Abstract
一种沉香树子萃取物制备皮肤抗发炎组成物的用途,其由数个沉香树(Aquilaria malaccensis)子经粉碎后,以90%乙醇进行超音波萃取,每批粉碎后的沉香树子萃取三次,每天萃取1次,萃取完毕后,借由减压浓缩机及真空泵移除所有醇水溶剂,使获得可应用于皮肤抗发炎用途的沉香树子萃取物。
Description
技术领域
本发明为一种沉香树子萃取物制备皮肤抗发炎组成物的用途。
背景技术
由于皮肤是人类身体直接暴露在外部环境中,皮肤不仅作为保护我们身体重要器官的保护层,且可调节水分蒸发、并保护身体免受外部感染;另,皮肤的表皮还可预防人体水分蒸发,表皮从外部依次为:角质层(stratum corneum)、颗粒层(stratum granulosum)、有棘层(stratum spinosum)、和基底层(stratum basale),健康的人的角质层细胞具有高浓度的天然保湿因子(Natural moisturzing factor,NMF),因此有效地与水分结合以防止皮肤的水分干燥。
然而,当皮肤过度暴露于紫外线或污染物会引起皮肤刺激,造成皮肤的细胞会有损伤,尤其角质层很容易因细胞受损而会有皮肤发炎、异位性皮肤炎、牛皮癣,甚至使皮肤细胞产生病变而导致有癌症或肿瘤的事情发生。
发明内容
本发明所要解决的主要技术问题在于,克服现有技术存在的上述缺陷,而提供一种沉香树子萃取物制备皮肤抗发炎组成物的用途,防止皮肤的水分干燥。
本发明解决其技术问题所采用的技术方案是:
一种沉香树子萃取物制备皮肤抗发炎组成物的用途,其将数个沉香树子分批以逆渗透流动水清洗,每次清洗10分钟,每一批沉香树子重复洗涤三次后于室温下沥干,并平铺于吸水性材质上阴干两天;然后,使用粉碎机粉碎该些沉香树子,并使粉碎后的沉香树子的粒径不大于2mm,同时控制粉碎机的粉碎时间,使连续粉碎时间不超过20分钟,接着将该些粉碎后的沉香树子分批以90%乙醇进行超音波萃取,每批粉碎后的沉香树子萃取三次,每天萃取1次,萃取完毕后,借由减压浓缩机及真空泵移除所有醇水溶剂,使获得沉香树子萃取物;
借此,该沉香树子萃取物确具有可制备皮肤抗发炎组成物的用途。
本发明的有益效果是,防止皮肤的水分干燥。

具体实施方式
有关本发明为达到目的所运用的技术手段及其构造,兹谨再配合图1至图6所示的实施例,详细说明如下:
如图1所示,将数个沉香树(Aquilaria malaccensis)子分批以逆渗透流动水清洗,每次清洗10分钟,每一批沉香树子重复洗涤三次后于室温下沥干,并平铺于吸水性材质上阴干两天;然后,使用粉碎机粉碎该些沉香树子,并使粉碎后的沉香树子的粒径不大于2mm,同时控制粉碎机的粉碎时间,使连续粉碎时间不超过20分钟,借以避免因机器过热改变沉香树内含的化学成分。接着,将该些粉碎后的沉香树子分批以90%乙醇进行超音波萃取,每批粉碎后的沉香树子萃取三次,每天仅萃取1次,萃取完毕后,借由减压浓缩机及真空泵移除所有醇水溶剂,使得到沉香树子萃取物(Crude-EtOH),且该沉香树子萃取物(Crude-EtOH)的型态为油状,使形成沉香子油。
承上述,较佳的实施例是,取2.9公斤的沉香树(Aquilaria malaccensis)子经上述步骤粉碎、沥干、阴干后,以15公升的90%的乙醇进行萃取后,可得102克的沉香树子萃取物(Crude-EtOH),亦即得到102克的沉香子油。
接着,将上述的沉香子油以4℃冷藏及避光储存一个月,该沉香子油的气味、颜色及酸碱值测等物理性质并未有变化;同时,透过核磁共振仪的评估,如图2所示,第一天的1HNMR(CDCl3,400MHz)与一个月后同一检品均存在佛波酯及脂肪酸的特征讯号,且于成分的比例上没有明显变化,1H NMR呈现稳定吻合的结果。借此可知,4℃冷藏与避光的环境显然为适合该沉香子油的储存方式。
此外,借由下列实验的具体实施例,可进一步证明本发明用途的应用范围,但并不以此限制本发明的范围。
实验一:
本实验是因不论何种本养产品涂抹于皮肤上时,皮肤的角质层为最些接触该些产品,因此本实验选用人类皮肤角质株化细胞进行安全性检测,评估沉香子油对皮肤细胞是否具细胞毒性。
