Abstract
The invention discloses a preparation method of alkali-extracted polysaccharide for improving antioxidation of passion fruit peel polysaccharide, and particularly relates to application of the passion fruit peel alkali-extracted polysaccharide in the fields of antioxidation cosmetics and medicines. The invention obtains the water extraction polysaccharide of the passion fruit peel by a water extraction method, and uses NaOH solution with certain concentration to continuously extract the passion fruit peel residue after water extraction to obtain the alkali extraction polysaccharide of the passion fruit peel. Tests on DPPH free radical scavenging activity, reducing capacity, hydroxyl free radical scavenging activity and the like show that the prepared passion fruit peel alkali-extracted polysaccharide has good antioxidant activity, particularly has remarkable effect on the reducing capacity of the alkali-extracted polysaccharide, and provides theoretical basis for development and application of antioxidant cosmetics and medicines.
Description
Technical Field
The invention belongs to the field of antioxidant cosmetics and medicine research and development, and particularly relates to a preparation method and application of alkali-extracted polysaccharide for improving antioxidation of passion fruit peel polysaccharide.
Background
Passion fruit is also known as passion fruit, and the like. The fruit juice is orange yellow, bright in color and especially aromatic in smell. The passion fruit contains rich nutrients such as protein, reducing sugar, vitamins and 17 amino acids needed by human body. The passion flower is suitable for developing commercial processing production application, such as products with unique flavor and high nutrient content, such as beverages, preserved fruits, jam, fruit wine and the like. The root, stem and leaf of Passiflora edulis can be used as medicine, and has the curative effects of diminishing inflammation, relieving pain, promoting blood circulation, strengthening body, reducing blood fat, lowering blood pressure and the like. The passion flower can also inhibit the growth of harmful microorganisms in intestines and stomach of a human body, has obvious effects of eliminating vivotoxin, invigorating stomach and helping digestion and improving the organism, and has special curative effect of eliminating inflammation such as colitis, gastroenteritis and the like; the passion flower has various health effects, such as anti-relaxation and anti-addiction medical health care functions, can treat insomnia, induce natural falling asleep and deep sleep, eliminate headache, dizziness and other phenomena, and is a natural tranquilizer and an anxiolytic.
Generally, plant polysaccharides contain a large amount of intracellular polysaccharides or cell wall-bound polysaccharides in addition to water-soluble polysaccharides, and hot water extraction cannot dissolve the plant polysaccharides, while a certain concentration of alkali solution can extract the cell wall-bound polysaccharides or intracellular polysaccharides. The invention utilizes the passion fruit peel residue after hot water extraction to further carry out alkali extraction to obtain the passion fruit peel alkali-extracted polysaccharide, and comparatively researches the antioxidation effect of the passion fruit peel water extraction and the alkali-extracted polysaccharide.
Disclosure of Invention
The invention aims to provide a preparation method and application of an alkali-extracted polysaccharide for improving antioxidation of passion fruit peel polysaccharide.
A method for preparing alkali-extracted polysaccharide for improving antioxidant activity of Passiflora edulis pericarp polysaccharide is provided. The method specifically comprises the following steps: preparing water-extracted polysaccharide and alkali-extracted polysaccharide of passion fruit peel. Water extraction of passion fruit peel polysaccharide is carried out by a water extraction method, and NaOH solution with certain concentration is used for continuously extracting the passion fruit peel residue after water extraction to obtain the passion fruit peel alkali-extracted polysaccharide.
The technical scheme for realizing the aim of the invention is as follows:
preparation of an alkali-extracted polysaccharide for improving antioxidant activity of passion fruit peel polysaccharide, and testing the antioxidant effect of water-extracted polysaccharide and alkali-extracted polysaccharide of passion fruit peel.
The invention achieves the above purpose by the following technical scheme:
the invention provides a preparation method of passion fruit peel polysaccharide,
(1) taking fresh passion fruit peel, drying and crushing;
(2) weighing appropriate amount of above materials, extracting with 75% ethanol at 65 deg.C under reflux for 2 hr, and extracting for 3 times. Drying the passion fruit peel subjected to alcohol extraction, extracting for 6 hours with hot water at 100 ℃ according to the material-liquid ratio of 1:16(g/mL), concentrating the extracting solution under reduced pressure, precipitating with ethanol, centrifuging, and freeze-drying to obtain passion fruit peel water-extracted polysaccharide;
(3) drying the passion fruit peel residue after hot water extraction, adding 6% NaOH solution according to the material-liquid ratio of 1:40(g/mL), extracting for 2h at 65 ℃, repeating for 3 times, filtering and concentrating;
(4) precipitating the concentrated solution with ethanol until the ethanol concentration is 85%, standing for 48 hr, centrifuging (4500r/min, 15min), redissolving the precipitate with distilled water, neutralizing with 35% hydrochloric acid, dialyzing with running water for 48 hr, and freeze drying to obtain passion fruit peel alkali-extracted polysaccharide.

