Description

Scalp care composition containing hyaluronic acid and preparation method and application thereof
Technical Field
The invention relates to a scalp care composition containing hyaluronic acid, a preparation method and application thereof, and belongs to the technical fields of biological medicines and cosmetics.
Background
There are many kinds of facial skin care products on the market, but the maintenance type products for the scalp are not many, the scalp is exposed to the environment every day, and the scalp problems are more and more along with the aggravation of pollution, so that the development of a product capable of being used in the scalp is more important for protecting the scalp.
Hyaluronic acid, also known as hyaluronic acid, is a high-molecular polysaccharide naturally present in the intercellular spaces of human tissues, is formed of alternating units of D-glucuronic acid and N-acetylglucosamine in a linear chain, is widely used in the fields of medicines, skin care products, foods and the like, and has high safety. The hyaluronic acid molecule is in a rigid spiral column shape in space, strong water absorption is generated on the inner side of the column due to the existence of a large number of hydroxyl groups, and on the other hand, a hydrophobic region is formed on a hyaluronic acid molecular chain due to the continuous directional arrangement of the hydroxyl groups, so that the hyaluronic acid can form a three-dimensional network structure. Hyaluronic acid HAs a variety of different molecular weights, and HA of each molecular weight HAs different efficacy and performance. At present, hyaluronic acid also has good application in hair washing and protecting products. However, hyaluronic acid has strong water solubility, and the surface of skin and hair is generally negatively charged, and hyaluronic acid is also generally negatively charged, so that hyaluronic acid has poor adsorbability on skin or hair, and thus cannot be well acted on skin or hair.
In order to solve this problem, the most commonly used method at present is to modify hyaluronic acid with cations, and the hyaluronic acid is chemically modified by a cationic polymer to be charged with cations, so as to increase the adhesion on skin and hair, and to perform the function better. For example, CN200680044541.8 and CN2088001002.7 adopt chemical modification to cationize hyaluronic acid, but these methods are complicated in process and high in cost.
Hyaluronic acid can also be increased in association with living tissue (skin) by mixing or complexing hyaluronic acid with a cationic polymerSkin care productsOr hair), as disclosed in patent CN105853254A, at least one cationic polymer and at least one hyaluronic acid with molecular weight are mixed and subjected to complexation, and the obtained product can stably take on a liquid state through the control of temperature, pH and the like, which simplifies the preparation process and saves cost, however, such compositions are only suitable for surfactant systems, and when used in non-surfactant systems, a stable and uniform system cannot be formed due to the fact that no surfactant is dispersed, thereby precipitating or generating precipitates.
Disclosure of Invention
In order to solve the problems, the invention provides a scalp care composition containing hyaluronic acid, in particular to a scalp care composition suitable for a non-superficial living system, the composition is prepared by compounding a cationic surfactant, anionic hyaluronic acid and salt thereof and tetrahydro-methyl pyrimidine carboxylic acid, the components are matched with each other, a critical flocculation state can be formed in the non-superficial living system, the stability is good, and the obtained composition also has the effects of caring scalp and repairing skin barrier, and can well improve symptoms such as scalp pruritus, alopecia and the like.
The specific technical scheme of the invention is as follows:
a scalp care composition containing hyaluronic acid comprises hyaluronic acid and its salt, cationic surfactant and tetrahydro-methyl pyrimidine carboxylic acid.
Further, the hyaluronic acid and the salt thereof are a mixture of high molecular hyaluronic acid and the salt thereof and low molecular hyaluronic acid and the salt thereof, or a mixture of high molecular hyaluronic acid and the salt thereof, low molecular hyaluronic acid and the salt thereof, and hydrolyzed hyaluronic acid and the salt thereof. When two combination modes of hyaluronic acid and hyaluronic acid salt are selected, the composition can achieve good stability.