(1)本实验采用MTT assay检测。将皮肤角质细胞(1×104/well)培养在96-well盘,并在37℃及5%CO2培养箱中培养至少24小时,加入10μL的MTT(3-(4,5-cimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide)溶液反应,于37℃、5%CO2反应4小时后移除培养液,加入100μL的DMSO溶解formazan沉淀物,最后于波长570nm下测定吸光值(BioTek,SynergyTM2,USA)。
(2)实验结果如图3所示,将冻干的沉香子油,以DMSO溶解配置成10mg/mL备用。HaCaT细胞经过沉香子油处理24小时后,结果显示1~100μg/mL沉香子油对细胞无明显毒性且于10-100μg/mL对细胞有明显增生的现象。
实验二:
本实验是利用显微镜观察沉香子油是否会改变皮肤细胞的型态。
(1)将人类皮肤角质株化细胞(1×104/well)培养在96-well盘,并在37℃及5%CO2培养箱中培养至少24小时。加入1μL的沉香子油以及于指定的时间作用,达反应时间后,于显微镜(Nikon,TE2000-U,Japan)下观察细胞型态,并拍照记录。
(2)实验结果如图4所示,利用显微镜观察细胞型态,经过1-100μg/mL沉香子油作用后细胞型态无明显变化。
实验三:
本实验是因促炎性细胞因子(Proinflammatory cytokine)是一系列可以促进炎症的细胞因子的总称,比较常见的促炎细胞因子包括TNF-α和IL-1;肿瘤坏死因子TNF-α(tumor necrosis factor-α)是一个发炎因子,发炎反应发生时,其在血清中含量会大幅的增加,此为发炎反应的指标,利用测定TNF-α含量的变化来评估沉香子油是否具有抑制发炎因子的释放。
(1)Coating buffer(0.2M sodium phosphate buffer):秤取11.8g的Na2HPO4和16.1g的NaH2PO4,加入适量ddH2O,调整pH值至6.5后再定量至1L。
Assay diluent(PBS+10%FBS):900mL PBS加入100mL FBS,储存于2~8℃,其须在配制三天内使用完毕。
Wash buffer(0.05%PBST):999.5Ml的PBS加入0.5mL的tween-20,储存于2~8℃,其须在配制三天内使用完毕。
Stop solution:1M H3PO4或2N H2SO4。将3×105/mL细胞数培养在24well,置于在37℃及5%CO2培养箱中培养24小时。加入沉香子油反应30分钟后,再加入1μg/mL LPS反应。达反应时间后,收集上清液,以1200rpm进行5分钟离心,然后加到新的1.5ml离心管于–20℃储存。
(2)本实验使用BD OptEIATMSet进行细胞内TNF-α含量测定。首先,加入100μLcapture Ab(1:250)于96-well中,用保鲜膜密封盘子后于4℃保存至隔夜。接着用washbuffer清洗盘子三次,最后一次将盘子倒置在滤纸上以确定完全移除残留的液体,再加入200μL的Assay diluent在室温下反应1小时后,再重复清洗的步骤。然后加入100μL的上清液,密封盘子后于室温下反应2小时,之后清洗五次,然后加入100μL的working detector(1:500detection antibody和1:250enzyme reagent),密封盘子后于室温下反应1小时,之后清洗7次,接着加入50μL的substrate solution,并于室温下反应30分钟,最后加入50μL的Stop solution于450nm测吸光值(BioTek,SynergyTM2,USA)。
(3)实验结果:人类皮肤角质株化细胞经过1μg/mL的LPS作用后,测得上清液中TNF-α的含量为20.2pg/mL,而经沉香子油(0.1-100μg/mL)作用后,如图5所示,上清液中TNF-α的含量分别为19.3、18.3、16.2、17.2和14.5pg/mL,其显示沉香子油具有抑制细胞内发炎因子TNF-α释放能力。
实验四:
本实验为测定沉香子油的抑制细胞内白血球介素-1(IL-1)含量。
