Performance test 1
Safety test (human skin patch test)
24 healthy subjects with no history of skin allergy between the ages of 25 and 50 were selected for patch testing in a closed patch test.
The specific spot sticking method comprises the following steps:
selecting a qualified spot tester, dropping 15 mu L of the anti-aging composition of the invention in the examples 1-7 and the comparative examples 1-5 into the spot tester, externally sticking a special adhesive tape on the back of a subject, sticking 12 spot testers on each subject, respectively sticking the anti-aging composition essence, removing the subject after 24 hours, observing skin reactions after 0.5, 6, 12, 24 and 48 hours after removal, and recording the results according to the skin reaction grading standard in the cosmetic hygiene Specification (2015 edition).
The classification criteria are shown in table 1 below:

the results of the human skin patch test show that all the subjects pass the patch test and observe skin reactions at 0.5 hour, 6 hours, 12 hours, 24 hours and 48 hours respectively, wherein 0 case has adverse reactions such as erythema, pimple and blister, and the like, which indicates that the anti-aging compositions provided by the examples and the comparative examples in the invention have high safety and no stimulation.
Performance test 2
The anti-aging compositions of examples 1-7 and comparative examples 1-5 were tested for oxidation resistance, saccharification resistance, and contamination resistance, specifically as follows:
(1) oxidation resistance test
Evaluation of DPPH radical scavenging ability: the ability to scavenge DPPH (1, 1-diphenyl-2-trinitrophenylhydrazine) free radicals is to some extent the antioxidant ability of the reaction mass. The greater the free radical clearance rate, the stronger the antioxidant capacity and the stronger the anti-aging capacity. Therefore, the anti-aging effect of the anti-aging composition can be judged by studying the ability of the anti-aging composition to scavenge DPPH free radicals.
The samples were diluted with DMSO to 2% mass fraction of the sample to be tested and the following tests were performed:

the DPPH free radical clearance is calculated by the formula:

(2) Anti-contamination test
Detecting the content of Reactive Oxygen Species (ROS) in human keratinocytes treated with the particulate matter, specifically comprising the following steps:
cells were plated at 1.0X 105cells/mL were seeded onto a well plate at a density and after incubation for 2 hours at 37 deg.C, the cells were exposed to 100. mu.g/mL PM (particulate matter) and 2.5. mu.g/mL BaP. After incubation at 37 ℃ for 1 hour, detection and quantitation of fluorescence was performed using LS-5B spectrofluorescence (Perkinelmer, Waltham, MA, USA).
Since moderate and high doses of ROS induce apoptosis and even cause necrosis of cells through oxidative stress of cells, measuring the accumulation of ROS in epidermal keratinocytes reflects the anti-contamination ability of the test samples, and the test results are shown in Table 2.
(3) Anti-glycation assay
The advanced glycation end product removal experiment test is adopted for testing, and the following reagents are respectively prepared: 0.1mol/L NaOH solution, 0.05mol/L PBS phosphate buffer solution (pH7.4), 10mg/mL bovine serum albumin solution, 45mg/mL glucose solution, 0.25mmol/L aminoguanidine hydrochloride solution and 200mg/mL solution of a sample to be tested.
PBS phosphate buffer solution, bovine serum albumin solution and glucose solution are placed in a constant temperature and humidity incubator, then the sample solution to be tested and aminoguanidine hydrochloride solution are added, and incubation is carried out for 40h at the temperature of 60 ℃. And taking out each sample, detecting the samples by using an enzyme-linked immunosorbent assay, wherein the fluorescence excitation/emission wavelength is 370/440nm, and recording the fluorescence absorption value. 3 replicates of each sample were taken and the results averaged, where the inhibition of AEGs was calculated using the following formula:

The results obtained are shown in table 2 below:

