Description

Feed additive for relieving oxidative damage of piglet liver caused by oxidized oil in feed of lactating sow
Technical Field
The invention belongs to the field of agricultural and livestock application, and particularly relates to a feed additive for relieving oxidative damage of piglet liver caused by oxidation oil in the feed of a lactating sow.
Background
Good feeding management is important for improving the production performance of animals, and high-quality feed is an important guarantee for successful cultivation. The fat is added into the feed, and because the energy content is higher, the fat-soluble vitamins are beneficial to transportation, absorption and utilization in blood, the heat consumption is reduced, dust is prevented, the palatability of the feed is improved, and the nutritional requirements of pigs can be better met. The value of oil in feed has been increasingly recognized. By adding the grease into the feed of the lactating sow, the content of energy substances in milk can be increased, the lactation yield can be increased, the energy intake of piglets can be increased, the daily gain and the survival rate of the piglets can be improved, and the phenomenon of oestrus failure caused by the excessive weight loss of the sow during lactation can be reduced.
However, the polyunsaturated fatty acids contained in the oil are easily oxidized during storage and transportation, and generate various toxic primary and secondary oxidation products, so that the oil is deteriorated and smelled, and the nutritional value of the feed is reduced. The oxidation products of the oil can be combined with biological macromolecules such as protein, nucleic acid, lipid and the like in an animal body to destroy the biological activity of the oil, disturb the oxidation-reduction state of the organism, influence the reproductive performance, reduce the production performance of the animal and influence the quality of animal products.
In order to prevent the adverse effect caused by the oxidation of oil and fat when the oil and fat are added into the feed, the addition of an antioxidant into the feed is an effective measure, and the antioxidant applied to the feed comprises an artificial synthetic antioxidant and a natural antioxidant. With the increasing demand for green, organic, and pollution-free foods, the demand for antioxidants has gradually shifted from synthetic antioxidants to natural antioxidants. Resveratrol, as a natural antioxidant, is widely present in plants such as grapes, plums, berries, peanuts and the like, is easy to absorb by oral administration, has multiple health-promoting effects such as oxidation resistance, inflammation resistance and the like in terms of both the resveratrol and metabolic derivatives thereof, and has been widely concerned by people.
Disclosure of Invention
Based on the defects, the invention provides the feed additive for relieving oxidative damage of the piglet liver caused by the oxidation of the oil in the feed of the lactating sow, and aims to enhance the oxidation resistance of the piglet liver and deal with the invasion of oxidative stress to the piglet under the condition that the oil in the feed is oxidized.
The technical scheme adopted by the invention is as follows: a feed additive for relieving oxidative damage of piglet liver caused by oxidation oil in the feed of a lactating sow comprises the following components in percentage by weight: consists of 85 percent of resveratrol, 10 percent of DL-alpha-tocopheryl acetate, 1 percent of saccharin sodium, 1 percent of citric acid, 2 percent of L-carnitine and 1 percent of zeolite powder.
The invention also has the following features:
1. the purity of the resveratrol in the formula of the feed additive is more than or equal to 98 percent.
2. The purity of DL-alpha-tocopherol acetate in the formula of the feed additive is more than or equal to 93 percent.
3. The preparation method of the feed additive comprises the following steps:
1) crushing resveratrol, saccharin sodium, citric acid, L-carnitine and DL-alpha-tocopherol acetate, and sieving by a 60-mesh sieve for later use;
2) pulverizing zeolite powder, and sieving with 40 mesh sieve;
3) weighing the raw materials according to the proportion of the formula, uniformly mixing the raw materials in a mixer for 5-10 minutes, and quantitatively packaging to obtain the finished product.
4. The feed additive is used from 107 days of gestation of the sow to the weaning period of the piglet, and the addition amount is 0.04 percent by weight.
The invention has the advantages that: the feed additive can enhance the activity of the antioxidant enzyme of the liver of the suckling piglet and simultaneously reduce the accumulation of oxidation products in the liver tissue.
Detailed Description
The invention is further illustrated by the following examples:
example 1
A feed additive for relieving oxidative damage of piglet liver caused by oxidation oil in the feed of lactating sow is used in the feed of sow from 107 days of gestation to weaning period of piglet.
1. Test diet composition
The basal feed for the sows is prepared by referring to nutritional needs of sows at the later gestation stage and the lactation stage of NRC (2012) in the United states, pig feeding standards issued by the Ministry of agriculture in China in 2004 and Chinese feed ingredient and nutritional value table (31 th edition in 2020). The composition and main nutrient level of each diet are shown in table 1. The first and second test groups used oxidized soybean oil instead of fresh soybean oil in the control group. Meanwhile, the second test group adds 0.04% of the feed additive of the invention to replace corn on the basis of the first test group.
TABLE 1 basic diet composition and nutritional levels (air-dried basis)
Figure BDA0003116565900000031
Note: the vitamin and trace element premix in the formula is as follows: each kilogram of daily ration comprises: 10mg of copper; 108mg of zinc; 0.3mg selenium; 116mg of manganese; 180mg of iron; 0.4mg iodine. Each kilogram of daily ration comprises: vitamin a,14000 IU; vitamin D32000 IU; vitamin E,140 IU; vitamin K33.6 mg; vitamin B12 mg; vitamin B25.2 mg; vitamin B63.6 mg; vitamin B120.015 mg; 0.03mg of vitamin H; pantothenic acid, 15 mg; niacin, 24 mg; folic acid, 0.9 mg.
2. Design of experiments
The experiment adopts single-factor design, 18 sows with similar weight and gestation times for 107 days are selected and randomly divided into 3 groups, each group has 6 repetitions, and each repetition has 1 sow. The control group is fed with basic diet, the test group uses oxidized soybean oil (POV of 220mEq/kg) to replace fresh soybean oil (POV of 11.5mEq/kg) in the control group, the test group II is added with the feed additive (the addition amount is 0.04%) on the basis of the test group I, and the influence of the addition of resveratrol on the oxidation resistance of the livers of piglets is researched under the condition that the oxidized soybean oil exists in the diet of lactating sows.
3. Data statistics
SPSS 23.0 software is adopted for data statistics to carry out one-factor variance analysis, a Duncan method is used for carrying out multiple comparison, and the result is expressed by the mean value +/-standard error.
4. Test results

Figure BDA0003116565900000041
Note: the same row is shouldered with different letters to indicate significant difference (P < 0.05).
As can be seen from table 2, in the first test group, compared to the control group, i.e., when oxidized soybean oil is contained in the feed for lactating sows, the activities of SOD, GSH-Px and CAT in the liver tissue of piglets are significantly reduced (P <0.05), but when 300mg/kg of resveratrol is added to the feed containing oxidized soybean oil, the activities of SOD, GSH-Px and CAT in the liver tissue are restored to the levels of the corresponding indexes in the control group. The result shows that the feed additive is beneficial to improving the problem that the activity of the antioxidant enzyme of the offspring is low because the sows oxidize the feed.
TABLE 3 suckling piglet liver oxidation product content
Figure BDA0003116565900000042
Note: the same row is shouldered with different letters to indicate significant difference (P < 0.05).
As can be seen from Table 3, the oxidation products MDA and H in the liver tissue of the piglets of the first test group are significantly increased compared with those of the control group2O2Level (P)<0.05), resveratrol helps to reduce liver tissue MDA and H2O2At a level, this result indicates that the feed supplement of the present invention helps to reduce the accumulation of oxidation products in the liver tissue of the offspring.