The invention selects cationic surfactant, tetrahydro-methyl pyrimidine carboxylic acid, hyaluronic acid and salt thereof as effective components. In the research, the cationic surfactant contains cations, hyaluronic acid and salts thereof and tetrahydro-methyl pyrimidine carboxylic acid carry anions, when hyaluronic acid and salts thereof and the cationic surfactant coexist in a non-surface active system such as water, precipitation is formed immediately, the appearance and the stability of the product are influenced, and the application of the components in the non-surface active system is also prevented. However, when the three components are compounded, the composition obtained by compounding the cationic surfactant, the tetrahydromethyl pyrimidine carboxylic acid, the hyaluronic acid and the salt thereof can form a critical flocculation state in a non-surface active system, and does not generate precipitates. In addition, the adhesion of hyaluronic acid and its salt, and tetrahydro-methyl pyrimidine carboxylic acid on hair or scalp are poor, when the cationic surfactant exists, the cationic content of the system is increased, the adhesion of each component in the composition on scalp and hair is increased, the retention time of the composition on scalp and hair is increased, and the functions of each component can be better played.
Further, the high molecular hyaluronic acid and the salt thereof refer to high molecular hyaluronic acid, high molecular hyaluronate, or a combination thereof. The molecular weight of the high molecular hyaluronic acid and the salt thereof is 1000-1800 kDa. The mass percentage of the polymer hyaluronic acid and the salt thereof in the scalp care composition is 0.01 to 10%, preferably 0.01 to 5%, and more preferably 0.5 to 5%. When the preferable content is selected, the scalp care effect is more excellent.
Further, the low molecular hyaluronic acid and the salt thereof refer to low molecular hyaluronic acid, low molecular hyaluronate, or a combination thereof. The molecular weight of the low molecular hyaluronic acid and the salt thereof is 30kDa-100 kDa. The mass percentage of the low molecular weight hyaluronic acid and the salt thereof in the scalp care composition is 0.5-10%, preferably 0.5-5%. When the preferable content is selected, the scalp care effect is more excellent.
Further, the hydrolyzed hyaluronic acid and salts thereof refer to hydrolyzed molecular hyaluronic acid, hydrolyzed molecular hyaluronate, or a combination thereof. The molecular weight of the hydrolyzed hyaluronic acid and the salt thereof is 1kDa-10 kDa. The mass percentage of the hydrolyzed hyaluronic acid and the salt thereof in the scalp care composition is 0 to 10%, preferably 0.05 to 10%, and more preferably 0.1 to 2%. When the preferable content is selected, the scalp care effect is more excellent.
Further, the salts of the high molecular hyaluronic acid, the low molecular hyaluronic acid and the hydrolyzed hyaluronic acid are sodium salts, potassium salts, calcium salts or zinc salts of the high molecular hyaluronic acid, the low molecular hyaluronic acid and the hydrolyzed hyaluronic acid.
Further, the cationic surfactant means that the hydrophilic group linked to the lipophilic group is a surfactant having a cation. Cationic surfactants are widely used in hair products, and are mainly used for conditioning hair, improving the combing property of hair, increasing the softness of hair and eliminating static electricity. The cationic surfactant may be various types of cationic surfactants reported in the art, and may be, for example, one or both of cationic guar gum and polyquaternium. The cationic guar gum is guar gum hydroxypropyl trimethyl ammonium chloride, and the polyquaternary ammonium salt can be one or two of polyquaternary ammonium salt-7 and polyquaternary ammonium salt-10.
Further, the mass percent of the cationic surfactant is 0.05-5%, preferably 0.1-2%.
Furthermore, the systematic name of the tetrahydromethylpyrimidine carboxylic acid, namely the ectoine, is 2-methyl-1, 4,5, 6-tetrahydropyrimidine-4-carboxylic acid, and the tetrahydromethylpyrimidine carboxylic acid is not only an important osmotic pressure compensation solute, but also has good protection effect on cells and biological macromolecules (biological membranes, proteins, enzymes and nucleic acids) under the stimulation of adverse environments such as high temperature, high salt, freezing, drying, radiation and the like. In the composition system, the adhesion of the tetrahydro-methyl pyrimidine carboxylic acid and the hyaluronic acid and the salt thereof of the invention on hair and scalp is increased, and the tetrahydro-methyl pyrimidine carboxylic acid and the hyaluronic acid and the salt thereof of the invention can stay on scalp hair for a longer time.