The anti-aging composition provided by the invention has obvious antioxidant, anti-photoaging and anti-saccharification effects, the DPPH free radical clearance rate of the anti-aging composition is 17.6-23.6%, the ROS epidermal keratinocyte accumulation amount is 15.6-16.8%, and the AEGs inhibition rate is 48.5-52.3%, which shows that the anti-aging composition can effectively eliminate free radicals, inhibit the generation of ROS in human epidermal keratinocytes under pollution conditions, further protect skin cells from the pressure of environmental pollution, and inhibit saccharification reaction at the same time. Therefore, the anti-aging composition provided by the invention can realize anti-aging effect from multiple aspects, and enables the skin to be young and healthy.
Application example 1
The application example provides an all-dimensional anti-aging face cream.
Mixing the anti-aging composition provided in example 1 with a cream base to prepare an all-round anti-aging cream A, wherein the anti-aging composition accounts for 30% by mass;
the cream matrix is a basic skin care product without other effects, and comprises the following main components in percentage by mass: 3% of glycerol, 2% of butanediol, 1% of acrylic acid (ester)/C10-30 alkanol acrylate cross-linked polymer, 0.5% of behenyl alcohol, 0.5% of stearyl alcohol, 0.5% of PEG-60 glyceryl isostearate, 0.5% of PEG-5 glyceryl stearate, 0.02% of phenoxyethanol, 0.02% of potassium hydroxide, 0.5% of disodium EDTA and the balance of water.
Application example 2
The application example provides the all-round anti-aging toning lotion.
Mixing the anti-aging composition provided in example 1 with a lotion base to prepare an all-round anti-aging lotion, wherein the anti-aging composition accounts for 30% by mass;
the cosmetic water matrix is a basic skin care product without other effects, and comprises the following main components in percentage by mass: 3% glycerol, 2% disodium EDTA, 1% citric acid, 1% sodium citrate, 1.5% 1, 2-pentanediol, 0.1% polysorbate-20 and 0.02% phenoxyethanol, the balance being water.
Application example 3
The application example provides an all-dimensional anti-aging face cream.
Mixing the anti-aging composition provided in example 1 with an emulsion matrix to obtain an all-directional anti-aging emulsion, wherein the anti-aging composition accounts for 30% by mass;
the emulsion matrix is a basic skin care product without other effects, and comprises the following main components in percentage by mass: 3% glycerol, 2% butylene glycol, 2% disodium EDTA, 1% carbomer, 0.5% PEG-5 glyceryl stearate, 0.5% acrylic/C10-30 alkanol acrylate crosspolymer, 0.5% citric acid, 0.5% sodium citrate, 0.1% polysorbate-20, 0.1% stearyl alcohol, 0.1% xanthan gum, 0.02% potassium hydroxide, and 0.02% phenoxyethanol, with the balance being water.
Application examples 4 to 7
An all-round anti-aging cream was provided, which was different from application example 1 in that the anti-aging composition provided in example 1 was replaced with the anti-aging compositions provided in examples 4 to 7, to obtain anti-aging cream B (application example 4), cream C (application example 5), cream D (application example 6), and cream E (application example 7).
Application comparative examples 1 to 5
An anti-aging cream is provided, which is different from application example 1 in that the anti-aging composition provided in example 1 is replaced with the anti-aging compositions provided in comparative examples 1 to 5 to obtain anti-aging cream F (application comparative example 1), cream G (application comparative example 2), cream H (application comparative example 3), cream I (application comparative example 4) and cream J (application comparative example 5).
Performance test 3
Clinical test of crow's feet
200 female volunteers aged 35-50 years and showing wrinkles around the eyes were selected as the test subjects, and were randomly divided into 10 groups of 20 persons each.
The wrinkle mark of the tested part around the eyes of the subject is obtained by using silicone resin, and then the cream A-J is respectively coated on one side of the eye of the subject, and the cream substrate is coated on the other side of the eye of the subject, 2 times a day, 0.3g each time, and 30 days are continued.
Wrinkle imprints were taken at periocular test sites of subjects 15 days and 30 days after treatment, respectively, and wrinkle depth was measured using a confocal microscope. The percentage reduction in wrinkle depth after 15 days and 30 days of treatment was calculated and the results are shown in table 3 below.