Detailed Description
The technical solution of the present invention is further illustrated by the following examples.
(1) Preparation of water-extracted passion fruit peel polysaccharide
Drying passion fruit peel, and crushing for later use; extracting with 75% ethanol at 65 deg.C under reflux for 2 hr for 3 times. Oven drying the alcohol extracted passion fruit peel, extracting with 100 deg.C hot water at a material-liquid ratio of 1:16(g/mL) for 6h, concentrating the extractive solution under reduced pressure, precipitating with ethanol until the ethanol concentration is 60%, standing for 48h, centrifuging, and freeze drying to obtain passion fruit peel water-extracted polysaccharide (WPEP).
(2) Preparation of passion fruit peel alkali-extracted polysaccharide
Drying the residue of the passion fruit peel after hot water extraction, preparing 6 percent NaOH solution, and mixing the solution according to the material-to-liquid ratio (m)Fruit peel∶VNaOH)1:40, condensing and refluxing at 65 ℃ for 2h, extracting for 3 times, filtering, combining extracting solutions, concentrating, precipitating concentrated solution with ethanol until the ethanol concentration is 85%, standing for 48h, centrifuging (4500r/min, 15min), redissolving precipitates with distilled water, neutralizing with 35% hydrochloric acid, dialyzing with running water for 48h, freeze-drying, and collecting a dried sample, namely passionflower pericarp alkali-extracted polysaccharide (APEP).
The passion fruit peel polysaccharide prepared by the embodiment is applied to antioxidant cosmetics and medicines, and has the following specific effects:
1. DPPH free radical scavenging action of water extraction and alkali extraction polysaccharide of passion fruit peel
The scavenging capacity of passion fruit peel polysaccharide on DPPH free radical increases with the concentration of the sample solution (figure 1), wherein the scavenging rate of APEP on DPPH free radical is up to 72.25%, and the scavenging rate of WPEP on DPPH free radical is up to 79.28%.
2. Reducing power of water extraction and alkali extraction of passion fruit peel polysaccharide
With the increase of the concentration of the polysaccharide, the reducing abilities of the APEP and the WPEP are in positive correlation with the concentration of the solution thereof (figure 2), and the reducing ability of the APEP is obviously higher than that of the WPEP, which shows that the reducing ability of the passion fruit peel polysaccharide can be obviously improved by alkali extraction.
3. Hydroxyl radical scavenging effect of water extraction and alkali extraction of passion fruit peel polysaccharide
As the concentration of polysaccharide increased, the hydroxyl radical scavenging capacity of APEP and WPEP also increased (fig. 3).
Claims (3)
1. A preparation method of passion fruit peel polysaccharide with an antioxidant effect is characterized by comprising the following specific steps:
(1) taking fresh passion fruit peel, drying and crushing;
(2) weighing appropriate amount of the above raw materials, extracting with 75% ethanol at 65 deg.C under reflux for 2 hr for 3 times, oven drying the ethanol extracted passion fruit peel, extracting with 100 deg.C hot water at a ratio of 1:16g/mL for 6 hr, concentrating the extractive solution under reduced pressure, precipitating with ethanol, centrifuging, and freeze drying to obtain passion fruit peel water extract polysaccharide;
(3) drying the passion fruit peel residue after hot water extraction, adding 6% NaOH solution according to the feed-liquid ratio of 1:40g/mL, extracting for 2h at 65 ℃, repeating for 3 times, filtering and concentrating;
(4) precipitating the concentrated solution with ethanol until the ethanol concentration is 85%, standing for 48h, centrifuging at 4500r/min for 15min, redissolving the precipitate with distilled water, neutralizing with 35% hydrochloric acid, dialyzing with running water for 48h, and freeze drying to obtain passion fruit peel alkali-extracted polysaccharide.
2. The method for preparing the passion fruit peel alkali-extracted polysaccharide with the antioxidation effect as claimed in claim 1, is characterized in that: and (3) further extracting cell wall bound polysaccharide or intracellular polysaccharide of the passion fruit peel residues after hot water extraction by using 6% NaOH as a material.

The application of the passion flower pericarp alkali-extraction polysaccharide prepared by the preparation method of claim 1 is characterized in that the passion flower pericarp alkali-extraction polysaccharide is applied to the research and development fields of antioxidant cosmetics and medicines.