Further, the weight percentage of the tetrahydromethylpyrimidine carboxylic acid is 0.1 to 5%, preferably 0.1 to 3%, more preferably 0.1 to 1%, and still more preferably 0.5 to 1%.
Further, the scalp care composition further comprises a preservative, a polyhydric alcohol and water. The preservative may be selected from phenoxyethanol, benzoic acid or potassium sorbate. The polyol may be selected from propylene glycol, glycerol, butylene glycol or pentylene glycol.
Further, the preservative is 0.1-2% by mass, preferably 0.1-1% by mass.
Further, the mass percent of the polyhydric alcohol is 1-5%, preferably 2-5%.
Further, the scalp care composition described above contains water to make up 100% in addition to other components.
Furthermore, the preparation method of the scalp care composition containing hyaluronic acid is simple, and the method can adopt the following steps: mixing the cationic surfactant with water, stirring until the cationic surfactant and the water are completely dissolved, then sequentially adding hyaluronic acid and salt thereof and tetrahydro-methylpyrimidine carboxylic acid into the solution, and uniformly stirring to obtain the hyaluronic acid.
Furthermore, the preparation method comprises the following steps: mixing the cationic surfactant with water, stirring to dissolve completely, sequentially adding high molecular hyaluronic acid and its salt, low molecular hyaluronic acid and its salt, hydrolyzed hyaluronic acid and its salt (if any), and tetrahydro-methylpyrimidine carboxylic acid, and stirring.
Further, when the composition also contains a preservative and a polyhydric alcohol, the preparation method comprises the following steps: mixing the cationic surfactant with water, stirring to completely dissolve, adding antiseptic and polyalcohol into the solution, stirring, sequentially adding high molecular hyaluronic acid and its salt, low molecular hyaluronic acid and its salt, hydrolyzed hyaluronic acid and its salt (if any) and tetrahydro-methyl pyrimidine carboxylic acid, and stirring.
The scalp care composition containing hyaluronic acid well solves the problem that when a cationic surfactant coexists with hyaluronic acid and tetrahydro-methyl pyrimidine carboxylic acid, the cationic surfactant is easily precipitated when added into a non-surfactant system, can be used in the non-surfactant system, and is more suitable for hair care products of the non-surfactant system; meanwhile, the adhesion of cations on the scalp or the hair can be obviously improved, the adhesion of hyaluronic acid and tetrahydro-methyl pyrimidine carboxylic acid on the scalp or the hair can be improved, so that the adsorbability of the composition on the skin or the hair can be effectively improved, the retention time of the composition on the scalp and the hair can be prolonged, the scalp-care shampoo has good effects of nourishing the scalp, improving deep water environment of the scalp and repairing damaged scalp barriers, and has good application prospects. Based on the excellent performance and effect, the invention also provides the application of the scalp care composition containing the hyaluronic acid in hair products.

Wherein the high molecular sodium hyaluronate, the low molecular sodium hyaluronate, the hydrolyzed sodium hyaluronate and the tetrahydro-methylpyrimidine carboxylic acid are all from Huaxi Biotechnology corporation.
Examples 1 to 13 and comparative examples 1 to 4
The scalp care composition containing hyaluronic acid consists of high molecular sodium hyaluronate (1000 kDa-1800 kDa), low molecular sodium hyaluronate (30 kDa-100 kDa), hydrolyzed sodium hyaluronate (1 kDa-10 kDa), a cationic surfactant, tetrahydromethyl pyrimidine carboxylic acid, phenoxyethanol, glycerol and water. Wherein, the content of phenoxyethanol is 0.9wt%, the content of glycerin is 4wt%, water is used as solvent to make up the balance, and the other 5 effective components are shown in the following table 1.
Figure 443761DEST_PATH_IMAGE001
The preparation method comprises the following steps:
weighing the components according to a formula table for later use; adding a cationic surfactant with a formula amount into a beaker containing a certain amount of water, stirring until the cationic surfactant is completely dissolved, then adding phenoxyethanol with the formula amount, then adding glycerol, stirring for 5 minutes, adding high-molecular-weight sodium hyaluronate, stirring until the high-molecular-weight sodium hyaluronate is completely dissolved, adding low-molecular-weight sodium hyaluronate, stirring until the low-molecular-weight sodium hyaluronate is completely dissolved, adding hydrolyzed sodium hyaluronate (if any), stirring until the high-molecular-weight sodium hyaluronate is completely dissolved, adding tetrahydro-methyl pyrimidine carboxylic acid, and uniformly stirring to obtain the hyaluronic acid scalp care composition.