The results in the table show that the facial creams A to E provided by the invention can obviously reduce the wrinkle depth of users, can reduce the wrinkle depth by 7.3 to 8.1 percent when used for 15 days, can be used continuously, have better effect, and can reduce the wrinkle depth by 18.6 to 20.6 percent when used for 30 days.
The applicant declares that the above description is only a specific embodiment of the present invention, but the scope of the present invention is not limited thereto, and it should be understood by those skilled in the art that any changes or substitutions that can be easily conceived by those skilled in the art within the technical scope of the present invention are within the scope and disclosure of the present invention.
Claims (10)
1. An anti-aging composition, comprising: tea leaf extract, watercress extract, horsetail extract, nettle leaf extract, silybum marianum fruit extract, tetrahydro-methyl pyrimidine carboxylic acid and ergothioneine.
2. The anti-aging composition as claimed in claim 1, wherein the mass ratio of the watercress extract, the equisetum arvense extract and the nettle leaf extract is 1 (0.5-1.5) to (1.5-3);
preferably, the mass ratio of the tea extract to the tetrahydro-methylpyrimidine carboxylic acid to the ergothioneine is 1 (2-5) to 0.8-2.
3. The anti-aging composition according to claim 1 or 2, further comprising a skin conditioner;
preferably, the skin conditioning agent comprises any one or a combination of at least two of vitamin E, carnosine, niacinamide, or sodium hyaluronate;
preferably, the anti-aging composition further comprises a humectant;
preferably, the humectant comprises any one or a combination of at least two of glycerol, ethanol or butylene glycol.
4. The anti-aging composition according to any one of claims 1 to 3, further comprising an emulsifier;
preferably, the anti-aging composition further comprises a penetrant;
preferably, the anti-aging composition further comprises a thickener;
preferably, the anti-aging composition further comprises a pH adjuster;
preferably, the anti-aging composition further comprises a preservative.
5. The anti-aging composition according to any one of claims 1 to 4, characterized by comprising, in mass percent:
1-10% of tea extract, 1-5% of watercress extract, 1-5% of field horsetail extract, 1-5% of nettle leaf extract, 3-10% of silybum marianum fruit extract, 1-10% of tetrahydro-methylpyrimidine carboxylic acid, 1-5% of ergothioneine, 0.1-5% of vitamin E, 0.1-3% of carnosine, 0.5-1% of hydrogenated lecithin, 0.5-1% of lecithin, 1-3% of thickening agent, 5-30% of humectant, 0.1-2% of penetrating agent, 1-3% of pH regulator, 0.5-1% of preservative and the balance of water.
6. A method for preparing the anti-aging composition according to any one of claims 1 to 5, wherein the method comprises the following steps:
dispersing the formula amount of tea extract, watercress extract, horsetail extract, nettle leaf extract, silybum marianum fruit extract, tetrahydro-methyl pyrimidine carboxylic acid and ergothioneine in water to obtain the anti-aging composition;
preferably, the temperature of the dispersion is 40 ℃ to 45 ℃.
7. The method of claim 6, comprising the steps of:
(1) adding the humectant, the penetrant and the emulsifier into water, heating to 70-90 ℃, stirring for dissolving, and keeping the temperature for 30-35 min;
(2) cooling to 40-45 ℃, adding a thickening agent and a preservative, stirring, and adding a pH regulator;
(4) cooling to 25-35 ℃, adding a tea extract, a watercress extract, an equisetum arvense extract, an isourtica leaf extract, a silybum marianum fruit extract, tetrahydro-methyl pyrimidine carboxylic acid, ergothioneine and a skin conditioner, stirring and dissolving, and keeping the temperature for 10-15 min to obtain the anti-aging composition.
8. A cosmetic comprising the anti-aging composition according to any one of claims 1 to 5.
9. The cosmetic according to claim 8, wherein the anti-aging composition is contained in the cosmetic in an amount of 10 to 20% by mass;
preferably, the cosmetic comprises any one of a lotion, essence, emulsion, cream, eye cream or mask.
10. Use of the anti-aging composition according to any one of claims 1 to 5 for the preparation of cosmetics.