(1)Coating buffer(0.2M sodium phosphate buffer):秤取11.8g的Na2HPO4和16.1g NaH2PO4加入适量ddH2O,调整pH值至6.5后再定量至1L。
Assay diluent(PBS+10%FBS):900mL的PBS加入100mL的FBS,储存于2~8℃,其须在配制三天内使用完毕。
Wash buffer(0.05%PBST):999.5mL的PBS加入0.5mL tween-20,储存于2~8℃,其须在配制三天内使用完毕。
Stop solution:1M H3PO4或2N H2SO4。将3×105/mL细胞数培养在24well,置于在37℃及5%CO2培养箱中培养24小时。加入沉香子油反应30分钟后,再加入1μg/mL的LPS反应。达反应时间后,收集上清液,以1200rpm进行5分钟离心,然后加到新的1.5ml离心管于–20℃储存。
(2)本实验使用BD OptEIATMSet进行细胞内IL-1含量测定。首先,加入100μLcapture Ab(1:250)于96-well中,用保鲜膜密封盘子后于4℃保存至隔夜。接着用washbuffer清洗盘子三次,最后一次将盘子倒置在滤纸上以确定完全移除残留的液体,再加入200μL的Assay diluent在室温下反应1小时后,再重复清洗的步骤。加入100μL的上清液,密封盘子后于室温下反应2小时,之后清洗五次,然后加入100μL的working detector(1:500detection antibody和1:250enzyme reagent),密封盘子后于室温下反应1小时,之后清洗7次,接着加入50μL的substrate solution室温下反应30分钟,最后加入50μL的Stopsolution于450nm测吸光值(BioTek,SynergyTM2,USA)。
(3)实验结果如图6所示,人类皮肤角质株化细胞经过1μg/mL的LPS作用后,测得上清液中IL-1的含量为14.0pg/mL,而经沉香子油(0.1-100μg/mL)作用后,上清液中IL-1的含量为9.4、9.6、10.5、12.1、13.8pg/mL,显示沉香子油具有些许抑制细胞内发炎因子IL-1释放能力。
借由上述实验可知,本发明的沉香树子萃取物(即上述的沉香子油)确具有可抑制细胞内发炎因子TNF-α及白血球介素-1的释放能力,且作用后细胞型态具有增生的现象,意即本发明的沉香树子萃取物(即上述的沉香子油)应用于皮肤抗发炎的用途,确实具有明显的效果。
又,为提升本发明沉香树子萃取物(即上述的沉香子油)的应用范围,可将本发明上述实验中沉香树子萃取物(即上述的沉香子油)最高作用浓度50~100μg/mL(0.005~0.01%)放大100倍,将有效作用浓度(0.5~1%)的沉香树子萃取物(即上述的沉香子油)添加油相中可抗氧化的维他命E、可滋润肤感的橄榄油、玫瑰果油,亦可添加水相添加济中可保湿嫩肤的玻尿酸、可保湿增稠的三仙胶、可光滑肤感及增稠的海藻胶,以及可提高保存的氯苯甘醚(Chlorphenesin)等成分,进而获得沉香子复合油、沉香子油精华霜等产品;因此,本发明的沉香树子萃取物可使皮肤有效抗发炎,本发明确可用于制备皮肤抗发炎用途的组成物。
以上所述,仅是本发明的较佳实施例而已,并非对本发明作任何形式上的限制,凡是依据本发明的技术实质对以上实施例所作的任何简单修改、等同变化与修饰,均仍属于本发明技术方案的范围内。
Claims (2)
1.一种沉香树子萃取物制备皮肤抗发炎组成物的用途,其特征在于,由数个沉香树子分批以逆渗透流动水清洗,每次清洗10分钟,每一批沉香树子重复洗涤三次后于室温下沥干,并平铺于吸水性材质上阴干两天;然后,使用粉碎机粉碎该些沉香树子,并使粉碎后的沉香树子的粒径不大于2mm,同时控制粉碎机的粉碎时间,使连续粉碎时间不超过20分钟,接着将该些粉碎后的沉香树子分批以90%乙醇进行超音波萃取,每批粉碎后的沉香树子萃取三次,每天萃取1次,萃取完毕后,借由减压浓缩机及真空泵移除所有醇水溶剂,使获得沉香树子萃取物,而该沉香树子萃取物的型态为油状,使形成沉香子油。
2.根据权利要求1所述的沉香树子萃取物制备皮肤抗发炎组成物的用途,其特征在于,所述沉香树子萃取物的有效作用浓度为50~100μg/mL。