Verification example 1 stability test
5g of the composition of example 1 was added with 95g of water, and stirred uniformly to obtain a 5% essence of example 1. According to this method, 5% solutions of each example and comparative example were prepared. An accelerated test was conducted at 45 ℃ to observe whether or not there was precipitation.
Figure 104550DEST_PATH_IMAGE002
From the experimental results, under the condition that a certain content of the tetrahydromethyl pyrimidine carboxylic acid exists, the composite sodium hyaluronate, the cationic surfactant and the tetrahydromethyl pyrimidine carboxylic acid are added into a non-surfactant system, namely the system is stable without precipitation, which shows that the composition can be suitable for the non-surfactant system. The combination of comparative example 1 without the tetrahydro-methyl pyrimidine carboxylic acid and the combination of comparative example 3 without the compound sodium hyaluronate cannot form a critical flocculation state because the ion balance among the three is not achieved, so that precipitation is not suitable for a non-surface active system, and is most obvious in the precipitation of the comparative example 1; comparative example 2 also generated white flocculent precipitate in one month due to the lower content of the tetrahydro-methylpyrimidine carboxylic acid, and comparative example 4 instantly generated white flocculent precipitate in the preparation process due to the excessively high content of the cationic surfactant.
Verification example 2 skin barrier repair experiment
Cell wound repair regular scratch regions were formed in 24-well culture plates using ibidi chambers, and the effect of the samples on the ability of human epidermal keratinocytes (HaCaT) to migrate to the scratch regions was examined to evaluate whether the samples had the ability to repair wounds.
In a new 24-well plate, an ibidi cell was placed, and human epidermal keratinocytes HaCaT in logarithmic growth phase were taken at 3X 105Density of one/mLThe cells were inoculated into an ibidi chamber in a 24-well plate, 70. mu.L of each cell suspension was added to each of the left and right wells of the chamber, 400. mu.L of complete medium was supplemented outside the chamber, and the cells were cultured at 37 ℃ under 5% CO2 for 24 hours. The chamber was removed, the old culture medium was aspirated off, and 1mL of sample solution was added. The sample solution was: the samples of examples 1 to 13 and comparative examples 1 to 3 were prepared at a concentration of 5% (v/v) using DMEM high-sugar medium, and were sterilized by filtration using a 0.22 μm filter, wherein the sample of comparative example 1 was stirred to disperse flocs at the time of use. The control group was replaced with culture medium without sample. After further incubation for 24h, photographs were observed. Results mobility was calculated by analysis using Image J software.
Mobility = area of scratch area of experimental group/area of scratch area of control group × 100%
Figure 13862DEST_PATH_IMAGE003
The data in the table show that the composition prepared by the embodiment of the invention can well repair damaged skin barriers, and particularly, the composition has a good repairing effect on the skin barriers when the high-molecular sodium hyaluronate, the low-molecular sodium hyaluronate, the hydrolyzed sodium hyaluronate and the tetrahydro-methyl pyrimidine carboxylic acid are in a certain proportioning range, and the cationic surfactant has no obvious influence on the repairing effect on the skin barriers.

After cleaning, respectively drying by using a blower, fixing one end of the hair bundle on a wood board, allowing the hair bundle to naturally droop, combing for 10 times by using a comb, and recording the opening angle (namely the upwarping angle) of the hair bundle, wherein the larger the opening angle is, the larger the static electricity is, and the more the cations are.

3. Results of the experiment
From the above results, the angle of opening the hair is the largest in example 1, which shows that the hair comb has a large amount of charges, strong repulsive force and more cations, and the comparison shows that the examples can better increase the adhesion of the cations, thereby effectively increasing the adsorption of the composition to the skin or hair, improving the retention time of the composition on the scalp and hair, and being beneficial to the absorption and utilization of the composition.
