的生长性能没有太大的影响;而试验2组断奶仔猪的生长性能明显要优于对照组和试验1组,说明基础日粮中添加1%的含5-氨基乙酰丙酸的猪饲料能显著提高断奶仔猪的生长性能。
⑵含5-氨基乙酰丙酸的猪饲料对断奶仔猪血液指标的影响
对于血液学指标,饲喂试验结束后,将断奶仔猪的血液样本收集到非肝素化和K3EDTA中,分别用真空管获得血清和全血样。血样随后4℃,2000×g离心30min后放入4℃冰箱内保存备用。
RBC(红血球)、WBC(白血球)用全自动血细胞分析仪(瑞典麦道尼克CA620)测定,Hb(血红蛋白,hemoglobin)和HCT(血细胞比容,hematocrit)采用全自动生化分析仪(日立7180)测定;用流式细胞仪(Guave6HT)测定Lymphocyte(淋巴细胞)的数量(表4)。对照组和试验1组血液中各指标差异不大,试验2组血液中各指标含量较对照组和试验1组相比,均有较明显的提高,说明在基础饲料中添加1%的含5-氨基乙酰丙酸的猪饲料可以显著提高动物血液中Hb、HCT、RBC、WBC和Lymphocyte的含量。
【实施例4】含5-氨基乙酰丙酸的猪饲料对致病菌的抑菌效果
⑴于培养皿中挑取致病菌金黄色葡萄球菌ATCC6538、大肠杆菌ATCC25922和肠炎沙门氏菌ATCC13076,在接种于新鲜LB液体培养基中,置于37℃恒温箱中培养了18~24h时制成菌液备用;
⑵称取1g含5-氨基乙酰丙酸的猪饲料,溶于5mL蒸馏水中,用漩涡振荡仪振荡混匀后,4000r/min离心10min,取离心后的上清液,即为含5-氨基乙酰丙酸的猪饲料的浸出液,用打孔法检测浸出液对3种致病菌的抑菌效果。
由表5可知,含5-氨基乙酰丙酸的猪饲料对致病菌具有良好的抑制作用。说明,在日粮中添加含5-氨基乙酰丙酸的猪饲料可以很好地抑制动物肠道中的致病菌,增加动物免疫,减少动物疾病的发生。

1.一种固体发酵含5-氨基乙酰丙酸的猪饲料,其特征在于,按重量份由以下组分组成:玉米粉50~60份,玉米蛋白粉2.5~5份,麸皮2.5~5份,酒糟20~25份,豆粕2.5~5份。
2.根据权利要求1所述固体发酵含5-氨基乙酰丙酸的猪饲料,其特征在于,还包括营养添加剂复合包,所述营养添加剂复合包按重量份由以下组分组成:黄芪提取物10~15份,单宁酸0.2~1份,纳米壳聚糖1~3份,丁酸钠3~5份,二氧化硅2~8份,余量为沸石粉载体。
3.一种固体发酵含5-氨基乙酰丙酸的猪饲料的制备方法,其特征在于,包括如下步骤:
⑴将各原料通过粉碎机粉碎后混匀;
⑵制备酵母菌种子液后,将其转接入酵母菌液体发酵培养基中,制备成酵母菌发酵液,将设定量的酵母菌发酵液加入水中,并加入设定量的葡萄糖、甘氨酸、丁二酸后,制备成固体发酵用菌液;
⑶固体发酵原料与固体发酵用菌液按照设定比例混合均匀,固体发酵36~72h后,发酵完成,烘干至水分12%以下;
⑷营养添加剂复合包与固体发酵饲料按照设定比例混匀,即获得固体发酵含5-氨基乙酰丙酸的猪饲料。
4.根据权利要求3所述固体发酵含5-氨基乙酰丙酸的猪饲料的制备方法,其特征在于,所述步骤⑵进一步包括:
(2.1)制备酵母菌种子液时,将菌种在固体培养基中活化处理;固体培养基成分,按重量份由以下组分组成:葡萄糖1~3份,硫酸铵0.1~3份,无氨基酵母氮源0.1~1份,琼脂粉1~3份,30℃培养48~72h;
(2.2)活化后,菌种接入液体培养基,其培养基成分,按重量份由以下组分组成:葡萄糖1~3份,胰蛋白胨1~3份,酵母浸粉1~3份,氯化钠0.2~0.5份,30℃,220r/min培养24~48h。
5.根据权利要求4所述固体发酵含5-氨基乙酰丙酸的猪饲料的制备方法,其特征在于,所述步骤(2.2)接续:
(2.3)制备酵母菌发酵液时,将培养好的种子液接入发酵罐中的液体发酵培养基,其发酵培养基成分,按重量份由以下组分组成:黄豆饼粉0.5~1.5份,柠檬酸铵1~2份,无水硫酸镁0.1~0.5份,七水合硫酸锌0.01~0.05份,无水葡萄糖1~3份,30℃,150r/min培养24~48h。
6.根据权利要求5所述固体发酵含5-氨基乙酰丙酸的猪饲料的制备方法,其特征在于,所述步骤(2.3)接续:
(2.4)制备固体发酵用菌液时,于混合罐中加入相对于固体发酵原料重量份的水50份,加入葡萄糖0.1~1份,甘氨酸0.1~1份,丁二酸0.1~1份,混匀后加入1~3份的酵母发酵液。

(3.1)固体发酵原料与固体发酵用菌液为1:0.3~1:0.6的比例。
8.根据权利要求3所述固体发酵含5-氨基乙酰丙酸的猪饲料的制备方法,其特征在于,所述步骤⑷进一步包括:
(4.1)营养添加剂复合包与固体发酵饲料为1:9~2:8的比例。
9.一种含5-氨基乙酰丙酸的猪饲料在断奶仔猪上的应用,其特征在于,包括如下步骤:
⑴含5-氨基乙酰丙酸的猪饲料对断奶仔猪生长性能的影响:
